Species-Specific Adaptations of Trypanosome Morphology and Motility to the Mammalian Host.

African trypanosomes thrive in the bloodstream and tissue spaces of a wide range of mammalian hosts. Infections of cattle cause an enormous socio-economic burden in sub-Saharan Africa. A hallmark of the trypanosome lifestyle is the flagellate's incessant motion. This work details the cell motility behavior of the four livestock-parasites Trypanosoma vivax, T. brucei, T. evansi and T. congolense. The trypanosomes feature distinct swimming patterns, speeds and flagellar wave frequencies, although the basic mechanism of flagellar propulsion is conserved, as is shown by extended single flagellar b

Distribution and abundance of hematophagous flies (Glossinidae, Stomoxys, and Tabanidae) in two national parks of Gabon.

In order to minimize risks of pathogen transmission with the development of ecotourism in Gabon, a seasonal inventory has been performed in five contrasted biotopes in Ivindo (INP) and Moukalaba-Doudou (MDNP) National Parks. A total of 10,033 hematophagous flies were captured. The Glossinidae, with six different species identified, was the most abundant group and constitutes about 60% of the captured flies compared to the Stomoxys (6 species also identified) and Tabanidae with 28% and 12%, respectively. The Glossinidae showed a higher rate of capture in primary forest and in research camps. In

Control of Glossina longipennis (Diptera: Glossinidae) by insecticide-treated targets at Galana ranch, Kenya, and confirmation of the role of G. longipennis as a vector of cattle trypanosomiasis.

Glossina longipennis Corti was studied in Galana Ranch, Kenya over a four year period, in two areas (Tank E and Lali) where the species was abundant and other species were absent or scarce. There was active transmission of trypanosomiasis to cattle in both areas, the parasite species being Trypanosoma vivax Ziemann and T. congolense Broden. Mean infection rates of the G. longipennis were 1.1% and 0. 55% for T. vivax and T. congolense respectively at Tank E, and 0.88% and 0.15% at Lali. Experimental transmission studies showed that cattle in fly-proof enclosures challenged with wild G. longipen

Effect of tsetse control on trypanosome prevalence in livestock: problems of experimental design and statistical interpretation--a case study in northern Côte d'Ivoire.

As part of a study on livestock productivity under trypanosomosis risk in the region of Boundiali, northern Côte d'Ivoire, 21 herds of cattle (N'Dama, Baoulé and Zebu crosses) and 20 flocks of Djallonké and Djallonké x Sahel sheep were monitored monthly for body weight, packed red cell volume and trypanosomal parasitaemia over various periods between January 1984 and December 1992. A tsetse control campaign using biconical traps impregnated with alpha-cypermethrin started in December 1987. Tsetse control reduced the relative tsetse density by over 95% between 1988 and 1992, and this was associ

Pathogenicity of tsetse-transmitted Trypanosoma congolense for waterbuck (Kobus defassa) and Boran cattle (Bos indicus).

Five waterbuck (Kobus defassa) and four Boran cattle (Bos indicus) were infected with Trypanosoma congolense IL2895 using Glossina morsitans morsitans. At the same time, two waterbuck and two cattle were inoculated intravenously with bloodstream forms. With both methods of challenge, cattle had short prepatent periods followed by a continuous high parasitaemia. All cattle became severely anaemic and had to be treated with trypanocidal drugs to prevent death. In contrast, tsetse and intravenous challenge of waterbuck resulted in a long prepatent period, followed by brief, intermittent levels of

Production of metacyclic forms by cyclical transmission of west African Trypanosoma (T.) brucei isolates from man and animals.

Fifteen West African Trypanosoma (T.) brucei isolates from man and animals were cyclically transmitted. Five stocks, belonging to the non-gambiense group, could easily be transmitted through Glossina morsitans morsitans or Glossina m. centralis infected on mice, whereas successful transmission of the 10 isolates, identified as Trypanosoma brucei gambiense, was performed using G. palpalis gambiensis as vector. Glossina p. gambiensis was infected with culture-derived procyclic trypanosomes by repeated membrane feeding. In both cases, metacyclic forms could normally be detected in saliva samples

Sleeping sickness and the factors affecting it in Botswana.

From the first appearance of sleeping sickness in Botswana in 1934 outbreaks increased in severity up to 1971. All populated areas around the fly belt were affected. Rates of infection are highest in the hot wet season and males aged between 30 and 50 years most affected. Rates vary considerably between tribes and have greatly increased in one tribe since 1966. Between 5 and 20% of cases die each year. Increase both in area of tsetse fly infestation and in human population size can account for the increase in severity of outbreaks. Timing of outbreaks correlates with years of low rainfall when

Chemical Inhibition of Bromodomain Proteins in Insect-Stage African Trypanosomes Perturbs Silencing of the Variant Surface Glycoprotein Repertoire and Results in Widespread Changes in the Transcriptome.

The eukaryotic protozoan parasite Trypanosoma brucei is transmitted by the tsetse fly to both humans and animals, where it causes a fatal disease called African trypanosomiasis. While the parasite lacks canonical DNA sequence-specific transcription factors, it does possess histones, histone modifications, and proteins that write, erase, and read histone marks. Chemical inhibition of chromatin-interacting bromodomain proteins has previously been shown to perturb bloodstream specific trypanosome processes, including silencing of the variant surface glycoprotein (VSG) genes and immune evasion. Global distribution, host range and prevalence of Trypanosoma vivax: a systematic review and meta-analysis.

BACKGROUND: Trypanosomosis caused by Trypanosoma vivax is one of the diseases threatening the health and productivity of livestock in Africa and Latin America. Trypanosoma vivax is mainly transmitted by tsetse flies; however, the parasite has also acquired the ability to be transmitted mechanically by hematophagous dipterans. Understanding its distribution, host range and prevalence is a key step in local and global efforts to control the disease. METHODS: The study was conducted according to the methodological recommendations of the Preferred Reporting Items for Systematic Reviews and Meta-A

Drug reformulation for a neglected disease. The NANOHAT project to develop a safer more effective sleeping sickness drug.

BACKGROUND: Human African trypanosomiasis (HAT or sleeping sickness) is caused by the parasite Trypanosoma brucei sspp. The disease has two stages, a haemolymphatic stage after the bite of an infected tsetse fly, followed by a central nervous system stage where the parasite penetrates the brain, causing death if untreated. Treatment is stage-specific, due to the blood-brain barrier, with less toxic drugs such as pentamidine used to treat stage 1. The objective of our research programme was to develop an intravenous formulation of pentamidine which increases CNS exposure by some 10-100 fold, le

Genetic diversity and population structure of the tsetse fly Glossina fuscipes fuscipes (Diptera: Glossinidae) in Northern Uganda: Implications for vector control.

Uganda is the only country where the chronic and acute forms of human African Trypanosomiasis (HAT) or sleeping sickness both occur and are separated by < 100 km in areas north of Lake Kyoga. In Uganda, Glossina fuscipes fuscipes is the main vector of the Trypanosoma parasites responsible for these diseases as well for the animal African Trypanosomiasis (AAT), or Nagana. We used highly polymorphic microsatellite loci and a mitochondrial DNA (mtDNA) marker to provide fine scale spatial resolution of genetic structure of G. f. fuscipes from 42 sampling sites from the northern region of Uganda wh

The role of domestic animals in the epidemiology of human African trypanosomiasis in Ngorongoro conservation area, Tanzania.

BACKGROUND: Trypanosomiasis is a neglected tropical disease caused by the trypanosome parasite and transmitted by the tsetse fly vector. In Sub-saharan Africa, both the human and animal variants of the disease are a great obstacle towards agriculture, development, and health. In order to better understand and therefore combat Trypanosomiasis, characterizing disease hotspots across species is critical. METHODS: In this study, 193 samples from cattle, sheep, and goats were collected from eight sites. Samples were taken from animals belonging mostly to Maasai herdsmen in the Ngorongoro Crater Co

Population genetics of Trypanosoma brucei and the epidemiology of human sleeping sickness in the Lambwe Valley, Kenya.

Numerical taxonomy was used to review isoenzyme variation in isolates of Trypanosoma brucei obtained from cattle, tsetse, humans and wildlife from the Lambwe Valley, Kenya. From isoenzyme information alone, it was possible to classify isolates as to source through the use of linear discriminant functions analysis, with an error rate of only 2% in humans, and 14% over all groups. Differentiation was mostly dependent on patterns in the enzymes ASAT, PEP1, and ICD. Parasites from non-human sources, especially tsetse, were characterized by high isoenzyme diversity, and many unique zymodemes. Obser

The sequential cellular changes in the local skin reaction produced in goats by Glossina morsitans morsitans infected with Trypanosoma (Trypanozoon) brucei.

Sequential biopsies of the skin reaction elicited in goats by Glossina morsitans morsitans infected with Trypanosoma (Trypanozoon) brucei were examined histologically to identify and to quantify the cellular populations involved in the reaction. The peak of the tissue response occurred 7-8 days after challenge with infected tsetse and preceded the initial detection of parasitaemia by 4-5 days. Microscopically, the cellular reaction was characterized initially by a marked infiltration of polymorphonuclear leucocytes (PMN) which were replaced by lymphoid cells. Plasma cells and macrophages were

[Glossina, domestic livestock and wild fauna: is a conciliation possible?].

Animal trypanosomiasis is one of the major obstacles to livestock production in Africa. South of the Sahara. Tsetse fly control is considered to be the best method of overcoming the disease. The adverse effect of Glossina on cattle rearing and their role in the protection of the environment in West Africa is discussed. Meat production possibilities in infested areas, difficulties encountered in tsetse control and related problems are reviewed. Lastly solutions which would assure both livestock development and at the same time protection of the wildlife are examined.

Control of the African trypanosomiases with special reference to land use.

There is a need to reassess the role of the trypanosomiases, both human and animal, in African ecology and economy. Withdrawal of control services from the Congo basin was followed by a resurgence of infection from ancient foci, many of them thought to have been extinguished. Successful elimination of Glossina in savanna habitats has sometimes been followed by reinvasion causing massive epizootics. Elsewhere continued recession of the tsetse fly has been followed by cattle population growth which is periodically interrupted by catastrophic pasture famines.Efficiency of control rather than larg
