An attenuated African swine fever virus expressing the E2 glycoprotein of classical swine fever virus protects pigs against challenge of both viruses.

African swine fever (ASF) and classical swine fever (CSF) are highly contagious diseases with high morbidity and mortality rates resulting in an enormous impact on the global pig industry. A bivalent vaccine that simultaneously protects against both ASF and CSF is highly desirable. We previously developed a seven-gene-deleted African swine fever virus (ASFV) attenuated vaccine candidate (HLJ/18-7GD) that provides complete protection against homologous strains. Herein, we constructed a recombinant virus HLJ/18-7GD-E2 by inserting the classical swine fever virus (CSFV) E2 gene into the HLJ/18-7G

Immune responses induced by a recombinant C-strain of classical swine fever virus expressing the F317L protein of African swine fever virus.

African swine fever (ASF), a highly infectious and devastating disease affecting both domestic pigs and wild boars, owes its etiology to African swine fever virus (ASFV). ASFV encodes more than 165 proteins. However, novel immunogenic proteins remain unknown. This study aimed to determine the antigenicity of the F317L protein (pF317L) of ASFV. The results revealed that pF317L was able to react with convalescent pig sera, indicating that pF317L could be a candidate antigen. The antigenic potential of pF317L expressed by rHCLV-F317L, a recombinant virus in the backbone of C-strain (a lapinized l

An update on active and passive surveillance for African swine fever in the Dominican Republic.

African swine fever (ASF) is a viral, hemorrhagic disease of swine that is reportable to the World Organisation for Animal Health. Since 2007, ASF has been expanding globally and causing severe disruption to the global swine industry. In 2021, ASF was detected in the Dominican Republic, prompting an emergency response from local and international officials. Three years later, ASF is still present in the country despite control efforts. This study used data from January 2023 to March 2024 from government-mandated sampling of commercial farms, a newly initiated active surveillance program of bac

The Effect of Partial and Temporary Vaccination on African Swine Fever Eradication Rates.

African swine fever (ASF) is one of the most severe diseases of pigs, with drastic impact on pig industry. Wild boar populations play the role of ASF virus epidemiological reservoir. No effective and safe ASF vaccine is available, yet, but a future vaccine will not be 100% effective and will provide protection for no more than a few months. We present an individual-based spatially explicit model of wild boar demography and ASF epidemiology, allowing to simulate a vaccination campaign. We tested how many animals should be vaccinated in relation to vaccine efficacy and to the duration of vaccine

Structural insights into the DNA topoisomerase II of the African swine fever virus.

Type II topoisomerases are ubiquitous enzymes that play a pivotal role in modulating the topological configuration of double-stranded DNA. These topoisomerases are required for DNA metabolism and have been extensively studied in both prokaryotic and eukaryotic organisms. However, our understanding of virus-encoded type II topoisomerases remains limited. One intriguing example is the African swine fever virus, which stands as the sole mammalian-infecting virus encoding a type II topoisomerase. In this work, we use several approaches including cryo-EM, X-ray crystallography, and biochemical assa

African swine fever. IV. Demonstration of the viral antigen by means of immunofluorescence.

African swine fever immunofluorescent conjugates were prepared in swine and used successfully in the demonstration of viral antigen in frozen tissue sections and in inoculated tissue culture cells. Cross reactivity was observed with the six strains used in the inoculation of swine. The high antibody content of the serum of immune swine did not interfere with demonstration of the antigen in frozen tissue sections of certain of their organs. The localisation and extent of antigen varied with the stage of infection. The virus was demonstrated in spleen and other organs as early as after one day o

Biosecurity indemnification and attitudes of United States swine producers towards the prevention of an african swine fever outbreak.

Animal disease outbreaks, such as the recent outbreak of African Swine Fever in 2018, are a major concern for stakeholders across the food supply chain due to their potential to disrupt global food security, cause economic losses, and threaten animal welfare. As a result of their transboundary nature, discussions have shifted to preventive measures aimed at protecting livestock while ensuring food security and safety. Emergency assistance has been a critical response option during pandemics. However, this may not be sustainable in the long run because the expectation of government bailout may

Transboundary Animal Diseases (TADs) affecting domestic and wild African ungulates: African swine fever, foot and mouth disease, Rift Valley fever (1996-2018).

Transboundary Animal Diseases (TADs) affecting ungulates are potentially epidemic diseases that are contagious or transmissible, and a concern for humans worldwide. While globalization has led to increased containment efforts at a global scale, additional local attention is needed as well for vulnerable pastoralist communities that are in close proximity to livestock. We reviewed and then summarized outbreaks, cases, and deaths for 3 TADs (African Swine Fever (ASF), Foot and Mouth Disease (FMD), and Rift Valley Fever (RVF)) affecting domestic and wild ungulates for all African countries from 1

Enhancing immune resilience in swine: a comprehensive meta-review of preventive and host-directed strategies against African swine fever and swine influenza.

African swine fever (ASF) and swine influenza (SI) are among the most consequential infectious threats to global swine production because they combine major economic burden with fundamentally different transmission ecology, tissue tropism, and immune-pathological behavior. ASF is a highly lethal systemic disease characterized by macrophage tropism, immune dysregulation, environmental persistence, and severe trade and production consequences, whereas SI is a highly transmissible respiratory infection associated with recurrent herd-level morbidity, viral evolution, and zoonotic significance. A new vaccination regimen using adenovirus-vectored vaccine confers effective protection against African swine fever virus in swine.

African swine fever (ASF) is an acute and highly contagious lethal infectious disease in swine that severely threatens the global pig industry. At present, a safe and efficacious vaccine is urgently required to prevent and control the disease. In this study, we evaluated the safety and immunogenicity of replication-incompetent type-2 adenoviruses carrying African swine fever virus (ASFV) antigens, namely CP204L (p30), E183L (p54), EP402R (CD2v), B646L (p72), and B602L (p72 chaperone). A vaccine cocktail delivered by simultaneous intramuscular (IM) and intranasal (IN) administration robustly el

Proteome Analysis of Swine Macrophages after Infection with Two Genotype II African Swine Fever Isolates of Different Pathogenicity.

Since the introduction of a highly pathogenic genotype II isolate of the African swine fever virus (ASFV) into Georgia in 2007, African swine fever (ASF) has gone panzootic. Outbreaks have been reported in Europe, Asia and, more recently, Latin America. Thus, ASFV has become a major threat to the pig industry worldwide, as broadly applicable vaccines are not available. While the majority of ASFV strains show high virulence in domestic pigs and wild boar, variations within the ASFV genome have resulted in the emergence of attenuated strains with low or moderate virulence. However, the molecular

A host-virus arms race over FAM134B in African swine fever virus infection.

Although reticulophagy, the selective autophagy of the endoplasmic reticulum (ER), restricts several RNA viruses, whether this pathway constrains DNA virus infection remains unknown. Recently, we found that FAM134B (family with sequence similarity 134 member B), an ER-resident cargo receptor that mediates reticulophagy, restricts African swine fever virus (ASFV), a large cytoplasmic DNA virus, by targeting the major capsid proteins p72 and pA137R for lysosomal degradation, thereby blocking capsid assembly at perinuclear viral factories. ASFV counters this restriction through the virulence fact

Designing a multi-epitope vaccine against African swine fever virus using immunoinformatics approach.

African swine fever (ASF) is a highly contagious and fatal haemorrhagic disease affecting domestic and wild pigs, with no effective vaccine currently available. The lack of an effective vaccine has hindered global ASF control efforts, leading to devastating economic losses in the swine industry. Traditional vaccine development approaches have faced challenges due to ASFV's genetic complexity and immune evasion strategies. Therefore, this study aims to leverage immunoinformatic approaches to facilitate acceleration of the early stages of vaccine development, optimizing resource utilization and

Attenuated African swine fever viruses and the live vaccine candidates: a comprehensive review.

The African swine fever virus (ASFV) is spreading worldwide and causing huge economic losses to the global pig industry. The ASFV genome is 170-193 kb in length, contains approximately 150 open reading frames, and encodes more than 200 proteins, most of which have unknown functions. Owing to the unique viral structure, replication strategy, large number of genes of unknown function, and complicated pathogenesis, vaccine development research is challenging. Several naturally attenuated ASFV isolates have been extensively investigated and many genetically manipulated, gene-deleted, and cell-adap

Clinicopathological and ultrastructural study of African swine fever outbreak in North-East India.

The present investigation focuses on examining the clinical, histopathological, and ultrastructural changes that occurred in pig, during an outbreak of African swine fever (ASF) in 2022 in Assam, India. The disease initially manifested as a per-acute case with high mortality but without any evident clinical signs. Subsequently, some animals exhibited an acute form of the disease characterized by high fever (104-106 °F), anorexia, vomiting, respiratory distress, and bleeding from the anal and nasal orifices. During acute African swine fever virus (ASFV) infections, elevated levels of pro-inflam

Detection of African swine fever virus utilizing the portable MatMaCorp ASF detection system.

African swine fever (ASF) has emerged as a major threat to domestic and wild suid populations, and its continued spread threatens commercial swine production worldwide. The causative agent of ASF, African swine fever virus (ASFV), possesses a linear, double stranded DNA genome. Traditional detection of ASFV relies on laboratory-based virus isolation or real-time PCR of samples, typically blood or spleen, obtained from suspect cases. While effective, these methodologies are not easily field deployable, a major limitation during disease outbreak and response management scenarios. In this report,

African swine fever: A re-emerging viral disease threatening the global pig industry.

African swine fever (ASF) recently has spread beyond sub-Saharan Africa to the Trans-Caucasus region, parts of the Russian Federation and Eastern Europe. In this new epidemiological scenario, the disease has similarities, but also important differences, compared to the situation in Africa, including the substantial involvement of wild boar. A better understanding of this new situation will enable better control and prevent further spread of disease. In this article, these different scenarios are compared, and recent information on the pathogenesis of ASF virus strains, the immune response to i

BacMam immunization partially protects pigs against sublethal challenge with African swine fever virus.

Lack of vaccines and efficient control measures complicate the control and eradication of African swine fever (ASF). Limitations of conventional inactivated and attenuated virus-based vaccines against African swine fever virus (ASFV) highlight the need to use new technologies to develop efficient and safe vaccines against this virus. With this aim in mind, in this study we have constructed BacMam-sHAPQ, a baculovirus based vector for gene transfer into mammalian cells, expressing a fusion protein comprising three in tandem ASFV antigens: p54, p30 and the extracellular domain of the viral hemag

[Pathology of lymphoid tissue cells infected by African swine fever virus in vitro].

The authors studied the pathology of bone marrow (BM) lymphoid cell from pigs infected by African swine fever virus (ASFV) in vitro. Monocytes were shown to be primarily afflicted in unstimulated BM culture. These cells disappeared completely 72 hours after infection. Just 24 hours following ASFV infection, there were atypical lymphocytes amounting to 12% of the general lymphoid population at hour 72 after inoculation. The area and perimeter of minor, middle, and large lymphocytes tended to reduce during both BM cell cultivation and inoculation. Lymphoblasts and monocytes were generally triploi
