Immunomodulatory inhibition of osteoclastogenesis by a marine microalgal ethanol fraction targeting T-cells, antigen presentation, and macrophage fate.

BACKGROUND: Targeting immune pathways to prevent bone loss represents a promising, yet underexplored therapeutic strategy. METHODS: An ethanol-soluble fraction derived from the freeze-dried biomass of the marine microalga Skeletonema costatum (SKLT) was tested for its ability to modulate immune responses and inhibit osteoclastogenesis. Its effects were evaluated in a zebrafish model of bone regeneration, a medaka model of RANKL-induced osteoporosis, and in vitro using murine RAW 264.7 macrophages. Transcriptomic profiling of regenerating fin blastemas at 24 hours post-amputation was performed

Indirect photochemical transformations of acyclovir and penciclovir in aquatic environments increase ecological risk.

Acyclovir and penciclovir, 2 antiviral drugs, are increasingly detected in aquatic environments. The present study explores the natural photochemical transformation mechanisms and fate of these drugs, examining direct and indirect photochemical transformation under simulated sunlight irradiation. The 2 antiviral drugs are photostable under certain conditions but significantly degrade in the presence of chromophoric dissolved organic matter (DOM). The degradation rate associated with the drugs' indirect photochemical transformation scaled with chromophoric DOM concentration. Quenchers and sensi

Comparative risk assessment of permethrin, chlorothalonil, and diuron to coastal aquatic species.

The precise application of risk assessment can lead to different conclusions about risk depending on how species are grouped in the assessment. We compared the use of different risk assessment methods for three different classes of pesticide, the herbicide diuron, the fungicide chlorothalonil, and the insecticide permethrin for marine and estuarine species. Permethrin was the most toxic pesticide to marine and estuarine crustaceans. Diuron was the most toxic pesticide to algae, and chlorothalonil was most toxic to early life stages of molluscs and other invertebrates. Toxicity data (96 h LC50/

Microalgal proteins as sustainable alternatives: CRISPR opportunities and challenges.

Against the backdrop of risinggrowing global demand for high-quality protein and increasing constraints on natural resources and the environment, microalgae are regarded as a promising alternative protein source, owing to their nutritional value, resource efficiency, and relatively low environmental footprint. However, their market adoption remains constrained by the limited availability of industrially robust strains combining high protein productivity with desirable processing traits. Recent advances in CRISPR-based genome engineering offer new opportunities for targeted microalgal strain im

Aquatic Toxicological Assessment of Solid Pyrolysis Product (SPP) from Synthetic Textile Feedstock Relative to Biochar, Carbon Black, and Activated Carbon.

Pyrolysis as a waste conversion process appears relatively feedstock-agnostic, yielding carbonaceous solid pyrolysis products (SPPs) plus condensable gases. Ground SPPs may substitute for or improve upon other carbon commodity particles, yet the environmental hazards of textile-derived SPPs have not been characterized. Here, we produced SPP particles from textile waste feedstock and from wood (biochar) using a pyrolysis process that could scale to waste management. We used electron microscopy, spectroscopy, gas adsorption, and standardized biochar analyses to characterize the chemical, morphol

Potential Bioactive Function of Microbial Metabolites as Inhibitors of Tyrosinase: A Systematic Review.

Tyrosinase (EC 1.14.18.1) is a binuclear copper enzyme responsible for the rate-limiting steps of melanogenesis, catalyzing the hydroxylation of L-tyrosine and oxidation of L-DOPA into o-quinones that polymerize melanin. Beyond its physiological role in pigmentation, tyrosinase is also implicated in food browning and oxidative stress-related disorders, making it a key target in cosmetic, food, and biomedical industries. This systematic review, conducted following PRISMA guidelines, aimed to identify and analyze microbial metabolites with tyrosinase inhibitory potential as sustainable alternati

Effects of elevated atmospheric carbon dioxide to Microcystis aeruginosa under different forms of phosphorus sources.

Human activities have led to an increase in atmospheric carbon dioxide (CO2) concentration, which can enhance the flux of CO2 from air to water, thus impacting algal growth. Phosphorus (P) is a key factor influencing the formation of cyanobacteria blooms. Nutrient utilization is closely related to carbon (C) metabolism, but the effects of elevated CO2 on microalgae under different P sources are rarely studied. In this study, we investigated the growth and physiological and biochemical responses of Microcystis aeruginosa (M. aeruginosa) under ambient (400 ppm) and elevated (550 ppm) CO2 levels

Bacterial ι-CAs.

Recent research has identified a novel class of carbonic anhydrases (CAs), designated ι-CA, predominantly found in marine diatoms, eukaryotic algae, cyanobacteria, bacteria, and archaea genomes. This class has garnered attention owing to its unique biochemical properties and evolutionary significance. Through bioinformatic analyses, LCIP63, a protein initially annotated with an unknown function, was identified as a potential ι-CA in the marine diatom Thalassiosira pseudonana. Subsequent biochemical characterization revealed that LCIP63 has CA activity and its preference for manganese ions over

Removal of nutrients from synthetic wastewater by different Brazilian chlorophyte strains in batch bioreactors under various light regimes.

With the increase in pollution and improper waste disposal, aquatic ecosystems are experiencing escalating degradation leading to various detrimental effects, including eutrophication and adverse impacts on the health of the population reliant on these water resources. Consequently, microalgae have demonstrated efficacy in nutrient removal, minimal environmental disruption, and superior cost-effectiveness in comparison to traditional treatment methods. Thus, this study aimed to investigate wastewater treatment in an aerobic batch system, using two strains of non-axenic mixotrophic chlorophytes

Cell morphology engineering enhances grazing resistance of Synechococcus elongatus PCC 7942 for non-sterile large-scale cultivation.

In the practical scale of cyanobacterial cultivation, the golden algae Poterioochromonas malhamensis is a well-known predator that causes devastating damage to the culture, referred to as pond crash. The establishment and maintenance of monoculture conditions are ideal for large-scale cultures. However, this is a difficult challenge because microbial contamination is unavoidable in practical-scale culture facilities. In the present study, we unexpectedly observed the pond crash phenomenon during the pilot-scale cultivation of Synechococcus elongatus PCC 7942 using a 100-L photobioreactor. This

Dual functionalized brush copolymers as versatile antifouling coatings.

Polymer coatings containing both fouling-resistant and fouling-release components have been reported to show synergistic antifouling properties. However, it remains unclear how the polymer composition influences the antifouling performance, particularly regarding foulants of different sizes and biological natures. Herein, we prepare dual functionalized brush copolymers containing fouling-resistant poly(ethylene glycol) (PEG) and fouling-release polydimethylsiloxane (PDMS) and examine their antifouling performances against different biofoulants. We utilize poly(pentafluorophenyl acrylate) (PPFP

The Microalgal Diatoxanthin Inflects the Cytokine Storm in SARS-CoV-2 Stimulated ACE2 Overexpressing Lung Cells.

Contact between SARS-CoV-2 and human lung cells involves the viral spike protein and the human angiotensin-converting enzyme 2 (ACE2) receptor on epithelial cells, the latter being strongly involved in the regulation of inflammation as well as blood pressure homeostasis. SARS-CoV-2 infection is characterized by a strong inflammatory response defined as a "cytokine storm". Among recent therapeutic approaches against SARS-CoV-2 targeting the dramatic inflammatory reaction, some natural products are promising. Diatoms are microalgae able to produce bioactive secondary metabolites, such as the xan

Variation in allelopathy of extracellular compounds produced by Cylindrotheca closterium against the harmful-algal-bloom dinoflagellate Prorocentrum donghaiense.

Allelopathy between algae is an ecological strategy that can facilitate or inhibit the occurrence of algal blooms. The role of allelopathic effects of marine microalgae Cylindrotheca closterium in other phytoplankton population dynamics are still limited. In the current study, the effects of cell-free filtrates of diatom Cylindrotheca closterium on two common dinoflagellates (Prorocentrum donghaiense and Prorocentrum cordatum), a chrysophyceae (Isochrysis galbana) and a diatom (Chaetoceros curvisetus) were investigated within controlled laboratory experiments. It was observed that the growth o

Cross-species transcriptomics and transgenic analyses implicate major LHCII genes in microalgal responses to ammonium stress.

INTRODUCTION: Nitrogen is an essential macronutrient for photosynthetic organisms, but excessive ammonium causes ammonium toxicity and severely restricts growth. Although ammonium stress has been widely documented, the conserved response modules shared by microalgae and higher plants remain poorly understood. METHODS: We performed a cross-species transcriptomic comparison using Auxenochlorella pyrenoidosa, Chlamydomonas reinhardtii and rice (Oryza sativa) to identify conserved pathways associated with ammonium stress. To test its functional relevance, we generated overexpression lines of the

Illuminating the path to oncolysis: rise of microbial phototherapy.

Phototherapy, which includes photothermal therapy (PTT) and photodynamic therapy (PDT), has attracted significant interest in the field of cancer treatment due to its spatiotemporal controllability and minimal invasiveness. Nonetheless, the utilization of phototherapy as a standalone treatment faces several formidable challenges. These encompass the non-specific distribution and rapid clearance of phototherapeutic agents, leading to inadequate antitumor efficacy and potential systemic toxicity. Additionally, the hypoxic and immunosuppressive nature of the tumor microenvironment (TME) substanti

Phycoremediation of methylene blue mediated by algal coculture: insights into morpho-physiological and metabolic responses.

Synthetic dyes are persistent environmental pollutants that pose significant ecological and toxicological risks. This study investigates the remediation potential of co-cultivated microalgae Chlorella vulgaris and Chlamydomonas reinhardtii for methylene blue (MB) removal. Optimal decolourization (100%) was achieved at 1-10 mg/L dye concentration and pH 7, with efficiency increasing over time. Morphological assessments revealed that C. vulgaris remained structurally stable with a slight change in the cell size, while C. reinhardtii exhibited reversible stress responses including palmella format

Natural Phycocyanin-Based Hydrogel for Endometritis Therapy via Endometrial Repair and Microbiota Homeostasis Restoration.

Endometritis is a prevalent inflammatory disorder of the uterine lining that is primarily triggered by microbial infection and poses a significant threat to the reproductive health of women of childbearing age worldwide. Current clinical management relies heavily on empirical antibiotic administration, which is increasingly challenged by antibiotic resistance and disruption of the vaginal microbiota. Herein, we developed a thermosensitive poly(lactic acid-co-glycolic acid)-poly(ethylene glycol)-poly(lactic acid-co-glycolic acid) (PLGA-PEG-PLGA) hydrogel as a targeted delivery system for natura
