Trials of the anticoagulant rodenticides bromadiolone and difenacoum against the house mouse (Mus musculus L.).

Laboratory and field trials were conducted to determine the efficacy of the anticoagulant rodenticide bromadiolone against the house mouse (Mus musculus). In laboratory feeding tests, family groups of warfarin-resistant mice maintained in pens and conditioned to feeding on plain foods were offered pinhead oatmeal bait containing bromadiolone at 0.005%. Overall mortality in replicated 21-day poison treatments was 55/58 or 94.8%. Six field trials were carried out, using the same poison bait, against mice infesting farm buildings. Treatment success, estimated from the results of census baitings c

Laboratory evaluation of bromadiolone as a rodenticide for use against warfarin-resistant and non-resistant rats and mice.

Laboratory feeding tests were carried out to determine the efficacy of the anticoagulant rodenticide bromadiolone against Rattus norvegicus, R. rattus and Mus musculus. Using 0.005% bromadiolone, complete kills of R. norvegicus and R. rattus not resistant to warfarin were obtained after exposure to the poison for 1 and 5 days respectively. Warfarin-resistant R. norvegicus were all killed in 4 days, and resistant M. musculus in 12 days. In general, the results resembled those obtained with difenacoum. Acceptance of bromadiolone was very good.

Field trials of brodifacoum (WBA 8119) against the house mouse (Mus musculus L.).

The anticoagulant rodenticide brodifacoum was tested against house mice (Mus musculus L.) infesting farm buildings. In six trials, treatment success was assessed from the results of census baitings conducted before and after treatment. With 0.005% brodifacoum in canary seed/corn oil bait, the control achieved ranged between 92.7% and 100%, mean 98.8%. Two mouse populations were eradicated in 3 to 4 weeks but a few individuals survived each of the other four treatments which lasted 6 weeks. The effectiveness of brodifacoum against mice is compared with that of 0.1% calciferol and 0.025% warfari

Laboratory evaluation of WBA 8119 as a rodenticide for use against warfarin-resistant and non-resistant rats and mice.

Feeding tests were carried out in the laboratory to evaluate WBA 8119 as a potential new rodenticide against wild common rats (Rattus norvegicus), ship rats (R. rattus) and house mice (Mus musculus). The results obtained are compared with data previously obtained for difenacoum, another member of the same series of 4-hydroxycoumarin anticoagulants. With warfarin-resistnat and non-resistant common rats, complete kills were obtained using a concentration of 0-0005% for 2 days, or 0-001% for 1 day: a 1-day test at 0-0005% killed 6 out of 10 and 17 out of 20 of the two types respectively. At 0-000

A cellular system for quantitation of vitamin K cycle activity: structure-activity effects on vitamin K antagonism by warfarin metabolites.

Warfarin and other 4-hydroxycoumarins inhibit vitamin K epoxide reductase (VKOR) by depleting reduced vitamin K that is required for posttranslational modification of vitamin K-dependent clotting factors. In vitro prediction of the in vivo potency of vitamin K antagonists is complicated by the complex multicomponent nature of the vitamin K cycle. Here we describe a sensitive assay that enables quantitative analysis of γ-glutamyl carboxylation and its antagonism in live cells. We engineered a human embryonic kidney (HEK) 293-derived cell line (HEK 293-C3) to express a chimeric protein (F9CH) co

Photo-mediated gene activation using caged RNA/DNA in zebrafish embryos.

We report a new and simple technique for photo-mediated temporal and spatial control of gene activation in zebrafish embryos as an alternative to the gene 'knockdown' approach using antisense, morpholino-modified oligonucleotides (morpholinos). The synthetic compound 6-bromo-4-diazomethyl-7-hydroxycoumarin (Bhc-diazo) forms a covalent bond with the phosphate moiety of the sugar-phosphate backbone of RNA, a process known as caging. The 6-bromo-7-hydroxycoumarin-4-ylmethyl (Bhc) group binds to approximately 30 sites on the phosphate moieties per 1 kb of RNA sequence. Bhc-caged mRNA undergoes pho

Recent findings in understanding the biological function of vitamin K.

Vitamin K is required for the posttranslational modification of liver precursors of prothrombin, blood coagulation Factors VII, IX, and X and additional proteins of undetermined functions in plasma and other tissues. This modification involves the formation of gamma-carboxyglutamic acid, an acidic amino acid needed for the interaction of these proteins with calcium ions. The vitamin is a cofactor of a unique microsomal carboxylase which requires the reduced form of vitamin K, CO2, and molecular oxygen. The vitamin is apparently oxidized to its 2,3-epoxide during the carboxylation reaction. Liv

Enantioselective synthesis of coumarins catalyzed by an isosteviol-derived tertiary amine-squaramide catalyst.

A newly tertiary amine-squaramide organocatalyst has been successfully developed and applied into the asymmetric Michael addition of 4-hydroxycoumarin to β,γ-unsaturated α-ketoesters. The catalyst system performed well with a low catalyst loading of 1 mol% under mild reaction conditions. A series of coumarin derivatives were obtained in good to high yields (up to 97%) with high enantioselectivities (up to 96% ee).

A new tool to assess ceramide bioactivity: 6-bromo-7-hydroxycoumarinyl-caged ceramide.

The bioactivity of natural, long-chain ceramides has until now been studied after its delivery to cells in organic solvent mixtures containing dodecane. We have synthesized ceramides conjugated to a (6-bromo-7-hydroxycoumarin-4-yl)methyl group. The photocaged ceramide is efficiently released with 350 nm light in aqueous solution at neutral pH, thus providing a promising new tool to study ceramide's properties.

[Rodenticidal action of warfarin/4th communication: Binding to serum albumin of mice and rats (author's transl)].

The binding properties of 3-(1'-phenyl-2'-acetylethyl)-4-hydroxycoumarine (warfarin-Na) to serum albumin of mice and rats were determined by using the equilibrium dialysis. The binding percentage (beta), the binding capacity (K), and the association constant (K1) were calculated. The results are characteristic for a strong binding of warfarin to serum albumin of rats but only a small one to serum albumin of mice, caused probably by the different albumin structures of these species.

Spontaneous hemoperitoneum from brodifacoum overdose.

Brodifacoum is a 4-hydroxycoumarin derivative that is commonly used as a rodenticide. Human exposures have produced severe coagulopathies resulting in hematuria, gastrointestinal bleeding, intracranial hemorrhage, and death. This is the first report of spontaneous hemoperitoneum secondary to brodifacoum ingestion. The patient was successfully managed with fresh frozen plasma, packed red blood cells, and vitamin K1. No surgical intervention was performed. The patient required ongoing daily vitamin K1 therapy for longer than 6 months.

Anticoagulant residues associated with an attempted rodent eradication from a subtropical coral atoll.

The use of rodenticides is a primary method for eradicating rodents from islands for conservation purposes. Rodenticide residue monitoring is often incorporated into rodent eradication project planning to understand the potential effects on nontarget species, but robust long-term sampling is often challenging due to logistical and financial constraints. We documented more than two years of rodenticide residues at fine-scale intervals with over 570 samples associated with a rodent eradication attempt. Brodifacoum-25D Conservation was applied in an attempt to eradicate house mice (Mus musculus)

Anticoagulant rodenticide poisoning confirmed by vitamin K1 depletion in raptors from Patagonia.

The use of second-generation anticoagulant rodenticides (SGARs) raises a dilemma regarding the regulation of persistent bioaccumulative toxic substances (PBTs), given that these products have applications in high demand by society, such as the control of zoonotic pests, for which there are no clear alternatives. In this work, we describe for the first time SGAR contamination in birds of prey from Argentina and South America. We found concerning levels in the liver and prevalence of SGAR (brodifacoum, bromadiolone, difenacoum, flocoumafen and difethialone) bioaccumulation (18.2%), particularly

Sanitary measures considerably improve the management of resistant Norway rats on livestock farms.

BACKGROUND: Norway rats (Rattus norvegicus) need to be controlled to prevent transmission of pathogens and damages to stored products and material, leading to considerable economic risks and losses. Given increasing resistance in Norway rats, the most persistent, bio-accumulative and toxic anticoagulant rodenticides are widely used for management, which presents hazards to the environment especially for non-target species. We investigated how sanitary measures improved management of Norway rats on 12 paired livestock farms in a region of Germany with a high population of resistant rats for red

Bromadiolone poisoning leading to subarachnoid haemorrhage: A case report and review of the literature.

WHAT IS KNOWN AND OBJECTIVE: Many cases of rodenticide poisoning have been reported. Bromadiolone, often called a super-warfarin, is a second-generation dicoumarin rodenticide with long half-life. The main clinical manifestations of bromadiolone poisoning are excessive or inappropriate bleeding of skin mucosa, digestive tract and urinary tract. However, the phenomenon of central nervous system (CNS) toxicity is an uncommon medical emergency. We present a case of SAH and intracerebral haematoma mediated by bromadiolone intoxication, revealing that bromadiolone poisoning might cause intracerebra

Detection of Brodifacoum and other Rodenticides in Drug Mixtures using Thermal Desorption Direct Analysis in Real Time Mass Spectrometry (TD-DART-MS).

In recent months, there has been increased reporting of seized drug and toxicology cases containing rodenticides, the active ingredient in rat poisons. Seeing as rodenticides are not scheduled substances, they are not commonly screened for in seized drug analysis. This work investigates the use of TD-DART-MS for the simultaneous detection of rodenticides and drugs. Six rodenticides were evaluated, an optimal method was established, and limits of detection in the tens of nanograms were calculated. Additional studies highlight that detection at less than 1% by weight in mixtures with AB-FUBINACA

Surreptitious superwarfarin ingestion with brodifacoum.

Physicians must have a high index of suspicion when patients have unexplained prolongation of the prothrombin time and bleeding in the absence of detectable warfarin. Several common rodenticides contain modified versions of warfarin that are not detectable in standard warfarin assays. We present a case of surreptitious brodifacoum ingestion in a patient who had years of unexplained bleeding and negative warfarin levels.
