Due to anthropic environmental changes, vector-borne diseases are emerging worldwide. Ticks are known vectors of several pathogens of concern among humans and animals. In recent decades, several examples of tick-borne emerging viral diseases have been reported (Crimean Congo hemorrhagic fever virus, Powassan virus, encephalitis virus, heartland virus, severe fever with thrombocytopenia syndrome virus). Unfortunately, few studies addressing the presence of viruses in wild ticks have been carried out in South America. With the aim of detecting flaviviruses and orthobunyaviruses in ticks, we carr

A new genotype phylogenetically close to Ehrlichia canis named as Ehrlichia minasensis was identified infecting cattle and deer in Canada, as well as Rhipicephalus microplus ticks and cattle in Brazil. Although it was detected in R. microplus, little is known about the epidemiology of this ehrlichiosis, especially in other tick species. This study evaluated the minimum infection rate of E. minasensis in Amblyomma sculptum and R. microplus ticks from locations where naturally infected cattle were previously detected. Overall, 45 engorged R. microplus ticks after molting [43 pools of adults (13.

The lone star tick, Amblyomma americanum, vectors pathogens of emerging diseases of humans and animals in the United States. Currently, measures are not available for effective control of A. americanum infestations. Development of vaccines directed against tick proteins may reduce tick infestations and the transmission of tick-borne pathogens. However, the limiting step in tick vaccine development has been the identification of tick protective antigens. Herein, we report the application of RNA interference (RNAi) for screening an A. americanum cDNA library for discovery of tick protective anti

The responses of adult Amblyomma variegatum ticks released from various distances to different doses of the synthetic attraction-aggregation-attachment pheromone (AAAP) (made up of orthonitrophenol, methyl salicylate and nonanoic acid in paraffin oil), dispensed from the center of circular field plots, were studied in the presence or absence of elevated levels of CO2. Up to 90% of the ticks released were attracted to the pheromone source in the presence of CO2 within 3h. CO2 alone was unattractive, similar to previous findings in Zimbabwe, but unlike results from a Caribbean A. variegatum popu

Unfed adults and nymphs of the bont tick Amblyomma hebraeum Koch are attracted to hosts on which fed males, emitting an aggregation-attachment pheromone (AAP), are attached. Pheromone/acaricide mixtures have the potential to selectively attract and kill these ticks. We have investigated the effects of three acaricides, amitraz (an amidine), flumethrin (a synthetic pyrethroid) and chlorfenvinphos (an organophosphate), combined with AAP, on the attraction and attachment of the unfed adults. Attraction, measured in field assays involving AAP and CO2, was not inhibited by any of the acaricides. At

Experimental transmission of the yellow fever virus by the tick Amblyomma variegatum (F.) (The Tropical Bont Tick) is demonstrated for the first time. The tick transmitted the virus from one monkey to another. Trans-stasial passage from nymph to adult stage was also demonstrated. The possible role of Amblyomma variegatum as a vector and a potential reservoir of yellow fever is discussed.

Amblyomma ovale is a widespread hard tick in Neotropics and it is an important vector of pathogens, particularly Rickettsia parkeri strain Atlantic rainforest, which causes a spotted fever illness in humans. Adult ticks primarily parasitize medium and large mammals, such as carnivores, while larvae and nymphs feed on small mammals and occasionally on birds. In Brazil, the species is found in all six major biomes (Amazon, Atlantic forest, Cerrado, Caatinga, Pantanal, Pampa), and is generally found in humid, forested areas. Despite its broad range, large-scale genetic studies have been lacking.

Rickettsia parkeri is an emerging eschar-causing human pathogen in the spotted fever group of Rickettsia and is transmitted by the Gulf coast tick, Amblyomma maculatum. Tick saliva has been shown to alter both the cellular and humoral components of the innate and adaptive immune systems. However, the effect of this immunomodulation on Rickettsia transmission and pathology in an immunocompetent vertebrate host has not been fully examined. We hypothesize that, by modifying the host immune response, tick feeding enhances infection and pathology of pathogenic spotted fever group Rickettsia sp. In

Rickettsia parkeri Luckman (Rickettsiales: Rickettsiaceae), a member of the spotted fever group of Rickettsia, is the tick-borne causative agent of a newly recognized, eschar-associated rickettsiosis. Because of its relatively recent designation as a pathogen, few studies have examined the pathogenesis of transmission of R. parkeri to the vertebrate host. To further elucidate the role of tick feeding in rickettsial infection of vertebrates, nymphal Amblyomma maculatum Koch (Acari: Ixodidae) were fed on C3H/HeJ mice intradermally inoculated with R. parkeri (Portsmouth strain). The ticks were al

This study reports rickettsial infection in Amblyomma cajennense and Amblyomma dubitatum ticks collected in an area of the state of Minas Gerais, Brazil, where Brazilian spotted fever is considered endemic. For this purpose, 400 adults of A. cajenennse and 200 adults of A. dubitatum, plus 2,000 larvae and 2,000 nymphs of Amblyomma spp. were collected from horses and from the vegetation. The ticks were tested for rickettsial infection through polymerase chain reaction (PCR) protocols targeting portions of three rickettsial genes (gltA, ompA, and ompB). Only two free‑living A. cajennense adult t

The Cayenne tick Amblyomma cajennense infests preferably horses in its adult form but other mammal species in its immature stages and is the main vector of Rickettsia rickettsii, the Brazilian spotted fever pathogen. As wooless lambs are often raised on pasture together with horses, an experiment was performed to investigate their possible acquisition of resistance to A. cajennense after experimental infestations. Seven naïve wooless lambs were infested thrice at 60 days interval with immature and adult instars of A. cajennense from a laboratory colony, the tick biotic potential being determin

The present study aimed to evaluate the occurrence of immune resistance in dogs successively infested with Amblyomma cajennense nymphs. Five animals were submitted to four consecutive infestations with A. cajennense nymphs, at fourteen-day intervals. For each infestation, 50 nymphs were used per animal and data on the parasitic and non-parasitic periods were recorded. The average recovering rate of engorged nymphs in the successive infestations were 52.0, 29.2, 9.6 and 12.8%, respectively, with a significant reduction (p<0.05) of this parameter from the second infestation onwards. Laboratory-reared larval Gulf Coast ticks (GCTs) (Amblyomma maculatum) were exposed experimentally and found to acquire Hepatozoon americanum infection while feeding on parasitemic dogs. These ticks supported gamogonic and sporogonic development of the apicomplexan, and oocysts from newly molted nymphs were infectious for a dog. Other nymphs from this cohort that were allowed to feed on a blood-parasite naive sheep molted normally; the resulting adult ticks contained oocysts that were infectious for another dog. Merogonic development of H. americanum in the dogs and the resulting lesions/disea

UNLABELLED: Jingmenviruses are a group of segmented flaviviruses detected in arthropods and vertebrates that have attracted growing public health interest due to the recognition of some members as emerging human arboviral pathogens. As part of a study aimed at deciphering the virome of ticks of medical and veterinary importance in Costa Rica, we detected Jingmen tick virus (JMTV) in host-feeding Amblyomma mixtum ticks collected from horses. We assembled three complete genome segments and one partial segment from tick pools. Phylogenetic analyses revealed that JMTV from Costa Rica (JMTV Costa R

The three-host tick Amblyomma variegatum, commonly known as the tropical bont tick, poses a major threat to livestock health and productivity in tropical and subtropical regions worldwide. The inability to control the tick and prevent the diseases it transmits is partly due to the necessity of rearing the tick on animals. Thus, the goal of this study was to adapt a previously developed artificial membrane feeding system to complete the life cycle of A. variegatum. All life stages of A. variegatum were fed using fresh goat blood at 38 °C and blood replacement occurred every 12 h. Key parameters

A novel Borrelia species, Candidatus Borrelia javanense, was found in ectoparasite ticks, Amblyomma javanense, from Manis javanica pangolins seized in anti-smuggling operations in southern China. Overall, 12 tick samples in 227 (overall prevalence 5.3%) were positive for Candidatus B. javanense, 9 (5.1%) in 176 males, and 3 (5.9%) in 51 females. The phylogenetic analysis, based on the 16S rRNA gene and the flagellin gene sequences of the Borrelia sp., exhibited strong evidence that Candidatus B. javanense did not belong to the Lyme disease Borrelia group and the relapsing fever Borrelia group

Tick-borne rickettsiae are undergoing epidemiological changes in the eastern United States while human encounters with lone star ticks (Amblyomma americanum L.) have increased substantially. We used real-time polymerase chain reaction assays to test for three species of spotted fever group rickettsiae (SFGR) (Rickettsiales: Rickettsiaceae) in 1,858 nymphal A. americanum collected from Monmouth County, New Jersey, a coastal county with endemic Lyme disease and established tick surveillance. Out of the 1,858 tested, 465 (25.0%) were infected with Rickettsia amblyommatis Karpathy, a species of un

Owing to the sanitary importance of the tick Amblyomma parvum, this study evaluated the infection by Ehrlichia, Anaplasma and Rickettsia species of questing A. parvum collected in northwestern Argentina. Our results showed that A. parvum ticks in this region are infected with the recently reported Ehrlichia sp. strain San Luis, closely related to Ehrlichia chaffeensis. A high prevalence of Candidatus Rickettsia andeanae was observed. Most of the infected ticks presented rickettsial loads lower than those previously reported for other spotted fever group rickettsiae. The presence of Ehrlichia s

The predominant human-biting tick throughout the southeastern United States is Amblyomma americanum. Its ability to transmit pathogens causing Lyme disease-like illnesses is a subject of ongoing controversy. Results of previous testing by the Department of Defense Human Tick Test Kit Program and other laboratories indicated that it is highly unlikely that A. americanum transmits any pathogen that causes Lyme disease. In contrast, a recent publication by Clark and colleagues (K. L. Clark, B. Leydet, and S. Hartman, Int. J. Med. Sci. 10:915-931, 2013) reported detection of Lyme group Borrelia in

Arenaviridae are a family of single stranded RNA viruses of mammals and boid snakes. Twenty-nine distinct mammalian arenaviruses have been identified, many of which cause severe hemorrhagic disease in humans, particularly in parts of sub-Saharan Africa, and in Central and South America. Humans typically become infected with an arenavirus through contact with excreta from infected rodents. Tacaribe virus (TCRV) is an arenavirus that was first isolated from bats and mosquitoes during a rabies surveillance survey conducted in Trinidad from 1956 to 1958. Tacaribe virus is unusual because it has ne
