[Analysis of Pagumogonimus skrjabini antigen and its application in serodiagnosis].

OBJECTIVE: To analyse the soluble antigens of different developmental stages of Pagumogonimus skrjabini and develop a specific and sensitive serodiagnostic method for pagumogonimiasis. METHODS: The soluble antigens of P. skrjabini of various stages were separated by SDS-PAGE. The specific antigen of the adult fluke was recognized immunologically by immunoblot assay. The protein bands between 10-30 kDa purified by SDS-PAGE and electrophoretic elution were used in dot-ELISA. RESULTS: Using dot-ELISA, the soluble antigens of adult were recognized by sera infected with P. skrjabini. More reactiv

Studies on host specificity in Paragonimus westermani: I. Ultrastructural characterization of metacercariae and juvenile and adult worms from abnormal and definitive hosts.

Comparative ultrastructural studies have been made of newly excysted metacercariae and worms recovered from abnormal (rats) and normal definitive hosts (dogs). Worms from rats survived for prolonged periods without any growth or development in muscles of the host. They appeared to resemble metacercariae in general features, although ultrastructural observations revealed differences in the composition of tegumental granules and the development of the excretory bladder and caeca. Worms from dogs showed well-developed morphology in association with active energy metabolism and biosynthesis.

[Biological diagnosis of pulmonary parasitoses and mycoses].

Pulmonary involvement in parasitic disease is now due above all to the visceral Larva migrans syndrome, the diagnosis of which is serological, and to imported distomatosis due to Paragonimus sp, confirmed by the discovery of eggs in the sputum. An immature immunological status or immunodepression is necessary for the development of Pneumocystis carinii, where all of the stages of diagnosis, including that of the obtaining of sample material, are difficult. Pulmonary mycoses are represented above all by candidiosis, aspergillosis and rarely cryptococcosis which also commonly occur in the presen

[A new focus of human paragonimosis discovered in Côte d'Ivoire (West Africa): the case of the Lauzoua Island].

Fifteen cases of human paragonimosis were detected in Ivory Coast from 1974 to 1999. Since no further cases have been reported, an epidemiological survey was carried out in local health centers. The purpose of this paper is to described a new focus of paragonimosis discovered on Lauzoua Island. Clinical and parasitological examinations were performed on 17 patients presenting chronic cough, haemoptysis and/or epilepsy. Stools belonging to cats, dogs and pigs as well as river crustaceans were also examined to identify parasite eggs and metacercariae respectively. Paragonimus eggs were found in

Identities of two Paragonimus species from Sri Lanka inferred from molecular sequences.

Metacercariae of Paragonimus spp. were obtained from field-collected freshwater crabs in Sri Lanka. Genomic DNA was extracted from single metacercariae. Two gene regions (partial mitochondrial cytochrome c oxidase subunit 1 (CO1) and the second internal transcribed spacer of the nuclear ribosomal gene repeat (ITS2)) were amplified using the polymerase chain reaction. Two differing sequences were obtained for each of these gene regions. Phylogenetic analyses placed the type 1 sequences as sister to a clade containing P. westermani and P. siamensis whereas the type 2 sequences were close to publ

Identification of African Paragonimidae by multivariate analysis of the eggs.

A study of metacercariae and adult Paragonimidae revealed the existence in West-Africa of four species, of which two are largely recorded in literature (Paragonimus africanus and Paragonimus uterobilateralis), one was suspected (Paragonimnus westermani-like) and one remained unrecorded in Africa (Euparagonimus sp). Among the two last ones, P. westermani-like was probably confused with P. africanus, and Euparagonimus sp. with P. uterobilateralis. P. westermani-like adult worms differed from P. africanus by the morphology of ovary, testes and the size of the metraterm eggs. Euparagonimus could b

Assessment of the diagnostic efficacy of enolase as an indication of active infection of Schistosoma japonicum.

Schistosomiasis is a common zoonoses affecting humans. The atypical clinical symptoms, low morbidity, and low degree of infection impede diagnosis and assessment of epidemics. Detecting circulating antigens from adult worms in patients' body fluids should be diagnostically superior to examining eggs in feces. Herein, the excretory-secretory proteins of adult worms were analyzed by using 2-D protein electrophoresis and mass spectrometry. The Schistosoma japonicum enolase (Sj enolase) was identified as the most abundant excretory-secretory antigen. Purified recombinant Sj enolase was prepared, a

Molecular variation in the Paragonimus heterotremus complex in Thailand and Myanmar.

Paragonimiasis is an important food-borne parasitic zoonosis caused by infection with lung flukes of the genus Paragonimus. Of the 7 members of the genus known in Thailand until recently, only P. heterotremus has been confirmed as causing human disease. An 8th species, P. pseudoheterotremus, has recently been proposed from Thailand, and has been found in humans. Molecular data place this species as a sister species to P. heterotremus, and it is likely that P. pseudoheterotremus is not specifically distinct from P. heterotremus. In this study, we collected metacercariae of both nominal species

Pleuropulmonary infection by Paragonimus westermani in the United States: a rare cause of Eosinophilic pneumonia after ingestion of live crabs.

Infections caused by the parasite Paragonimus westermani are endemic to Southeast Asia. Most infections reported in the United States are among immigrants who acquired the disease abroad. Due to the nonspecific nature of its presentation and rarity in the United States, the diagnosis may first be suggested by the pathologist on biopsy review. Definitive diagnosis may need serologic testing for confirmation. We report 4 cases of pleuropulmonary disease caused by United States-acquired P. westermani, which were identified in the consultation files of the authors. Patients (3 men and 1 woman; age

Immunoglobulin G (IgG) subclass and IgE responses in human paragonimiases caused by three different species.

In 40 cases of human paragonimiases caused by Paragonimus westermani (20 cases), P. miyazakii (10 cases), and P. skrjabini (10 cases), responses of serum immunoglobulin G (IgG), IgG subclasses, and IgE were analyzed by immunoblotting with crude antigens prepared from egg, 4-week-old juvenile, and adult forms of P. westermani. The 32- and 35-kDa proteins in the adult extracts showed specific reactions regardless of the causative species (39 of 40 cases; 98%). Sera of patients infected with P. westermani and P. miyazakii reacted strongly with the 28-, 46-, and 94-kDa proteins of egg extracts, wh

[Preparation and diagnostic application of monoclonal antibodies against Schistosoma japonicum].

The present paper reported on an anti-CCA monoclonal antibody, McAb-IIID 10, which could be used in determinations of both parasite-oriented circulating antigens and specific anti-CCA antibodies. The established competitive ELISA (C-ELISA) using McAb-IIID 10 to detect schistosome-antibodies showed high sensitivity and specificity in the diagnosis of schistosomiasis with few cross-reactions. In field trials, coincident rates in 3 separate batches of serum samples when subjected to double-blind detections were obtained. A total of 1,915 serum samples had been determined by C-ELISA, among them 11

Serial CT findings of Paragonimus infested dogs and the Micro-CT findings of the worm cysts.

OBJECTIVE: To investigate the serial CT findings of Paragonimus westermani infected dogs and the microscopic structures of the worm cysts using Micro-CT. MATERIALS AND METHODS: This study was approved by the committee on animal research at our institution. Fifteen dogs infected with P. westermani underwent serial contrast-enhanced CT scans at pre-infection, after 10 days of infection, and monthly thereafter until six months for determining the radiologic-pathologic correlation. Three dogs (one dog each time) were sacrificed at 1, 3 and 6 months, respectively. After fixation of the lungs, both

A Young Man With Hemoptysis and Cavitary Lung Lesions.

A man in his 20s presented with 2 months of mild fatigue and intermittent hemoptysis of less than a tablespoon per episode. He was previously healthy and was on no medications. He denied fevers, night sweats, weight loss, wheezing, dyspnea, musculoskeletal symptoms, and rashes. He had emigrated from a South American country to the United States 3 years earlier. He worked as a groundskeeper but had no exposures to animals, mold, or dusts. He reported rare prior cigarette smoking with no history of alcohol or drug use. He was unsure whether he had received the Bacillus Calmette-Guérin vaccine.

The American lung fluke, Paragonimus kellicotti, in a cat model.

Twenty-four specific-pathogen-free domestic cats were infected orally with from 2 to 30 Paragonimus kellicotti metacercariae and followed for 2-61 wk. Following infection by metacercarial cysts dissected from crayfish, the lungs of the cats undergo changes of intense eosinophilic pneumonia, granulomatous pneumonitis, squamous epithelial-lined cyst formation of bronchogenic origin, and finally (late in infection) partial resolution of the host response. Bronchograms and mechanical probing illustrate the patency of cyst communication with bronchi. The modulation of the host response and adaptive

Studies on host specificity in Paragonimus westermani: II. Histochemical and cytochemical characterization of metacercariae and worms from rats and dogs.

Histochemical tests were done on newly excysted metacercariae and worms recovered from an abnormal host (rat) and the definitive host (dog) for some oxidoreductases, phosphatases and glycosidases. The results demonstrate that rat worms have enzymatic distribution and intensities more similar to those of metacercariae than to adult worms from dogs. Ultracytochemical examination of acid and alkaline phosphatase and Mg-ATPase activity was also carried out. Acid phosphatase activity occurred exceptionally in the excretory bladder and caeca of dog worms. No activity was observed in rat worms except
