Third-stage juveniles (L3) of Anisakis simplex, surgically implanted into the abdominal cavity of CBA/J mice and necropsied at 7, 14, or 21 days postinfection (PI), embedded in the gut mesentery and only rarely invaded viscera. Histologically, intense aggregations of neutrophils adjacent to the parasites were noted at Day 7 PI. At Day 14 PI, mature granulomata consisting mostly of eosinophils and large numbers of fibroblasts and associated collagen were observed. Granulocytes and occasionally multinucleate giant cells occupied the host-parasite interface. At 21 day PI, lesions displayed the pr

PURPOSE: This study aimed to investigate whether proteins present in the molting membranes of third-stage (L3) Anisakis larvae could serve as potential risk factors for allergic reactions.
METHODS: Third-stage larvae (L3) of Anisakis spp. were primarily collected from mackerels and cultured in vitro to yield both molting membranes and fourth-stage (L4) larvae. Major soluble proteins in the molting membranes were identified using SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis). Crude antigens extracted from L3, L4, and the molting membranes were subsequently evaluated by w

OBJECTIVE: Anisakis simplex is a fish-borne zoonotic parasite. Although this parasite has been known since the 19th century, publications on anisakiasis have increased in recent years. Despite this, this subject has yet to be well studied by bibliometric analysis. This study was conducted to show the research points and trends of A. simplex.
METHODS: The Web of Science Core Collection (WoSCC) was mined for articles on A. simplex. The VOSviewer software visually evaluated countries, institutions, authors, references, and keywords in this field.
RESULTS: A total of 1362 publications were inclu

BACKGROUND: So far, the frequency of Anisakis simplex-specific IgE antibodies has been determined by skin prick tests (SPTs) and the ImmunoCAP system. These commercial methods have good sensitivity, but their specificity is poor because they use complete parasite extracts. Our aim was to determine the frequency of sensitization to A. simplex using recombinant Ani s 1, Ani s 3, Ani s 5, Ani s 9 and Ani s 10 and to evaluate these allergens for diagnosis, comparing their performance with the commercial methods.
PATIENTS AND METHODS: We conducted a descriptive, cross-sectional validation study pe

BACKGROUND: Laboratory diagnosis of anisakidosis is based on specific serum IgE detection. Recently, detection of allergen-induced basophil activation by flow cytometry has been proposed as a valuable tool for allergy diagnosis.
OBJECTIVE: To evaluate if detection of Anisakis-induced basophil activation by flow cytometry is a useful tool in the diagnosis of Anisakis allergy.
METHODS: Patients with Anisakis allergy (A.s.+, n = 37), patients reporting chronic urticaria or abdominal pain unrelated to fish ingestion (A.s.-, n = 51), and healthy controls (n = 12) were studied. Specific IgE to Ani

Anisakis simplex is a seafish nematode, which is responsible for the well-known human infection (anisakiasis) and can induce IgE-mediated reactions. IgE sensitization to Anisakis simplex can be frequent in particular countries and should be suspected in patients with acute allergic symptoms after ingestion of fish. The etiological role of Anisakis simplex was evaluated in 49 adult subjects with acute allergic symptoms after ingestion of anchovies. Serum-specific IgE and prick tests to anchovy were negative in each patient. Specific IgE reactions to Anisakis were positive in 45 patients and ski

The diagnosis of allergic reactions to Anisakis remains challenging due to the lack of specific allergens available for routine clinical use. However, the latest version of the multiplex macroarray ALEX-2 now allows the detection of specific IgE against Ani s 1, the major species-specific allergen, as well as Ani s 3 (tropomyosin), a highly cross-reactive molecule with homologs in other allergenic sources. This study aimed to evaluate the potential role of ALEX-2 in diagnosing Anisakis sensitization by comparing it with a previously validated diagnostic algorithm. Serum samples from patients w

BACKGROUND: An abundance of vitamins, minerals, and vital amino acids can be found in fish. The parasitic zoonotic disease anisakiasis is caused by ingesting the third larval stage of Anisakis spp., which is carried by nematodes called anisakids. Serious health problems, such as abdominal pain, nausea, vomiting, and allergic reactions that could approach anaphylactic level, are brought to the human body by ingesting undercooked fish, where this parasite is present.
AIM: Raw herrings, smoked herrings, raw sardines, salted sardines, raw mackerel, and raw saurus were all tested for the presence

Patient aged 71 with a history of type 2 diabetes mellitus. He came to the emergency department for abdominal pain and vomiting. Laboratory tests showed an increase in acute phase reactants. Abdominal CT scan showed dilated jejunal loops, compatible with intestinal occlusion. Urgent intervention was performed, resecting the affected segment. The pathology report showed a prominent transmural inflammatory infiltrate and interstitial oedema, with moderate villous atrophy, identifying parasitic structures compatible with anisakis larvae (family Anisakidae). Given the mechanism of tissue invasion,

BACKGROUND: Allergy to Anisakis simplex (s.) is spreading due to the increased consumption of raw, smoked or marinated fish. In man, Anisakis s. can directly attack the gastrointestinal mucosa, provoking a parasitosis known as anisakiasis, or giving rise to the formation of IgE and, finally, inducing IgE-mediated reactions like urticaria, angioedema and anaphylactic shock. During recent years, a dietary approach to Anisakis s. infestation has also been addressed.
METHODS: A total of 620 patients with urticaria, angioedema, or both and a history of anaphylaxis following consumption of raw, smo

BACKGROUND: Anisakis simplex can elicit allergic reactions when ingested in raw or marinated fish. The prevalence of A simplex hypersensitivity and allergy in Sicily (Italy), an area where the consumption of raw or marinated fish is very common, has not been investigated thus far.
OBJECTIVE: To investigate the prevalence of A simplex sensitization and its clinical relevance in a large group of unselected patients.
METHODS: All consecutive patients referred to the authors' allergy clinic during a 22 month-period were included in the study, evaluated for sensitization to A simplex and other al

Gastrointestinal anisakidosis is a nematode infection caused by the ingestion of larvae-infected raw or undercooked fish. The Japanese like to eat raw or undercooked fish, so gastric anisakiasis is a common disease in Japan. However, reports of anisakiasis with gastrointestinal cancer are rare. A 63-year-old Japanese male was diagnosed with a small early gastric cancerous lesion associated with gastric anisakiasis. From our experience and based on a review of the literature, the attachment of an anisakis larva to early gastric cancer is not considered accidental.

Nematodes of the genera Anisakis and Pseudoterranova (family Anisakidae) are zoonotic parasites for which marine mammals (e.g., whales, dolphins, porpoises, seals, sea lions, walruses) act as final hosts, and crustaceans, cephalopods and fish as intermediate and/or paratenic hosts. In humans, the ingestion of Anisakidae larvae can result in infection with live larvae, an allergic reaction to Anisakidae allergens (even when dead larvae are ingested), or both. Worldwide, more than 2000 infections are diagnosed in humans every year, yet most of the infections and allergic reactions are undiagnose

A gene coding for a 24 kDa protein (22 U homologous; As22U) was isolated from the Anisakis simplex third-stage larvae cDNA library during expressed sequence tag analysis. As22U was 636 bp long, and was found to code for 212 amino acid residues with a calculated mass of 23.5 kDa and a PI of 9.06. The As22U deduced amino acid sequence harbored a signal peptide region and 16 highly conserved cysteine residues, and it was identified in both the total extracts and excretory secretory (ES) protein of A. simplex. Its molecular weight was measured at 24 kDa via western blot analysis. The expression le

Anisakis simplex has been recognized as an important cause of disease in man and as a foodborne allergen source. Actually, this food-borne was recently identified as an emerging food safety risk including allergenic symptoms. This parasite contains a large variety of allergenic proteins enforcing the necessity to detect new allergens. Commonly, these efforts have been focused on the developing of cDNA libraries, where virtually all expressed mRNAs are present, by using immunoreactive patient serum or polyclonal antibodies. Phage display system is an alternative strategy which permits the physi

To facilitate improved diagnosis and detection of the third stage larva (L3) of Anisakis pegreffii from the Minnan-Taiwan bank fishing ground in Taiwan Strait, a real-time PCR method for the detection in situ and differentiation was developed to amplify a region of the second internal transcribed spacer (ITS-2) of this parasite. The real-time PCR assay was capable of detecting 1/3 of a single L3 in 30 mg of marine fish tissue, and also exhibited a high level of specificity for A. pegreffii, no fluorescence signals were observed in other five major larval anisakid species found in commercial ma

Anisakis spp. L3 larvae commonly found in marine fish species are an important biological hazard when raw or inadequately thermally processed fishery products are consumed with live larvae, highlighting the need for effective control strategies. This study evaluated the efficacy and mechanism of photodynamic inactivation (PDI) of Anisakis L3 larvae, using the cationic photosensitizer (PS) 5,10,15,20-tetrakis(1-methylpyridinium-4-yl)porphyrin tetraiodide (TMPyP). Larvae were collected from Atlantic horse mackerel and grouped by size: batch A (larvae ≈ 1.5 cm) and batch B (larvae ≈ 2.0 cm). Inac
