Acrylamide formation in low-fat potato snacks and its correlation with colour development.

A recipe and technological process for the production of low-fat potato snacks (7% fat) was developed at the Institute of Food Technology of Plant Origin (Agricultural University of Poznan) using a combination of extrusion and a roasting process. Due to the character of the product, the level of acrylamide was analysed. At the same time, the influence of temperature and time of heating on acrylamide levels were monitored, as well as the correlation between colour development and acrylamide content. The level of acrylamide in low-fat potato snacks was 489 +/- 26 microg kg(-1), which is comparab

How starch molecular fine structure influences starch graft copolymerization with acrylamide.

Fifteen starches with distinct molecular structures were used to synthesize starch-g-polyacrylamide copolymers via horseradish-peroxidase-catalyzed graft copolymerization. Starch fine structures, including amylose content and the chain-length distributions (CLDs) of both amylose and amylopectin, were quantitatively analyzed using size-exclusion chromatography, and the data interpreted with biosynthesis-based mathematical models. Successful grafting was confirmed by Fourier-transform infrared spectroscopy, while the degree of branching (DB), degree of substitution and grafting ratio (GR) were q

AC Electrokinetics-Enhanced Capacitive Aptasensor for Point-of-Care Testing of Acrylamide in Coffee.

Acrylamide (AA) is a common contaminant in foods processed at high temperatures and has attracted significant attention due to its potential neurotoxicity and carcinogenicity. Therefore, the development of a highly sensitive, highly selective sensing technology suitable for on-site detection is of great importance for ensuring food safety. In this study, an aptamer (Apt)-based capacitive AA sensor was developed based on the alternating current electrokinetics (ACEK) effect. The sensor utilizes an aptamer as the biomimetic recognition element, which can specifically recognize AA, thereby enabli

Beyond acrylamide: a structure-guided investigation of covalent warheads in the development of CDK7 inhibitors.

To achieve covalent CDK7 inhibitors with superior antiproliferative activity and enhanced selectivity, we designed and synthesized a series of covalent CDK7 inhibitors bearing diverse warheads based on the THZ1 scaffold. Experimental validation confirmed that 2-chloroacrylamide-based covalent inhibitors possess potent CDK7 kinase inhibitory activity. HRMS results revealed that the interaction between these compounds and the cysteine residue of CDK7 extends beyond a simple Michael addition, potentially involving nucleophilic substitution at the covalent warhead. Among them, compound B19 exhibit

ATRA-mediated RAR-α activation attenuates acrylamide-induced testicular toxicity.

Acrylamide (ACR) is an environmental reproductive toxicant with unclear testicular toxicity mechanisms. Retinoids are a group of vitamin A-related compounds that function by activating retinoid receptors. We aimed in this study to further explore All-trans retinoic acid's (ATRA) protective response against an acrylamide-induced testicular insult model and the underlying possible mechanisms. Fifty male rats were allocated into control, DMSO, ACR (40 mg/kg bwt, i.p. daily for 14 days), ATRA (7.5 mg/kg bwt, i.p. daily), and ACR + ATRA groups. Body and testes weights, sperm parameters, testosteron

Visually evoked potentials as an early indicator of acrylamide-induced visual dysfunction in rats.

Humans rely heavily on visual function to gather information, and loss of vision has a significant impact on quality of life. However, it is difficult to quantitatively evaluate the effects of chemical compounds on visual function in toxicity studies using animals (such as OECD guideline studies). Consequently, evaluation including ophthalmological and histopathological examinations has played a major role to date. Visually evoked potential (VEP) is a type of brain wave that reflects the activity of the entire visual pathway, including the retina, optic nerve, and visual cortex. We investigate

Purification of vegetable oils from acrylamide pollutants using metal-organic frameworks.

Acrylamide is a chemical compound that can form in certain foods during high-temperature cooking processes like frying, roasting, and baking. The presence of acrylamide in used cooking oil has environmental impact; therefore, the purification of used oils may reduce the environmental risks. Nowadays, biochar, a porous carbonaceous material derived from biomass pyrolysis can added to crystalline porous materials called metal-organic framework (MOF), these combination lead to fantastic properties for adsorption of contaminants. Here, ZIF-8@Biocharas adsorbents were synthesized for purification o

Computational Insights into Acrylamide Fragment Inhibition of SARS-CoV-2 Main Protease.

The pathogen of COVID-19, SARS-CoV-2, has caused a severe global health crisis. So far, while COVID-19 has been suppressed, the continuous evolution of SARS-CoV-2 variants has reduced the effectiveness of vaccines such as mRNA-1273 and drugs such as Remdesivir. To uphold the effectiveness of vaccines and drugs prior to potential coronavirus outbreaks, it is necessary to explore the underlying mechanisms between biomolecules and nanodrugs. The experimental study reported that acrylamide fragments covalently attached to Cys145, the main protease enzyme (Mpro) of SARS-CoV-2, and occupied the subs

Mitochondrial dysfunction promotes the necroptosis of Purkinje cells in the cerebellum of acrylamide-exposed rats.

Acrylamide (ACR) is a common neurotoxicant that can induce central-peripheral neuropathy in human beings. ACR from occupational setting and foods poses a potential threat to people's health. Purkinje cells are the only efferent source of cerebellum, and their output is responsible for coordinating motor activity. Recent studies have reported that Purkinje cell injury is one of the earliest neurotoxicity at any dose rate of ACR. However, the mechanism underlying ACR-mediated damage to Purkinje cells remains unclear. This research aimed to investigate whether necroptosis is involved in ACR-induc

Vitexin inhibits acrylamide-induced neuroinflammation and improves behavioral changes in zebrafish larvae.

Neuroinflammation is crucial for the pathophysiological hallmarks of many neurodegenerative disorders. Hyperactivated microglia has long been implicated as a detrimental player in regulating unresolvable inflammatory insults which cause damage to neurons. In the context of acrylamide (ACR) neurotoxicity, microglia activation is documented to correlate with ACR-adduct formation in the presynaptic neurons. Thus, inhibition of inflammatory mediators through vital candidate is greatly warranted to retard the disease progression. In the present study, we investigated, whether vitexin, a C-glycosyla
