Function of orthologous fibroblast growth factor 11 protein in angiogenesis and immunomodulatory after spinal cord injury.

Intracellular fibroblast growth factors (iFGFs) are emerging as promising therapeutic agents in tissue engineering and regenerative medicine. Although FGF11, a typical iFGF, plays crucial roles in cellular hypoxia adaptation, its roles in angiogenesis and immunomodulation post-spinal cord injury (SCI) remain poorly characterized. This study investigated four FGF11 orthologs: ZFgf11a/ZFgf11b from zebrafish (Danio rerio) and MFGF11a/MFGF11b from mouse (Mus musculus). In vitro, intracellularly delivered ZFgf11a and MFGF11b exhibited stronger pro-angiogenic effects compared to ZFgf11b or MFGF11a.

Omentin-1 promotes wound healing in diabetic mice by improving vascular endothelial cell function.

Diabetes is a metabolic disease that affects global human health, with 25% of diabetic patients suffering from chronic non-healing ulcers. Omentin-1, also referred to as intelectin-1 and encoded by the itln1 gene, is a recently discovered adipokine that modulates inflammation, cellular activities, and vascular tone. Research indicates that omentin-1 provides protective benefits in several conditions, including atherosclerosis and diabetic retinopathy. Nevertheless, the therapeutic potential of omentin-1 in promoting diabetic wound healing is still not well understood. Our results revealed a si

3',4'-Dihydroxyflavonol Inhibits Fibrotic Response in a Rabbit Model of Glaucoma Filtration Surgery.

Post-operative fibrosis of the filtering bleb limits the success of glaucoma filtration surgery (GFS). To minimise subconjunctival scarring following GFS, treatment with antimetabolites such as Mitomycin C (MMC) has become standard practice; however, their use is associated with considerable side effects. This study aimed to investigate the anti-scarring properties of 3',4'-dihydroxyflavonol (DiOHF). GFS was performed in New Zealand white rabbits who received eye drops of DiOHF three times daily and vehicle eye drops after surgery (n = 5) or a single intraoperative treatment of MMC (n = 5). Bl

Punicalagin Promotes Peripheral Nerve Repair by Enhancing Mitophagy via the TLR4/MAPK Signaling Pathway and Facilitating Angiogenesis.

Peripheral nerve injury (PNI) often leads to functional impairment, and current therapeutic options are limited. Punicalagin (PUN), a polyphenolic compound derived from Punica granatum , exhibits antioxidant and anti-inflammatory properties. This study investigated the therapeutic potential of PUN for PNI and its underlying mechanisms, focusing on mitochondrial autophagy (mitophagy), TLR4/MAPK signaling, and angiogenesis. In vitro, Schwann cells were treated with PUN under oxidative stress induced by tert-butyl hydroperoxide to evaluate cell viability, mitochondrial function, mitophagy, reacti

Small-molecule and antibody approaches to molecular chemotherapy of primary brain tumors.

High-grade primary brain tumors remain refractory to conventional treatment approaches, including radiotherapy and cytotoxic chemotherapy. Molecular neuro-oncology has now begun to clarify the transformed phenotype of these malignant tumors and identify oncogenic pathways that might be amenable to small-molecule and antibody 'targeted' therapy. Growth factor signaling pathways are often upregulated in these tumors and contribute to oncogenesis through autocrine and paracrine mechanisms. Excessive growth factor receptor stimulation can also lead to overactivity of the downstream Ras signaling p

Phytochemical-driven angiogenesis and mucosal regeneration in gastric ulcer healing: molecular mechanisms, preclinical evidence, and translational perspectives.

Gastric ulcer remains a significant global gastrointestinal condition characterized by disruption of the gastric mucosal barrier, attributed to oxidative stress, inflammation, Helicobacter pylori infection, nonsteroidal anti-inflammatory drugs (aspirin, indomethacin, diclofenac), and compromised tissue repair. While traditional therapies effectively reduce acid secretion and eliminate H. pylori, they often do not actively promote mucosal regeneration and angiogenesis, both of which are crucial for the complete healing of ulcers and the prevention of recurrence. This review integrates molecular

Discovery of VEGFR2 inhibitors by integrating naïve Bayesian classification, molecular docking and drug screening approaches.

The high morbidity and mortality of cancer make it one of the leading causes of global death, thus it is an urgent need to develop effective drugs for cancer therapy. Vascular endothelial growth factor receptor-2 (VEGFR2) acts as a central modulator of angiogenesis, and is therefore an important pharmaceutical target for developing anti-angiogenic agents. In this study, ligand-based naïve Bayesian (NB) models and structure-based molecular docking were combined to develop a virtual screening (VS) pipeline for identifying potential VEGFR2 inhibitors from FDA-approved drugs. The best validated na

Metformin and oncogenic pathways: Crosstalk between energy sensing and tumor progression.

Metformin, a widely prescribed oral biguanide for type 2 diabetes mellitus, has gained significant attention as a potential anti-cancer agent. Beyond its established role in improving insulin sensitivity and lowering blood glucose, preclinical and epidemiological studies suggest that metformin exerts anti-tumor effects through both insulin-dependent and insulin-independent mechanisms. Central to its activity is the activation of AMP-activated protein kinase (AMPK), which disrupts the mammalian target of rapamycin (mTOR) pathway, leading to cell cycle arrest, apoptosis, and reduced tumor growth

Extracellular vesicles in pharmacology: Innovations in drug delivery and therapeutic applications in cancer.

Extracellular vesicles (EVs) are a diverse population of membrane nanoparticles secreted by nearly all cell types, playing a key role in intercellular communication by transferring bioactive macromolecular cargo. In cancer, EVs shape both the local tumour microenvironment and distant premetastatic niches. They are essential regulators of tumour initiation, progression, immune modulation, angiogenesis and metastatic dissemination. Due to their abundance in biofluids, EVs have attracted attention as diagnostic and prognostic biomarkers for early detection and assessment of therapeutic response.

Pros and Cons of the Cannabinoid System in Cancer: Focus on Hematological Malignancies.

The endocannabinoid system (ECS) is a composite cell-signaling system that allows endogenous cannabinoid ligands to control cell functions through the interaction with cannabinoid receptors. Modifications of the ECS might contribute to the pathogenesis of different diseases, including cancers. However, the use of these compounds as antitumor agents remains debatable. Pre-clinical experimental studies have shown that cannabinoids (CBs) might be effective for the treatment of hematological malignancies, such as leukemia and lymphoma. Specifically, CBs may activate programmed cell death mechanism

LRP5: A Multifaceted Co-Receptor in Development, Disease, and Therapeutic Target.

Low-density lipoprotein receptor-related protein 5 (LRP5) is a multifunctional transmembrane coreceptor that plays a pivotal role in development and disease. Wnt/β-catenin signaling is the primary downstream signaling pathway activated by LRP5. Furthermore, some LRP5 functions are mediated by noncanonical pathways, such as AKT/P21 and TGF-β/Smad signaling. Pathologically, both loss-of-function and gain-of-function mutations in LRP5 produce distinct phenotypes, ranging from osteoporosis-pseudoglioma syndrome to high bone mass disorders. Beyond the skeletal system, LRP5 has emerged as a key regu

Biologically based strategies for overcoming in vivo barriers with functional nano-delivery systems.

Nanomedicine has been developed to reduce or eliminate the side effects and toxicity upon systemic therapy of chemotherapeutic agents and to improve their therapeutic efficacy. However, the translation of non-sized or nano-encapsulated drugs is hampered by the low penetration and accumulation of engineered nanoparticles (NPs) in sites of tumors as well as their poor pharmacokinetics. This may be due to the synthetic structure of NPs and also complicated and unknown characteristics of the solid tumor microenvironment (TME). As a result, the TME is being better identified, and the interactions b
