The histidine kinase NahK in Pseudomonas aeruginosa is essential for nitric oxide-stress resistance in nutrient starved media.

UNLABELLED: Biofilms are communities of bacteria growing within a matrix composed of polymeric substances that act as a barrier to antimicrobials, making bacteria within a biofilm recalcitrant to conventional antibiotic treatments. This lifestyle of bacteria is especially relevant during its pathogenesis, as observed in the case of Pseudomonas aeruginosa infections of immunocompromised patients. However, pico- to nanomolar concentrations of nitric oxide (NO) have been shown to be efficient in triggering P. aeruginosa to disperse from biofilms, suggesting that a combination of NO exposure and a

Metatranscriptomic insights into the effect of Lactiplantibacillus plantarum inoculation on fermentation metabolism, bacterial community function and antibiotic resistome of corn silage.

Lactic acid bacteria inoculants are widely applied in silage production; however, it remains unclear how Lactiplantibacillus plantarum inoculation shapes temporal changes in microbial function during ensiling, and whether it accelerates normal successional process or is associated with changes in antibiotic resistome dynamics. In this study, we applied an integrated metatranscriptomic approach combining microbial community profiling, differential expression analysis, carbohydrate-active enzyme (CAZy) profiling, and resistome characterization to analyze whole-crop corn silage inoculated with L.

Nitrobacter and Nitrospira genera as representatives of nitrite-oxidizing bacteria: detection, quantification and growth along the lower Seine River (France).

Pollution from agriculture and urban effluents influences the ecology and biochemical functioning of the Seine River. Nitrification dominates nitrogen transformations downstream of the effluents of the Paris wastewater treatment plant (WWTP) at Achères, treating, by activated sludge the wastewater of 6.5 million inhabitant equivalents from Paris and its suburbs, without nitrification and denitrification treatment. It discharges effluents containing large amounts of nitrogen, ammonium mostly (approximately 30 mg L(-1) N-NH(4+) L(-1)), on average 45 mg L(-1) of suspended particulate matter, high

The nitrogen isotopic composition of biotic components in Northern Biscayne Bay and Associated Waterways, South Florida USA: implications for sources of nitrogen.

Over an approximate twelve-month period, between June 2021 and June 2022, the δ15N values of aquatic vegetation and sediments and the δ18O and δ15N values of the NO3- in water samples were analyzed in order to understand the sources and processes affecting nitrogen entering the northern portion of Biscayne Bay, an urban area located to the west of Miami, in the southern United States. Samples were collected from 22 sites, primarily located in waterways feeding into the Bay (Miami River, Little River, Biscayne Canal, and Snake Creek). Additional samples were also collected in the central region

Microbial community structure and antibiotic resistance profiles in sediments with long-term aquaculture history.

The aim of this investigation was to examine the microbial populations and their resistance patterns towards antibiotics, including the impact of nitrogen metabolism in response to the reintroduction of antibiotics, as well as the presence of resistance genes in sediments from shrimp ponds that have been utilized for extended periods of 5, 15, and over 30 years. Results showed that the sediments exhibited a high prevalence of Proteobacteria, Bacteroidetes, Planctomycetes, Chloroflexi, and Oxyphotobacteria as the most abundant bacterial phyla, accounting for 70.35-77.43% of the total bacterial

Removal of nitric oxide in a biotrickling filter under thermophilic condition using Chelatococcus daeguensis.

UNLABELLED: The development of a thermophilic biotrickling filter (BTF) system to inoculate a newly isolated strain of Chelatococcus daeguensis TAD1 for the effective treatment of nitric oxide (NO) is described. A bench-scale BTF was run under high concentrations of NO and 8% O2 in thermophilic aerobic environment. A novel aerobic denitrifier Chelatococcus daeguensis TAD1 was isolated from the biofilm of an on-site biotrickling filter and it showed a denitrifying capability of 96.1% nitrate removal rate in a 24 h period in aerobic environment at 50 degrees C, with no nitrite accumulation. Enhancing aerobic granulation for biological nutrient removal from domestic wastewater.

This study focuses on the enhancement of aerobic granulation and biological nutrient removal maintenance treating domestic wastewater. Two sequencing batch reactors (SBRs) were inoculated with either only floccular sludge (100%-floc SBR) or supplemented with 10% crushed granules (90%-floc SBR). Granules developed in both reactors. The 100%-floc SBR achieved 75% of nitrogen and 93% of phosphorus removal at the end of the performance, but some floccular sludge remained in the system. The 90%-floc SBR became fully granulated and finished with 84% and 99% of nitrogen and phosphorus removal, respec

Judging the sources of inferior groundwater quality and health risk problems through intake of groundwater nitrate and fluoride from a rural part of Telangana, India.

Evaluation of groundwater quality and related health hazards is a prerequisite for taking preventive measures. The rural region of Telangana, India, has been selected for the present study to assess the sources and origins of inferior groundwater quality and to understand the human health risk zones for adults and children due to the consumption of nitrate ([Formula: see text])- and fluoride (F-)-contaminated groundwater for drinking purposes. Groundwater samples collected from the study region were determined for various chemical parameters. Groundwater quality was dominated by Na+ and [Formu

Conceptual Model of Biofilm Antibiotic Tolerance That Integrates Phenomena of Diffusion, Metabolism, Gene Expression, and Physiology.

Transcriptomic, metabolomic, physiological, and computational modeling approaches were integrated to gain insight into the mechanisms of antibiotic tolerance in an in vitro biofilm system. Pseudomonas aeruginosa biofilms were grown in drip flow reactors on a medium composed to mimic the exudate from a chronic wound. After 4 days, the biofilm was 114 μm thick with 9.45 log10 CFU cm-2 These biofilms exhibited tolerance, relative to exponential-phase planktonic cells, to subsequent treatment with ciprofloxacin. The specific growth rate of the biofilm was estimated via elemental balances to be app

Temporal dynamics of soil microbial C and N cycles with GHG fluxes in the transition from tropical peatland forest to oil palm plantation.

Tropical peatlands significantly influence local and global carbon and nitrogen cycles, yet they face growing pressure from anthropogenic activities. Land use changes, such as peatland forests conversion to oil palm plantations, affect the soil microbiome and greenhouse gas (GHG) emissions. However, the temporal dynamics of microbial community changes and their role as GHG indicators are not well understood. This study examines the dynamics of peat chemistry, soil microbial communities, and GHG emissions from 2016 to 2020 in a logged-over secondary peat swamp forest in Sarawak, Malaysia, which

Fifty years of environmental progress for United States dairy farms.

Dairy farms in the United States have changed in many ways over the past 50 yr. Milk production efficiency has increased greatly, with ∼30% fewer cows producing about twice the amount of milk today. Other improvements include increases in crop yields, fuel efficiency of farm equipment, and efficiency in producing most resources used on farms (e.g. electricity, fuel, fertilizer). These improvements have led to changes in the environmental impact of farms. Through simulation of representative dairy farms in 1971 and 2020, changes in nutrient losses and farmgate life cycle assessments of greenhou

Environmental nitrogen losses in winter wheat across yield zones under long-term precipitation variability.

Nitrogen (N) loss dynamics from winter wheat in humid agroecosystems remain uncertain due to variability in soil characteristics across yield zones and high interannual precipitation variability. We hypothesized that dominant N loss pathways vary significantly across yield zones, driven primarily by the interaction of soil hydrology and rainfall patterns. The objective of this study was to integrate field experimentation with process-based modeling to identify and quantify environmental N loss (Nenvloss) pathways across contrasting yield zones under long-term climatic variability. In this stud

Soil moisture dynamics, fertiliser timing and fertiliser nitrogen form (nitrate: ammonium ratio) drive nitrous oxide (N2O) emission patterns in a temperate grassland.

Although both straight N and NPK (nitrogen, phosphorus, potassium) compound fertilisers are widely used, the influence of nitrate (NO3--N) to ammonium (NH4+-N) ratios in these fertilisers on nitrous oxide (N2O) emission dynamics remains poorly quantified. This study examined how fertilisers with contrasting NO3--N to NH4+-N ratios influence N2O emissions under varying environmental conditions, over a growing year in an Irish grassland soil. A randomised block field experiment was conducted on a moderately drained luvisol with a perennial ryegrass (Lolium perenne) sward. Fertilisers with varyin

Effect of Fe2+-activated persulfate combined with biodegradation in removing gasoline BTX from karst groundwater: A box-column experimental study.

In-situ chemical oxidation with persulfate (PS-ISCO) is a preferred approach for the remediation of fuel-contaminated groundwater. Persulfate (PS) can be activated by various methods to produce stronger sulfate radicals for more efficient ISCO. Despite karst aquifers being widespread, there are few reports on PS-ISCO combined with Fe2+-activated PS. To better understand the effects of Fe2+-activated PS for the remediation of gasoline-contaminated aquifers in karst areas, a box-column experiment was conducted under flow conditions, using karst groundwater and limestone particles to simulate an

Diversity of dirammox microorganisms.

UNLABELLED: Our knowledge of the nitrogen cycle has been advanced by the discovery of anammox bacteria, ammonia-oxidizing archaea, and comammox bacteria, along with their high diversity. Direct ammonia oxidation (dirammox), a recently discovered heterotrophic nitrification pathway, converts ammonium and organic nitrogen directly to N2 through the DnfABC multienzyme system, further expanding our understanding of the nitrogen cycle. However, the taxonomic distribution, prevalence, and nitrogen metabolic potential of microorganisms encoding dirammox genes remain poorly understood. Here, large-sca

Prescribed burns drive lasting changes in soil nitrogen cycling and microbial function.

Fire is a major pulse disturbance to soil microbial communities, with broad implications for nutrient cycling; however, regular burning is also a natural and often-essential process maintaining biodiversity in unique and imperiled fire-dependent ecosystems. Prescribed fire is widely used to promote this biodiversity and simultaneously reduce wildfire risk. Although such repeated burning is known to alter surface biodiversity, belowground soil geochemistry, and soil microbial community structure, the functional consequences (i.e., the metabolic capabilities that underlie the ecosystem services

Nitrous oxide is the main product during nitrate reduction by a novel lithoautotrophic iron(II)-oxidizing culture from an organic-rich paddy soil.

Microbial nitrate reduction coupled to iron(II) oxidation (NRFeOx) occurs in paddy soils due to high levels of dissolved iron(II) and regular application of nitrogen fertilizer. However, to date, there is no lithoautotrophic NRFeOx isolate or enrichment culture available from this soil environment. Thus, resulting impacts on greenhouse gas emissions during nitrate reduction (i.e., nitrous oxide [N2O]) and on toxic metalloid (i.e., arsenic) mobility can hardly be investigated. We enriched a lithoautotrophic NRFeOx culture, culture HP (Huilongpu paddy, named after its origin), from a paddy soil
