Machine Learning and Statistical Shape Modelling Methodologies to Assess Vascular Morphology before and after Aortic Valve Replacement.

Introduction: Statistical shape modelling (SSM) is used to analyse morphology, discover qualitatively and quantitatively unique shape features within a population, and generate mean shapes and shape modes that show morphological variability. Hierarchical agglomerative clustering is a machine learning analysis used to identify subgroups within a given population in relation to shape features. We tested the application of both methods in the clinically relevant scenario of patients undergoing aortic valve repair (AVR). Every year, around 5000 patients undergo surgical AVR in the UK. Aims: Evalua

Tree-based machine learning models recover rule-based flare labels from virtual symptom diaries in pediatric Behçet's disease: an in silico proof-of-concept study.

Behçet's disease (BD) in childhood is characterised by recurrent inflammatory flares that can result in significant morbidity, most notably with ocular, mucocutaneous, and musculoskeletal manifestations. Timely detection of impending flares is difficult in routine practice, and digital health technologies coupled with patient-reported symptom diaries have been proposed as a candidate route to real-time disease surveillance. As a first methodological step before any prospective evaluation in real patients, we set out to demonstrate that a diary-based machine learning (ML) pipeline can recover a

An annotated dataset of Building Management System periodic alarms.

This article presents alarm events data collected from 22 Building Management Systems (BMS) from 9 countries (mainly European) and 7 market segments (Commercial, Education, Entertainment, Government, Healthcare, Residential, Transport) over a period of 1 month. 321 alarm event timeseries were collected. Each series corresponds to a single (unknown) user-defined alarm rule switching from "inactive" (reset) to "active" (alarm) state and back. Such a rule usually consists in a simple upper or lower threshold applied to an observed variable such as a room temperature sensor, a valve pressure, etc.

Transcriptomic signatures of classical monocytes reveal pro-inflammatory modules and heterogeneity in polyarticular juvenile idiopathic arthritis.

INTRODUCTION: Polyarticular juvenile idiopathic arthritis (pJIA) is a childhood-onset autoimmune disease. Immune cells contribute to persistent inflammation observed in pJIA. Despite the crucial role of monocytes in arthritis, the precise involvement of classical monocytes in the pathogenesis of pJIA remains uncertain. Here, we aimed to uncover the transcriptomic patterns of classical monocytes in pJIA, focusing on their involvement in disease mechanism and heterogeneity. METHODS: A total of 17 healthy subjects and 18 premenopausal women with pJIA according to ILAR criteria were included. Cla

Comprehensive Analysis Based on the Cancer-Immunity Cycle Identifies a Novel Immunosuppressive Subtype of Bladder Cancer.

BACKGROUND: Immune checkpoint blockade (ICB) has reshaped the approach to treating a broad spectrum of cancers, including bladder cancer (BLCA). However, a substantial fraction of BLCA patients still exhibit resistance despite high programmed death-ligand 1(PD-L1) expression in the tumor. Consequently, it is imperative to characterize the tumor immunosuppressive microenvironment (TME) and ascertain key biomarkers associated with immune evasion in order to stratify patients according to their propensity for immunotherapy resistance. METHODS: A retrospective analysis of RNA sequencing data sour

Comparison of Pleomorphic Dermal Sarcoma to Advanced Cutaneous Squamous Cell Carcinoma and Soft-Tissue Sarcoma.

IMPORTANCE: Pleomorphic dermal sarcomas (PDSs) are classified as sarcomas; however, they share features with cutaneous carcinomas, such as presentation on sun-exposed areas and high tumor mutational burden (TMB). Data regarding management of PDS are limited, and it is unclear whether sarcoma or cutaneous cancer treatment paradigms should apply. At present, there are no published data comparing the mutational genetics of PDSs to other cutaneous tumors and to other soft-tissue sarcomas (STSs). OBJECTIVE: To compare TMB, UV signature, and single-nucleotide and pathway-level mutational profiles o

An archetypal analysis of lumbopelvic profiles could help predict adverse spinopelvic mobility after total hip arthroplasty.

BACKGROUND: The hip‒spine relationship (HSR) plays a crucial role in the outcomes of total hip arthroplasty (THA) as it influences spinopelvic mobility and the risk of prosthetic impingement. Traditional preoperative assessments, which rely on static spinopelvic parameters, often fail to predict postoperative changes. To overcome this limitation, we developed an unsupervised machine learning model on the basis of an archetype analysis that revealed seven distinct lumbopelvic profiles, independent of modifiable postoperative factors. The aim of this study was to apply the trained model to a new

Interactions between NAD+ metabolism and immune cell infiltration in ulcerative colitis: subtype identification and development of novel diagnostic models.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory disease of the colonic mucosa with increasing incidence worldwide. Growing evidence highlights the pivotal role of nicotinamide adenine dinucleotide (NAD+) metabolism in UC pathogenesis, prompting our investigation into the subtype-specific molecular underpinnings and diagnostic potential of NAD+ metabolism-related genes (NMRGs). METHODS: Transcriptome data from UC patients and healthy controls were downloaded from the GEO database, specifically GSE75214 and GSE87466. We performed unsupervised clustering based on differentially exp

Developing models for the diagnosing of ulcerative colitis and prognosis of anti-TNF-α non-response based on neutrophil extracellular trap-associated genes.

BACKGROUND: Neutrophil extracellular traps (NET) play a pivotal role in the pathogenesis of ulcerative colitis (UC) and may contribute to the impaired response to anti-tumor necrosis factor alpha (TNF-α) therapies. However, the functional implications of NET-associated genes in UC remain poorly understood. This study aims to identify key NET-associated molecular signatures in UC, develop diagnostic models based on NET-related biomarkers, and construct predictive models for response to anti-TNF-α therapies (infliximab and golimumab). METHODS: NET-associated genes were obtained from the Kyoto E

Uncovering distinct motor development trajectories in infants during the first half year of life.

Infants undergo significant developmental changes in the first few months of life. While some risk factors increase the risk of developmental disability, such as preterm birth, the developmental trajectories of infants born pre-term (PT) and full-term (FT) present with individual variability. This study aims to investigate whether the utilization of data-driven unsupervised machine learning can identify patterns within groups of infants and categorize infants into specific developmental trajectories. Thirty-four infants, 19 FT and 15 PT, were assessed with the gross and fine motor subscales of

The knowns and unknowns of phlebotomine sand flies (Diptera: Psychodidae) in selected countries of Central Europe.

BACKGROUND: Sand flies are vectors of the protozoan Leishmania spp. and phleboviruses. In Europe, several species are widely distributed in the Mediterranean region and a northward spread can be observed. They can be found regularly also in some regions of Central Europe, with Phlebotomus mascittii being the most cold-tolerant and northerly distributed species, but the knowledge on their distribution in countries such as Germany, Austria, Czechia, Slovakia, and Hungary remains fragmentary because of a lack of comprehensive field studies and a poor understanding of the ecological requirements a

Peripheral HLA-DRhiCD141+ Classical Monocytes Predict Relapse Risk and Worsening in Multiple Sclerosis.

BACKGROUND AND OBJECTIVES: Multiple sclerosis (MS) is an immune-mediated demyelinating disease of the CNS characterized by a heterogeneous disease trajectory, highlighting the need for biomarkers to predict disease activity. Current disease-monitoring tools primarily reflect existing disease damage rather than impending activity. Peripheral blood mononuclear cells (PBMCs) are an ideal source of potential biomarkers due to their accessibility and their known role in MS pathology. Among PBMCs, myeloid cells are key players in MS pathogenic processes, yet they have not been as extensively studied

Deriving phenotype-representative left ventricular flow patterns by reduced-order modeling and classification.

BACKGROUND: Extracting phenotype-representative flow patterns and their associated numerical metrics is a bottleneck in the clinical translation of advanced cardiac flow imaging modalities. We hypothesized that reduced-order models (ROMs) are a suitable strategy for deriving simple and interpretable clinical metrics of intraventricular flow suitable for further assessments. Combined with machine learning (ML) flow-based ROMs could provide new insight to help diagnose and risk-stratify patients. Anomaly detection in the veterinary antibiotic prescription surveillance system (IS ABV).

Antibiotic resistance is one of the major concerns in veterinary and human medicine and poses a considerable threat to both human and animal health. It has been shown that over- or misuse of antibiotics is one of the primary drivers of antibiotic resistance. To develop the surveillance of antibiotic use, Switzerland introduced the "Informationssystem Antibiotika in der Veterinärmedizin" (IS ABV) in 2019, mandating electronic registration of antibiotic prescriptions by all veterinarians in Switzerland. However, initial data analysis revealed a considerable amount of implausible data entries, po
