Role of microRNAs in cardiac hypertrophy and heart failure.

MicroRNAs (miRNAs) are a class of endogenous, highly conserved, small noncoding RNAs that regulate gene expression post-transcriptionally. Recent studies have demonstrated that miRNAs are aberrantly expressed in the cardiovascular system. The implications of miRNAs in cardiovascular disease have recently been recognized, representing the most rapidly evolving research field. Gain- and loss-of-function studies in mice models have identified distinct roles for specific miRNAs during cardiac hypertrophy, heart failing, and myocardial infarction. In the present article, the currently relevant find

MicroRNA-mediated regulation of inflammatory gene expression in asthma.

Asthma is a long-term inflammatory condition of the airways that includes complex immune dysregulation, epithelial barrier issues, and structural remodelling. The critical function of the miRNAs as modulators of inflammatory gene expression in asthma is highlighted by recent studies. It regulates immune cell development, cytokine production, and important signalling pathways like MAPK, JAK/STAT, and NF-κB. The let-7 family, miR-21, miR-155, and miR-126aresignificantin Th2-driven eosinophilic asthma and inflammation caused by neutrophils. Smooth muscle cell growth and epithelial cell transforma

Expanding Therapeutic Horizons in Parkinson's Disease Through MicroRNA-Based Interventions: A Comprehensive Review.

Parkinson's disease (PD) is a neurodegenerative disorder with rapidly increasing prevalence, characterized by dopaminergic neuronal loss and α-synuclein aggregation. Current therapies provide only symptomatic benefit and do not alter the disease course. MicroRNAs (miRNAs), small noncoding RNAs that regulate gene expression, are increasingly recognized as key modulators of PD pathogenesis, influencing oxidative stress, mitochondrial dysfunction, neuroinflammation, and proteostasis. Therapeutic modulation of miRNAs in PD has centered on two complementary approaches: miRNA inhibitors (antagomiRs)

MiRNA as a Potential Target for Multiple Myeloma Therapy-Current Knowledge and Perspectives.

Multiple myeloma (MM) is the second most common hematological malignancy. Despite the huge therapeutic progress thanks to the introduction of novel therapies, MM remains an incurable disease. Extensive research is currently ongoing to find new options. MicroRNAs (miRNAs) are small, non-coding RNA molecules that regulate gene expression at a post-transcriptional level. Aberrant expression of miRNAs in MM is common. Depending on their role in MM development, miRNAs have been reported as oncogenes and tumor suppressors. It was demonstrated that specific miRNA alterations using miRNA mimics or ant

MicroRNAs in the diagnosis and prevention of drug-induced cardiotoxicity.

Drug-induced cardiotoxicity is a serious problem associated with the administration of many drugs. MicroRNAs (miRNAs) have been reported to be affected by drugs and other xenobiotics, and the potential of miRNAs as biomarkers and diagnostic tools has been considered. In recent years, an association of certain miRNAs with the cardiotoxicity of some drugs, namely anthracyclines, bevacizumab, cyclosporine A and isoprenaline, has already been found. This review article summarizes available information about the changes in miRNA levels induced by cardiotoxic drugs. Three aspects are discussed: the

MicroRNAs in cardiovascular disease.

PURPOSE OF REVIEW: Noncoding RNAs regulate many aspects of cardiovascular biology and are potential therapy targets. In this review, we summarize and highlight current discoveries in the field of microRNAs, a class of noncoding RNAs. RECENT FINDINGS: miRNAs regulate posttranscriptional gene expression and have been shown to control cardiac development, hypertrophy, fibrosis, and regeneration. Of note are the miRNAs that regulate cardiac contractility (for example, miR-25 and miR-22), cardiac regeneration (like miR-302-367 and miR99/100 families), and fibrosis (as miR-125b). Consistently with

MiR-21-5p promotes RPE cell necroptosis by targeting Peli1 in a rat model of AMD.

Nonexudative age-related macular degeneration (dry AMD) is characterized by the progressive degeneration of retinal pigment epithelial (RPE) cells and photoreceptors, resulting in central vision loss. The disease is primarily marked by the accumulation of drusen and RPE atrophy. Given the emerging role of miR-21-5p in various ocular diseases, including diabetic retinopathy, glaucoma, pterygium, and choroidal neovascularization, we hypothesized that miR-21-5p may also impact RPE cell integrity in AMD. To test this hypothesis, we employed a rat model of dry AMD induced by sodium iodate (NaIO3) a

Metabolic and energetic benefits of microRNA-22 inhibition.

INTRODUCTION: We previously demonstrated in primary cultures of human subcutaneous adipocytes and in a mouse model of diet-induced obesity that specific microRNA-22-3p antagomirs produce a significant reduction of fat mass and an improvement of several metabolic parameters. These effects are related to the activation of target genes such as KDM3A, KDM6B, PPARA, PPARGC1B and SIRT1 involved in lipid catabolism, thermogenesis, insulin sensitivity and glucose homeostasis. RESEARCH DESIGN AND METHODS: We now report a dedicated study exploring over the course of 3 months the metabolic and energetic

IL-8 induces miR-424-5p expression and modulates SOCS2/STAT5 signaling pathway in oral squamous cell carcinoma.

Suppressor of cytokine signaling (SOCS) proteins are negative feedback regulators of the Janus kinase/signal transducer and activator of transcription (JAK/STAT) pathway. Dysregulation of SOCS protein expression in cancers can be one of the mechanisms that maintain STAT activation, but this mechanism is still poorly understood in oral squamous cell carcinoma (OSCC). Here, we report that SOCS2 protein is significantly downregulated in OSCC patients and its levels are inversely correlated with miR-424-5p expression. We identified the SOCS2 protein, which modulates STAT5 activity, as a direct tar

Down-regulation of miR-133a contributes to up-regulation of Rhoa in bronchial smooth muscle cells.

RATIONALE: Augmented bronchial smooth muscle (BSM) contraction is one of the causes of bronchial hyperresponsiveness. The protein RhoA and its downstream pathways have now been proposed as a new target for asthma therapy. MicroRNAs (miRNAs) play important roles in normal and diseased cell functions, and a contribution of miR-133 to RhoA expression has been suggested in cardiomyocytes. OBJECTIVES: To make clear the mechanism(s) of up-regulation of RhoA observed in the BSMs of experimental asthma, the role of miR-133a in RhoA expression was tested. METHODS: Total proteins and RNAs (containing
