Molecular characterization of Mannheimia haemolytica isolates associated with pneumonic cases of sheep in selected areas of Central Ethiopia.

BACKGROUND: Mannheimia haemolytica has been recognized as the principal cause of pneumonic pasteurellosis in sheep and goats. It is one of the important diseases of small ruminants in Ethiopia. While annual vaccination using a monovalent vaccine (inactivated Pasteurella multocida biotype A) is common, respiratory diseases are still reported in various parts of Ethiopia. This suggests the need for further investigation into the species and strains responsible for the disease, which is vital information for development of a multivalent vaccine. The objective of the current study was to isolate M

A rapid minor groove binder PCR method for distinguishing the vaccine strain Brucella abortus 104M.

BACKGROUND: Brucellosis is a widespread zoonotic disease caused by Gram-negative Brucella bacteria. Immunisation with attenuated vaccine is an effective method of prevention, but it can interfere with diagnosis. Live, attenuated Brucella abortus strain 104M has been used for the prevention of human brucellosis in China since 1965. However, at present, no fast and reliable method exists that can distinguish this strain from field strains. Single nucleotide polymorphism (SNP)-based assays offer a new approach for such discrimination. SNP-based minor groove binder (MGB) and Cycleave assays have b

A novel multiplex PCR-electronic microarray assay for rapid and simultaneous detection of bovine respiratory and enteric pathogens.

Respiratory and enteric diseases continue to be two major causes of economic losses to the cattle industry worldwide. Despite their multifactorial etiology, the currently available diagnostic tests for bovine respiratory disease complex (BRDC) and bovine enteric disease (BED) are single-pathogen-tests. DNA microarray when combined with multiplex polymerase chain reaction (PCR) is a powerful tool in detection and differentiation of multiple pathogens in a single sample. This study reports development and initial validation of two independent highly sensitive and specific multiplex PCR-electroni

Identification of antibiotic resistance genes and their bacterial hosts in samples collected from wastewater treatment plants in Palestine.

UNLABELLED: Wastewater treatment plants are considered disseminators of antibiotic resistance bacteria (ARB) and antibiotic resistance genes (ARGs). In the West Bank, Palestine, there are four working wastewater treatment plants (WWTPs) located in Jenin, Tulkarem, Ramallah (Al-Terah), and Al-Bierh. Most of which have secondary treatment facilities that depend on the activated sludge process. In this study, Illumina high-throughput sequence analysis was used to determine the profile of ARB and ARGs in Al-Bierh and Al-Terah WWTPs. Raw wastewater sample (influent) and secondary treated water samp

High-throughput microfluidic real-time PCR as a promising tool in disease ecology.

Among recent advances in molecular biology for studying infectious diseases, the microfluidic high-throughput real-time polymerase chain reaction (Htrt PCR) has emerged as an efficient first-line tool for the detection of a wide range of infectious agents (IA) in a host system. This technology allows large batches of samples to be screened simultaneously for tens of targeted IA by real-time PCR. It represents a promising tool in disease ecology. As a proof of concept, we present here the development steps and initial application of a microfluidic Htrt PCR system for the detection of DNA from 2

The use of oral fluids and sock samples for monitoring key pathogens in pig populations for surveillance purposes.

Despite the prevalence of co-infections and the association of over 50 viral and 46 bacterial pathogens with pig diseases, little is known about their simultaneous occurrence, particularly in commercial pig farming environments where health programs are in place. To address this knowledge gap, this study aimed to evaluate the pathogen threshold of respiratory and enteric pathogens in pig herds using the Pork MultiPath™ (PMP1 and PMP2, respiratory and enteric respectively) technology, which detects multiple pathogens simultaneously in a single reaction with high sensitivity and specificity. Feeding Saccharomyces cerevisiae fermentation products lessens the severity of a viral-bacterial coinfection in preweaned calves.

We have previously reported that supplementation with Saccharomyces cerevisiae fermentation products (SCFP) ameliorates clinical signs and lung pathology following experimental bovine respiratory syncytial virus (BRSV) infection in preweaned dairy calves. The objectives of this study were to determine the effect of SCFP supplementation on the metabolic and endocrine responses, and disease outcome of a viral-bacterial coinfection in preweaned calves. Twenty-seven, 1- to 2-d-old Holstein-Angus cross calves were enrolled in the study; one SCFP calf was removed from the trial during the pre-challe

Systemic granulomatous and haemorrhagic syndrome in New Zealand dairy cattle.

CASE HISTORY: Cases were obtained through passive surveillance reporting by veterinary pathologists, via the Ministry for Primary Industries Exotic Pest and Disease Hotline. They included ill or dead cows that had evidence of frank haemorrhage, petechial haemorrhages on mucous membranes, wasting or dermatitis of unknown cause, and were reported between 2009-2014. Affected cows (n=16) were from nine seasonally calving dairy farms, aged ≥3 years, and were predominantly in their mid-to-late non-lactating period. A brassica crop was identified in 15/16 cases as part of the current or recent ration

[Infectious burdens of reproduction of female dogs].

The results of gynecological investigations in 142 bitches were evaluated and the complexity of infectious affections is discussed. High proportions of infectious cases were found in cases of limitation of fertility (67.5%), in vaginal discharge in the estrus (60.8%), in cases of mastitis/pseudopregnancy (61.5%) and in mortality of newborn puppies. St. aureus and E. coli were often isolated. There is a high etiological correlation in epidemiology of diseases of the reproductive tract in the single bitch and especially in kennel bitches.

Full genome sequence and some biological properties of reticuloendotheliosis virus strain APC-566 isolated from endangered Attwater's prairie chickens.

Reticuloendotheliosis virus (REV) causes runting, high mortality, immunosuppression, and chronic neoplasia associated with T and/or B cell lymphomas in a variety of domestic and wild birds, including Attwater's prairie chickens (APC) (Tympanuchus cupido attwateri). The complete proviral sequence of a recent REV isolate from APC (REV APC-566) was determined. This virus was isolated from an APC maintained in captivity in a reproduction program intended to avoid its extinction. REV APC-566 was determined to be oncogenic in Japanese quail (Coturnix coturnix japonica), chickens (Gallus gallus) and

[A personal view of the history of the genus Yersinia].

The first recorded experience Australia had of the genus Yersinia was the arrival in 1889 of a French expedition led by Pasteur's nephew, Dr. Adrien Loir. At that time Australia was in the grips of an epidemic of rabbits, and Loir's purpose was to eradicate the rabbits by means of fowl plague (Pasteurella multocida). Sadly, bureaucratic and political obstacles prevailed, and Loir was never granted permission to release his biological control agent. Alexander Yersin had been tempted to join Loir's expedition, but elected in the end to travel to Hong Kong, where he discovered the plague bacillus

Exploring cervicovaginal microbiome differences between single and multiple endometrial polyps: implications for non-invasive classification.

Single and multiple endometrial polyps (EP) are common gynecological conditions with differing recurrence rates, influencing clinical treatment decisions. This study aimed to characterize the reproductive tract microbiome in both subtypes to support the development of methods for the non-invasive categorization of EPs. Using metagenomic sequencing, we analyzed vaginal and cervical samples from 27 reproductive-aged patients with single EP and 22 with multiple EP. Compared with controls and multiple EP cases, single EP vaginal and cervical samples exhibited a lower percentage of community state

NqrM (DUF539) Protein Is Required for Maturation of Bacterial Na+-Translocating NADH:Quinone Oxidoreductase.

UNLABELLED: Na(+)-translocating NADH:quinone oxidoreductase (Na(+)-NQR) catalyzes electron transfer from NADH to ubiquinone in the bacterial respiratory chain, coupled with Na(+) translocation across the membrane. Na(+)-NQR maturation involves covalent attachment of flavin mononucleotide (FMN) residues, catalyzed by flavin transferase encoded by the nqr-associated apbE gene. Analysis of complete bacterial genomes has revealed another putative gene (duf539, here renamed nqrM) that usually follows the apbE gene and is present only in Na(+)-NQR-containing bacteria. Expression of the Vibrio harvey
