[Human parasites and amoeba: lower warm-water temperature is favourable to Legionella bacteria].

Our knowledge of Legionella bacteria has increased since their discovery in 1977. Legionnaires' disease is an underestimated diagnosis in Sweden. Most cases are community acquired and probably due to the presence of legionella bacteria in the water distribution systems of buildings, though the precise source of infection often remains unknown. Although a number of putative virulence factors have been identified, to date few of them have been shown to be significant. The replication of legionella in human macrophages is similar to that in protozoa. Their interaction with protozoa might explain

Emerging patents versus brain eating amoebae, Naegleria fowleri.

Primary Amoebic Meningoencephalitis (PAM) is a severe and often fatal infection caused by the free-living amoebae Naegleria fowleri. This condition typically results from exposure to contaminated warm freshwater/inadequately treated recreational water/or ablution/nasal irrigation with contaminated water. The management of PAM is hindered by the absence of effective treatment coupled with challenges in early diagnosis. This review explores emerging patents that could be utilized for the treatment, diagnosis of PAM, as well as water treatment. Recent patents from the past five years, along with

Nanovehicles in the improved treatment of infections due to brain-eating amoebae.

Pathogenic free-living amoebae are known to cause fatal central nervous system infections with extremely high mortality rates. High selectivity of the blood-brain barrier hampers delivery of drugs and untargeted delivery of drugs can cause severe side effects. Nanovehicles can be used for targeted drug delivery across the blood-brain barrier. Inorganic nanoparticles have been explored as carriers for various biomedical applications and can be modified with various ligands for efficient targeting and cell selectivity while lipid-based nanoparticles have been extensively used in the development

The Effect of Different Environmental Conditions on the Viability of Naegleria fowleri Amoebae.

Naegleria fowleri, a free-living amoeba found in soil and freshwater environments, is the causative agent of Primary Amoebic Meningoencephalitis. Infection occurs when amoebae enter the nasal cavity, attach to the nasal mucosa and travel along olfactory neurons towards the olfactory bulb. Upon reaching the central nervous system, the amoebae replicate very rapidly and can cause death in 3-10 days. Little is known about the conditions in which the amoeba can survive in the environment. We have tested conditions beyond the known boundaries on the viability of amoebae by introducing them into mod

Drug Target Discovery Methods In Targeting Neurotropic Parasitic Amoebae.

Neurotropic parasitic amoebal infections have imposed an enormous challenge to chemotherapy in patients who fall victims to the infections caused by them. Conventional antibiotics that are given to treat these infections have a low patient compliance because of the serious adverse effects that are associated with their use. Additionally, the growing incidence of the development of drug resistance by the neurotropic parasites like Naegleria fowleri, Balamuthia mandrillaris, and Acanthamoeba spp has made the drug therapy more challenging. Recent studies have reported some cellular targets in the

Influence of climate and geography on the occurrence of Legionella and amoebae in composting facilities.

BACKGROUND: The incidence of Legionnaires' disease (LD) in southern Switzerland is three times higher than in northern Switzerland. Climatic and geographic factors may be potential causes for this difference. We studied the prevalence of Legionella and free-living amoebae (FLA) in compost and bioaerosol in two Swiss regions to understand the role of climate and geography in the transmission of LD. We also tried to investigate whether or not compost storage duration would influence the composition of Legionella and FLA communities. RESULTS: A larger proportion of compost heaps in facilities fro

Fresh-water amoebae from four aquatic sites in Egypt.

Water samples from four aquatic sites in Egypt were examined for the presence of free-living amoebae. Amoebae were isolated and cultured on 1.5% non-nutrient agar with bacteria. They were studied morphologically and tested for production of flagellated forms, encystment, excystment, nuclear division, temperature tolerance and pathogenicity in young mice (8 - 10 gm). Didascalus thorntoni and Naegleria gruberi were dominant in all surface waters. Naegleria fowleri, Acanthamoeba glebae, A. culbertsoni and A. rhysodes were found in Lower Egypt, while Vahlkampfia avara and V. thornata were found in

Preliminary report on the pathogenicity of Legionella pneumophila for freshwater and soil amoebae.

Legionella pneumophila, the causative organism of Legionnaires' disease, is pathogenic for free living, ubiquitous, freshwater, and soil amoebae of the genera Acanthamoeba and Naegleria. Some species support the growth of strains from serogroups 1 to 6, others only strains from certain serogroups. Initial studies with seeded material indicate that amoebal enrichment could be utilised for the isolation of legionellae from clinical specimens and natural habitats. It is suggested that a vacuole, or amoeba, full of legionellae, rather than free legionellae, could be the infective particle for man.

Vannella russkaya sp. nov. (Amoebozoa, Discosea) - a new marine amoeba from a terrestrial habitat.

A new amoeba species, Vannella russkaya sp. nov., is described based on morphological (light and transmission electron microscopy) and molecular (SSU rRNA and Cox1) analyses. The strain was isolated from soil near a brackish stream, Solonik (Novgorod Region, Russia), and was cultivated under marine conditions. Phylogenetic analysis placed the new species within a clade comprising predominantly marine Vannella, with the closest affinity to V. salarenaria, which was also found in soil and cultivated under marine conditions. The studied strain exhibits a broad tolerance to salinity from 5‰ to 50‰

Drug susceptibility of a clinical isolate of Balamuthia mandrillaris, a pathogenic free-living amoeba.

Balamuthia amoebic encephalitis (BAE) is a highly fatal infection caused by Balamuthia mandrillaris, an amoeba that lives in soil and water. In Thailand, three fatal cases of BAE have been documented, but no survivors have been reported, raising questions about current treatment regimens. Previous drug repurposing studies reveal some potent pharmacological compounds, but the drug susceptibility of the clinical isolate of pathogenic amoeba remains variable. Given the success in isolating B. mandrillaris from the human biopsied brain, this study aims to assess the amoebicidal effect of several p

Fluvastatin and atorvastatin induce programmed cell death in the brain eating amoeba Naegleria fowleri.

Naegleria fowleri is the causative agent of a type of encephalitis called Primary Amoebic Encephalitis (PAM). Almost 98 % of PAM cases reported worldwide are fatal and affect mostly immunocompetent children and young adults. The current therapeutic option against PAM cases includes a combination of miltefosine, amphotericin B and other drugs which are unfortunately associated with severe side effects. In a recent study in our group, statins were tested in vitro against Naegleria fowleri trophozoites showing activity against these pathogens at low concentrations causing low toxicity. Consequent

Neuroleptic Drug Targets a Brain-Eating Amoeba: Effects of Promethazine on Neurotropic Acanthamoeba castellanii.

Acanthamoeba spp. has recently been reported to express diverse group of ion channels and receptors that are expressed by human cells which bind drugs that are used in noninfectious diseases. Bioinformatics computational tools, growth assays, and 3D structural modeling have enabled the discovery of primitive muscarinic receptors, voltage-gated calcium channels, and ion transport pumps such as Na-K ATPase in this protist pathogen. The significance of the reported receptors and ion channels in the biology of Acanthamoeba is yet to be determined. We selected promethazine, which is a known antagon

Cryptococcus neoformans Escape From Dictyostelium Amoeba by Both WASH-Mediated Constitutive Exocytosis and Vomocytosis.

Cryptococcus neoformans is an environmental yeast that can cause opportunistic infections in humans. As infecting animals does not form part of its normal life-cycle, it has been proposed that the virulence traits that allow cryptococci to resist immune cells were selected through interactions with environmental phagocytes such as amoebae. Here, we investigate the interactions between C. neoformans and the social amoeba Dictyostelium discoideum. We show that like macrophages, D. discoideum is unable to kill C. neoformans upon phagocytosis. Despite this, we find that the yeast pass through the

Evaluation of solvation free energies for small molecules with the AMOEBA polarizable force field.

The effects of electronic polarization in biomolecular interactions will differ depending on the local dielectric constant of the environment, such as in solvent, DNA, proteins, and membranes. Here the performance of the AMOEBA polarizable force field is evaluated under nonaqueous conditions by calculating the solvation free energies of small molecules in four common organic solvents. Results are compared with experimental data and equivalent simulations performed with the GAFF pairwise-additive force field. Although AMOEBA results give mean errors close to "chemical accuracy," GAFF performs s

Effects of antacid therapy in asymptomatic patients with presence of amoeba in stool report.

Amoebic infection caused by the protozoan parasite Entamoeba histolytica is a prevalent infection in the developing countries. Milder form of this infection is associated with loose stool, flatulence and borborygmi, may or may not be associated with pain in abdomen and treated symptomatically by some physicians by antacid. To find out the effects of antacid (sorbacid) therapy in patients with amoeba in stools by examining the changes in the stool report, a study was conducted among 25 patients enrolled in the study with complaints of "gas" in the abdomen with stool reports positive for amoeba.
