Urea supplementation is associated with holistic changes in suprathreshold non-volatile flavour perception through oral microbiome carbon metabolism.

Urea is a small inorganic compound, readily available in saliva, that can be metabolised by the oral microbiome. We hypothesised that dietary supplementation with urea would increase salivary levels, alter microbial metabolism and, potentially, modify suprathreshold non-volatile flavour perception. A semi-trained panel of 20 participants completed taste evaluations using visual analogue scales for nine tastants, both before and during a 5-day urea supplementation regimen. System-level analysis revealed a significant shift in suprathreshold non-volatile flavour perception during supplementation

Nanomotor-Assisted Intravesical Chemotherapy for Bladder Tumor Reduction and Suppression of Early Tumor Regrowth.

Nanoparticles are widely used in nanomedicine for controlled drug delivery and improved bioavailability. However, their effectiveness is often limited by passive diffusion, especially in confined, fluid-filled environments like the bladder, where rapid drug clearance and uneven distribution reduce therapeutic impact. These challenges contribute to high recurrence in bladder cancer despite intravesical chemotherapy. To address this limitation, we present urease-powered nanomotors (NM) based on mesoporous silica nanoparticles loaded with Mitomycin C (MMC), the standard chemotherapeutic for nonmu

L-asparaginase by filamentous fungi and yeasts from lichens and soils across maritime Antarctic islands.

This study aimed to evaluate L-asparaginase (L-ASNase) production by fungi isolated from soils across different Antarctic islands. Out of 138 isolates, 33 filamentous fungi and 5 yeasts exhibited enzymatic activity in solid culture media. Filamentous fungi were identified by sequencing the Internal Transcribed Spacer (ITS) region, while yeasts were identified using the D1/D2 domains of the 26 S rDNA gene. The blue filamentous fungus Antarctomyces F13.CR.LASP exhibited a maximum L-ASNase productivity of 0.76 U/L. Regarding the yeasts, the isolate Vishniacozyma victoriae G.L11 reached a producti

Detection and characterization of pathogenic Bacillus haynesii from Tribulus terrestris extract: ways to reduce its levels.

Plant parts such as roots, bark, leaves, flowers, and fruits that hold ethnopharmacological significance are naturally prone to microbial contamination, influenced by environmental factors like moisture and humidity. This study focuses on assessing the microbial load in the raw material of Tribulus terrestris (TT). The primary bacterium isolated from the pulverized raw material was identified as Bacillus haynesii through 16S rRNA sequencing. Biochemical assays revealed the organism's ability to utilize lysine and ornithine, produce urease, and generate hydrogen sulfide. The bacterium exhibited

Struvite ureterolithiasis associated with ureolytic bacterial pyelonephritis in a cat.

CASE SUMMARY: A 2-year-old female spayed domestic shorthair cat with a 1-week history of lethargy and dysorexia was presented to a veterinary teaching hospital. Physical examination identified severe left nephromegaly, hyperthermia and abdominal pain. Abdominal ultrasound revealed a left ureteral dilation due to obstruction by a ureterolith, associated with marked subcapsular effusion. Urinalysis showed alkaline urine with crystals of struvite. Culture of urine from the renal pelvis and the urinary bladder was positive for Staphylococcus pseudintermedius. An infected struvite ureterolith due t

A mucoadhesive-to-penetrating nanomotors-in-hydrogel system for urothelium-oriented intravesical drug delivery.

Intravesical therapy (IT) is widely used to tackle various urological diseases. However, its clinical efficacy is decreased by the impermeability of various barriers presented on the bladder luminal surface, including the urinary mucus layer and the densely packed tissue barrier. In this study, we report a mucoadhesive-to-penetrating nanomotors-in-hydrogel system for urothelium-oriented intravesical drug delivery. Upon intravesical instillation, its poloxamer 407 (PLX) hydrogel gelated and adhered to the urothelium to prolong its intravesical retention. The urea afterwards diffused into the hy

Gastric coinfection with thiopeptide-positive Cutibacterium acnes decreases FOXM1 and pro-inflammatory biomarker expression in a murine model of Helicobacter pylori-induced gastric cancer.

H. pylori infects half of the world population and is the leading cause of gastric cancer. We previously demonstrated that gastric cancer risk is associated with gastric microbiota. Specifically, gastric urease-positive Staphylococcus epidermidis and Streptococcus salivarius had contrasting effects on H. pylori-associated gastric pathology and immune responses in germ-free INS-GAS mice. As gastritis progresses to gastric cancer, the oncogenic transcription factor Foxm1 becomes increasingly expressed. In this study, we evaluated the gastric commensal C. acnes, certain strains of which produce t

GC/MS analysis and potential synergistic effect of mandarin and marjoram oils on Helicobacter pylori.

Helicobacter pylori can cause chronic gastritis, peptic ulcer, and gastric carcinoma. This study compares chemical composition and anti-H. pylori activity of mandarin leaves and marjoram herb essential oils, and their combined oil. GC/MS analysis of mandarin oil revealed six compounds (100% identified), mainly methyl-N-methyl anthranilate (89.93%), and 13 compounds (93.52% identified) of marjoram oil, mainly trans-sabinene hydrate (36.11%), terpinen-4-ol (17.97%), linalyl acetate (9.18%), and caryophyllene oxide (8.25%)). Marjoram oil (MIC = 11.40 µg/mL) demonstrated higher activity than manda

Hyperammonemic encephalopathy associated with post-radiotherapy vesicointestinal fistula following cervical cancer.

The majority of hyperammonemic encephalopathy is due to liver cirrhosis. However, urinary tract infection caused by urease-producing bacteria increases ammonia in urine and can lead to hyperammonemic encephalopathy, especially in cases with obstructive uropathy and vesicointestinal fistula. This is the first case report of hyperammonemic encephalopathy in a cervical cancer patient associated with postradiotherapy vesicointestinal fistula. A 52-year-old woman developed diarrhea due to vesicosigmoidal fistula 14 years after radical hysterectomy and radiotherapy to treat cervical cancer. She refu

Helicobacter pylori factors associated with disease development.

Although certain factors appear to predispose the host to infection by Helicobacter pylori, clearly the bacterium possesses a well-defined battery of virulence factors that allow the organism to: (1) colonize the gastric mucosa (urease, flagella, adhesins, acid-inhibitory protein, iron acquisition proteins, and heat shock proteins); (2) evade host defense (shedding of surface proteins, catalase, superoxide dismutase, and poorly reactive lipopolysaccharide); and (3) damage host tissue (vacuolating cytotoxin, protease, CagA-related factors, inducers of cytokines, and chemotaxins). Together these
