Copper-catalyzed (4+1) and (3+2) cyclizations of iodonium ylides with alkynes.

The copper(ii)-catalyzed (4+1) cyclizations and copper(i)-catalyzed (3+2) cycloadditions of iodonium ylides and alkynes were successfully developed by employing efficient and safe iodonium ylides instead of traditional diazo compounds. Highly functionalized dimethyl (E)-3-benzylideneindoline-2,2-dicarboxylates and methyl 5-(2-hydroxyphenyl)-2-methoxy-4-phenylfuran-3-carboxylates were conveniently prepared in moderate to excellent yields. The possible reaction mechanisms were also discussed.

Intermolecular cyclotrimerization of haloketoalkynes and internal alkynes: facile access to arenes and phthalides.

A highly chemo- and regioselective cyclo(co)trimerization between 3-halopropiolamides and symmetrical internal alkynes is reported. The reaction is catalyzed by CpRuCl(COD) and proceeds under air at ambient temperature in ethanol with no additional precautions. Iodo-, bromo-, and chloropropiolamides, esters, and ketones are viable coupling partners and, in a 2 : 1 stoichiometry relative to internal alkyne, yield fully-substituted arenes in a single step. The highest regioselectivities (96% single isomer) were observed when employing 2° and 3°-halopropiolamides. A mechanistic hypothesis account

Synthesis and cycloaddition reactions of strained alkynes derived from 2,2'-dihydroxy-1,1'-biaryls.

A series of strained alkynes, based on the 2,2'-dihydroxy-1,1'-biaryl structure, were prepared in a short sequence from readily-available starting materials. These compounds can be readily converted into further derivatives including examples containing fluorescent groups with potential for use as labelling reagents. The alkynes are able to react in cycloadditions with a range of azides without the requirement for a copper catalyst, in clean reactions with no observable side reactions.

Strain-promoted cycloadditions involving nitrones and alkynes--rapid tunable reactions for bioorthogonal labeling.

The development and applications of strain-promoted alkyne-nitrone cycloaddition (SPANC) reactions have brought about new tools for rapid and specific functionalization of biomolecules in different settings. While a number of strain-promoted reactions have been successfully developed, SPANC reactions offer high reactivity with bimolecular rate constants of k2 that are as fast as 60M(-1)s(-1). SPANC reactions also offer stability of starting materials, particularly in the case of endocyclic nitrones, as well as stereoelectronic tunability of the nitrone moiety to optimize reactivity towards dif

Heterogeneously catalyzed efficient hydration of alkynes to ketones by tin-tungsten mixed oxides.

The Sn-W mixed oxide prepared by calcination of the Sn-W mixed hydroxide precursor with a Sn/W molar ratio of 2:1 at 800 °C (SnW2-800) acts as an efficient heterogeneous catalyst for the hydration of alkynes. Structurally diverse terminal and internal alkynes, including aromatic, aliphatic, and double-bond-containing ones, can be converted into the corresponding ketones in moderate to high yields. The catalytic activity of SnW2-800 is much higher than those of previously reported heterogeneous catalysts and commonly utilized acid catalysts. The observed catalysis was truly heterogeneous, and t

Flexible Alkyne-Linked Thymidine Phosphoramidites and Triphosphates for Chemical or Polymerase Synthesis and Fast Postsynthetic DNA Functionalization through Copper-Catalyzed Alkyne-Azide 1,3-Dipolar Cycloaddition.

Two alternative flexible alkyne-linked thymine nucleosides (propargyl-diethylene glycol- or undecyn-linked 5-hydroxymethyluracil derivatives), as well as their phosphoramidites and triphosphates, were designed and synthesized. The nucleoside 3'- O-phosphoramidites were successfully incorporated into oligonucleotides on a solid support, whereas the nucleoside triphosphates served as good substrates for polymerase synthesis of modified DNA, which underwent fast and efficient copper-catalyzed alkyne-azide 1,3-dipolar cycloaddition (CuAAC) reactions.

Photocatalytic Semireduction of Propiolamides in Microheterogeneous Solutions via Alkyne Radical Anions.

A photocatalytic protocol for generating alkyne radical anions from propiolamides in aqueous micellar solutions is reported. Using ascorbic acid and Ru(bpy)32+ under blue light, hydrated electrons mediate single-electron reduction. Alkyne radical anions undergo Z-selective semireduction or reductive hydroalkylation with styrenes. The method shows a broad scope, excellent functional group tolerance, and gram-scale scalability, replacing hazardous reagents and organic solvents with vitamin C and water.

Experimental and theoretical studies on the (co)cyclotrimerization of alkynes (and ethylene) in a TpRh compound.

The reactions between the Rh(I) ethylene complex TpRh(C2H4)2 (1; Tp = hydrotris(pyrazolyl)borate), and the alkynes di-tert-butylacetylene dicarboxylate (DTBAD), acetylene, phenylacetylene, and methyl propiolate (MP) have been studied, and the results compared with those obtained previously for dimethylacetylene dicarboxylate (DMAD). Electron withdrawing groups (DTBAD) at both termini of the triple bond stabilize η4-diene-Rh(I) species, while terminal alkynes (acetylene, phenylacetylene) easily lead to η3-allyl-Rh(III) species. The terminal alkyne methyl propiolate, with only one electron withd

Nickel-Catalyzed Reductive Alkyne Hydrocyanation Enabled by Malononitrile and a Formaldehyde Additive.

The development of a nickel-catalyzed reductive alkyne hydrocyanation is described using 2-methyl-2-phenylmalononitrile (MPMN), a C-bound electrophilic transnitrilation reagent. Reproducibility issues led to the detection of oxidized hemiaminal impurities within N,N-dimethylacetamide. These impurities release formaldehyde in situ, which was ultimately identified as a critical reaction additive. A range of diaryl and aryl-alkyl alkynes underwent hydrocyanation. Mechanistic experiments revealed that formaldehyde and MPMN undergo a Ni-catalyzed reductive coupling of two π-components, leading to t

Practical Considerations, Challenges, and Limitations of Bioconjugation via Azide-Alkyne Cycloaddition.

Interrogating biological systems is often limited by access to biological probes. The emergence of "click chemistry" has revolutionized bioconjugate chemistry by providing facile reaction conditions amenable to both biologic molecules and small molecule probes such as fluorophores, toxins, or therapeutics. One particularly popular version is the copper-catalyzed azide-alkyne cycloaddition (AAC) reaction, which has spawned new alternatives such as the strain-promoted azide-alkyne cycloaddition reaction, among others. This focused review highlights practical approaches to AAC reactions for the s

Polyhedral Cu2 O Crystals for Diverse Aryl Alkyne Hydroboration Reactions.

Cu2 O cubes, octahedra, and rhombic dodecahedra have been used to examine facet-dependent catalytic activity in aryl alkyne hydroboration reactions. Although the reaction can proceed by using ethanol or other alcohols as solvent, the use of 1,4-dioxane gave the best product yield. All particle shapes gave exclusively the E-product, but the rhombic dodecahedra exposing {110} surfaces were consistently far more reactive than the other particle morphologies. A product yield of 99 % was achieved by using Cu2 O rhombic dodecahedra to catalyze the hydroboration of phenylacetylene at 60 °C for 5 h. T

Profiling protein methylation with cofactor analog containing terminal alkyne functionality.

Enzymatic transmethylation from the cofactor S-adenosyl-L-methionine (SAM) to biological molecules has recently garnered increased attention because of the diversity of possible substrates and implications in normal biology and diseases. To reveal the substrates of protein methyltransferases (PMTs), the present article focuses on an alkyne-containing SAM mimic, Se-adenosyl-L-selenomethionine (ProSeAM), and a cleavable azido-azo-biotin probe to profile the targets of endogenous PMTs in cellular contexts. This article describes the stepwise preparation of cell lysates containing active, endogeno

Alkyne insertion at a nickel-aluminum heterometallic complex: implications for pyridine alkenylation.

Cooperative Ni-Lewis acid catalysis has emerged as an effective strategy for the selective C-H functionalisation of heteroarenes, yet the elementary steps following C-H activation remain poorly understood. In particular, direct experimental insight into the migratory insertion step, which governs C-C bond formation and potentially product regioselectivity, has been lacking. Herein, we demonstrate that a heterobimetallic {Ni-Al} complex derived from the C-H activation of a pyridine undergoes efficient and selective insertion of alkynes under mild conditions. This reaction provides direct access

Reshaping Chiral Self-Assemblies Through Alkyne-Involved Click Chemistry.

Highly efficient click reactions in the solid state or space-confined condition potentially induce in situ compositional and structural evolution, offering opportunities for smart materials and nanofabrication technologies. Herein, we report a strategy for reshaping self-assembled chiral nanomaterials mediated by a [2+2] cycloaddition reaction. Amino acids grafted with large π-conjugated systems are employed as the primary building blocks, into which the electron-deficient tetracyanoquinodimethane (TCNQ) and ethynylaniline (EDA) are orthogonally introduced. These components form ternary co-ass

On-Surface Azide-Alkyne Cycloaddition Reaction: Does It Click with Ruthenium Catalysts?

Owing to its simplicity, selectivity, high yield, and the absence of byproducts, the "click" azide-alkyne reaction is widely used in many areas. The reaction is usually catalyzed by copper(I), which selectively produces the 1,4-disubstituted 1,2,3-triazole regioisomer. Ruthenium-based catalysts were later developed to selectively produce the opposite regioselectivity─the 1,5-disubstituted 1,2,3-triazole isomer. Ruthenium-based catalysis, however, remains only tested for click reactions in solution, and the suitability of ruthenium catalysts for surface-based click reactions remains unknown. Al

Polyphenylene-Based Solid Acid as an Efficient Catalyst for Activation and Hydration of Alkynes.

Porous polymer catalysts possess the potential to combine the advantages of heterogeneous and homogeneous catalysis, namely, easy postreaction recycling and high dispersion of active sites. Here, we designed a -SO3H functionalized polyphenylene (PPhen) framework with purely sp2-hybridized carbons, which exhibited high activity in the hydration of alkynes including challenging aliphatic substrates such as 1-octyne. The superiority of the structure lies in its covalent crosslink in the xy-plane with a π-π stacking interaction between the planes, enabling simultaneously high swellability and poro

The Utility of Sonogashira Coupling Reaction for the Derivatization of Aryl Halides with Fluorescent Alkyne.

Aryl halides are a very important category of compounds that include many vital drugs and key industrial additives, such as clofibrate and bromobenzene, respectively. Due to their importance, our research group previously developed a novel fluorescence labeling approach for their analysis using a fluorescent aryl boronic acid as a reagent, based on the Suzuki coupling reaction. This coupling reaction was successfully applied for the determination of aryl halides in biological fluids; however, there was a limitation of low reactivity towards ortho-substituted aryl halides. In the present study,

Electrochemical Bromo-Alkyloxylation of Conjugated Alkenes: A Method to Access Bromo-Alkyloxy-Alkenes/Alkynes.

A highly efficient bromo-alkyloxylation of conjugated alkenes through direct electrolysis under mild reaction conditions has been developed. This protocol provides an efficient approach to a series of bromo-alkyloxy-alkenes/alkynes bearing various functional groups, achieving moderate to high yields. Mechanistic studies revealed that the charge and ionization effects induce the high selectivity observed in the reaction of 1,3-dienes and 1,3-enynes.

Promoting CO2 Conversion With Terminal Alkynes Over ZSM-5 Zeolite-Confined Uniform Ag Nanoparticles.

Heterogeneous catalysis-mediated terminal alkyne carboxylation enabled the construction of stable C─C bonds via direct C─H bond activation, achieving efficient conversion of CO2 into unsaturated carboxylic multi-carbon derivatives, and providing a new route for artificial carbon fixation and the synthesis of multifunctional chemicals. However, designing heterogenous catalysts with highly efficient, uniformed, stable active sites for this solid-liquid-gas three-phase systems remained a significant challenge. Herein, we proposed a solvent-assist hydrothermal method to synthesis 6-7 nm uniformed

Cascade Coupling of Dinitrogen, Carbon Monoxide, Carbon Dioxide, and Alkynes in a Dititanium Framework.

Dinitrogen (N2), carbon monoxide (CO), and carbon dioxide (CO2) are all abundant and readily available chemical feedstocks. The functionalization of N2 with CO and CO2 is of considerable interest and importance, but remains a formidable challenge, owing to their high thermodynamic and kinetic stability. Here, we report an unprecedented cascade coupling of N2, CO, CO2, and alkynes within a dititanium framework. Sequential reactions of N2, CO, alkynes (R1C≡CR2), and CO2 with a dititanium tetrahydride complex at room temperature selectively afforded the corresponding four-component coupling produ
