beta-glycosidase from the hyperthermophilic archaeon Sulfolobus solfataricus: structure and activity in the presence of alcohols.

beta-Glycosidase from the extreme thermophilic archaeon Sulfolobus solfataricus is a tetrameric protein with a molecular mass of 240 kDa, stable in the presence of detergents, and with a maximal activity at temperatures above 95 degrees C. Understanding the structure-activity relationships of the enzyme under different conditions is of fundamental importance for both theoretical and applicative purposes. In this paper we report the effect of methanol, ethanol, 1-propanol, and 1-butanol on the activity of S. solfataricus beta-glycosidase expressed in Escherichia coli. The alcohols stimulated th

Alcohols as reactive species modulators: Rethinking quenching in UV/chlorine process.

Alcohols are widely used as quenchers in UV/chlorine process currently to evaluate the roles of radicals, principally relying on the premise that they selectively react with target radicals (e.g., HO• and Cl•) without affecting other reactions. However, this study revealed several unrecognized alcohol-dependent phenomena in this system that clearly challenge this principle. Specifically, tert‑butanol inhibited chlorine decay (kobs = 0.14 min-1), whereas methanol, ethanol, and isobutanol accelerated it by 1.2-2.3 folds (kobs = 0.23-0.45 min-1). Mechanistic analysis indicated that alcohols quenc

Ozone Fine-Bubble Oxidation of Water-Soluble Alcohols via Electron Transfer across the Gas-Liquid Interface, Quantified by Kinetic Studies.

A novel fine-bubble reactor was developed to enable stable and quantitative kinetic studies of reactions at the gas-liquid interface, a process previously challenging to elucidate. Using this advanced analytical platform, we investigated ozone fine-bubble oxidation of a series of water-soluble alcohols, revealing fundamentally new insights into interfacial reactivity. Under single-substrate conditions, ozone fine bubbles exhibited comparable reactivity toward primary 1-butanol and secondary 2-butanol. However, under competitive conditions with a 1:1 mixture, ozone fine bubbles preferentially o

Bat2p is essential in Saccharomyces cerevisiae for fusel alcohol production on the non-fermentable carbon source ethanol.

Branched-chain amino acids (BCAAs) are key substrates in the formation of fusel alcohols, important flavour components in fermented foods. The first step in the catabolic BCAA degradation is a transaminase step, catalyzed by a branched-chain amino acid transaminase (BCAAT). Saccharomyces cerevisiae possesses a mitochondrial and a cytosolic BCAAT, Bat1p and Bat2p, respectively. In order to study the impact of the BCAATs on fusel alcohol production derived from the BCAA metabolism, S. cerevisiae BCAAT-deletion mutants were constructed. The BCAA l-leucine was exogenously supplied during cultivati

A Highly Sensitive Low-Temperature N-Butanol Gas Sensor Based on a Co-Doped MOF-ZnO Nanomaterial Under UV Excitation.

Volatile organic compounds (VOCs) are presently posing a rather considerable threat to both human health and environmental sustainability. Among these, n-butanol is commonly identified as bringing potential hazards to environmental integrity and individual health. This study presents the creation of a highly sensitive n-butanol gas sensor utilizing cobalt-doped zinc oxide (ZnO) derived from a metal-organic framework (MOF). A series of x-Co/MOF-ZnO (x = 1, 3, 5, 7 wt%) nanomaterials with varying Co ratios were generated using the homogeneous co-precipitation method and assessed for their gas-se

Modeling Postmortem Ethanol Production/Insights into the Origin of Higher Alcohols.

The forensic toxicologist is challenged to provide scientific evidence to distinguish the source of ethanol (antemortem ingestion or microbial production) determined in the postmortem blood and to properly interpret the relevant blood alcohol concentration (BAC) results, in regard to ethanol levels at death and subsequent behavioral impairment of the person at the time of death. Higher alcohols (1-propanol, 1-butanol, isobutanol, 2-methyl-1-butanol (isoamyl-alcohol), and 3-methyl-2-butanol (amyl-alcohol)) are among the volatile compounds that are often detected in postmortem specimens and have

Weaponizing volatiles to inhibit competitor biofilms from a distance.

The soil bacterium Bacillus subtilis forms beneficial biofilms that induce plant defences and prevent the growth of pathogens. It is naturally found in the rhizosphere, where microorganisms coexist in an extremely competitive environment, and thus have evolved a diverse arsenal of defence mechanisms. In this work, we found that volatile compounds produced by B. subtilis biofilms inhibited the development of competing biofilm colonies, by reducing extracellular matrix gene expression, both within and across species. This effect was dose-dependent, with the structural defects becoming more prono

Effect of the Pulsatilla decoction n-butanol extract on vulvovaginal candidiasis caused by Candida glabrata and on its virulence factors.

Vulvovaginal candidiasis (VVC) caused by Candida glabrata (C. glabrata) is more persistent and resistant to treatment than when caused by Candida albicans (C. albicans) and has been on the rise in recent years. The n-butanol extract of Pulsatilla Decoction (BEPD) has been shown to be effective in treating VVC caused by C. glabrata, but the underlying mechanism of action remains unclear. In this study, the experimenter conducted in vitro and in vivo experiments to explore the effects of BEPD on the virulence factors of C. glabrata, as well as its efficacy, with a focus on possible immunological

Immunochemical evidence for a role of cytochrome P-450 in liver microsomal ethanol oxidation.

Antibodies to cytochrome P-450 isozyme 3a, the ethanol-inducible isozyme in rabbit liver, were used to determine the role of this enzyme in the microsomal oxidation of alcohols and the p-hydroxylation of aniline. P-450 isozymes, 2, 3b, 3c, 4, and 6 did not crossreact with anti-3a IgG as judged by Ouchterlony double diffusion, and radioimmunoassays indicated a crossreactivity of less than 1%. Greater than 90% of the activity of purified form 3a toward aniline, ethanol, n-butanol, and n-pentanol was inhibited by the antibody in the reconstituted system. The catalytic activity of liver microsomes

Impact of butanol and hexanol on RCCI engine efficiency and emission characteristics using sapota oil methyl ester and response surface methodology.

This research explores the performance, emissions, and combustion behavior of a dual-fuel Reactivity Controlled Compression Ignition (RCCI) engine fueled with sapota oil methyl ester (SOME) blended with butanol, hexanol, and diesel. Sapota oil methyl ester, a still less-researched biodiesel, is evaluated for its synergistic effect when blended with oxygenated additives. Using Response Surface Methodology (RSM), some of the fuel blends like a base blend (B20D80) and others like B20BU10D70 and B20HEX10D70 were compared to enhance engine efficiency and emissions. The findings show that Brake Ther

Effects of constituents of Amomum xanthioides on gastritis in rats and on growth of gastric cancer cells.

In this study we investigated the effects of constituents of Amomum xanthioides (AX) on gastritis in rats and on the growth of human gastric cancer cells. The ethanol extract of Amomum xanthioides significantly inhibited HCl ethanol-induced gastric lesions and the growth of Helicobacter pylori (H. pylon). The ethanol extract of AX was further fractionated with hexane, chloroform, butanol and H20. Among these fractions, oral treatment with the butanol fraction at a dose of 350 mg/kg was the most effective at preventing HCl* ethanol-induced gastric lesions. In pylorus ligated rats, the butanol f

Volatile Profiling and Variety Discrimination of Leather Using GC-IMS Coupled with Chemometric Analysis.

Volatile fingerprint analysis using Gas Chromatography-Ion Mobility Spectrometry (GC-IMS) was applied to differentiate cowhide (22 samples), sheepskin (6 samples), and pigskin (6 samples). A total of 126 signal peaks were detected from the whole GC-IMS dataset, with 96 volatile compounds identified. Principal Component Analysis (PCA) revealed distinct clustering: cowhide exhibited unique volatile profiles, separating itself clearly from sheepskin and pigskin, which showed significant similarity. This was confirmed by Hierarchical clustering, K-means clustering (optimal k = 2), and Partial Leas

A novel method to produce solid lipid nanoparticles using n-butanol as an additional co-surfactant according to the o/w microemulsion quenching technique.

Solid Lipid Nanoparticles (SLN) and Nanostructured Lipid Carriers (NLC) are novel medicinal carriers for controlled drug release and drug targeting in different roots of administration such as parenteral, oral, ophthalmic and topical. These carriers have some benefits such as increased drug stability, high drug payload, the incorporation of lipophilic and hydrophilic drugs, and no biotoxicity. Therefore, due to the cost-efficient, proportionally increasable, and reproducible preparation of SLN/NLC and the avoidance of organic solvents used, the warm microemulsion quenching method was selected

Hydration shell parameters of aqueous alcohols: THz excess absorption and packing density.

Solvation in water requires minimizing the perturbations in its hydrogen bonded network. Hence solutes distort water molecular motions in a surrounding domain, forming a molecule-specific hydration shell. The properties of those hydration shells impact the structure and function of the solubilized molecules, both at the single molecule and at higher order levels. The size of the hydration shell and the picoseconds time-scale water dynamics retardation are revealed by terahertz (THz) absorption coefficient measurements. Room-temperature absorption coefficient at f = 0.28 [THz] is measured as a

Purification and characterization of a primary-secondary alcohol dehydrogenase from two strains of Clostridium beijerinckii.

Two primary alcohols (1-butanol and ethanol) are major fermentation products of several clostridial species. In addition to these two alcohols, the secondary alcohol 2-propanol is produced to a concentration of about 100 mM by some strains of Clostridium beijerinckii. An alcohol dehydrogenase (ADH) has been purified to homogeneity from two strains (NRRL B593 and NESTE 255) of 2-propanol-producing C. beijerinckii. When exposed to air, the purified ADH was stable, whereas the partially purified ADH was inactivated. The ADHs from the two strains had similar structural and kinetic properties. Each

A recombinant inbred strain analysis of sleep-time responses to several sedative-hypnotics.

The results of previous studies suggested that the sleep-time differential between long-sleep (LS) and short-sleep (SS) mice decreases for a series of sedative-hypnotic drugs as lipid solubility increases. Using the LS x SS recombinant inbred (RI) strains, we have tested whether this relationship arises because of common genetic influences on the sleep-time responses or was simply a fortuitous difference between LS and SS mice. Mice were sleep-time tested after intraperitoneal injections of a variety of sedative-hypnotic drugs that vary in lipid solubility including alcohols, barbiturates, chl
