In mammals, IFN-γ acts as a critical cytokine that triggers intracellular signaling by binding to specific IFNGR1 and IFNGR2 receptors. Unlike mammals, teleosts possess two members of type II IFN genes (IFN-γ and IFN-γrel), two IFNGR1 paralogs (CRFB13 and CRFB17) and a single IFNGR2 homolog (CRFB6). Despite the identification of type II IFN receptors in multiple fish species, the molecular features, expression profiles and signaling-associated function remain limited. In this study, three type II interferon receptor genes, AjCRFB6, AjCRFB13 and AjCRFB17 were identified in Japanese eel, Anguill

In this study, two stable cell lines were established from the kidney (AJK) and spleen (AJSP) tissues of Japanese eel (Anguilla japonica) using the tissue explant method, with the aim of supporting immunological research in this species. Following isolation, AJK and AJSP cells were cultured in DMEM/F-12 medium supplemented with 20 % fetal bovine serum (FBS) at 28 °C, with passaging every 3-4 days. After the fifth passage, the cells were maintained in DMEM/F-12 containing 15 % FBS for passage culture. Ribosomal RNA gene sequencing confirmed the origin of both cell lines as A. japonica. Karyotyp

Vibrio harveyi (Vh) and Vibrio vulnificus (Vv) are major bacterial pathogens affecting farmed eels, but their comparative virulence mechanisms remain poorly characterized. This study combined histopathology and transcriptomic profiling to investigate organ-specific damage and host responses in American eels (Anguilla rostrata, 20 g per fish, for a total of 60 fish) following experimental infection with LD50 doses of Vh (strain HA_1, 7.5 × 104 CFU/fish) and Vv (strain FJ_4, 5.0 × 105 CFU/fish). Tissue samples from liver, kidney, and spleen were collected at 0, 36, and 60 h post-infection (hpi).

Edwardsiellosis outbreaks cause significant losses in Japanese eel aquaculture. The causative agent, Edwardsiella tarda, is an intracellular pathogen, and the use of antibiotics has a limited effectiveness. As Japanese eels are sensitive to stress, injection vaccines are not recommended for treatment; immersion methods are less stressful, but not cost-effective. Alternatively, oral vaccination methods are more promising. The aim of this study was to develop a starch hydrogel-based oral (SHO) vaccine against edwardsiellosis in Japanese eel, using formalin-killed cells. To assess the protective

Dermocystidium is a pathogenic fungus-like protist that infects many fish species, causing lesions on the skin, gills and other tissues, which may result in systemic disease. In the present study, a new Dermocystidium anguillae infection in the skin and eyes of the koi carp (Cyprinus carpio) from Iran was described using a comprehensive analysis of morphological, histological and molecular data. Filiform hyphal-like cysts were visible on wet mounts. The cysts were filled with spherical to round spores. Wet mount preparations of fresh cyst contents of Dermocystidium anguillae revealed a massive

Aeromonas hydrophila is a commonly pathogenic bacterium in cultivated eels, but its pathogenicity to American eel (Anguilla rostrata) and the molecular mechanism of host anti-A. hydrophila infection remains uncertain. In this study, LD50 of A. hydrophila to American eels was determined and bacterial load in the liver and kidney of eels was assessed post 2.56 doses of LD50 of A. hydrophila infection. The results showed that the LD50 of A. hydrophila to American eels was determined to be 3.9 × 105 cfu/g body weight (7.8 × 106 cfu/fish), and the bacterial load peaked at 36 h post the infection (h

The evidence for androgens having a pivotal role in the functioning of the female reproductive axis--such as initiating puberty or vitellogenesis--is mounting. However, the use of aromatizable androgens and the tissue-specific focus of most studies often make it unclear if androgenic effects throughout the axis proceed via androgen or estrogen signalling mechanisms. In this study, we assessed the effects of 11-ketotestosterone (11KT, a non-aromatizable androgen) on the pituitary and ovary of previtellogenic (PV) freshwater eels Anguilla australis, comparing them with eels naturally undergoing

Anguillid herpesvirus 1 (AngHV-1) is a pathogen that causes hemorrhagic disease in various farmed and wild freshwater eel species, resulting in significant economic losses. Although AngHV-1 has been detected in the American eel (Anguilla rostrata), its pathogenicity has not been well characterized. In this study, an AngHV-1 isolate, tentatively named AngHV-1-FC, was isolated from diseased American eels with similar symptoms as those observed in AngHV-1-infected European eels and Japanese eels. AngHV-1-FC induced severe cytopathic effects in the European eel spleen cell line (EES), and numerous

Vibrio harveyi, a recently discovered pathogenic bacterium isolated from American eels (Anguilla rostrata), poses uncertainties regarding its pathogenesis in American eel and the molecular mechanisms underlying host defense against V. harveyi infection. This study aimed to determine the LD50 of V. harveyi in American eel and assess the bacterial load in the liver, spleen, and kidney post-infection with the LD50 dose. The results showed that the LD50 of V. harveyi via intraperitoneal injection in American eels over a 14d period was determined to be 1.24 × 103 cfu/g body weight (6.2 × 104 cfu/fi

Anguillid herpesvirus 1 (AngHV), the causative agent of "mucus sloughing and hemorrhagic septicemia disease", causes serious infectious diseases in farmed eel. Among the effective prevention and control strategies, vaccination is one of the most effective approaches. However, no vaccine for AngHV is available. Our study developed a formalin-inactivated AngHV vaccine and evaluated its performance in American eels. Initially, AngHV-FJ, a strain of AngHV, was inactivated completely by 0.1 % formaldehyde, mixed with adjuvant Montanide ISA 763 A VG (763A). Then, vaccines containing different amount

Vibrio harveyi is a highly pathogenic bacterium causing severe septicemia in American eel (Anguilla rostrata), yet the regulatory roles of long non-coding RNAs (lncRNAs) in the host immune response remain largely unexplored. In this study, American eels were challenged with V. harveyi, and kidney tissues were collected at 0-, 12-, 24-, 36-, 48 and 60-h post-infection (hpi) for bacterial load detection. Histopathological lesions and transcriptomic profiles (strand-specific RNA sequencing) were systematically analyzed at 0-, 24-, 36 and 48 hpi. Co-expression and co-location networks were constru

Two areas that have received substantial attention in investigations of muscle activity during fish swimming are (1) patterns of fiber type recruitment with swimming speed and (2) the timing of muscle activation in relation to muscle strain. Currently, very little is known about either of these areas in eels, which represent an extreme body form among fishes and utilize a mode of locomotion found at one end of the undulatory spectrum (anguilliform locomotion). To assess how this swimming mode and body form influence the neuromuscular control of swimming, I recorded electromyographic data from

Understanding the feeding habits of predatory fish is essential for unravelling food web structures and implementing conservation strategies in riverine ecosystems. However, conventional lethal stomach content analysis methods are not necessarily appropriate for long-term dietary studies, particularly for threatened species, as they require large sample sizes due to the inability to repeatedly analyse stomach contents from the same individuals. This study aimed to develop and validate a non-lethal stomach content analysis method using tubes for the giant mottled eel Anguilla marmorata, a repre

Dopamine plays a crucial role in controlling reproduction in eels, and its action is mediated through D2-type dopamine receptors. D2A and D2B receptors in the Japanese eel Anguilla japonica were cloned and characterized in the present study. Attention (daily expression patterns in the brain and endogenous regulation) was paid to D2B receptor because it is considered to play a crucial role in eel reproduction. The cDNAs of D2A and D2B receptors had open reading frames comprising 456 and 454 amino acid residues, respectively, which were phylogenetically clustered with those of other teleost spec

The Japanese eel (Anguilla japonica) is a commercially valuable catadromous species widely cultured in East Asia. However, large-scale artificial propagation remains challenging. Activin B, a dimeric protein belonging to the transforming growth factor-β (TGF-β) superfamily, plays key roles in vertebrate gonadal development, yet its function in eel ovarian development has not been characterized. In this study, we cloned the inhbb gene from Japanese eel, expressed its recombinant Activin B protein, and evaluated its effects on ovarian development under artificial induction conditions. Adult fema

This study was conducted to record the histological features of the gastrointestinal tract of wild Indonesian shortfin eel, Anguilla bicolor bicolor (McClelland, 1844), captured in Peninsular Malaysia. The gastrointestinal tract was segmented into the oesophagus, stomach, and intestine. Then, the oesophagus was divided into five (first to fifth), the stomach into two (cardiac and pyloric), and the intestine into four segments (anterior, intermediate, posterior, and rectum) for histological examinations. The stomach had significantly taller villi and thicker inner circular muscles compared to t

The Japanese eel (Anguilla japonica) is a major aquaculture species in South Korea. However, a limited number of officially approved antimicrobial agents have led to frequent off-label drug use, raising concerns about therapeutic efficacy, food safety, and regulatory compliance. Flumequine (FQ), a first-generation quinolone effective against gram-negative pathogens, has been used in eels without species-specific pharmacokinetic or residue data. This study evaluated the pharmacokinetics, therapeutic efficacy, and residue depletion of FQ in the Japanese eel. The oral administration of 20 mg/kg s
