Discovery of New Pyrazolone Derivatives for Treating Invasive Fungal Infections by Iron Homeostasis Disruption.

Invasive fungal infections caused by Cryptococcus neoformans and Candida glabrata pose a severe threat to global public health due to high mortality, urgently requiring the development of antifungal agents with novel mechanisms. Herein, a series of new pyrazolone derivatives were designed and evaluated. In particular, compound D2 exhibited subnanomolar antifungal activity against Cryptococcus neoformans and Candida glabrata strains (MIC range: 0.00012-0.0078 μg/mL). Mechanistically, compound D2 disrupted fungal iron homeostasis, impaired mitochondrial structure and function, and triggered oxid

Valorization of Sea Buckthorn (Hippophae rhamnocaides L.) Leaves: Polyphenol-Rich Extract Targeting Helicobacter pylori Virulence and Gastric Mucosal Homeostasis.

Sea buckthorn (Hippophae rhamnoides L.) leaves are rich in polyphenols and exhibit potent activity against Helicobacter pylori. This study found that its extract demonstrates a dual mechanism of action, inhibiting bacterial virulence while protecting the gastric mucosa. For multidrug-resistant strains, the minimum inhibitory concentration (MIC) range of this extract was 0.08-0.64 mg/mL. At 0.32 mg/mL, it significantly suppressed H. pylori motility and urease activity by 72% and 56%, respectively. In an infected mouse model, the extract lowered gastric pro-inflammatory cytokines (TNF-α, IL-1β,

Regression of Advanced Gastric MALT Lymphoma after the Eradication of Helicobacter pylori.

A 66-year-old female presented with a 1-month history of dyspepsia. An initial upper gastrointestinal endoscopy with biopsy revealed a low-grade mucosa-associated lymphoid tissue (MALT) lymphoma. A rapid urease test was positive for Helicobacter pylori. Endoscopic ultrasound (EUS) and computed tomography (CT) revealed a 30×15-mm lymph node (LN) in the subcarinal area. Histopathologic and phenotypic analyses of the biopsy specimens obtained by EUS-guided fine-needle aspiration revealed a MALT lymphoma, and the patient was diagnosed with a stage 4E gastric MALT lymphoma. One year after H. pylori

Identification of Proteins Related to Nickel Homeostasis in Helicobater pylori by Immobilized Metal Affinity Chromatography and Two-Dimensional Gel Electrophoresis.

Helicobacter pylori (H. pylori) is a widespread human pathogen causing peptic ulcers and chronic gastritis. Maintaining nickel homeostasis is crucial for the establishment of H. pylori infection in humans. We used immobilized-nickel affinity chromatography to isolate Ni-related proteins from H. pylori cell extracts. Two-dimensional gel electrophoresis and mass spectrometry were employed to separate and identify twenty two Ni-interacting proteins in H. pylori. These Ni-interacting proteins can be classified into several general functional categories, including cellular processes (HspA, HspB, Ts

Novel Copper (II) Complexes with Fluorine-Containing Reduced Schiff Base Ligands Showing Marked Cytotoxicity in the HepG2 Cancer Cell Line.

Several novel copper (II) complexes of reduced Schiff bases containing fluoride substituents were prepared and structurally characterized by single-crystal X-ray diffraction. The complexes exhibited diverse structures, with the central atom in distorted tetrahedral geometry. The biological effects of the products were evaluated, specifically their cytotoxicity, antimicrobial, and antiurease activities, as well as affinity for albumin (BSA) and DNA (ct-DNA). The complexes showed marked cytotoxic activities in the HepG2 hepatocellular carcinoma cell line, considerably higher than the standard ci

Advances in Helicobacter pylori vaccine research: From candidate antigens to adjuvants-A review.

BACKGROUND: Helicobacter pylori is a Gram-negative, spiral-shaped bacterium that infects approximately 50% of the world's population and has been strongly associated with chronic gastritis, peptic ulcers, gastric mucosa-associated lymphoma, and gastric cancer. The elimination of H. pylori is currently considered one of the most effective strategies for the treatment of gastric-related diseases, so antibiotic therapy is the most commonly used regimen for the treatment of H. pylori infection. Although this therapy has some positive effects, antibiotic resistance has become another clinically pro

Plant Growth-Promoting Rhizobacteria Promote Growth of Seedlings, Regulate Soil Microbial Community, and Alleviate Damping-Off Disease Caused by Rhizoctonia solani on Pinus sylvestris var. mongolica.

As the excessive use of chemical fertilizers harms organisms and adversely affects the soil environment, the replacement of chemical fertilizers with biological fertilizers has attracted widespread attention as an environmental protection strategy. In this study, the effects of rhizosphere bacteria inoculation on growth of Pinus sylvestris var. mongolica seedlings, soil parameters, soil microbial community structure, and the biocontrol of damping-off were studied by pot experiments. The results showed that all three rhizosphere bacteria (Pseudomonas chlororaphis, Pseudomonas extremaustralis, a

Systems Approaches for Unveiling the Mechanism of Action of Bismuth Drugs: New Medicinal Applications beyond Helicobacter Pylori Infection.

Metallodrugs have been widely used as diagnostic and therapeutic agents. Understanding their mechanisms of action may lead to advances in rational drug design. However, to achieve this, diversified approaches are required because of the complexity of metal-biomolecule interactions. Bismuth drugs in combination with antibiotics as a quadruple therapy show excellent success rates in the eradication of Helicobacter pylori, even for antibiotic-resistant strains, and in fact, they have been used in the clinic for decades for the treatment of infection. Understanding the mechanism of action of bismu

Study of molecular interactions with 13C DNP-NMR.

NMR spectroscopy is an established, versatile technique for the detection of molecular interactions, even when these interactions are weak. Signal enhancement by several orders of magnitude through dynamic nuclear polarization alleviates several practical limitations of NMR-based interaction studies. This enhanced non-equilibrium polarization contributes sensitivity for the detection of molecular interactions in a single NMR transient. We show that direct (13)C NMR ligand binding studies at natural isotopic abundance of (13)C gets feasible in this way. Resultant screens are easy to interpret a

Helicobacter pylori and the pathogenesis of gastroduodenal disease: Implications for the management of peptic ulcer disease.

The discovery of Helicobacter pylori has changed our understanding of the pathophysiology of peptic ulcer disease. An estimated one billion people harbour the organism worldwide but the highest prevalence is found in developing countries with up to 80% of people infected. The most favoured modes of transmission are faeco-oral and oral-oral. The mechanisms of H pylori-induced gastroduodenal disease include the provocation of local inflammatory reaction with the release of toxic cytokines, elevation of gastrin concentration and cytotoxic epithelial injury from the activity of urease and other en

The physiologic approach to the medical management of stone disease.

The cause of urinary stone disease can now be detected in approximately 80% of patients. Effective treatment can substantially reduce the recurrence of urinary calculi. Proper therapy depends on a thorough understanding of the physiology of calcium, oxalate, uric acid, cystine, and struvite formation and the medication developed for prevention. This article reviews the physiologic basis of urinary stone management in a straightforward, understandable fashion.

Inhibitory Effect of Fucoidan Combined with Bifidobacteria on Helicobacter pylori.

In the study, fucoidan (FUC) was shown to have antigastric digestive properties and promote the growth of B. animalis BD7, a gastric fluid-resistant, highly aggregative Bifidobacterium. The combination of FUC and B. animalis BD7 significantly inhibited the adhesion of H. pylori SS1 to gastric mucosal epithelial cells (GES-1), decreased urease activity (71.24 ± 3.29%), and promoted the synthesis of bacterial inhibitory substances in vitro, while in vivo, it could alleviate the inflammation of the gastric mucosa of the infected mice, regulate the level of inflammatory factors, and repair the muc

Pathogenicity and virulence of Helicobacter pylori: A paradigm of chronic infection.

Infection with Helicobacter pylori is one of the most common infections of mankind. Infection typically occurs in childhood and persists for the lifetime of the host unless eradicated with antimicrobials. The organism colonizes the stomach and causes gastritis. Most infected individuals are asymptomatic, but infection also causes gastric and duodenal ulceration, and gastric cancer. H. pylori possesses an arsenal of virulence factors, including a potent urease enzyme for protection from acid, flagella that mediate motility, an abundance of outer membrane proteins that can mediate attachment, se

Enzyme Immobilization over Polystyrene Surface Using Cysteine Functionalized Copper Nanoparticle as a Linker Molecule.

The work focus on the development of a simple and efficient method of enzyme immobilization over a polystyrene surface using cysteine functionalized copper nanoparticle as linker molecule. The polystyrene surface is activated by generating -NO2 groups by the process of nitration reaction. The nitrated polystyrene plate then is silanized with (3-mercaptopropyl) trimethoxysilane (MPTS) followed with the coupling of cysteine-capped copper nanoparticles on the silanized surface through thiol moiety. A nanoparticle layer is thus created over the polystyrene surface which is efficiently used for cov

The role of acid inhibition in Helicobacter pylori eradication.

Infection of the stomach by the gastric pathogen Helicobacter pylori results in chronic active gastritis and leads to the development of gastric and duodenal ulcer disease and gastric adenocarcinoma. Eradication of H. pylori infection improves or resolves the associated pathology. Current treatments of H. pylori infection rely on acid suppression in combination with at least two antibiotics. The role of acid suppression in eradication therapy has been variously attributed to antibacterial activity of proton pump inhibitors directly or through inhibition of urease activity or increased stabilit

A study of the structure of the crystalline bacterial biofilms that can encrust and block silver Foley catheters.

The aim of this study was to examine the structure of the crystalline bacterial biofilms that encrust and block silver/hydrogel-coated latex catheters. Scanning electron microscopy was used to examine the crystalline deposits that were found encrusting catheters obtained from six patients undergoing long-term catheterization in a community setting. Large populations of bacilli and cocci were seen on all catheters developing on a basal foundation layer of crystalline material. These observations show that in patients prone to catheter encrustation, crystalline material formed in the urine can c

New insights on the pathogenesis of invasive Cryptococcus neoformans infection.

Disseminated cryptococcosis begins with infection of the lungs via inhalation. This is followed by escape from the lungs and entry into the bloodstream allowing dissemination to the brain and central nervous system. We discuss the steps involved in dissemination and the host and microbial factors that influence each step. For the host, containment in the lung is accomplished with a combination of cell-mediated and antibody responses. Dissemination occurs when these systems fail and/or when phagocytic cells that fail to kill the yeast instead act as a niche for replication. One of the main micr
