Identifying potential nutrient acquisition mechanisms for long-term survival: adaptive evolution of Halomonas isolated from subseafloor crustal fluids.

In nature, microbes must often survive for long periods of time under conditions of nutrient and carbon limitation while also facing extremes in temperature, pressure, and competition with other microbes. One low-carbon, cold, and high pressure environment is the subseafloor crustal aquifer, where fluids circulate through old ocean crust. While microbial communities are known to be present in these fluids and contribute to biogeochemical cycling, the survival strategies of microbes in these communities is poorly constrained. In this study, multiple Halomonas strains were isolated from subseafl

Screening and comparison of in vitro-induced resistant and clinically resistant Stenotrophomonas maltophilia strains for eravacycline resistance-related genes.

OBJECTIVE: To identify genes related to eravacycline resistance in Stenotrophomonas maltophilia (Sm) and to provide a theoretical basis for the study of eravacycline resistance mechanisms in Sm and the development of new antibiotics. METHODS: The study employed an integrated omics approach: (1) In vitro antimicrobial susceptibility profiling via broth microdilution to determine baseline MICs for eravacycline and comparator drugs; (2) Induction of resistance in clinical Sm isolates (WJ_4, WJ_14, WJ_18) with low eravacycline MICs through serial passage in escalating drug concentrations; (3) Tra

Natural variation of the streptococcal group A carbohydrate biosynthesis genes impacts host-pathogen interaction.

Streptococcus pyogenes is a leading cause of infection-related mortality in humans globally. The characteristic cell wall-anchored group A carbohydrate (GAC) is expressed by all S. pyogenes strains and consists of a polyrhamnose backbone with alternating N-acetylglucosamine (GlcNAc) side chains, of which 25% are decorated with glycerol phosphate (GroP). The genes in the gacA-L cluster are critical for GAC biosynthesis, with gacH-L being responsible for the characteristic GlcNAc-GroP decoration, which confers the agglutination in rapid test diagnostic assays and contributes to S. pyogenes patho

A novel population of myeloid cells responding to coxsackievirus infection assists in the dissemination of virus within the neonatal CNS.

Enterovirus infection in newborn infants is a significant cause of aseptic meningitis and encephalitis. Using a neonatal mouse model, we previously determined that coxsackievirus B3 (CVB3) preferentially targets proliferating neural stem cells located in the subventricular zone within 24 h after infection. At later time points, immature neuroblasts, and eventually mature neurons, were infected as determined by expression of high levels of viral protein. Here, we show that blood-derived Mac3(+) mononuclear cells were rapidly recruited to the CNS within 12 h after intracranial infection with CVB

Humoral immune response to outer surface protein C of Borrelia burgdorferi in Lyme disease: role of the immunoglobulin M response in the serodiagnosis of early infection.

We determined the humoral immune response to outer surface protein C (OspC) of Borrelia burgdorferi in patients with early or late manifestations of Lyme disease and investigated the use of this antigen in the serodiagnosis of early infection. The ospC gene from the low-passage human isolate 297, a North American B. burgdorferi strain, was used to make a recombinant maltose-binding protein (MBP)-OspC fusion protein for serologic tests. This gene showed 84 to 85% nucleotide sequence identity and 76 to 79% amino acid identity with ospC of B. burgdorferi B31 and 2591. The antibody responses to MB

Cobamide-based interactions between soil bacteria can be predicted based on monoculture growth.

Interactions between microbes shape the structure and function of microbial communities. While studying interactions is key to understanding microbial communities as a whole, gaining a detailed mechanistic view is challenging due to the number of co-occurring interactions. Here, we focus on a single class of shared nutrients, the cobalamin (vitamin B12) family of enzyme cofactors known as cobamides, to study nutrient interactions in laboratory consortia of increasing complexity. Measuring cobamide-dependent growth of two soil Caulobacter isolates in monoculture revealed that each grows faster

Genomic analysis of Enterococcus faecium co-carrying optrA and poxtA from a swine farm: dissemination across the human-animal-environment interface.

BACKGROUND: The transferable resistance genes optrA and poxtA mediate cross-resistance to florfenicol and linezolid, posing serious challenges to both veterinary and human healthcare. Swine farms serve as critical ecological niches for the development and dissemination of multidrug-resistant (MDR) Enterococcus faecium (E. faecium) strains. However, the mechanisms by which E. faecium harboring optrA and poxtA disseminates and persists across the human-animal-environment interface remain unclear. RESULTS: In this study, 61 multidrug-resistant E. faecium isolates carrying optrA and/or poxtA were

Development and clinical application of a primary cancer organoid-PBMC co-culture system for personalized immunotherapy.

AIM: Many patients with non-small cell lung cancer (NSCLC) do not derive clinical benefit from immune checkpoint inhibitors (ICIs), and a reliable method for identifying potential responders is lacking. This study aims to establish an optimized patient-derived organoids (PDOs) culture system and develop a novel organoid-peripheral blood mononuclear cell (PBMC) co-culture platform to evaluate response to ICIs. METHODS: We optimized culture conditions for patient-derived NSCLC organoids by integrating conditional reprogramming techniques. ICIs response was assessed with an organoid-PBMC co-cult

Establishment and characterization of patient-derived mucinous gastric cancer organoids with faithful histopathological features and reproducible drug response patterns.

BACKGROUND: Mucinous gastric cancer (MGC) is a rare subtype of gastric cancer (GC) characterized by distinct histological features and clinical behavior. However, the lack of specific in vitro models for MGC has hindered progress in understanding its carcinogenic mechanisms and developing effective therapeutic strategies. METHODS: Patient-derived MGC organoids were established from paired primary tumor (MGC-T) and synchronous peritoneal metastatic nodule (MGC-ND) tissues obtained from a patient with MGC. To validate the generalizability of the findings, two additional independent MGC organoid

Genomic characterization and stability of mcr-carrying plasmids in Klebsiella pneumoniae from Taiwanese hospitals: Identification of a distinct mcr-9-harboring plasmid in the high-risk ST11-KL47 clone.

BACKGROUND: The spread of colistin-resistant Klebsiella pneumoniae carrying mobilized colistin resistance (mcr) genes via plasmids threatens last-line treatment options and complicates infection control in healthcare settings. Understanding the transferability and stability of these plasmids is crucial for assessing their potential for long-term persistence and hospital dissemination. This study characterizes the genetic contexts, transferability, and stability of mcr-carrying plasmids among colistin-resistant K. pneumoniae clinical isolates from tertiary hospitals in Taiwan, evaluating their

Clinical and in vitro models identify distinct adaptations enhancing Staphylococcus aureus pathogenesis in human macrophages.

Staphylococcus aureus is a facultative intracellular pathogen of human macrophages, which facilitates chronic infection. The genotypes, pathways, and mutations influencing that phenotype remain incompletely explored. Here, we used two distinct strategies to ascertain S. aureus gene mutations affecting pathogenesis in macrophages. First, we analyzed isolates collected serially from chronic cystic fibrosis (CF) respiratory infections. We found that S. aureus strains evolved greater macrophage invasion capacity during chronic human infection. Bacterial genome-wide association studies (GWAS) ident

Dengue and Zika virus NS4B proteins differ in topology and in determinants of ER membrane protein complex dependency.

Flaviviruses utilize the cellular endoplasmic reticulum (ER) for all aspects of their lifecycle. Genome replication and other viral activities take place in structures called replication organelles (ROs), which are invaginations induced in the ER membrane. Among the required elements for RO formation is the biogenesis of viral nonstructural proteins NS4A and NS4B. We have previously shown that NS4A and NS4B from Dengue virus (DENV) and Zika virus (ZIKV) depend on the cellular ER membrane protein complex (EMC) for biogenesis. Here, we find that this dependency extends to the NS4A and NS4B prote

Overcoming Culture Restriction for SARS-CoV-2 in Human Cells Facilitates the Screening of Compounds Inhibiting Viral Replication.

Efforts to mitigate the coronavirus disease 2019 (COVID-19) pandemic include the screening of existing antiviral molecules that could be repurposed to treat severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections. Although SARS-CoV-2 replicates and propagates efficiently in African green monkey kidney (Vero) cells, antivirals such as nucleos(t)ide analogs (NUCs) often show decreased activity in these cells due to inefficient metabolization. SARS-CoV-2 exhibits low viability in human cells in culture. Here, serial passages of a SARS-CoV-2 isolate (original-SARS2) in the human he

The evaluation of attenuated Neospora caninum by long-term passages on murine macrophage cell line in prevention of vertical transmission in mice.

To date, there is no effective vaccine to prevent abortion or vertical transmission associated with neosporosis in cattle. In the present study, the efficacy of a live experimental vaccine of Neospora caninum attenuated (NCa) by long-term serial passages on a murine macrophage cell line was evaluated in the prevention of vertical transmission and abortion in the mouse model. Forty non-pregnant mice were randomly divided into four equal groups including non-immunized/challenged (injected with PBS); positive control (inoculated with un-attenuated NC-1 tachyzoites); immunized/challenged (inoculat

A Molecular Determinant of West Nile Virus Secretion and Morphology as a Target for Viral Attenuation.

West Nile virus (WNV), a member of the Flavivirus genus and currently one of the most common arboviruses worldwide, is associated with severe neurological disease in humans. Its high potential to reemerge and rapidly disseminate makes it a bona fide global public health problem. The surface membrane glycoprotein (M) has been associated with Flavivirus-induced pathogenesis. Here, we identified a key amino acid residue at position 36 of the M protein whose mutation impacts WNV secretion and promotes viral attenuation. We also identified a compensatory site at position M-43 whose mutation stabili
