Bovine In Vitro Fertilization Embryo Development Predicted by Capacitation-Induced Zinc Ion Flux and Acrosome Remodeling.

Sire-dependent variation in bovine in vitro fertilization (IVF) success is poorly explained by conventional semen parameters once samples meet standard quality thresholds. Sperm capacitation is required for acquisition of fertilization competence, yet its contribution to sire-dependent variation in bovine IVF outcomes remains unclear. Fifteen bulls (Bos taurus) were evaluated across five IVF replicates using 30 abattoir-derived oocytes per bull per replicate. Sperm were analyzed before (0 h) and after 3 h of in vitro capacitation using image-based flow cytometry to quantify zinc ion localizati

Characterization of expression patterns and dynamic relocation of Notch proteins during acrosome reaction of bull spermatozoa.

Notch is a conserved cell-signaling pathway involved in spermatogenesis regulation. This study firstly evaluated the presence, localization patterns, acquisition origin and relation to acrosome reaction of Notch proteins in bull sperm. Western Blot analysis detected all Notch proteins in ejaculated bull sperm, and immunostaining described their specific sperm localization. Recovery of sperm from different segments showed that Notch proteins have testicular origin (NOTCH1, NOTCH2, DLL4), are sequentially acquired during sperm maturation along epididymal transit (NOTCH3, DLL3, JAGGED1-2), or pos

Starvation induces an increase in intracellular calcium and potentiates the progesterone-induced mouse sperm acrosome reaction.

We have recently reported two different methodologies that improve sperm functionality. The first method involved transient exposure to the Ca2+ ionophore A23187 , and the second required sperm incubation in the absence of energy nutrients (starvation). Both methods were associated with an initial loss of motility followed by a rescue step involving ionophore removal or addition of energy metabolites, respectively. In this work, we show that starvation is accompanied by an increase in intracellular Ca2+ ([Ca2+ ]i ). Additionally, the starved cells acquire a significantly enhanced capacity to u

Sperm from hyh mice carrying a point mutation in alphaSNAP have a defect in acrosome reaction.

Hydrocephalus with hop gait (hyh) is a recessive inheritable disease that arose spontaneously in a mouse strain. A missense mutation in the Napa gene that results in the substitution of a methionine for isoleucine at position 105 (M105I) of alphaSNAP has been detected in these animals. alphaSNAP is a ubiquitous protein that plays a key role in membrane fusion and exocytosis. In this study, we found that male hyh mice with a mild phenotype produced morphologically normal and motile sperm, but had a strongly reduced fertility. When stimulated with progesterone or A23187 (a calcium ionophore), sp

Characterization of zona pellucida glycoprotein 3 (ZP3) and ZP2 binding sites on acrosome-intact mouse sperm.

There is considerable evidence that mouse fertilization requires the binding of sperm to two of the three glycoproteins that form the zona pellucida (ZP), ZP3 and ZP2. Despite the biologic importance of this binding, no one has demonstrated that sperm express separate, saturable, and specific binding sites for ZP3 and for ZP2. Such a demonstration is a prerequisite for defining the distribution, numbers, affinities, and regulation of function of ZP3 and ZP2 binding sites on sperm. The experiments reported herein used fluorochrome-labeled ZP3 and ZP2 and quantitative image analysis to character

Enhancement of motility and acrosome reaction in human spermatozoa: differential activation by type-specific phosphodiesterase inhibitors.

Inhibition of sperm phosphodiesterase (PDE) has been shown to increase cAMP concentrations and stimulate motility and the acrosome reaction. While several PDE genes exist in mammals, little is known about the physiological role of PDE forms expressed in human spermatozoa. Using type-selective inhibitors, we identified two of the PDE forms expressed in human spermatozoa and studied their involvement in sperm function. Selective inhibitors of calcium-calmodulin-regulated PDE1 (8-methoxy-isobutyl-methylxanthine) and cAMP-specific PDE4 (RS-25344, Rolipram) were used to study PDE forms in human spe

Restoration of Motility in Acrosome-Reacted Boar Spermatozoa Induced by the Ionophore 4-Br-A23187.

The acrosome reaction (AR) is a calcium-dependent exocytotic event required for successful fertilization. Calcium ionophores are commonly used to induce AR in vitro; however, this treatment is associated with a significant loss of sperm motility. The aim of this study was to evaluate the recovery of motility in boar spermatozoa after ionophore-induced AR and to assess the role of extracellular calcium. Boar spermatozoa were incubated with 10 μM 4-Br-A23187 to induce AR. Then, spermatozoa were washed and incubated for 30 min in a recovery medium containing either 1 mM calcium or 5 mM EGTA. The

Rheb ubiquitination drives LC3 lipidation via non-canonical autophagy to promote porcine sperm acrosome reaction.

The coordinated interaction between ubiquitination and autophagy is essential for maintaining cellular homeostasis; however, their precise roles in regulating the acrosome reaction (AR) remain unclear. To determine whether Rheb ubiquitination modulates LC3 lipidation through the mTORC1/ULK1-Beclin 1 signaling axis during sperm capacitation and thereby influences the AR, we conducted a series of molecular analyses in porcine sperm samples. These included quantitative ubiquitin proteomics, co-immunoprecipitation (CO-IP), Western blot (WB), and immunofluorescence. Our results revealed that capaci

Cordycepin improves hyperactivation and acrosome reaction through adenosine receptors during human sperm capacitation in vitro.

BACKGROUND: Sperm capacitation is a prerequisite for natural or in vitro fertilization. After capacitation, sperm become hyperactivated and undergo an acrosome reaction, which helps them penetrate the oocyte. Cordycepin, a bioactive compound first isolated from Cordyceps militaris, is an adenosine analog with numerous physiological activities. However, its effects on sperm capacitation remain unclear. This study aims to elucidate the effects and mechanisms of cordycepin on human sperm capacitation. METHODS: During in vitro capacitation culture, healthy human sperm were treated with cordycepin

Spastin is an essential regulator of male meiosis, acrosome formation, manchette structure and nuclear integrity.

The development and function of male gametes is dependent on a dynamic microtubule network, yet how this is regulated remains poorly understood. We have recently shown that microtubule severing, via the action of the meiotic AAA ATPase protein clade, plays a crucial role in this process. Here, we sought to elucidate the roles of spastin, an as-yet-unexplored member of this clade in spermatogenesis. Using a SpastKO/KO mouse model, we reveal that spastin loss resulted in a complete loss of functional germ cells. Spastin plays a crucial role in the assembly and function of the male meiotic spindl

Changes in fertilization medium viscosity using hyaluronic acid impact bull sperm motility and acrosome status.

The female reproductive tract, in particular the composition of the uterine and oviduct fluids, is responsible, at least in part, for triggering sperm cell modifications, essential for the acquisition of fertilization ability. Hyaluronic acid (HA) is a glycosaminoglycan present in these fluids, and its role in the fertilization process and sperm functionality is still barely understood. This work was designed to (a) determine the rheological characteristics of the fertilization medium by the addition of HA and (b) determine the HA influence on sperm motility and functional status. To that end,

Lysine acetylation participates in boar spermatozoa motility and acrosome status regulation under different glucose conditions.

Efficient artificial insemination (AI) with liquid preserved boar semen is essential for the swine industry. Glucose is the most widely utilized energy source in refrigerated boar semen extenders. However, the relationship between glucose concentration and spermatozoa quality is not fully understood. In the present study, boar spermatozoa were used as a model to investigate the impact of different glucose concentrations on spermatozoa motility, mitochondrial activity, and acrosome integrity at physiological temperature (37 °C) and during refrigeration (17 °C). The proportion of progressively m

Manchette-acrosome disorders during spermiogenesis and low efficiency of seminiferous tubules in hypercholesterolemic rabbit model.

Hypercholesterolemia is a marker for several adult chronic diseases. Recently we demonstrated that sub/infertility is also associated to Hypercholesterolemia in rabbits. Seminal alterations included: abnormal sperm morphology, decreased sperm number and declined percentage of motile sperm, among others. In this work, our objective was to evaluate the effects of hypercholesterolemia on testicular efficiency and spermiogenesis, as the latter are directly related to sperm number and morphology respectively. Tubular efficiency was determined by comparing total number of spermatogenic cells with ea

Automatic classification of the acrosome status of boar spermatozoa using digital image processing and LVQ.

We consider images of boar spermatozoa obtained with an optical phase-contrast microscope. Our goal is to automatically classify single sperm cells as acrosome-intact (class 1) or acrosome-damaged (class 2). Such classification is important for the estimation of the fertilization potential of a sperm sample for artificial insemination. We segment the sperm heads and compute a feature vector for each head. As a feature vector we use the gradient magnitude along the contour of the sperm head. We apply learning vector quantization (LVQ) to the feature vectors obtained for 320 heads that were labe

Protein kinase C plays an important role in the human zona pellucida-induced acrosome reaction.

To investigate the involvement of protein kinases in signal transduction in the human zona pellucida (ZP)-induced acrosome reaction (AR), the effects of protein kinase (PK) activators, dibutyryl cAMP (PKA) and cGMP (PKG), phorbol 12-myristate 13-acetate (PMA, PKC), and the PKC inhibitor, staurosporine were studied. Sperm samples were obtained from normozoospermic men with normal sperm-ZP binding. Oocytes were obtained from other patients with failure of fertilization in vitro. Motile spermatozoa selected by a swim-up technique were pre-incubated with 2.5-20 microM PMA, 1 mM dibutyryl cAMP or c

Fertilization failure after subzonal sperm insertion associated with defective fusional capacity of acrosome-reacted spermatozoa.

A case of an isolated impairment of the ability of acrosome-reacted spermatozoa to fuse with the oocyte is reported. This impairment was associated with fertilization failure after subzonal sperm insertion even though elevated numbers of spermatozoa were inserted per oocyte. This observation shows the need for further studies into the relationship between the acrosome reaction and the sperm-oocyte fusion to better understand the advantages and the limitations of subzonal sperm insertion in different types of sperm pathology.

A Novel Homozygous ZPBP Frameshift Variant Is Associated With Severe Acrosomal Structural Abnormalities in Asthenoteratozoospermia.

BACKGROUND: Experimental disruption of Zpbp homologs in mice causes defective acrosomal biogenesis and male infertility. Human evidence remains limited to a small number of association-based or case-level reports linking ZPBP alterations to abnormal sperm-head or acrosomal phenotypes, without integrated characterization across protein abundance, ultrastructure, sperm chromatin integrity, and proteome-wide changes. OBJECTIVES: Our study aimed to characterize the genetic, structural, and molecular sperm phenotype associated with a novel homozygous frameshift variant in ZPBP. MATERIALS AND METH

Role of the Ca2+-ATPase Pump (SERCA) in Capacitation and the Acrosome Reaction of Cryopreserved Bull Spermatozoa.

Cryopreservation of sperm is a resource used for artificial insemination. In the case of bovines, it ensures the reproduction of animals through cells from males with a highlighted genetic background. The fertilisation capacity of sperm cells is achieved after capacitation, a process that includes subcellular modifications that could be altered by cryopreservation. Intracellular calcium handling plays a crucial role in the development of capacitation, culminating in the acrosomal reaction. SERCA protein is responsible for calcium reuptake into the acrosome, which is one of the main calcium res

Mouse spermatogenesis-associated protein 1 (SPATA1), an IFT20 binding partner, is an acrosomal protein.

BACKGROUND: Intraflagellar transport is a motor-driven trafficking system that is required for the formation of cilia. Intraflagellar transport protein 20 (IFT20) is a master regulator for the control of spermatogenesis and male fertility in mice. However, the mechanism of how IFT20 regulates spermatogenesis is unknown. RESULTS: Spermatogenesis associated 1 (SPATA1) was identified to be a major potential binding partner of IFT20 by a yeast two-hybrid screening. The interaction between SPATA1 and IFT20 was examined by direct yeast two-hybrid, co-localization, and co-immunoprecipitation assays.
