Anhedonia in Semantic Dementia-Exploring Right Hemispheric Contributions to the Loss of Pleasure.

Semantic dementia (SD) is a younger-onset neurodegenerative disease characterised by progressive deterioration of the semantic knowledge base in the context of predominantly left-lateralised anterior temporal lobe (ATL) atrophy. Mounting evidence indicates the emergence of florid socioemotional changes in SD as atrophy encroaches into right temporal regions. How lateralisation of temporal lobe pathology impacts the hedonic experience in SD remains largely unknown yet has important implications for understanding socioemotional and functional impairments in this syndrome. Here, we explored how l

Bacopa monnieri as augmentation therapy in the treatment of anhedonia, preclinical and clinical evaluation.

Bacopa monnieri (L.) is widely used in Ayurvedic medicine as a neural tonic for improving intelligence and memory. Several studies highlighted its efficacy in neuropsychiatric diseases but there is no evidence regarding anhedonia. Aim of the present work was to preclinically and clinically test against anhedonia a standardized B. monnieri extract (20% bacosides). In a mouse model of a depressive-like syndrome induced by lipopolysaccharide (LPS), the daily administration of the extract (50-200 mg kg-1 , p.o.) for 1 week, dose-dependently counteracted the immobility time in Porsolt and Tail susp

Anhedonia to Gentle Touch in Fibromyalgia: Normal Sensory Processing but Abnormal Evaluation.

Social touch is important for interpersonal interaction. Gentle touch and slow brushing are typically perceived as pleasant, the degree of pleasantness is linked to the activity of the C-tactile (CT) fibers, a class of unmyelinated nerves in the skin. The inability to experience pleasure in general is called anhedonia, a common phenomenon in the chronic pain condition fibromyalgia. Here, we studied the perception and cortical processing of gentle touch in a well-characterized cohort of fibromyalgia. Patients and controls participated in functional brain imaging while receiving tactile stimuli

Structural connectivity and risk for anhedonia after trauma: A prospective study and replication.

Anhedonia emerges in some people after psychological trauma, reflected by a loss of interest, diminished affect, and detachment. Structural abnormalities in specific neural pathways at the time of trauma may influence the development of these posttraumatic anhedonia (PTA) symptoms. In this prospective study, we determined whether white matter connectivity at around one month post-trauma predicts PTA and other PTSD symptoms at six months post-trauma. Thirty men and women aged 19-62 were recruited from the emergency department of a Level I trauma center. Participants received diffusion tensor im

Genes associated with anhedonia: a new analysis in a large clinical trial (GENDEP).

A key feature of major depressive disorder (MDD) is anhedonia, which is a predictor of response to antidepressant treatment. In order to shed light on its genetic underpinnings, we conducted a genome-wide association study (GWAS) followed by investigation of biological pathway enrichment using an anhedonia dimension for 759 patients with MDD in the GENDEP study. The GWAS identified 18 SNPs associated at genome-wide significance with the top one being an intronic SNP (rs9392549) in PRPF4B (pre-mRNA processing factor 4B) located on chromosome 6 (P = 2.07 × 10-9) while gene-set enrichment analysi

Stress-induced anhedonia is associated with an increase in Alzheimer's disease-related markers.

BACKGROUND AND PURPOSE: Stress is believed to be associated with the development of neuropsychiatric disorders, including Alzheimer's disease (AD). We have studied mechanisms implicated in vulnerability to stress and the relationship with changes in AD-related markers. EXPERIMENTAL APPROACH: Anhedonia induced by a chronic mild stress (CMS) procedure, applied for 6 weeks, was used to select rats vulnerable or resistant to stress. Sucrose intake, the Porsolt forced swimming test and cognitive deficits in the novel object recognition test (NORT) were used to characterize vulnerable and resilient

Anhedonia and amotivation in psychiatric outpatients with fully remitted stimulant use disorder.

This study evaluated whether psychiatric outpatients with a past stimulant use disorder in full remission for >/= 2 months (STIM+, n = 204) and those with no history of stimulant use disorder (STIM-, n = 2070) differed in the prevalence of current anhedonia and amotivation. Results showed that a significantly greater proportion of STIM+ participants reported anhedonia and amotivation than STIM- participants. The relation between stimulant use disorder history and anhedonia remained robust after controlling for other relevant clinical and demographic factors. These findings suggest that anhedon

Depression, anhedonia and anxiety in temporomandibular joint and other facial pain syndromes.

Depression, anhedonia, state anxiety (A-state), trait anxiety (A-trait), and self-reported pain estimate were measured in almost 500 facial pain patients. These patients were divided into 3 diagnostic categories: myofacial pain dysfunction syndrome (MPD) [18], arthritis of the temporomandibular joints (TMJ arthritis), and trigeminal neuralgia. Three control groups were measured for comparison. They consisted of an normal, or non-patient group, a group of arthritis patients, and a group of movement disorder patients attending a neurology clinic. Among the facial pain patients and the normal con

Subregional amygdala functional connectivity abnormalities and anhedonia impairments in first-episode schizophrenia.

BACKGROUND: Although abnormal amygdala functional connectivity (FC) has been reported in schizophrenia, most studies have treated the amygdala as a single structure. This study aimed to explore the association between amygdala subregional FC and anhedonia in patients with first-episode schizophrenia (FES). METHODS: Resting-state functional magnetic resonance imaging (fMRI) was conducted in 31 FES patients (including 11 drug-naïve) and 33 matched healthy controls (HCs). Clinical symptoms were assessed using the Positive and Negative Syndrome Scale (PANSS), and anhedonia was evaluated via the S

Diminished anticipatory pleasure predicts anhedonia in women with endometrisois: a case-control study.

BACKGROUND: Anhedonia, a diminished response to pleasurable stimuli extensively studied in mental disorders and chronic pain, has been reported as prevalent among patients with deep endometriosis. Anhedonia is considered a multifaceted transdiagnostic construct involving cognitive, affective, and behavioral processes related to the anticipation and experience of pleasure. In this study, we aim to expand on previous findings by exploring the presence of anhedonia in patients with ultrasound diagnosed (UD) endometriosis compared to a control group, and by examining how its anticipatory and consu

Cocaine use disorder: anhedonia and emotional flattening in an emotional picture rating task.

OBJECTIVES: Anhedonia, the reduced capacity to experience or pursue pleasure, is common among individuals with cocaine use disorder (CUD). This blunted emotional experience may extend beyond diminished positive affect to include reduced sensitivity to negative emotions. This narrowing of the emotional spectrum, if also present in individuals with CUD, may offer a target for treatment strategies. We hypothesized that individuals with greater anhedonia would rate images more neutrally and that this emotional flattening would extend to physiological responses, including heart rate reactivity and

Associations between polygenic risk for anhedonia and functional brain activity during reward processing.

Anhedonia is an important symptom in major mental disorders and is difficult to address due to a limited understanding of its neurobiological and genetic underpinnings. Here, we investigated the effect of a polygenic risk score (PRS) for anhedonia on brain activation during a well-established monetary incentive delay (MID) task, while performing functional magnetic resonance imaging. We derived an anhedonia PRS from a large genome wide analysis (n = 375,275) in our sample of 517 subjects. This sample consisted of four subgroups (healthy controls (n = 373), major depressive disorder (n = 57), s

Natural language processing to identify suicidal ideation and anhedonia in major depressive disorder.

BACKGROUND: Anhedonia and suicidal ideation are symptoms of major depressive disorder (MDD) that are not regularly captured in structured scales but may be captured in unstructured clinical notes. Natural language processing (NLP) techniques may be used to extract longitudinal data on suicidal behaviors and anhedonia within unstructured clinical notes. This study assessed the accuracy of using NLP techniques on electronic health records (EHRs) to identify these symptoms among patients with MDD. METHODS: EHR-derived, de-identified data were used from the NeuroBlu Database (version 23R1), a lon

Abnormal Structure-Function Coupling in Major Depressive Disorder Patients With and Without Anhedonia.

Background: As a core symptom of major depressive disorder (MDD), previous magnetic resonance studies have demonstrated that MDD with anhedonia may exhibit distinctive brain structural and functional alterations. Nevertheless, the impact of anhedonia on synchronized alterations in the structure and function of brain regions in MDD remains uncertain. Methods: A total of 92 individuals were enrolled in the study, including 29 MDD patients with anhedonia, 33 MDD patients without anhedonia, and 30 healthy controls (HCs). All subjects underwent structural and resting-state functional magnetic reson

Behavioral Phenotyping of Dopamine Transporter Knockout Rats: Compulsive Traits, Motor Stereotypies, and Anhedonia.

Alterations in dopamine neurotransmission are generally associated with diseases such as attention-deficit/hyperactivity disorder (ADHD) and obsessive-compulsive disorder (OCD). Such diseases typically feature poor decision making and lack of control on executive functions and have been studied through the years using many animal models. Dopamine transporter (DAT) knockout (KO) and heterozygous (HET) mice, in particular, have been widely used to study ADHD. Recently, a strain of DAT KO rats has been developed (1). Here, we provide a phenotypic characterization of reward sensitivity and compuls

Contrasting contributions of anhedonia to obsessive-compulsive, hoarding, and post-traumatic stress disorders.

Anhedonia is a transdiagnostic construct that can occur independent of other symptoms of depression; its role in neuropsychiatric disorders that are not primarily affective, such as obsessive compulsive disorder (OCD), hoarding disorder (HD), and post-traumatic stress disorder (PTSD) has received limited attention. This paper addresses this gap. First, the data revealed a positive contribution of anhedonia, beyond the effects of general depression, to symptom severity in OCD but not in HD or PTSD. Second, anhedonia was operationalized as a reduced sensitivity to rewards, which allowed employin

Early-life adversity facilitates acquisition of cocaine self-administration and induces persistent anhedonia.

Early-life adversity increases the risk for emotional disorders such as depression and schizophrenia. Anhedonia, thought to be a core feature of these disorders, is provoked by our naturalistic rodent model of childhood adversity (i.e., rearing pups for one week in cages with limited bedding and nesting, LBN). Drug use and addiction are highly comorbid with psychiatric disorders featuring anhedonia, yet effects of LBN on drug-seeking behavior and the reward and stress-related circuits that underlie it remain unknown. Here we examined the effects of LBN on cocaine intake and seeking, using a ba

High-Fat Diet Induced Anxiety and Anhedonia: Impact on Brain Homeostasis and Inflammation.

Depression and type 2 diabetes (T2D) are highly comorbid disorders that carry a large public health burden. However, there is a clear lack of knowledge of the neural pathological pathways underlying these illnesses. The present study aims to elucidate the molecular mechanisms by which a diet rich in fat can cause multiple complications in the brain, thereby affecting intracellular signaling and gene expression that underlie anxiety and depressive behaviors. The results show that a high-fat diet (HFD; ~16 weeks) causes anxiety and anhedonic behaviors. Importantly, the results also show that 4 m

Neural correlates of change in major depressive disorder anhedonia following open-label ketamine.

Anhedonia is a cardinal symptom of major depression and is often refractory to standard treatment, yet no approved medication for this specific symptom exists. In this exploratory re-analysis, we assessed whether administration of rapid-acting antidepressant ketamine was associated specifically with reduced anhedonia in medication-free treatment-refractory patients with major depressive disorder in an open-label investigation. Additionally, participants received either oral riluzole or placebo daily beginning 4 hours post-infusion. A subgroup of patients underwent fluorodeoxyglucose positron e
