Developmental dynamics : an official publication of the American Association of Anatomists2026eng
BACKGROUND: Mechanistic target of rapamycin (mTOR) is a serine/threonine kinase with diverse roles in development and homeostasis, but its spatiotemporal dynamics in the embryonic spinal cord are not fully defined. Here, we map mTOR activity across mouse spinal cord development and test its function using a conditional gene mutant.
RESULTS: Immunohistochemistry for phosphorylated readouts of mTORC1 and mTORC2 revealed distinct, stage-dependent patterns. In the early neural tube, phosphorylated S6 (p-S6; Ser235/236 and Ser240/244) marked dorsal progenitors and ventral motor neurons, while p-p7

BACKGROUND: Ambystoma mexicanum is renowned for its extraordinary regenerative capacity. Regardless of whether progenitor cells arise via dedifferentiation or stem cell activation, these cells must replicate DNA to proliferate before they ultimately differentiate. This rapid cell division increases the likelihood of DNA damage, requiring efficient repair mechanisms.
RESULTS: We focused on the non-homologous end joining pathway, responsible for repairing double-strand DNA breaks, and investigated the effects of 2 key components-KU70/80 and LIGIV- using chemical inhibitors STL127705 and SCR130,

BACKGROUND: Advances in reproductive and cellular biotechnology have significantly enhanced the ability to study, conserve, and manipulate species within the Bovinae family, including domestic cattle and the endangered European bison (wisent). Central to the success of such technologies, including in vitro fertilization, somatic cell nuclear transfer, and the culture of pluripotent stem cells (PSCs), is the chromosome stability of cultured cells and embryos.
RESULTS: Here, we present the design and application of a specific centromere Cot-1 DNA probe for fluorescence in situ hybridization (FI

BACKGROUND: Gaining finer spatiotemporal control over gene expression is critical for unraveling the regulatory networks that coordinate embryogenesis. While the heat-shock inducible gene expression system is a widely used tool for controlling temporal transgene expression, its applicability in spatiotemporal control is limited.
RESULTS: We present the BOOPTHAT (Batch-Operating Optically Powered Targeted Heater for Activating Transgenes) as a low-cost system for activating heat-shock inducible transgenes with spatiotemporal control in multiple zebrafish embryos at a time. The BOOPTHAT system

Hypermobile Ehlers-Danlos syndrome (hEDS) and hypermobility spectrum disorders (HSD) are common yet underrecognized connective tissue disorders characterized by joint hypermobility and multi-organ system involvement. Current diagnostic criteria inadequately capture the clinical heterogeneity of hEDS/HSD. Consequently, patients often face diagnostic delays and fragmented care, which exacerbate underlying comorbidities such as chronic pain and psychological distress. This narrative review explores the key limitations of existing diagnostic criteria, the burden of underrecognized comorbidities, a

Myocyte enhancer factor 2C (MEF2C) is a critical transcription factor and plays a pivotal role in muscle development, cardiac reprogramming, cellular differentiation, and disease pathogenesis. In this review, we have systematically described the structural characteristics of the MEF2C protein, particularly its DNA-binding MADS-box and transcriptional regulatory domains, and outlined the multi-level regulation mechanism of its expression and activity. We further have highlighted the functional roles of MEF2C in heart and skeletal muscle development, including its involvement in cardiomyocyte di

Sox8, Sox9, and Sox10 arose by multiple rounds of genome duplications from a single SoxE gene in ancestral vertebrates. In this review, we will briefly discuss the molecular structure and function of SoxE transcription factors and their evolutionary origin. We will then discuss their expression, function, and developmental disorders. SoxE proteins play critical roles during the development of multiple tissues in vertebrate embryos, including the neural crest, inner ear, cartilages, and glia cells of diverse origins, heart, gonads, and gastrointestinal tract. Because they recognize the same DNA

BACKGROUND: Rubinstein Taybi syndrome (RSTS), a rare congenital disease, is caused by mutations in lysine acetyl transferase type 3 (KAT3) genes, EP300 and CREBBP. Many of the tissues affected in RSTS are derived from the neural crest (NC). Hence, we proposed that NC development would be perturbed in RSTS.
RESULTS: Zebrafish RSTS models generated by knocking down or mutating ep300a and crebbpa genes reveal defects in NC migration. This effect on migration is conserved in NC cells generated from human RSTS patient-derived induced pluripotent stem cells (iPSC) cells. The defects in NC migration

BACKGROUND: Human mutations in INVERSIN are associated with nephronophthisis, variable penetrance of situs inversus and congenital heart disease. Inversin has been shown to localize to cilia and many of the patient phenotypes are attributed to disrupted cilia function. We set out to characterize the invssa12246 allele in zebrafish to investigate its utility in understanding patient disease.
RESULTS: The left-right axis is established correctly in invssa12246 mutants, kidneys appear to develop normally without any cysts and cilia appear normal, however mutants are significantly shorter. In pos

BACKGROUND: Fate maps relate progenitor cell positions to later fates and locations of their progeny, revealing early embryonic organization. Previous zebrafish fate maps identified the origin of germ layers and derivative cell fates, but ventral gastrula progenitor domains were not fully resolved. In particular, the neural crest, a multipotent cell type central to vertebrate development and evolution, was not characterized in the zebrafish fate map.
RESULTS: We generated a region-based fate map to resolve the origin of the neural crest and overall organization of the ventral gastrula. This f

BACKGROUND: Testicular development is a highly orchestrated process essential for male fertility, but its temporal genetic regulation remains incompletely resolved in many mammalian models. This study re-analyses rat testis transcriptomes across 15 stages from embryonic day 12 to postnatal week 16 using bulk RNA-sequencing data from a multispecies organ development atlas, generating a continuous, testis-focused temporal framework, and pinpointing key developmental transition windows and regulatory modules.
RESULTS: Of 23,748 annotated genes, 17,717 were differentially expressed (DEGs), and hi

Neural crest cells are multipotent, migratory stem-like cells essential for vertebrate development that contribute broadly to many tissues including the craniofacial skeleton, peripheral nervous system, and pigment-producing cells. Their development progresses through phases of induction, specification, delamination, migration, and differentiation. Importantly, advancement through these phases requires detecting and responding to extracellular cues through the plasma membrane. While the gene regulatory networks and the signals that govern neural crest development are well characterized, plasma

BACKGROUND: The continuously growing mouse incisor serves as a model for stem cell-driven tooth renewal. Although mice possess maxillary and mandibular incisors, most studies have focused on the mandible. Potential differences in structure and renewal between these serially homologous organs remain insufficiently characterized.
RESULTS: Mandibular incisors are longer and contain greater enamel volume, mineral density, and mineral content than maxillary incisors, whereas maxillary incisors are more curved and display a larger cross-sectional area. Mineral density increases along the apical-to-

BACKGROUND: RSPO1 and FOXL2 are female sex-determining genes involved in the differentiation and organization of the ovary in some eutherian mammals. Mutations or loss of function of these genes are associated with partial to full sex reversal in mice, humans, and goats. RUNX1 may also play a role in ovarian development, but its expression in marsupials has not been examined as yet. We studied the conservation and protein localization of RSPO1, FOXL2, and RUNX1 orthologs in the marsupial tammar wallaby (Notamacropus eugenii) compared to other vertebrates.
RESULTS: RSPO1, FOXL2, and RUNX1 were

BACKGROUND: Tadpoles of the clawed frog Xenopus laevis can regenerate their tails following partial amputation, replacing the missing spinal cord, muscles, and fin. However, for a brief period of development this response becomes unstable, leading to a proportion of tadpoles that undergo wound healing rather than regenerative programme. Inspired by a growing number of links between the microbiome and human inflammatory disease, we asked how the tadpole skin microbiome and innate immunity influence the regeneration of a complex appendage. We previously showed that tadpoles raised in antibiotics

BACKGROUND: Diabetes is a condition characterized by a loss of pancreatic β-cell function, which results in the dysregulation of insulin homeostasis. Using a partial pancreatectomy model in axolotl, we aimed to observe the pancreatic response to injury.
RESULTS: Here we show a comprehensive histological characterization of pancreatic islets in axolotl. Following pancreatic injury, no apparent blastema-like structure was observed. We found a significant, organ-wide increase in cellular proliferation post-resection in the pancreas compared to sham-operated controls. This proliferative response

BACKGROUND: Diabetes is a group of diseases characterized by loss of β cell mass and/or function, resulting in hyperglycemia. With no established curative treatment, this has initiated research in β cell regeneration. Current animal models have either limited regenerative capacity (mice) or small size and evolutionary distance from humans (zebrafish). There is a need for new models to study endogenous regeneration pathways. This study proposes the axolotl salamander (Ambystoma mexicanum) as a model for studying the regeneration of β cells and aims to establish a protocol for STZ-induced hyperg

BACKGROUND: Perfluoroalkyl substances (PFAS) are persistent environmental contaminants previously used for industrial purposes as a non-stick coating and flame retardant. The stability of these molecules prevents their breakdown, which results in ground water contamination across the globe. Perfluoroalkyl substances molecules are known to bioaccumulate in various organisms. However, the health consequences remain unclear due to the large number of molecules in the PFAS family and different effects on various tissues. Here, we use the frog Xenopus laevis to investigate the developmental consequ

BACKGROUND: During ventral body wall closure in amniotes, the skeleton and muscles such as sternum, ribs, and pectoral muscles undergo a striking positional change from the dorso-lateral to mid-ventral region. The precise process of ventral body wall closure has poorly been explored. We have presented and examined two opposing models: the invading closure model, in which musculoskeletal components infiltrate into the mid-ventral tissue, and the replacing closure model, in which the original mid-ventral tissue degenerates to be replaced by the dorso-lateral body wall.
RESULTS: We developed a n

BACKGROUND: While heart function is broadly conserved across vertebrates, the cellular phenotype of muscle cells (cardiomyocytes) varies across taxa and throughout ontogeny. Emerging evidence suggests that some attributes may correlate with the capacity for spontaneous cardiomyocyte replacement following injury. For example, among non-regenerating taxa like adult mammals and birds, cardiomyocytes are polyploid, rarely proliferate, and are large in size. In contrast, in regeneration-competent zebrafish and amphibians, cardiomyocytes are diploid, spontaneously proliferate, and are comparatively
