Helicobacter pylori-induced cystathionine-γ-lyase drives redox imbalance and metabolic remodeling in gastric cancer.

BACKGROUND: Helicobacter pylori (H. pylori) infection is a major risk factor for gastric cancer (GC) and depends on nutrient and energy metabolism, particularly amino acids, for colonization. In host cells, cysteine is regulated by the transsulfuration (TSS) pathway, in which cystathionine-γ-lyase (CSE) generates cysteine, hydrogen sulfide (H2S), and reactive sulfur species (RSS). Dysregulation of this pathway may contribute to gastric carcinogenesis. This study aims to investigate the role of CSE in H. pylori-associated GC and to characterize the associated alterations in sulfur metabolism an

Antiviral and virucidal activities against SARS-CoV-2 and antibacterial properties of bile acids and their salts with naturally occurring organic cations of l-carnitine, creatinine, and choline.

Bile acids have many roles in biological systems, and they have received great attention recently. Bile from a cow, known as gall, together with garlic, wine, and leeks, is used in the traditional medicine recipe of Bald's Leechbook, a thousand-year-old Anglo-Saxon formula for the treatment of infected eyelash follicles. Different aspects of previous works on bile acids have been reported, and this work adds antiviral, virucidal, and antibacterial properties of bile acids and their salts against SARS-CoV-2. Four bile acids, lithocholic acid (LCA, 1), deoxycholic acid (DCA, 5), ursodeoxycholic

Diallyl thiosulfinate enhanced the anti-cancer activity of dexamethasone in the side population cells of multiple myeloma by promoting miR-127-3p and deactivating the PI3K/AKT signaling pathway.

BACKGROUND: Side population (SP) cells, which have similar features to those of cancer stem cells, show resistance to dexamethasone (Dex) treatment. Thus, new drugs that can be used in combination with Dex to reduce the population of SP cells in multiple myeloma (MM) are required. Diallyl thiosulfinate (DATS, allicin), a natural organosulfur compound derived from garlic, has been shown to inhibit the proliferation of SP cells in MM cell lines. Therefore, we investigated the effect of a combination of DATS and Dex (DAT + Dex) on MM SP cells. METHODS: SP cells were sorted from MM RPMI-8226 and

Neuroprotective mechanisms of S-allyl-L-cysteine in neurological disease.

S-allyl-L-cysteine (SAC) is a sulfur-containing amino acid present in garlic and exhibits a wide range of biological activities such as antioxidant, anti-inflammatory, and anticancer agent. An earlier study demonstrated that SAC ameliorates oxidative damage in a model of experimental stroke. However, the antioxidant property of SAC does not suffice to explain its beneficial effects in terms of the underlying mechanisms. Endoplasmic reticulum (ER) stress and ER stress-induced cell death have been shown to be involved in various neurological diseases such as brain ischemia, Alzheimer's disease,

S-allyl cysteine restores erectile function through inhibition of reactive oxygen species generation in diabetic rats.

Excessive production of reactive oxygen species (ROS) by an overactive nicotinamide adenine dinucleotide phosphate (NADPH) oxidase system in penile tissue is an important mechanism of erectile dysfunction (ED). S-allyl cysteine (SAC), a bioactive component derived from garlic, was recently reported to exert versatile antioxidant properties. We hypothesized that SAC would be able to resolve diabetes-related ED by reducing ROS generation, and designed this study to investigate this possibility as well as to determine the related underlying mechanisms. Complementary and alternative medicine use decreases adherence to HAART in HIV-positive women.

The use of complementary and alternative medicine (CAM) to treat chronic illnesses, especially HIV, is becoming increasingly widespread. Given this popularity, it is critical to understand how HIV-positive individuals use CAM and, more specifically, whether CAM use impacts their adherence to prescribed antiretroviral regimens (HAART). The present study examined the relationship between CAM use and HAART adherence among HIV+ women. Data were analysed from 366 HIV-positive, mostly African-American women, aged 18-50 years in Alabama and Georgia who were enrolled in an intervention to reduce high-

[Asthma and food allergy: report of 163 pediatric cases].

The prevalence of food as a cause for asthma is not well known. The aim of this study was to define with standardized tests the incidence of food-induced asthma, the distribution of foods allergens in asthmatic children with food allergy. The study was carried on 163 asthmatic children with food allergy followed during average of 5.5 years. Asthma has been identified with pulmonary function tests (reversibility of FEV1 to bronchodilators) and food allergy has been documented by double-blind placebo-controlled food challenge (DBPCFC). Familial atopic disease was found in 148 children (90.7%). I

Prevention of acetaminophen-induced cataract by a combination of diallyl disulfide and N-acetylcysteine.

Injection of acetaminophen (APAP) (350 mg/kg body weight) into C57BL/6 mice in which cytochrome P450 (CYP) 1A1/1A2 had been induced produced acute cataract and other ocular tissue damage. Treatment of APAP-injected mice with one of the major organosulfides in garlic oil, diallyl disulfide (DADS) (200 mg/kg body weight), prevented cataract development and prolonged survival time. N-acetyl L-cysteine (NAC) (500 mg/kg body weight), a prodrug that stimulates glutathione synthesis, also prolonged survival time but was effective only weakly to prevent cataract formation. Herbal Medicines and Drugs Interactions: Cytochrome P450 Responsibility.

Despite the global prevalence of herbal medicine use and the common perception of their safety, substantial evidence indicates a significant potential for clinically relevant herb-drug interactions that may affect the pharmacokinetics and pharmacodynamics of co-administered pharmaceuticals. This critical issue poses a considerable threat to patient safety and the effectiveness of conventional treatments, primarily through modulation of the cytochrome P450 (CYP450) enzyme system, a key metabolic pathway for many drugs. This research aims to elucidate how herbal remedies affect CYP450 enzyme act

An activatable naphthalimide-based fluorescent probe for hydrogen peroxide detection: visualizing microcystin-LR induced oxidative stress in zebrafish and hepatocyte injury mechanism.

Microcystin-LR (MC-LR), a toxic cyanotoxin, accumulates in fish through uptake, posing severe threats to fisheries' safety and human health. The interaction between MC-LR and aquatic fish induces oxidative stress, which triggers excessive production of reactive oxygen species (ROS), notably hydrogen peroxide (H2O2). Abnormal ROS fluctuations are closely linked to the onset of various diseases, making the development of in vivo H2O2 detection and regulation methods critically important. Herein, we designed and synthesized an activatable fluorescent probe using naphthalimide as the fluorophore.
