DREADD-induced silencing of the medial amygdala reduces the preference for male pheromones and the expression of lordosis in estrous female mice.

Sexually naïve estrous female mice seek out male urinary pheromones; however, they initially display little receptive (lordosis) behavior in response to male mounts. Vomeronasal-accessory olfactory bulb inputs to the medial amygdala (Me) regulate courtship in female rodents. We used a reversible inhibitory chemogenetic technique (Designer Receptors Exclusively Activated by Designer Drugs; DREADDs) to assess the contribution of Me signaling to females' preference for male pheromones and improvement in receptivity normally seen with repeated testing. Sexually naïve females received bilateral Me

Neuronal connections of the central amygdalar nucleus with refeeding-activated brain areas in rats.

Following fasting, satiety is accompanied by neuronal activation in brain areas including the central amygdalar nucleus (CEA). Since CEA is known to inhibit food intake, we hypothesized that CEA contributes to the termination of meal during refeeding. To better understand the organization of this satiety-related circuit, the interconnections of the CEA with refeeding-activated neuronal groups were elucidated using retrograde (cholera toxin-β subunit, CTB) and anterograde (phaseolus vulgaris leucoagglutinin, PHA-L) tracers in male rats. C-Fos-immunoreactivity was used as marker of neuronal acti

Association of stress-sensitive mid-insula activity with alcohol drinking and negative affect-like behavior during abstinence in mice.

Stress is central to many neuropsychiatric conditions, including alcohol use disorder (AUD). Stress influences alcohol initiation, escalation, progression to AUD, and relapse. Identifying stress-activated neurocircuits and individual variability in these responses is critical for developing new AUD treatment targets. This study investigates the relationship between adult stress response and AUD hyperkatifeia-a prolonged negative emotional state in protracted abstinence. In C57BL/6J mice, repeated restraint stress did not alter ethanol consumption but heightened aversive behavior during abstine

Selective spinal interneuron activation enhances the hypercapnic ventilatory response in chronic spinal cord injury.

The ability to sense and regulate breathing in response to changes in carbon dioxide (CO₂) levels is vital for maintaining respiratory and systemic homeostasis. Impaired CO2 chemosensitivity is a common feature of several debilitating conditions, including chronic obstructive pulmonary disease (COPD), central hypoventilation syndromes, and cervical spinal cord injury (cSCI) and contributes to significant respiratory complications such as reduced ventilatory drive, poor gas exchange, and diminished adaptability to metabolic demands. Chronic cSCI disrupts descending respiratory pathways, leading

The role of dorsomedial striatum and parvalbumin interneurons in schizophrenia-related cognitive deficits: insights from disrupted strategic decision-making in Akt1 heterozygous mice.

Schizophrenia, a debilitating disorder with genetic and neurobiological underpinnings, often manifests cognitive deficits, including impaired decision-making. Utilizing Akt1 heterozygous mutant (HET) mice as a model, which mimic schizophrenia due to AKT1's implication as a susceptibility gene, we investigated the involvement of Akt1 and its neural mechanisms influencing strategic decision-making to identify potential therapeutic targets for schizophrenia-associated cognitive impairments. In six experiments, we first revealed that lesions targeting the dorsomedial striatum (DMS) significantly i

Increased sympathetic outflow induced by ventromedial hypothalamic nucleus activation aggravates cardiac electrophysiology disturbance after myocardial ischemia-reperfusion injury.

BACKGROUND AND AIMS: The ventromedial hypothalamus (VMH) is the key brain region related to emotional stress and sympathetic activity, which can exacerbate myocardial ischemia-reperfusion (I/R) injury and induce severe ventricular arrhythmia (VA). We aim to investigate the role of increased sympathetic outflow induced by VMH activation after myocardial I/R injury, especially the changes of cardiac electrical activity. METHODS: Sprague-Dawley rats were divided randomly into the Sham, I/R, hM3d(Gq) and hM4d(Gi) groups, with VMH specifically activated or inhibited using Designer Receptors Exclus

Parabrachial Calca Neurons Influence Aversive and Appetitive Taste Function.

The parabrachial (PB) nucleus participates in taste processing and integration with other senses. PB neurons that express the Calca gene support sensory-integrative responses, albeit only limited data have addressed their influence on taste. Here we investigated how chemogenetic dampening of PB-Calca neurons affected mouse orosensory preferences for diverse taste stimuli in brief-access fluid exposure tests, which capture oral sensory/tongue control of licking behavior. Intracranial delivery of Cre-dependent viruses in female and male Calca Cre/+ mice induced expression of the inhibitory desig

Transcutaneous auricular vagus nerve stimulation inhibits food intake and body weight gain through the orexin dependent pathway in high fat diet mice.

Obesity represents a significant global health challenge, adversely affecting both physical and mental well-being. Neuromodulation techniques have demonstrated efficacy in promoting weight loss by regulating appetite. Recently, transcutaneous vagus nerve stimulation (taVNS) has garnered considerable attention. This study aims to investigate the effects of taVNS on food intake, body weight, and blood glucose levels in mice subjected to a high-fat diet (HFD), and to explore its potential mechanisms. Balb/c male mice were randomly assigned to three groups: control group, HFD + sham electroacupunc

Chronic Striatal Cholinergic Interneuron Excitation Causes Cerebral Palsy-Related Dystonic Behavior in Mice.

OBJECTIVE: Mouse models of genetic dystonias have demonstrated abnormal striatal cholinergic interneuron excitability, but do not consistently demonstrate subjective dystonic features. To determine whether striatal cholinergic interneuron excitation can cause potentially dystonic motor behaviors, we first determined features correlated specifically with dystonia severity in people and then determined whether these features emerged in mice following striatal cholinergic interneuron excitation. METHODS: Eight movement disorders experts rated dystonia severity in 193 videos of people with cerebr

Descending facilitation from rostral ventromedial medulla mu opioid receptor-expressing neurons is necessary for maintenance of sensory and affective dimensions of chronic neuropathic pain.

Pharmacological ablation of rostral ventromedial medulla (RVM) mu opioid receptor-expressing cells before peripheral nerve injury prevents the development of neuropathic pain. However, whether these neurons are required for the expression of established neuropathic pain is not known. Male Oprm1Cre heterozygous (MOR Cre ) or wild-type (MOR WT ) mice received AAV8-hSyn-DIO-hM4D(Gi)-mCherry in the RVM. After partial sciatic nerve ligation (PSNL), we evaluated pain behaviors and descending control of nociception in response to acute or sustained chemogenetic inhibition of RVM-MOR cells expressing

CCK-sensitive C fibers activate NTS leptin receptor-expressing neurons via NMDA receptors.

The hormone leptin reduces food intake through actions in the peripheral and central nervous systems, including in the hindbrain nucleus of the solitary tract (NTS). The NTS receives viscerosensory information via vagal afferents, including information from the gastrointestinal tract, which is then relayed to other central nervous system (CNS) sites critical for control of food intake. Leptin receptors (lepRs) are expressed by a subpopulation of NTS neurons, and knockdown of these receptors increases both food intake and body weight. Recently, we demonstrated that leptin increases vagal activa

Altered calcium signaling in Bergmann glia contributes to spinocerebellar ataxia type-1 in a mouse model of SCA1.

Spinocerebellar ataxia type 1 (SCA1) is a neurodegenerative disease caused by an abnormal expansion of glutamine (Q) encoding CAG repeats in the ATAXIN1 (ATXN1) gene and characterized by progressive cerebellar ataxia, dysarthria, and eventual deterioration of bulbar functions. SCA1 shows severe degeneration of cerebellar Purkinje cells (PCs) and activation of Bergmann glia (BG), a type of cerebellar astroglia closely associated with PCs. Combining electrophysiological recordings, calcium imaging techniques, and chemogenetic approaches, we have investigated the electrical intrinsic and synaptic

The lateral habenula is not required for ethanol dependence-induced escalation of drinking.

The lateral habenula (LHb) is an epithalamic nuclei that has been shown to signal the aversive properties of ethanol. The present study tested the hypothesis that activity of the LHb is required for the acquisition and/or expression of dependence-induced escalation of ethanol drinking and somatic withdrawal symptoms. Male Sprague-Dawley rats completed 4 weeks of baseline drinking under a standard intermittent access two-bottle choice (2BC) paradigm before undergoing 2 weeks of daily chronic intermittent ethanol (CIE) via vapor inhalation. Following this CIE exposure period, rats resumed 2BC dr

Regulation of food intake by astrocytes in the brainstem dorsal vagal complex.

A role for glial cells in brain circuits controlling feeding has begun to be identified with hypothalamic astrocyte signaling implicated in regulating energy homeostasis. The nucleus of the solitary tract (NTS), within the brainstem dorsal vagal complex (DVC), integrates vagal afferent information from the viscera and plays a role in regulating food intake. We hypothesized that astrocytes in this nucleus respond to, and influence, food intake. Mice fed high-fat chow for 12 hr during the dark phase showed NTS astrocyte activation, reflected in an increase in the number (65%) and morphological c

Separate vmPFC Ensembles Control Cocaine Self-Administration Versus Extinction in Rats.

Recent studies suggest that the ventral medial prefrontal cortex (vmPFC) encodes both operant drug self-administration and extinction memories. Here, we examined whether these opposing memories are encoded by distinct neuronal ensembles within the vmPFC with different outputs to the nucleus accumbens (NAc) in male and female rats. Using cocaine self-administration (3 h/d for 14 d) and extinction procedures, we demonstrated that vmPFC was similarly activated (indexed by Fos) during cocaine-seeking tests after 0 (no-extinction) or 7 extinction sessions. Selective Daun02 lesioning of the self-adm

Melanin-concentrating hormone neurons contribute to dysregulation of rapid eye movement sleep in narcolepsy.

The lateral hypothalamus contains neurons producing orexins that promote wakefulness and suppress REM sleep as well as neurons producing melanin-concentrating hormone (MCH) that likely promote REM sleep. Narcolepsy with cataplexy is caused by selective loss of the orexin neurons, and the MCH neurons appear unaffected. As the orexin and MCH systems exert opposing effects on REM sleep, we hypothesized that imbalance in this REM sleep-regulating system due to activity in the MCH neurons may contribute to the striking REM sleep dysfunction characteristic of narcolepsy. To test this hypothesis, we

Signaling pathways coordinating the alkaline pH response confer resistance to the hevein-type plant antimicrobial peptide Pn-AMP1 in Saccharomyces cerevisiae.

Genome-wide screening of Saccharomyces cerevisiae revealed that signaling pathways related to the alkaline pH stress contribute to resistance to plant antimicrobial peptide, Pn-AMP1. Plant antimicrobial peptides (AMPs) are considered to be promising candidates for controlling phytopathogens. Pn-AMP1 is a hevein-type plant AMP that shows potent and broad-spectrum antifungal activity. Genome-wide chemogenomic screening was performed using heterozygous and homozygous diploid deletion pools of Saccharomyces cerevisiae as a chemogenetic model system to identify genes whose deletion conferred enhanc

Bed nucleus of the stria terminalis neurons encode and retrieve cytokine-specific responses in male mice.

The brain monitors peripheral inflammatory states and coordinates adaptive immune and physiological responses to maintain homeostasis. Specific brain regions process motor and sensory information from defined body regions, forming a somatotopic homunculus. Whether an analogous brain organizational principle exists for decoding peripheral cytokine signals and coordinating physiological responses is unknown. Here, using targeted recombination in active populations combined with serial two-photon tomography in male mice, we show that systemically administered tumor necrosis factor (TNF), a pro-in

Negative allosteric modulation of α5-GABAA receptors engages dynamic cortical glutamatergic and GABAergic mechanisms underlying adaptive behavior in mice.

Chronic stress disrupts glutamatergic and GABAergic plasticity in the medial prefrontal cortex (mPFC), impairing circuit integration and contributing to the pathophysiology of stress-related disorders, such as Major Depressive Disorder (MDD). Rapid-acting antidepressants like ketamine can rapidly reverse these deficits, but its clinical use is limited by psychotomimetic side effects. Notably, the α5-GABAAR negative allosteric modulator (α5-NAM) Basmisanil (BSM), reproduces ketamine-like behavioral outcomes in preclinical models, although the cellular mechanisms underlying its actions remain un

Repeated treatment with short-term mild stress reverses aging- and stress-induced emotional and social behavioral deficits.

Aging increases vulnerability to stress-induced neuronal dysfunction, yet the underlying mechanisms remain unclear. Here, in young mice (2 months), chronic stress elevates basal serum glucocorticoid (GC) levels and induces despair-like behavior, as well as impaired sociability. By contrast, aged mice (14.5 months) naturally exhibit elevated basal GC levels, but do not display depressive-like behavior or sociability deficits, although further analysis reveals a social memory impairment. However, exposure to subthreshold stress in aged mice further elevates basal GC levels and induces both emoti
