The CD40 ligand (CD40L) is an activation-induced surface membrane protein expressed by CD4+ T helper cells in lymphoid follicles, and is involved in the contact-dependent signaling-mediated activation, proliferation, and differentiation of CD40+ B cells. Using immunohistochemistry, the present study analyzes the cell microenvironment of lymphoid tissues in two cases of X-linked hyper-IgM syndrome, a congenital immunodeficiency caused by mutations of the CD40L gene, and which represents a unique model to dissect the functional and morphologic consequences of disrupted CD40/CD40L interactions. Methamphetamine (MA) addiction is characterized by persistent drug-associated memory and high relapse risk. Perineuronal nets (PNNs), which mainly surround PV interneurons, are specialized extracellular matrix structures involved in synaptic stabilization and memory regulation. Our previous studies have shown that PNNs in the medial prefrontal cortex (mPFC) are essential for MA-induced conditioned place preference (CPP) memory, but the role of hippocampal PNNs in MA-associated contextual memory remains unclear. Here, a CPP paradigm was employed in female mice to investigate the role and underl

Postoperative cognitive dysfunction (POCD) is a common postoperative neurological complication, with memory consolidation deficits being one of its prominent features. However, the mechanisms underlying postoperative memory consolidation deficits are still unclear. The perineuronal net (PNN) is a specialized extracellular matrix that predominantly surrounds parvalbumin (PV) interneurons and is closely associated with cognitive functions, including learning and memory. Theta oscillations in the hippocampal CA1 region are crucial for memory consolidation. However, the relationships among postope

Prolonged exposure to motion in one direction often leads to the illusion of motion in the opposite direction for stationary objects. This motion aftereffect likely arises across several visual areas from adaptive changes in the balance of activity and competitive interactions. We examined whether or not the mouse was susceptible to this same illusion to determine whether it would be a suitable model for learning about the neural representation of the motion aftereffect. Under a classical conditioning paradigm, mice learned to lick when presented with motion in one direction and not the opposi

The nucleus accumbens (NAc) is critical for motivated behavior and is rewired following exposure to drugs of abuse. Medium spiny neurons (MSNs) in the NAc express either D1 or D2 receptors and project to distinct downstream targets. Differential activation of these MSNs depends on both excitation from long-range inputs and inhibition via the local circuit. Assessing how long-range excitatory inputs engage inhibitory circuitry is therefore important for understanding NAc function. Here, we use slice electrophysiology and optogenetics to study ventral hippocampal (vHPC)-evoked feedforward inhibi

The transcriptional properties of unliganded thyroid hormone receptors are thought to cause the misdevelopment during hypothyroidism of several functions essential for adult life. To specifically determine the role of unliganded thyroid hormone receptor alpha1 (TRalpha1) in neuronal tissues, we introduced a mutation into the mouse TRalpha1 gene that lowers affinity to thyroid hormone (TH) 10-fold. The resulting heterozygous mice exhibit several distinct neurological abnormalities: extreme anxiety, reduced recognition memory, and locomotor dysfunction. The anxiety and memory deficiencies were r

Throughout history, muscle research has led to numerous scientific breakthroughs that have brought insight to a more general understanding of all biological processes. Potentially one of the most influential discoveries was the role of the second messenger calcium and its myriad of handling and sensing systems that mechanistically control muscle contraction. In this review we will briefly discuss the significance of calcium as a universal second messenger along with some of the most common calcium binding motifs in proteins, focusing on the EF-hand. We will also describe some of our approaches

Brain injury due to neonatal hypoxia-ischemia (HI) is more homogenously severe in male than in female mice. Because, necrostatin-1 (nec-1) prevents injury progression only in male mice, we hypothesized that changes in brain-derived neurotrophic factor (BDNF) signaling after HI and nec-1 are also sex-specific providing differential conditions to promote recovery of those more severely injured. The increased aromatization of testosterone in male mice during early development and the link between 17-β-estradiol (E2) levels and BDNF transcription substantiate this hypothesis. Hence, we aimed to in

The neuronal ceroid lipofuscinoses (NCL) are progressive neurodegenerative disorders with onset from infancy to adulthood that are manifested by blindness, seizures, and dementia. In NCL, lysosomes accumulate autofluorescent proteolipid in the brain and other tissues. The mnd/mnd mutant mouse was first characterized as exhibiting adult-onset upper and lower motor neuron degeneration, but closer examination revealed early, widespread pathology similar to that seen in NCL. We used the autofluorescent properties of accumulated storage material to map which CNS neuronal populations in the mnd/mnd

Children are especially vulnerable to the neurotoxic effects of nerve agents, which can cause lasting neuronal dysfunction, including cognitive impairments, epilepsy, and related comorbidities. Current benzodiazepine anticonvulsants often fail to prevent long-term neuropathology following neurotoxic chemical exposure, underscoring the urgent need for more effective treatments. This study investigated the therapeutic potential of the synthetic neurosteroid ganaxolone (GX) in a pediatric rat model of the nerve agent soman exposure. Pediatric (postnatal day 21) rats were acutely exposed to soman

Endothelin-1 (ET-1) from renal-tubule-epithelial-cells inhibits NaCl reabsorption via ETB receptor in an autocrine manner, and inhibition of ETB receptors leads to salt-sensitive-hypertension. In the distal convoluted tubule (DCT), NaCl enters the cell via NaCl-cotransporter (NCC) and Cl- exits the cell in part by ClC-K2 channels, which play a role in regulating With-No-lysine kinase 4 (WNK4). The study aims to explore whether ET-1-induced inhibition of NaCl absorption is also achieved by inhibiting the basolateral Cl- channels in the DCT. Patch-clamp and immunoblotting assessed ET-1 effects o

Down syndrome is the most common genetic neurodevelopmental disorder associated with mild-to-moderate intellectual disability. A disturbed excitation-inhibition balance is thought to be a major cause for the intellectual deficits in DS. In this study, we used patch-clamp electrophysiology, optogenetic stimulation and immunohistochemistry to investigate synaptic inhibition from specific interneuron subpopulations onto granule cells of the dentate gyrus in Ts65Dn mice. Optogenetically evoked inhibitory postsynaptic currents (IPSCs) from somatostatin (SOM) interneurons onto dendrites of granule c

Inhibition in central auditory pathways declines in old age, which contributes to age-related auditory processing deficits. In auditory cortex, most inhibitory interneurons can be identified by one of three marker proteins: parvalbumin (PV), somatostatin (SOM), or vasoactive intestinal peptide (VIP). About 10% of interneurons contain the α2 nicotinic acetylcholine receptor (α2 cells), and most of these are layer 5 Martinotti cells (α2-MCs). We examined the distribution of these four interneuron subtypes in auditory cortex of FVB mice, from young adult through old age, and the extent to which α

BACKGROUND: Clinical studies showed that chronic pain conditions affected patients' cognitive function, but the specific mechanism remained unclear. Animal experiments revealed that compared with female mice, male mice experiencing chronic pain were more prone to hippocampal neuroinflammation and cognitive dysfunction. Parvalbumin-positive (PV) interneurons exerted a critical function in modulating cognitive performance. On the basis of these observations, the present study investigated the effects and underlying mechanisms of estrogen against chronic pain-triggered cognitive dysfunction.

Early-life adversity alters inhibitory circuit maturation across developing brain systems, but most studies have focused on corticolimbic regions involved in emotional and cognitive regulation. Whether early adversity also reshapes inhibitory extracellular matrix architecture in primary sensory cortex remains less clear. Here, we examined the effects of maternal separation on parvalbumin-positive (PV+ ) interneurons and Wisteria floribunda agglutinin-labeled perineuronal nets (PNNs) in the mouse primary somatosensory barrel cortex (S1BF), a highly organized sensory region in which laminar posi

Alzheimer's Disease is the most common neurodegenerative disease worldwide, but significant gaps in pathophysiological understanding have hampered development of disease-modifying therapies. In Alzheimer's disease, neurophysiological function is impaired, with gamma frequency oscillations - thought to be essential for higher-order cognitive processes - disrupted in both patients and animal models. However, the mechanisms driving these disruptions are unclear and, in particular, the role of neuroinflammation in these changes is poorly understood. In this study, we investigated neuronal network

Binge drinking continues to pose major societal and health challenges. Understanding the molecular mechanisms that drive problematic alcohol use is critical for developing novel therapeutics. Parvalbumin-expressing interneurons (PV-INs), a key subpopulation of interneurons within the prefrontal cortex (PFC) and other brain areas, have been implicated in a variety of alcohol-seeking behaviors. Recent work has identified the metabotropic glutamate 5 (mGlu5) receptor as a critical sex-specific regulator of PV-IN activity. Genetic deletion of mGlu5 from PV-INs alters PFC-dependent cognitive behavi

BACKGROUND: Failure of epilepsy pharmacotherapy is common, justifying efforts to identify optimal cell targets for advanced therapies. Genetic manipulations of the inhibitory system in principle offer a finely tuned intervention that cannot be achieved with a broad reduction of pyramidal neuron excitability. In this context, neuron-derived neurotrophic factor-expressing (NDNF+) interneurons evoke long-lasting GABAA and GABAB receptor-mediated inhibition. However, their contribution to restraining hyperexcitability during epileptiform activity remains unexplored.
METHODS: To address this, we e

The thalamus is now recognized as a dynamic regulator of cortical information flow, with the thalamic reticular nucleus (TRN) providing inhibitory control over the routing, gain, and timing of thalamocortical communication. Schizophrenia and obsessive-compulsive disorder (OCD) both involve thalamocortical abnormalities but produce distinct clinical phenotypes, psychosis versus compulsivity. In this narrative review, we synthesize evidence from neuroimaging, electrophysiology, postmortem studies, and animal models to compare thalamocortical gating mechanisms across these disorders. BACKGROUND: Fish are among the most frequent causes of immunoglobulin E (IgE)-mediated food allergies (Type I). Currently, there is no known cure for fish allergy and individuals who are sensitized have to practice strict, lifelong avoidance of fish products in their diets. The relationship between gut microbiome and food allergies is currently a major topic of discussion; these pathologies involve the development of dysbiosis, which is a microbial imbalance resulting from immune-related mechanisms. Recent studies have provided evidence that individuals suffering from food allergies, display a
