Advanced parental age in maternal uniparental disomy (UPD): implications for the mechanism of formation.

Uniparental disomy (UPD) describes the inheritance of a pair of chromosomes from only one parent. Meiotic nondisjunction followed by trisomy rescue is considered to be the major mechanism of formation. A literature search for cases with whole chromosome UPD other than UPD 15 was performed. Information on parental age was available in 111 cases with maternal UPD and in 34 cases with paternal UPD. In 52 out of 74 cases with maternal heterodisomy, information on the time of nondisjunction was also available. Around two-thirds of these cases were due to a maternal meiosis I error. Compared with th

The trend in the relationship of advanced maternal age to preterm birth and low birthweight.

PURPOSE: As women in developed countries tend to delay childbearing, it becomes more important to understand the relationship of advanced maternal age to birth outcomes. We aimed to estimate the trend in the relationship of advanced maternal age to preterm birth and low birthweight. MATERIALS AND METHODS: We analysed 4,264,417 ethnically homogeneous, singleton firstborns, born in hospitals to married couples in South Korea in 1997-2014. We regressed an indicator for preterm birth or low birthweight on advanced maternal age, the baby's sex, advanced paternal age, and a set of socioeconomic sta

[A case of maternally inherited diabetes with deafness (MIDD) occurring at an advanced age].

We report an elderly patient with maternally inherited diabetes with deafness (MIDD). A 69-year-old woman was found to be diabetic for the first time when she visited her local medical doctor for the symptoms of a common cold. Her casual plasma glucose level was 311 mg/dl and HbA1c was 8.3%. She had been aware of muscle atrophy of the lower extremities and hearing disturbance since age 66. As for her family history, her mother, older sister and younger brother were diabetic with hearing difficulty and all of them had died suddenly in their middle age. Her 45-year-old daughter was also diabetic

The paradox of declining fertility but increasing twinning rates with advancing maternal age.

BACKGROUND: Advancing female age is associated with declining fertility potential due to decreasing numbers and quality of oocytes but also with a distinct increase in dizygotic twinning rates, a phenomenon that has never been explained. METHOD: An analysis of follicle development was made in 959 spontaneous ovulatory cycles of 507 women. RESULTS: Multiple ovarian follicular development (>1 follicle >14 mm) and, by implication, multiple rather than single ovulations occurred in 105 women whose mean age (36.1 versus 34.6 years) and mean basal FSH concentrations (10.3 versus 7.7 IU/l) were sig

Assisted reproduction with advancing paternal and maternal age: an Ethics Committee opinion.

Ethical considerations for developing policies regarding providing assisted reproductive care to individuals of advanced reproductive age are complex; considerations include both reproductive autonomy and the well-being of the potential offspring. This document replaces the previously published document titled "Oocyte or embryo donation to women of advanced reproductive age: an Ethics Committee opinion" (Fertil Steril 2016;106:e3-7).

Sex-specific effects of advanced maternal age on cardiovascular function in aged adult rat offspring.

Pregnancy at an advanced maternal age has an increased risk of complications for both the mothers and their offspring. We have previously shown that advanced maternal age in a rat model leads to poor fetal outcomes, maternal vascular dysfunction, and hypertension, concordant with findings in humans. Moreover, offspring from aged dams had sex-specific cardiovascular dysfunction in young adulthood. However, the detrimental impact of aging on the cardiovascular system of the offspring in this model is unknown. We hypothesized that offspring born to aged dams (9.5-10 mo old) would have impaired ca

[Motherhood at the Age of 35 and Over: The Risk of Advanced Maternal Age as Perceived by Women - a Literature Study].

Background Delaying motherhood is an ongoing trend in industrialized countries worldwide. The higher the mother´s age, the greater the risks for fertility, pregnancy, childbearing, and for the newborn. The psychosocial situation of women who are conscious of pregnancy risks is thus negatively affected. This paper aims to answer the question as to how women experience the risks of age. Methodology An systematic search of the international literature was carried out between December 2015 and March 2016, using the electronic databases PubMed, CINAHL, PsycINFO, Sowiport, as well as a manual search

Placental trophoblast aging in advanced maternal age is related to increased oxidative damage and decreased YAP.

INTRODUCTION: The advanced maternal age (AMA) pregnancies escalate rapidly, which are frequently linked to higher risks of adverse outcomes. Advanced maternal age (AMA) placenta exhibited premature aging, presumably resulting in trophoblast dysfunction, inadequate placentation. However, the precise reasons and mechanisms of trophoblast aging in AMA placenta remain unclear, posing a significant limitation to provide effective guidance for prenatal healthcare in clinical settings. Notably, the organism shows heightened vulnerability to oxidative damage as it ages. YAP (Yes-associated protein) wa

Maternal and neonatal outcomes in full-term pregnancies of very advanced maternal age: a Japanese single-center retrospective cohort study.

BACKGROUND: Advanced maternal age (AMA), defined as pregnancy in people aged ≥ 35 years, is associated with an increased risk of adverse perinatal outcomes. Although the outcomes of AMA pregnancies have been reported, detailed information on the specific outcomes in pregnant individuals aged ≥ 45 years, particularly those reaching full term, remains limited. Therefore, this study aimed to investigate the perinatal outcomes of full-term pregnant individuals aged ≥ 45 years. METHODS: We conducted a retrospective cohort study involving full-term pregnant individuals aged ≥ 35 years who attempted

RICTOR Prevents Decidualization Disorder in Women With Advanced Maternal Age via Regulating FoxO1 Nuclear Export.

Miscarriage and abnormal embryonic development in women of advanced maternal age (AMA) are often associated with impaired decidualization. mTORC2 is an evolutionarily conserved protein kinase. As a core component of mTORC2, RICTOR has been implicated in nutrient sensing and is closely linked to implantation disorders; however, its role in regulating age-related decidualization disorders remains unreported. In this study, we identified pronounced decidualization defects in AMA foster mice, accompanied by a significant reduction in RICTOR in the decidual tissues of AMA women. Uterine-specific ge

Aberrant expression of Gremlin1 induces trophoblast senescence in the placenta of advanced maternal age pregnancy.

BACKGROUND: The global incidence of advanced maternal age (AMA) has risen substantially over the past two decades, accompanied by elevated risks of adverse perinatal outcomes. This phenomenon has been linked to premature placental aging, yet the precise molecular pathways have yet to be fully elucidated. Our study aims to delineate the impact of Gremlin1 (GREM1) aberrant expression in AMA pregnancies and elucidate its underlying molecular mechanisms. METHODS: This study employed a multi-level approach to investigate GREM1's role in AMA-associated placental aging. Analysis of 42 human placenta

Advanced maternal age increases the risk of adverse neonatal outcomes: a comparative study in Ethiopia.

BACKGROUND: Adverse neonatal outcomes (ANOs), including preterm birth, low birth weight, low Apgar scores, congenital malformations, and early neonatal death, remain a leading cause of neonatal and infant mortality worldwide. Pregnancy at advanced maternal age (> 35 years) increases the risk of ANOs, even in the absence of other socio-economic or medical risk factors. In developing countries such as Ethiopia, where neonatal mortality is high, evidence on the impact of advanced maternal age on neonatal outcomes is limited. This highlights the need for context-specific research to inform materna

Mitochondrial metabolism influences meiotic maturation in human oocytes of young and advanced maternal age women.

STUDY QUESTION: Is there a relationship between the mitochondrial activity and the meiotic progression of oocytes from germinal vesicle (GV) to metaphase II (MII) stages in young and advanced maternal age (AMA) women? SUMMARY ANSWER: Poor mitochondrial metabolism impairs the meiotic progression of human GV oocytes, contributing to a lower oocyte maturation capacity of AMA oocytes. WHAT IS KNOWN ALREADY: AMA oocytes are characterized by diminished quality, mostly due to the higher rates of chromosomal segregation errors occurring during meiosis I. Another hallmark of AMA oocytes is impaired m

Management of Pregnancy in Women of Advanced Maternal Age: Improving Outcomes for Mother and Baby.

Pregnancy at advanced maternal age (age >35 years old) is considered a risk factor for adverse maternal and perinatal outcomes. Yet, pregnancies of advanced maternal age have become more prevalent over the last few decades. Possible maternal complications of pregnancy at age 35 or older include increased risk of spontaneous miscarriage, preterm labor, gestational diabetes mellitus, pre-eclampsia, stillbirth, chromosomal abnormalities, and cesarean delivery. Possible adverse fetal outcomes include infants small for gestational age and intrauterine growth restrictions, low Apgar score, admission

Advanced maternal age and its association with placenta praevia and placental abruption: a meta-analysis.

This study aimed to investigate the existence and magnitude of the association between advanced maternal age (AMA) and occurrence of placenta praevia (PP) and placental abruption (PA) among nulliparous and multiparous women, by a systematic review and meta-analysis. We searched articles published between January 1, 2005 and December 31, 2015, in any language, in the following databases: PubMed, Scopus, Web of Science, and LILACS. Women were grouped into two age categories: up to 34 years old and 35 years or older. The Newcastle-Ottawa Scale was used to evaluate the methodological quality of th

Association between very advanced maternal age and adverse pregnancy outcomes: a cross sectional Japanese study.

BACKGROUND: While several studies have demonstrated the increased risk of pregnancy complications for women of advanced age, few studies have focused on women with very advanced age (≥ 45), despite the increasing rate of pregnancy among such women. Furthermore, how such risks of increase in age differ by maternal characteristics are also poorly understood. Thus, we aimed to clarify pregnant outcomes among women with very advanced age and how the effect of age differs by method of conception and parity. METHODS: We used the national multicenter Japan Society of Obstetrics and Gynecology perina
