Evaluating an extraction-free sample preparation method for multiplex detection of SARS-Cov-2, influenza A/B, and RSV with implementation on a microfluidic chip.

Following the relaxation of COVID-19 restrictions, other respiratory viruses such as influenza and respiratory syncytial virus (RSV), whose transmission were decreased due to COVID-19 precautions, are rising again. Because of similar clinical features and reported co-infections, multiplex detection of SARS-CoV-2, influenza A/B, and RSV is required to use specific treatments. This study assessed an extraction-free sample preparation (heat treatment at 95°C for 3 minutes) for multiplex detection using rRT-PCR. Despite an observed Ct-delay (∆Ct) averageing 1.26 compared to the standard method, an

Affinity-based 3D-printed microfluidic chip for clinical sepsis detection with CD69, CD64, and CD25.

Sepsis is a life-threatening immune response to infection in the body, eventually resulting in fatal organ failure. Current methods utilize blood cultures and quick-Sequential-Organ-Failure-Assessment (qSOFA), but there is a need for more accurate and time-sensitive diagnostic methods to improve survival rates. We present a 3D-printed microfluidic chip that bioconjugates antibodies CD69, CD64, and CD25 to channel surfaces to capture sepsis cells in blood samples and validate it with clinical samples (n = 125 septic, n = 10 healthy). Other variables were taken such as healthy volunteer blood sa

Detection of circulating tumor cells with a novel microfluidic system in malignant pleural mesothelioma.

Detection of rare tumor cells circulating in the blood (CTCs) presents technical challenges. CellSearch, the only approved system for clinical use, fails to capture epithelial cell adhesion molecule-negative CTCs such as malignant pleural mesothelioma (MPM). We have developed a novel microfluidic device (CTC-chip) in which any Ab to capture CTCs is conjugated. The CTC-chip was coated with an Ab against podoplanin that is abundantly expressed on MPM. Circulating tumor cell-detection performance was evaluated in experimental models in which MPM cells were spiked in blood sampled from a healthy v

A Novel High-Throughput Method for Molecular Detection of Human Pathogenic Viruses Using a Nanofluidic Real-Time PCR System.

Human enteric viruses are recognized as the main causes of food- and waterborne diseases worldwide. Sensitive and quantitative detection of human enteric viruses is typically achieved through quantitative RT-PCR (RT-qPCR). A nanofluidic real-time PCR system was used to develop novel high-throughput methods for qualitative molecular detection (RT-qPCR array) and quantification of human pathogenic viruses by digital RT-PCR (RT-dPCR). The performance of high-throughput PCR methods was investigated for detecting 19 human pathogenic viruses and two main process controls used in food virology. The c

Impact of adipose-derived stem cells on engineering hair follicle germ-like tissue grafts for hair regenerative medicine.

Hair regenerative medicine has emerged as a promising treatment strategy for severe hair loss, such as end-stage androgenetic alopecia. Various approaches to engineering three-dimensional tissue grafts have been explored since they drive the ability to regenerate hair follicles when transplanted. In the present study, we demonstrated the assembly of human adipose-derived stem cells (hASCs) into hair follicle germ (HFG)-like aggregates for de novo hair regeneration. We mixed human dermal papilla cells (hDPCs), murine embryonic epithelial cells, and hASCs in suspension, and allowed them to form

The reversible capillary field effect transistor: a capillaric element for autonomous flow switching.

New flow control elements in capillaric circuits are key to achieving ever more complex lab-on-a-chip functionality while maintaining their autonomous and easy-to-use nature. Capillary field effect transistors valves allow for flow in channels to be restricted and cut off utilising a high pressure triggering channel and occluding air bubble. The reversible capillary field effect transistor presented here provides a new element that can restore fluid flow in closed microchannels via autonomous circuit feedback. This allows new flow switching functionality without the need for direct user input.

This study presents a slidable paper-embedded plastic microdevice fully integrated with DNA extraction, loop-mediated isothermal amplification (LAMP), and colorimetric detection functionalities. The developed microdevice consists of three layers that allow a sliding movement to mix the sample and reagents for DNA purification, amplification, and detection in a sequential manner. An FTA card was employed in the main chamber for DNA extraction and purification from intact bacterial cells. Subsequently, LAMP reagents and fuchsin-stored chambers were pulled toward the main chambers for DNA amplifi

Simplifying Nucleic Acid Amplification from Whole Blood with Direct Polymerase Chain Reaction on Chitosan Microparticles.

Tremendous advances have been made in the development of portable nucleic acid amplification devices for near-patient use. However, the true limitation in the realization of nucleic acid amplification tests (NAATs) for near-patient applications is not the amplification reaction, it is the complexity of the sample preparation. Conventional approaches require several precise intervention steps during the protocol. There are numerous reports in the literature that mimic the sample preparation procedure within a lab-on-a-chip device or cartridge, but these systems require a high number of integrat

A microphysiological system HHT-on-a-chip platform recapitulates patient vascular lesions.

Hereditary Hemorrhagic Telangiectasia (HHT) is a rare congenital disease in which fragile vascular malformations (VM) focally develop in multiple organs. There are few treatment options and no cure. HHT patients inherit loss-of-function mutations affecting Alk1-Eng signaling; however, why this manifests as VMs remains poorly understood. Here we have developed a fully human cell-based microphysiological system of perfused vasculature in which inducible shRNA controls endogenous Alk1 in primary endothelial cells (EC). Resulting VMs develop over several days, recapitulate patient VM appearance, a

Quantitative single-cell imaging of efflux pump heterogeneity and antibiotic response dynamics.

Understanding how bacterial cells survive antibiotic treatment requires tools capable of resolving dynamic physiological heterogeneity at single-cell resolution. Here, we present a high-throughput, lineage-resolved microfluidic imaging platform for quantifying the expression and spatial distribution of the AcrAB-TolC efflux pump in Escherichia coli during antibiotic exposure. The system is built around a custom-designed multilayer microfluidic device, fabricated using a direct-write photolithography protocol, which ensures uniform delivery of nutrients and antibiotics while confining thousands

Nanomagnetic System for Rapid Diagnosis of Acute Infection.

Pathogen-activated antibody-secreting cells (ASCs) produce and secrete antigen-specific antibodies. ASCs are detectable in the peripheral blood as early as 3 days after antigen exposure, which makes ASCs a potential biomarker for early disease detection. Here, we present a magnetic capture and detection (MCD) assay for sensitive, on-site detection of ASCs. In this approach, ASCs are enriched through magnetic capture, and secreted antibodies are magnetically detected by a miniaturized nuclear magnetic resonance (μNMR) system. This approach is based entirely on magnetics, which supports high con

Artificial neural network analysis of electroosmotic triple diffusion Ree-Eyring nanofluid transport in a complex ciliated channel.

Complex cilia waves have significant applications in biomedical engineering and physiological fluid transport, where they accurately model the coordinated motion of cilia in respiratory tracts, reproductive systems, and microfluidic devices. They also enhance mixing, pumping efficiency, and targeted transport in drug delivery systems, bio-inspired micropumps, and lab-on-chip technologies. An electroosmotic MHD Ree-Eyring nanofluid transport through a symmetric transverse porous channel is investigated by incorporating triple diffusive convection, thermal radiation, viscous dissipation, and Jou

Future perspective: high-throughput construction of new ultrasensitive cytokine and virion liquid chips for high-throughput screening (HTS) of anti-inflammatory drugs or clinical diagnosis and treatment of inflammatory diseases.

Pathogen-host cell interactions play an important role in many human infectious and inflammatory diseases. Several pathogens, including Escherichia coli (E. coli), Mycobacterium tuberculosis (M. tb), and even the recent 2019 novel coronavirus (2019-nCoV), can cause serious breathing and brain disorders, tissue injury and inflammation, leading to high rates of mortality and resulting in great loss to human physical and mental health as well as the global economy. These infectious diseases exploit the microbial and host factors to induce serious inflammatory and immunological symptoms. Thus the

A three-dimensional in vitro dynamic micro-tissue model of cardiac scar formation.

In vitro cardiac models able to mimic the fibrotic process are paramount to develop an effective anti-fibrosis therapy that can regulate fibroblast behaviour upon myocardial injury. In previously developed in vitro models, typical fibrosis features were induced by using scar-like stiffness substrates and/or potent morphogen supplementation in monolayer cultures. In our model, we aimed to mimic in vitro a fibrosis-like environment by applying cyclic stretching of cardiac fibroblasts embedded in three-dimensional fibrin-hydrogels alone. Using a microfluidic device capable of delivering controlle

Enhanced neuroinvasion by smaller, soluble prions.

Infectious prion aggregates can propagate from extraneural sites into the brain with remarkable efficiency, likely transported via peripheral nerves. Yet not all prions spread into the brain, and the physical properties of a prion that is capable of transit within neurons remain unclear. We hypothesized that small, diffusible aggregates spread into the CNS via peripheral nerves. Here we used a structurally diverse panel of prion strains to analyze how the prion conformation impacts transit into the brain. Two prion strains form fibrils visible ultrastructurally in the brain in situ, whereas th

A ZnO-nanorod/PEDOT:PSS nanocomposite functionalized bridge-like membrane type nanomechanical sensing device for ultrasensitive blood lead detection.

Lead (Pb) ion detection poses a critical problem, particularly in environmental monitoring, industrial operations, and public health, especially for young children and expecting women. Determining lead levels in blood early on is essential to minimizing the long-term consequences of lead exposure. Several sophisticated detection instruments, such as mass spectrometers which perform with high sensitivity, specificity and accuracy, but require a lab-based setting, multi-step sample preparation, expensive payment and professional operation. It is evident that a highly sensitive, portable, low-cos

Portable Sensing Systems in Biological and Chemical Analyses: A Review of Sensor Technologies, Miniaturized Platforms, Data Processing, and Field Applications.

Portable sensing systems are increasingly important in biological and chemical analyses because they can provide analytical information at the point of decision-making. While traditional laboratory methods remain crucial for reference measurements, regulatory validation, and high-precision quantification, portable systems emphasize rapid response, convenience, cost-effectiveness, robustness, and relevance to decision-making. This paper views portable sensing systems as integrated analytical platforms rather than isolated sensing elements. The paper discusses recognition elements, including enz
