RATIONALE: Animal studies indicate that central acetaldehyde, dependent on catalase metabolism of ethanol (EtOH), modulates ethanol reinforcement. Brain catalase activity and acetaldehyde (ACD) production are significantly higher in rat pups compare d with adults. Interestingly, infant rats show high EtOH affinity for alcohol consumption and are particularly sensitive to the drug's reinforcing effects.
OBJECTIVES: We tested whether central ACD is necessary and sufficient to induce appetitive conditioning in newborn rats through the artificial nipple technique.
METHODS: Vehicle, EtOH (100 mg%

Acetaldehyde is one of the most prevalent carcinogens in cigarette smoke. It is also a major metabolite of ethanol and is found widely in the human diet and environment. Acetaldehyde DNA adducts are critical for its carcinogenic properties. The role of acetaldehyde DNA adducts in human cancer related to tobacco and alcohol exposure could be investigated with a suitable biomarker. Therefore, in this study, we have developed a method for analysis of the major DNA adduct of acetaldehyde, N2-ethylidene-dGuo (1), in human leukocyte DNA. Leukocyte DNA was subjected to enzyme hydrolysis in the presen

The contents of acetaldehyde (AA) in biological fluids obtained from the dead with the confirmed lethality causes, i.e. ischemic heart disease (IHD), alcoholic cardiomyopathy (ACMP) and mechanical traumas (MT), were examined on an actual forensic-medical material (AFMM). 14 death cases of males, aged 18 to 45, were studied. The method of gas-liquid chromatography (GLC), or rather its variation of vaporphase analysis, was used to state the presence and to assess the concentration of acetaldehyde. The results revealed differences between concentrations of acetaldehyde in the examined groups depe

In view of the increased anticoagulant effect of acetaldehyde-treated heparin, other glycosaminoglycans (GAGs) such as chondroitin sulfates A and C, dermatan sulfate (chondroitin sulfate B), heparan sulfate, and hyaluronic acid were tested for anticoagulant activity before and after exposure to acetaldehyde. Clotting times of human plasma Ci-Trol coagulation control, level I (Baxter Healthcare Corp.), were tested in the presence of 1.8, 3.0, 3.6, or 4.5 microg heparin (0.32, 0.54, 0.64, 0.81 units heparin). Additionally, 9, 27, or 90 microg of chondroitin sulfates A, B, or C was utilized in li

BACKGROUND: It has recently been reported that acetaldehyde induces bronchoconstriction indirectly via histamine release. However, no study has been performed to assess whether acetaldehyde worsens bronchial responsiveness in asthmatic subjects so this hypothesis was tested.
METHODS: Methacholine provocation was performed on three occasions: (1) after pretreatment with oral placebo and inhaled saline (P-S day), (2) after placebo and inhaled acetaldehyde (P-A day), and (3) after a potent histamine H1 receptor antagonist terfenadine and acetaldehyde (T-A day) in a double blind, randomised, cros

Considerable clinical and experimental evidence points to the importance of immune responses in the development of alcoholic liver disease. In the present study it was investigated whether circulating antibodies from patients with alcoholic liver disease recognize acetaldehyde-liver protein adducts. Cytosolic and microsomal fractions from livers of Wistar rats or from normal human liver were incubated with acetaldehyde (0.5-2.5 mmol/L) and/or cyanoborohydride (100 mmol/L) then analysed by immunoblotting. Cytosolic fractions that had been incubated with acetaldehyde and cyanoborohydride express

Rats which received acetaldehyde by intraperitoneal injection on a single occasion sustained neural degeneration in the cerebral cortex detectable with both light and electron microscopy. The degeneration was more intense and included the hippocampus when acetaldehyde exposure was given on 5 consecutive days in the form of ethanol vapor inhalation and disulfiram injections. Pretreatment of animals with a combination 2.5 g lysine/kg plus 50 mg pyridoxyl phosphate/kg did not alter the degree of degeneration response to either a single injection of acetaldehyde or 5 days acetaldehyde treatment. In a series of experimental studies involving ethanol vapour administration to rats, sustained blood alcohol levels in the range 89-115 mM for nine hours of each day over a two week period did not lead to neural degeneration detactable with either light or electron microscopy. A single nine hours exposure to ethanol and disulfiram giving rise to 20-41 mM alcohol and 52-76 microM acetaldehyde in the blood did lead to degeneration; and that with repeated exposures of this later type, the damage was found to be accumulative. The lowest levels of blood acetaldehyde which led to neural degeneration

The cardiovascular effects of ethanol ingestion after pretreatment with a new antialcohol drug, nitrefazole (2-methyl-4-nitro-1-(4-nitro-phenyl)imidazole, Altimol) were studied. Left ventricular function was examined by echocardiography and systolic time intervals in six healthy Finnish male volunteers who ingested 0.15-0.25 g of ethanol/kg, 24 hr after an 800 or 1600-mg peroral dose of nitrefazole. After ethanol ingestion, accumulation of acetaldehyde in blood (25-150 microM) was accompanied by a 1.5-2-fold increase in plasma noradrenaline, a 3-10-fold increase in plasma adrenaline, and a 0.5

Male Long-Evans rats consumed the alcohol and aldehyde dehydrogenase inhibitors pyrazole, cyanamide or disulfiram, for 6 days. No endogenous blood acetaldehyde could be detected in controls and pyrazole treated rats, endogenous blood concentrations up to 2-5 nmoles/ml were, however, measured in the cyanamide and disulfiram-treated animals. Other rats received daily ethanol gastric intubations in addition to the consumption of the inhibitors. Little or no acetaldehyde was detected in the controls and pyrazole treated animals during acute ethanol intoxication or on the subsequent days. High bloo

BACKGROUND: Fibrin(ogen), the primary structural component of blood clots, plays a critical role in thrombosis and hemostasis. Posttranslational modifications to fibrinogen that occur upon exposure to reactive endogenous metabolites or xenobiotics during inflammation and disease alter polymer formation, network structure, or susceptibility to lysis. Acetaldehyde is a reactive byproduct of hepatic ethanol metabolism and ubiquitous environmental pollutant that forms stable adducts with macromolecules. Previous studies report that acetaldehyde impairs the function of multiple coagulation factors,

Despite offering low-carbon and reliable energy, the utilization of nuclear energy is declining globally due to high upfront capital costs and longer returns on investments. Nuclear cogeneration of valuable chemicals from waste biomass-derived feedstocks could have beneficial impacts while harnessing the underutilized resource of ionizing energy. Here, we demonstrate selective methanol or acetaldehyde production from ethylene glycol, a feedstock derived from glycerol, a byproduct of biodiesel, using irradiations from a nuclear fission reactor. The influence of radiation quality, dose rate, and

BACKGROUND: Much of alcohol's toxicity is due to its product, acetaldehyde. The role of acetaldehyde derived from endogenous sources was assessed in alcoholic patients administered disulfiram, an inhibitor of aldehyde dehydrogenase.
METHODS: The first part of the study included 23 subjects without biochemical or clinical evidence of chronic liver disease who were abstinent for 2 weeks; 11 patients were started on disulfiram (250 mg/day), whereas the other 12 were not given disulfiram and served as controls. The second part of the study included 13 alcoholic patients with clinical or pathologi

The exaggerated blood acetaldehyde response that has been reported after ethanol administration to pregnant rats was found to be the beginning of a much larger alteration occurring during lactation. Indeed, at the end of pregnancy, we confirmed a 4-fold increase in the acetaldehyde values above nonpregnant values after an intragastric dose of 3 g/kg ethanol. During gestational days 1 to 17, the levels did not differ. After delivery, the exaggerated acetaldehyde response to ethanol was increased, producing acetaldehyde concentrations 15-fold greater than in nonlactating controls. This response

Formaldehyde and acetaldehyde, as volatile organic pollutants, can seriously harm human health if exposed for a long time. Developing efficient and favorable selective detection methods is of great practical significance. This paper presents a biomass probe, AN, which uses aminated chitosan as the carrier, 4-bromo-1,8-naphthalic anhydride as the fluorophore, and hydrazine groups as the reactive sites. This probe can selectively detect formaldehyde (FA) and acetaldehyde (AA) in pure aqueous solution. AN is a turn-on probe for formaldehyde and acetaldehyde. The reaction products of AN with forma

The dehydrogenation of ethanol to acetaldehyde and H2 represents an attractive synthetic route, owing to the versatile applications of acetaldehyde and the high atom economy. In this work, efficient ethanol dehydrogenation was achieved at the anode of a high-temperature polymer electrolyte membrane electrolyzer by integrating thermocatalytic ethanol dehydrogenation (EDHR) with electrochemical hydrogen oxidation (HOR) over Ru/C and PdAu/C composite catalytic layer. The Ru/C serves as a bifunctional catalyst for the EDHR and HOR, and the PdAu/C could rapidly electrooxidize retained acetaldehyde

The development of an enantioselective enamine-catalysed addition of masked acetaldehyde to nitroalkenes via a rational approach helped to move away from the use of chloroform. The presented research allows the use of water as a reaction medium, therefore improving the industrial relevance of a protocol to access very important pharmaceutical intermediates. Critical to the success is the use of chemometrics-assisted 'Design of Experiments' (DoE) optimisation during the development of the presented new synthetic approach, which allows to investigate the chemical space in a rational way.

Excessive alcohol consumption is one of the most significant causes of morbidity and mortality worldwide. Alcohol is oxidized to toxic and carcinogenic acetaldehyde by alcohol dehydrogenase (ADH) and further oxidized to a non-toxic acetate by aldehyde dehydrogenase (ALDH). There are two major ALDH isoforms, cytosolic and mitochondrial, encoded by ALDH1 and ALDH2 genes, respectively. The ALDH2 polymorphism is associated with flushing response to alcohol use. Emerging evidence shows that Lactobacillus and Bifidobacterium species encode alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (

The main aim of the present work is to study the accumulation of acetaldehyde and Strecker aldehydes (isobutyraldehyde, 2-methylbutanal, isovaleraldehyde, methional, phenylacetaldehyde) during the oxidation of red wines, and to relate the patterns of accumulation to the wine chemical composition. For that, eight different wines, extensively chemically characterized, were subjected at 25°C to three different controlled O2 exposure conditions: low (10 mg L-1) and medium or high (the stoichiometrically required amount to oxidize all wine total SO2 plus 18 or 32 mg L-1, respectively). Levels of vo

Alcohol drinking increases the risk for a number of cancers. Currently, the highest risk (Group 1) concerns oral cavity, pharynx, larynx, esophagus, liver, colorectum, and female breast, as assessed by the International Agency for Research on Cancer (IARC). Alcohol and other beverage constituents, their metabolic effects, and alcohol-related unhealthy lifestyles have been suggested as etiological factors. The aim of the present survey is to evaluate the carcinogenic role of acetaldehyde in alcohol-related cancers, with special emphasis on the genetic-epidemiological evidence. Acetaldehyde, as
