The diphosphination of alkenes through a radical pathway offers a promising approach for the rapid construction of aryl bisphosphines. However, such a synthetic strategy has not been successfully applied to the preparation of alkyl bisphosphines, partially due to the difficulties in the generation of phosphorus-centered radicals from common alkyl phosphine compounds. We herein demonstrate that this challenge can be overcome by hiring Janus-faced chlorophosphine as the phosphine source that can act as not only a radical precursor to generate phosphine-centered radicals but also a radicalphile t

The catalytic asymmetric inverse-electron-demand oxa-Diels-Alder (IODA) reaction is a highly effective synthetic method for creating enantioenriched six-membered oxygen-containing heterocycles. Despite significant effort in this area, simple α,β-unsaturated aldehydes/ketones and nonpolarized alkenes are seldom utilized as substrates due to their low reactivity and difficulties in achieving enantiocontrol. This report describes an intermolecular asymmetric IODA reaction between α-bromoacroleins and neutral alkenes that is catalyzed by oxazaborolidinium cation 1f. The resulting dihydropyrans are

Herein, we disclose a nickel/photo-cocatalyzed three-component alkyl-acylation of aryl-substituted alkenes with aldehydes and electron-withdrawing-group-activated alkyl bromides, providing straightforward access to various ketones under mild and ligand-free conditions. The photocatalyst TBADT plays a dual role in activating the acyl C-H bond of aldehydes via hydrogen atom transfer and reducing the C-Br bond of alkyl bromides via single-electron transfer. While the terminal C-C bond is forged through polarity-matched radical-type addition, nickel is likely involved in the acylation step.

Herein, we reported a method for iron-catalyzed, visible-light-induced hydrosilylation reactions of electron-deficient alkenes to produce value-added silicon compounds. Alkenes bearing functional groups with different steric properties were suitable substrates, as were derivatives of structurally complex natural products. Mechanistic studies showed that chlorine radicals generated by iron-catalyzed ligand-to-metal charge transfer in the presence of lithium chloride promoted the formation of silyl radicals.

The addition reaction between CuBpin and alkenes to give a terminal boron substituted intermediate is usually fast and facile. In this communication, a selectivity-reversed procedure has been designed and established. This selectivity-reversed borocarbonylation reaction is enabled by a cooperative action between palladium and copper catalysts and proceeds with complete regioselectivity. The key to the success of this transformation is the coordination of the amide group and slower CuBpin formation by using KHCO3 as the base. A wide range of β-boryl ketones were produced from terminal unactivat

Herein, we present a nickel-catalyzed three-component reductive alkylacylation of electron-deficient activated alkenes with tertiary alkyl bromides and acid anhydrides. This method enables the efficient preparation of a variety of ketones with broad substrate scope and high functionality tolerance starting from simple precursors. On the basis of the preliminary mechanistic investigations, a catalytic cycle involving the synergistic interaction of nickel, zinc, and MgCl2 is proposed as the major reaction pathway.

The first example of a metal-free direct carbotrifluoromethylation of alkenes using inexpensive TMSCF3 as the CF3 source is described. The methodology not only exhibits high chemoselectivity for this transformation but also expands the substrate scope that is difficult to access by known transition-metal-catalyzed methods.

The nephrotoxicity of halogenated alkenes is due to the beta-lyase mediated bioactivation of the hepatic glutathione (GS) conjugate to mutagenic or cytotoxic reactive species in kidney. Experimental evidence obtained for regioisomers and geometric isomers of haloalkene GS conjugates indicates that different isomers may be metabolized and excreted at different rates, follow different metabolic pathways, and exhibit different toxicities. Computational methods were applied in the present work to a conformational study of GS-haloalkene conjugates to determine the relative stabilities of possible r

Herein, we introduce a new synthetic protocol for azole alkylation. Our strategy first engages azoles using alkenylthianthrenium electrophiles to provide 1,2-azolothianthrenium salts, then selectively excises the sulfonium moiety via single electron transfer (SET). Given that alkenylthianthrenium salts can be readily accessed from alkenes, the net azole addition-hydrogenolysis sequence unlocks a versatile approach to formal Markovnikov alkene hydroazolation. Moreover, the key radical intermediate formed after reduction can be engaged in reactions beyond hydrogen atom transfer (HAT), providing

The development of efficient protocols for the construction of difluoromethylene (CF2) units has remained at the forefront of synthetic organic chemistry, owing to their widespread applications in pharmaceuticals, organic synthesis, and material science. Herein, we report a palladium-catalyzed redox-neutral difluoromethylation reaction of N-indolyl-tethered unactivated alkenes, via a visible-light-promoted defluorinative cascade of readily available trifluoromethylarenes. Mechanistic studies suggested that the reaction is likely to proceed through an efficient homolytic cleavage of a C(sp3)-F

The cyclopropyl group is an invaluable motif in both medicinal and synthetic chemistry, with 1,2-disubstituted halocyclopropanes serving as important building blocks for the preparation of more complex cyclopropane-containing structures. Herein, we report a Vitamin B12-photocatalyzed approach for the direct synthesis of 1,2-disubstituted halocyclopropanes, starting from alkenes and simple haloforms under visible-light irradiation. Our method yields a diverse range of 1,2-disubstituted halocyclopropanes with high cis-selectivity, providing efficient access to these traditionally difficult-to-pr

Organophosphorus compounds constitute an important class of functional molecules with broad relevance in synthesis, catalysis, and medicinal chemistry. Despite their importance, biocatalytic C-P bond formation remains rare, with only a limited number of examples identified from natural product biosynthesis. Here we report a previously unexplored biocatalytic platform for C-P bond construction through flavin-dependent enzyme-catalyzed intermolecular radical coupling involving phosphinoyl radical intermediates. The transformation is directly driven by ground-state FMNhq, and protein engineering

Oxidative cleavage of carbon-carbon double bonds often requires hazardous reagents and demanding conditions. In this study, we report a photocatalytic oxidative cleavage of alkenes using benign carbon dioxide (CO2) as an oxygen donor, producing ketones or carboxylic acids at atmospheric pressure and room temperature. A robust iron-based heterogeneous photocatalyst facilitates oxygen transfer to form an epoxide intermediate that subsequently undergoes ring opening and carbon-carbon bond cleavage to yield the oxidative products with high selectivity. Comprehensive mechanistic studies combine tim

A novel Mn-based single-atom photocatalyst is disclosed in this study, designed for the dichlorination of alkenes to achieve vicinal dichlorinated products using N-chlorosuccinimide as a mild chlorinating agent, which have widespread applications as pest controlling agents, polymers, flame retardants, and pharmaceuticals. In developing this innovative catalyst, we achieved the atomic dispersion of Mn on aryl-amino-substituted graphitic carbon nitride (f-C3N4). This marks the first instance of a heterogeneous version, offering an operationally simple, sustainable, and efficient pathway for dich

Transition-metal-catalyzed regio- and enantioselective cross-coupling of alkyl metallic species has emerged as a cornerstone in modern organic synthesis, which enables the construction of carbon-carbon and carbon-heteroatom bonds with high precision to facilitate rapid access to important organic targets with molecular complexity. However, the selective formation and utilization of different alkyl metallic intermediates from one precursor under identical conditions remain unknown and challenging. Herein, a Ni-catalyzed adaptive migratory asymmetric cross-hydroacylation of unactivated alkenes f

Polysubstituted cyclopropanes are valuable in complex molecule synthesis but challenging to access due to low atom economy and harsh conditions. Herein, we report a copper-mediated, peroxide-initiated cyclopropanation of activated alkenes with active methylene compounds via a radical pathway. This diazo-free, prefunctionalization-free method, integrated with continuous flow, enhances safety, scalability, and efficiency. Operable under mild conditions with broad substrate scope, it offers a practical and atom-economical route to diverse polysubstituted cyclopropanes.

Both fluorosulfonyl and cyano groups are important structural motifs in bioactive molecules. Herein, we report a new difunctionalization reaction of alkenes based on fluorosulfonyl radicals, which allows for the introduction of the fluorosulfonyl and cyano groups into unactivated alkenes in one step. This transformation is enabled by merging photoredox and copper catalysis, featuring visible light catalysis, mild conditions, and good functional group tolerance. Further transformation of products via SuFEx reactions is also demonstrated.

A general method for the light-driven intermolecular anti-Markovnikov hydroamination of alkenes with primary sulfonamides, sulfamides, and sulfamates is presented. The reaction is mediated by a ternary catalyst system composed of an iridium(III) chromophore, a fluorinated alkoxide base, and a thiol H-atom donor. We hypothesize that the reactions proceed through a proton coupled electron-transfer (PCET) mechanism wherein implementation of the alkoxide base imparts additional thermochemical driving force for the homolytic activation of strong N-H bonds that were previously inaccessible using thi

A visible-light-photocatalyzed 1,2-arylalkylation of N-(arylsulfonyl)acrylamides with ketone-based dihydroquinazolinones is described. The formal C-C bond cleavage of aliphatic ketones is unified with tandem radical alkylation/1,4-aryl migration/desulfonylation to forge two different types of vicinal C-C bonds and construct an all-carbon quaternary α-stereocenter, thus enhancing the carbogenic complexity and tolerating diverse functionalities. Additional to telescopic synthesis and product diversification, this method features a radical dicarbofunctionalization of conjugated N-(arylsulfonyl)ac
