Phytochemical, In Silico, In Vitro, and In Vivo Research on Piptadeniastrum africanum (Fabaceae) Unveiling Anti-Stereotypic, Anxiolytic, and Analgesic Effects in a Sodium Valproate-Induced Autistic Disorders Model.

OBJECTIVE: Individuals with autistic spectrum disorders (ASD) primarily exhibit deficits in communication and social interaction, along with repetitive behaviors and restricted interests. This disorder is often associated with anxiety, nociceptive disorders, and pain. While medical treatment generally focuses on treating the symptoms rather than addressing the underlying causes, traditional medicine is sometimes used as an alternative. Piptadeniastrum africanum is used in Cameroonian medicinal folks to treat cognitive disorders. However, its effects and mechanisms of action regarding the inhib

Netrin-1 Therapy Restores Partial Hindlimb Movement in a Rat Model of High-Severity Chronic Spinal Cord Injury.

Spinal trauma caused by contusion or compression is the leading cause of spinal cord injury (SCI) worldwide. These injuries frequently progress to a chronic phase, especially in cases of severe damage. This process results in permanent impairment, affecting both physiological functions and voluntary motor control below the lesion level. At cellular level, the formation of a glial scar, which delineates the cystic cavity, interrupts the connectivity between the central nervous system (CNS) and muscles, as well as the neural communication between the peripheral and CNSs. This process, combined w

Central monoamine dysfunction in Prader-Willi syndrome: a mechanistic framework for core clinical features.

Prader-Willi syndrome (PWS) is a rare genetic neurodevelopmental and neurobehavioral disorder characterized by hyperphagia and behavioral, endocrine, arousal, and cognitive dysregulation. Although caused by loss of paternally expressed, maternally imprinted genes in 15q11-q13, the mechanisms linking this lesion to the broad clinical phenotype remain incompletely understood. This review synthesizes evidence that central monoaminergic dysfunction, encompassing serotonin, dopamine, and norepinephrine, provides an integrative, though not exclusive, mechanistic framework for core features of PWS. Postnatal and Juvenile Fluoxetine Treatment Evokes Sex-Specific, Opposing Effects on Mood-Related Behavior, Gene Expression, Mitochondrial Function, and Dendritic Architecture in the Rat Medial Prefrontal Cortex.

BACKGROUND: Serotonin shapes emotional neurocircuit development, and serotonergic neurotransmission is implicated in both the pathophysiology and treatment of neuropsychiatric disorders. The selective serotonin reuptake inhibitor fluoxetine is a common first-line treatment for childhood and adolescent mood disorders due to a favorable risk-benefit profile. Using a rodent model, we addressed specific long-term behavioral, molecular, bioenergetic, and cytoarchitectural consequences of postnatal fluoxetine (PNFlx) and juvenile fluoxetine (JFlx) treatment. METHODS: Rat pups received PNFlx (postna

Structure-function coupling alterations in adolescent depression correlate with neurotransmitter systems and cell-type-specific transcriptomics.

BACKGROUND: Adolescent major depressive disorder (AMDD) emerges during a period of significant neurobiological reorganization, yet its specific pathophysiological mechanisms remain poorly understood. This study investigated structural-functional brain coupling (SC-FC coupling) in AMDD and its relationship with neurotransmitter systems and molecular profiles. METHODS: We examined 107 adolescents with AMDD and 78 healthy controls. Participants underwent multimodal neuroimaging (DTI, resting-state fMRI), clinical assessment, and cognitive testing. We analyzed regional SC-FC coupling abnormalitie

5-HT1B receptor activation produces rapid antidepressant-like effects in rodents.

Ketamine is noted for its rapid onset antidepressant response and effectiveness in patients with treatment resistant depression. While most research has focused on glutamatergic mechanisms, recent studies show that antidepressant-like effects in rodents are dependent upon the serotonergic (5-HT) system and suggest a potential contribution of the 5-HT1B receptor. In this study we utilized CP-94253 to examine whether 5-HT1B receptor agonism produces rapid and sustained antidepressant-like effects, focusing on rodent models and treatment approaches commonly used to demonstrate the differentiated

Identifying altered developmental pathways in human globoid cell leukodystrophy iPSCs-derived NSCs using transcriptome profiling.

BACKGROUND: Globoid cell leukodystrophy (GLD) is a devastating neurodegenerative disease characterized by widespread demyelination caused by galactocerebrosidase defects. Changes in GLD pathogenesis occurring at the molecular level have been poorly studied in human-derived neural cells. Patient-derived induced pluripotent stem cells (iPSCs) are a novel disease model for studying disease mechanisms and allow the generation of patient-derived neuronal cells in a dish. RESULTS: In this study, we identified gene-expression changes in iPSCs and iPSC-derived neural stem cells (NSCs) from a patient

Olanzapine Administration Reduces Chemotherapy-Induced Nausea Behavior in Rats.

Nausea and vomiting are consistently identified among the most distressing side effects of chemotherapy. In recent years, Olanzapine (OLZ) treatment was added to anti-emetic guidelines as a treatment for chemotherapy-induced nausea and vomiting (CINV), despite little available data supporting a mechanism behind the positive benefits of the drug. Here, we examine whether OLZ reduces cisplatin chemotherapy-induced side effects on food intake and pica behavior in rats (i.e., kaolin intake, a proxy for nausea/emesis). Behavioral experiments tested whether systemic or hindbrain administration of OL

Chronic paroxetine treatment: effects on other non-serotonergic neurotransmitter systems.

Due to its efficacy and acceptability, paroxetine is situated in the top ten of drugs prescribed for the treatment of major depression and essentially all anxiety disorders. Adults under paroxetine treatment report relief after 4-6 weeks of administration; furthermore, this drug can be prescribed for periods lasting longer than one year. Therefore, paroxetine treatment has a pattern of ingestion that is mainly chronic rather than acute. There is a considerable number of reviews in the literature concerning the effects of paroxetine on the serotonergic system; however, the alterations caused by

Inhibition of serotonin transporters by cocaine and meprylcaine through 5-TH2C receptor stimulation facilitates their seizure activities.

The present study examined whether the inhibition of serotonin transporters (SERT) contributes to cocaine- and other local anesthetics-induced convulsions, and which subtypes of 5-HT receptor are involved in the convulsions. For this purpose, cocaine, meprylcaine and lidocaine, all of which have different effects on SERT, were used as convulsants and the effects of serotonin reuptake inhibitors (SSRIs), specific agonists and antagonists for 5-HT receptor subtypes were evaluated in mice. Administration of SSRI, zimelidine, citalopram and fluoxetine, 5-HT(2A,2C) receptor agonist, R(-)-DOI and th

Anatomical distribution of brainstem sites where PGE1 induces hyperthermia in macaque species.

The neuroanatomical distribution of sites in the diencephalon and mesencephalon within which a prostaglandin (PG) of the E series elicits hyperthermia was characterized in Macaca mulatta and Macaca nemestrina. In 420 experiments undertaken in 13 animals, 225 loci were examined for their reactivity to PGE1 microinjected in a dose of 30 or 100 ng given in a volume of 1.0-1.5 microL. The regions of the brainstem for injection extended rostrally from the thermosensitive cells of the anterior hypothalamic, preoptic area (AH/POA) to the caudal border of the mesencephalon. Colonic and skin temperatur

Cocaine: on-line analysis of an accumbens amine neural basis for psychomotor behavior.

Dose-response studies on subcutaneous cocaine were done to ascertain its effects in nucleus accumbens in dopaminergic and serotonergic neuronal circuitry in the behaving rat with in vivo voltammetry. Simultaneously, and at each dose of cocaine, unconditioned psychomotor stimulant behavior induced by cocaine was studied in terms of multiple concurrent measures of spontaneous behavior and by activity pattern analysis, a study of spatial patterns of locomotion. Time course studies showed that the neurochemical effects of cocaine (10, 20, and 40 mg/kg SC) significantly (p less than 0.0001) increas

Multiscale signatures of cortical hierarchical alterations and dialysis-associated preservation in end-stage renal disease.

End-stage renal disease (ESRD) is a chronic condition associated with prominent cognitive deficits, potentially arising from disruptions in the brain's hierarchical organization. However, the underlying neurobiological mechanisms, as well as the extent to which hemodialysis-an effective treatment for ESRD-is associated with cognitive outcomes, remain unclear. In this study, we collected resting-state functional MRI data from 51 healthy controls, 54 hemodialysis-dependent ESRD patients (HD-ESRD), and 25 non-hemodialysis ESRD patients (NHD-ESRD). Functional connectivity gradients were derived an

Neurotransmitter-Defined Degeneration Patterns in Sporadic and C9orf72-Associated Amyotrophic Lateral Sclerosis: Predilection to GABAergic, Serotonergic, Opioid, Glutamatergic, Endocannabinoid, and Microglial Systems-Implications for Therapy Development.

OBJECTIVE: Amyotrophic lateral sclerosis (ALS) has a markedly distinctive clinical and neuroradiological signature, with the preferential involvement of specific brain networks and the apparent sparing of others. The molecular underpinnings of the strikingly selective anatomical vulnerability have not been fully elucidated to date despite the potential therapeutic relevance of characterizing neurotransmitter receptor-defined susceptibility to degeneration. METHODS: A large neuroimaging study was undertaken with 258 participants to systematically evaluate topological associations between neuro

Spinal glial remodeling: a critical driver of inhibitory circuit deterioration and central sensitization in inflammatory pain.

Peripheral inflammation triggers maladaptive remodeling of spinal inhibitory circuits, which sustains persistent pain hypersensitivity long after initial tissue injury resolves. Although glial activation is well recognized as a key driver of inflammatory pain, the sequential glial-mediated mechanisms that disrupt dorsal horn inhibitory synaptic function remain incompletely summarized. This review examines how sequential microglial and astrocyte activation dismantles inhibitory synaptic control in the dorsal horn during inflammatory pain. Activated microglia release interleukin-1 alpha, tumor n

Novel C. elegans models of Lewy body disease reveal pathological protein interactions and widespread miRNA dysregulation.

Lewy body diseases (LBD) comprise a group of complex neurodegenerative conditions originating from accumulation of misfolded alpha-synuclein (α-syn) in the form of Lewy bodies. LBD pathologies are characterized by α-syn deposition in association with other proteins such as Amyloid β (Aβ), Tau, and TAR-DNA-binding protein. To investigate the complex interactions of these proteins, we constructed 2 novel transgenic overexpressing (OE) C. elegans strains (α-synA53T;Taupro-agg (OE) and α-synA53T;Aβ1-42;Taupro-agg (OE)) and compared them with previously established Parkinson's, Alzheimer's, and Lew
