Actigraphy-based detection of isolated REM sleep behavior disorder: multicenter validation across devices and populations.

Scalable home-based detection of isolated REM sleep behavior disorder (iRBD) is essential for early care, prevention trials, and identifying candidates for neuroprotective interventions against synucleinopathies. We previously showed that high-resolution wrist actigraphy (Axivity AX6) could identify iRBD based on abnormal sleep (AUC of 0.916) and rest-activity-rhythms (RAR, AUC of 0.856) using machine learning. Here, we aimed to assess generalizability across: (1) other actigraphs using lower resolutions; (2) different populations. We tested the analysis pipeline directly in cohorts from the I

Effects of methamphetamine on actigraphy-based sleep parameters in female rhesus monkeys: Orexin receptor mechanisms.

BACKGROUND: The orexin system has been implicated as a mechanism underlying insomnia and methamphetamine-induced sleep disruptions, with a potential role for OX2 receptors in the sleep-modulating effects of orexin. The aim of the present study was to investigate the extent to which orexin receptors mediate the effects of acute methamphetamine administration on actigraphy-based sleep in female rhesus monkeys. METHODS: Actigraphy-based sleep measures were obtained in female rhesus monkeys (n=5) under baseline and acute test conditions. First, morning (10h) i.m. injections of methamphetamine (0.

Actigraphy evaluation before and after intravenous ferric carboxymaltose in 3 children with restless sleep disorder.

UNLABELLED: Restless sleep disorder has been described in the literature as a disorder affecting children and presenting with large muscle movements during sleep with an index of 5 events/h or more, leading to daytime impairment including sleepiness or behavioral problems. Children with restless sleep disorder have been found to have low ferritin levels. Studies with iron supplementation both oral or intravenous have been shown effective in clinically improving both nighttime and daytime symptoms. However objective data of the improvement is lacking. Repeating polysomnography is expensive, and

Blue Light and Temperature Actigraphy Measures Predicting Metabolic Health Are Linked to Melatonin Receptor Polymorphism.

This study explores the relationship between the light features of the Arctic spring equinox and circadian rhythms, sleep and metabolic health. Residents (N = 62) provided week-long actigraphy measures, including light exposure, which were related to body mass index (BMI), leptin and cortisol. Lower wrist temperature (wT) and higher evening blue light exposure (BLE), expressed as a novel index, the nocturnal excess index (NEIbl), were the most sensitive actigraphy measures associated with BMI. A higher BMI was linked to nocturnal BLE within distinct time windows. These associations were presen

Actigraphy-measured rest-activity circadian rhythm disruption in patients with advanced cancer: a scoping review.

PURPOSE: Patients with later-stage cancer have been reported to demonstrate more disrupted rest-activity circadian rhythms (RACR) than those with earlier-stage cancer, but consolidated evidence of this is lacking. The aim of this review was to examine and map the existing evidence on actigraphy-measured RACR in terms of their (1) pattern, (2) prevalence, (3) related factors, and (4) outcomes in advanced cancer patients. METHODS: A comprehensive scoping review was conducted using Arksey and O'Malley's framework. A literature search was performed using nine databases: MEDLINE, Embase, PsycINFO,

ActiTect: a generalizable machine learning pipeline for REM sleep behavior disorder screening through standardized actigraphy.

Isolated rapid eye movement sleep behavior disorder (iRBD) is a major prodromal marker of α-synucleinopathies, often preceding the clinical onset of Parkinson's disease, dementia with Lewy bodies, or multiple system atrophy. While wrist-worn actimeters hold significant potential for detecting RBD in large-scale screening efforts by capturing abnormal nocturnal movements, they require a reliable and efficient analysis pipeline. This study presents ActiTect, a fully automated, open-source machine learning tool to identify RBD from actigraphy recordings. To ensure generalizability across heteroge

Monitoring sleep duration, timing, and continuity among US youth and adults in NHANES using actigraphy.

OBJECTIVES: Sleep health differs across the life course and by sex, poverty-income ratio (PIR), and race and ethnicity. Monitoring sleep is important for informing interventions to improve sleep heath. Our objective was to explore and describe sleep characteristics among a nationally-representative US sample by age, sex, PIR, and race and ethnicity. METHODS: Data came from n=13,656 US residents aged 3-80 years in the National Health and Nutrition Examination Survey (2011-2014). Participants wore an ActiGraph GT3X+ on their wrist for ≤7 days to assess sleep. We used GGIR (v. 3.0.0) to derive s

Latent class analysis of actigraphy within the depression early warning (DEW) longitudinal clinical youth cohort.

BACKGROUND: Wearable-generated data yield objective information on physical activity and sleep variables, which, are in turn, related to the phenomenology of depression. There is a dearth of wearable-generated data regarding physical activity and sleep variables among youth with clinical depression. METHODS: Longitudinal (up to 24 months) quarterly collections of wearable-generated variables among adolescents diagnosed with current/past major depression. Latent class analysis was employed to classify participants on the basis of wearable-generated: Activity, Sleep Duration, and Sleep efficien

Does sleep aggravate tension-type headache?: An investigation using computerized ecological momentary assessment and actigraphy.

BACKGROUND: Both insufficient sleep and oversleeping have been reported as precipitating and aggravating factors of tension-type headache (TTH). However, previous studies relied on recalled self-reports, and the relationship has not been confirmed prospectively and objectively in a daily life situation. Recently, ecological momentary assessment (EMA) using electronic diaries, i.e., computerized EMA, is used to record subjective symptoms with the advantages of avoiding recall bias and faked compliance in daily settings. In addition, actigraphy has become an established method to assess sleep ou

Circadian Rhythm in Restless Legs Syndrome and Its Association With Symptom Severity: An Actigraphy-Based Study.

Restless legs syndrome (RLS) is characterized by an urge to move that occurs in the evening or night time, suggesting circadian involvement. However, objective evidence regarding the circadian phase in RLS remains limited, particularly in real-world settings. We conducted an exploratory pilot study to investigate circadian rhythm timing in patients with RLS using actigraphy-based dim light melatonin onset (DLMO) estimation and to explore its potential association with symptom severity. Patients with idiopathic RLS (n = 21) and age- and sex-matched chronic insomnia controls (n = 21) underwent 1

Actigraphy-based sleep outcomes in substance use disorders: A protocol for a systematic review and meta-analysis.

INTRODUCTION: Individuals with substance use disorders (SUDs) are at increased risk for sleep disturbances, creating a bidirectional relationship that may heighten relapse risk. While polysomnography is the gold standard for measuring sleep, many studies have used actigraphy, a noninvasive, wrist-worn device that estimates rest-activity patterns and sleep-wake characteristics. Despite its utility, the use of actigraphy in populations with SUDs remains limited, and findings vary across substances and methodologies. This protocol outlines a systematic review and meta-analysis aiming to synthesiz

Effects of methadone, buprenorphine, and naltrexone on actigraphy-based sleep-like parameters in male rhesus monkeys.

Opioid use disorder (OUD) has been associated with the emergence of sleep disturbances. Although effective treatments for OUD exist, evidence suggests that these treatments also may be associated with sleep impairment. The extent to which these effects are an effect of OUD treatment or a result of chronic opioid use remains unknown. We investigated the acute effects of methadone, buprenorphine, and naltrexone on actigraphy-based sleep-like parameters in non-opioid-dependent male rhesus monkeys (Macaca mulatta, n = 5). Subjects were fitted with actigraphy monitors attached to primate collars to

Efficacy of early warning signals and spectral periodicity for predicting transitions in bipolar patients: An actigraphy study.

Early-warning signals (EWS) have been successfully employed to predict transitions in research fields such as biology, ecology, and psychiatry. The predictive properties of EWS might aid in foreseeing transitions in mood episodes (i.e. recurrent episodes of mania and depression) in bipolar disorder (BD) patients. We analyzed actigraphy data assessed during normal daily life to investigate the feasibility of using EWS to predict mood transitions in bipolar patients. Actigraphy data of 15 patients diagnosed with BD Type I collected continuously for 180 days were used. Our final sample included e

Actigraphy in mechanically ventilated pediatric ICU patients: comparison to PSG and evaluation of behavioral circadian rhythmicity.

Sleep disruption is common in pediatric intensive care unit (PICU) patients, but measuring sleep in this population is challenging. We aimed to evaluate the utility of actigraphy for estimating circadian rhythmicity in mechanically ventilated PICU patients and its accuracy for measuring sleep by comparing it to polysomnogram (PSG). We conducted a single-center prospective observational study of children 6 months - 17 years of age receiving mechanical ventilation and standard, protocolized sedation for acute respiratory failure, excluding children with acute or historical neurologic injury. Sleep and psychotic symptoms: An actigraphy and diary study with young adults with low and elevated psychosis proneness.

Experimental research has shown that poor sleep triggers psychotic experiences, even in healthy participants. This warrants an in-depth investigation of this mechanism in a naturalistic environment, an exploration of which particular aspects of poor sleep trigger psychotic symptoms, and a test for reverse effects of symptoms on sleep. For this purpose, we conducted a 14-day ambulatory assessment study with 82 young adults (age: M = 21.24 years, SD = 1.54; 64.6% female), half of which were characterized by elevated psychosis proneness.
