African sleeping sickness is caused by Trypanosoma brucei, a parasite transmitted by the bite of a tsetse fly. Trypanosome infection induces a severe transcriptional downregulation of tsetse genes encoding for salivary proteins, which reduces its anti-hemostatic and anti-clotting properties. To better understand trypanosome transmission and the possible role of glycans in insect bloodfeeding, we characterized the N-glycome of tsetse saliva glycoproteins. Tsetse salivary N-glycans were enzymatically released, tagged with either 2-aminobenzamide (2-AB) or procainamide, and analyzed by HILIC-UHPL

INTRODUCTION: Human African Trypanosomiasis (HAT), a disease caused by protozoan parasites transmitted by tsetse flies, is an important neglected tropical disease endemic in remote regions of sub-Saharan Africa. Although the determination of the burden of HAT has been based on incidence, mortality and morbidity rates, the true burden of HAT goes beyond these metrics. This study sought to establish the socio-economic burden that households with HAT faced and the coping strategies they employed to deal with the increased burden.
MATERIALS AND METHODS: A mixed methods approach was used and data

Background: Human African trypanosomiasis (HAT) is a protozoal disease transmitted by tsetse flies. Infection with trypanosomes can lead directly to active HAT or latent infection with no detectable parasites, which may progress to active HAT or to spontaneous self-cure. Genetic variation could explain these differences in the outcome of infection. To test this hypothesis, polymorphisms in 17 candidate genes were tested ( APOL1 [ G1 and G2], CFH, HLA-A, HPR, HP, IL1B, IL12B, IL12RB1, IL10, IL4R, MIF, TNFA , IL6, IL4, IL8, IFNG, and HLA-G). Methods: Samples were collected in Democratic Republic

The protozoan parasite Trypanosoma brucei is responsible for sleeping sickness and alternates between mammal and tsetse fly hosts, where it has to adapt to different environments. We investigated the role of two members of the ALBA family, which encodes hypothetical RNA-binding proteins conserved in most eukaryotes. We show that ALBA3/4 proteins colocalize with the DHH1 RNA-binding protein and with a subset of poly(A+) RNA in stress granules upon starvation. Depletion of ALBA3/4 proteins by RNA interference in the cultured procyclic stage produces cell modifications mimicking several morphogen

The satellite DNA sequence of Trypanosoma (Nannomonas) godfreyi, a recently described parasite of Suidae, was determined. The sequence is 373bp in length, and contains two imperfect internal repeats of approximately 170bp. Like other trypanosome satellite DNAs, it has no extensive open reading frames and is probably non-coding. There is no significant homology with other major repetitive DNAs within subgenus Nannomonas. We have developed a PCR test that is specific for T. godfreyi and used it to identify the parasite in natural tsetse infections from Zimbabwe and Côte d'Ivoire. This test shows

Human African Trypanosomiasis is a devastating disease caused by the parasite Trypanosoma brucei. Trypanosomes live extracellularly in both the tsetse fly and the mammal. Trypanosome surface proteins can directly interact with the host environment, allowing parasites to effectively establish and maintain infections. Glycosylphosphatidylinositol (GPI) anchoring is a common posttranslational modification associated with eukaryotic surface proteins. In T. brucei, three GPI-anchored major surface proteins have been identified: variant surface glycoproteins (VSGs), procyclic acidic repetitive prote

Although tools to control sleeping sickness do exist, their use is difficult; areas where intervention is most required often cannot be targeted for lack of appropriate risk indicators. The importance of human behaviour and habits in the manifestation of the disease is clear. In the development of effective new approaches to the control of the disease, information must be gathered about human populations, and their interaction with the environment, in rural as well as in urban and peri-urban areas. The results of a study carried out in Daloa show that use of some methods for the development of

Investigations to identify the causes of high mortalities in cattle in the agropastoral zone (ZAP) of Yalé started in March 1993. African animal trypanosomosis (AAT) was found to be the major constraint, with incidence rates exceeding 30%, justifying a tsetse control programme, which started in March/ April 1994. The treatment of all cattle at bimonthly intervals with deltamethrin 1% pour on and the display of 1500 insecticide impregnated targets during the 6 months of the dry season each year helped to reduce the tsetse populations (Glossina tachinoides and G. morsitans submorsitans) by more

A set of 26 Trypanosoma brucei stocks from various African countries, previously characterized by multilocus enzyme electrophoresis (MLEE) for 18 polymorphic loci, have been selected to be representative of the three T. brucei classic subspecies. The kinetoplast DNA minicircle variable regions from these stocks have been amplified using the polymerase chain reaction (PCR) technique, and hybridized with the amplified variable regions of three T. brucei reference stocks, previously identified as T. brucei brucei, T. brucei gambiense, and T. brucei rhodesiense, respectively. Both T. b. brucei and

The pathogenesis and pathology of African animal trypanosomosis (AAT) in Baoulé, N'Dama/Baoulé-cross-bred and Zebu cattle was studied from 1987 to 1991 in a series of experiments conducted under natural and artificial conditions of challenge at the Centre de Recherches sur les Trypanosomoses Animales (CRTA) in Burkina Faso. This first paper reports on the clinical performance of 64 Baoulé, 10 N'Dama/Baoulé-cross-bred and 20 Zebu cattle, which were transferred to the pastoral zone of Satiri, 50 km northeast of Bobo-Dioulasso, a zone infested with Glossina palpalis gambiensis, G. morsitans submo

Vector-borne diseases are responsible for > 1 million deaths every year but genomic resources for most species responsible for their transmission are limited. This is true for neglected diseases such as sleeping sickness (Human African Trypanosomiasis), a disease caused by Trypanosoma parasites vectored by several species of tseste flies within the genus Glossina We describe an integrative approach that identifies statistical associations between trypanosome infection status of Glossina fuscipes fuscipes (Gff) flies from Uganda, for which functional studies are complicated because the species

Microbial pathogens use environmental cues to trigger the developmental events needed to infect mammalian hosts or transmit to disease vectors. The parasites causing African sleeping sickness respond to citrate or cis-aconitate (CCA) to initiate life-cycle development when transmitted to their tsetse fly vector. This requires hypersensitization of the parasites to CCA by exposure to low temperature, conditions encountered after tsetse fly feeding at dusk or dawn. Here we identify a carboxylate-transporter family, PAD (proteins associated with differentiation), required for perception of this d

The accurate identification of each of the three subspecies of Trypanosoma brucei remains a challenging problem in the epidemiology of sleeping sickness. Advances in molecular characterization have revealed a much greater degree of heterogeneity within the species than previously supposed. Only group 1 T. b. gambiense stands out as a separate entity, defined by several molecular markers. T. b. rhodesiense is generally too similar to sympatric T. b. brucei strains to be distinguished from them by any particular molecular markers. Nevertheless, characterization of trypanosome isolates from human

We have previously described a system for characterising the relationships between trypanosome stocks of the T.brucei group based on Southern blotting with repetitive DNA probes followed by cluster analysis of resultant banding patterns (G. Hide et al. Molec. Bioch. Parasitol. 39, 213-226, 1990). In this study, we extend this analysis to examine the relationships between trypanosome stocks isolated from major sleeping sickness foci in Zambia, Kenya, and Uganda. We show that the trypanosome strains responsible for disease in Zambia are quite distinct from those sampled from the Kenya/Uganda foc

Between 1976 and 1983 in the Busoga Region in Uganda 10,414 male and 9,560 female patients were detected with trypanosomiasis. Prior to the onset of the epidemic, Glossina f. fuscipes, the sole vector, had broken through the tsetse control barrier and had spread to cover most of the region. The fly was further observed to be resting in Lantana plants and coffee and banana plantations which are widespread in the area. A close and regular contact was thus established between man and fly. It was observed that the young and engergetic persons first became infected, but as the epidemic unfolded the

Trypanosomes cause disease in humans and livestock throughout sub-Saharan Africa. Although various species show evidence of clinical tolerance to trypanosomes, until now there has been no evidence of acquired immunity to natural infections. We discovered a distinct peak and decrease in age prevalence of T. brucei s.l. infection in wild African lions that is consistent with being driven by an exposure-dependent increase in cross-immunity following infections with the more genetically diverse species, T. congolense sensu latu. The causative agent of human sleeping sickness, T. brucei rhodesiense

Two FAO/IAEA indirect enzyme-linked immunosorbent assays (ELISA), which use microplates precoated with denatured crude Trypanosoma congolense or Trypanosoma vivax antigen for detecting anti-trypanosomal antibodies in bovine sera, were evaluated for their sensitivity, specificity and positive and negative predictive values, using 320 Ugandan field samples (known negative sera, n = 80; known positive sera, n = 80; cattle herds where control of tsetse and trypanosomosis was practiced, n = 80; and cattle herds where there was no such control, n = 80). Cut-off points of 30% and 25% positivity were

An overview of the evolution of sleeping sickness and control activities in the DRC during the period 1989-1998 is presented. A resurgence was already developing in the mid-1980s and, after a breakdown of active case-finding between 1990 and 1993, annual detection rates attained levels similar to those of the late 1920s. Although a staggering number of 150 591 new cases have been detected during the past decade, the problem is ignored by most of the international community. The major cause for the resurgence appears to be the interruption of active case-finding for a prolonged period of time.

Tsetse have been cleared from large areas of Zimbabwe during the past 65 years. In most areas, they are prevented from re-invading cleared areas by barriers of odour-baited, insecticide-treated targets. A trypanosomosis survey was conducted to determine the effectiveness of such barriers against re-invasion and to confirm the absence of tsetse in areas where they had previously been eradicated. Parasitological diagnostic methods and an anti-trypanosomal antibody detection enzyme-linked immunosorbent assay (antibody ELISA) were used. The prevalence of trypanosomal infections in the tsetse-clear
