Epithelial antimicrobial peptides: review and significance for oral applications.

Epithelial tissues provide the first line of defense between an organism and the environment. Disruption of this barrier leads to bacterial invasion and subsequent inflammation. This is precisely the situation existing in the human oral cavity, where tissues are constantly exposed to a variety of microbial challenges that can lead to bacterially induced periodontal diseases, and to infections of the oral mucosa by bacteria, fungi, and viruses. With the recent discoveries of host-derived peptide antibiotics in mammalian mucosal epithelium, a new line of investigation is emerging to test the hyp

Precision Antimicrobial Therapy Against Fusobacterium nucleatum Using Bioengineered Probiotics Expressing Guided Antimicrobial Peptides (gAMPs).

Colorectal cancer (CRC) is a leading cause of cancer-related mortality, with Fusobacterium nucleatum (F. nucleatum) identified as a key contributor to its progression. This study explores a novel therapy that targets this pathogen by using a bioengineered probiotic that expresses guided antimicrobial peptides (gAMPs) to selectively inhibit F. nucleatum. Lactococcus lactis MG1363 was engineered to express gAMPs derived from Ovispirin and Cathelin-related peptide SCF, linked to a Statherin-derived guide peptide that binds specifically to the F. nucleatum membrane porin FomA. The bacteria express

Novel formulation of antimicrobial peptides enhances antimicrobial activity against methicillin-resistant Staphylococcus aureus (MRSA).

Antimicrobial peptides (AMPs) have the ability to penetrate as well as transport cargo across bacterial cell membranes, and they have been labeled as exceptional candidates to function in drug delivery. The aim of this study was to investigate the effectiveness of novel formulation of AMPs for enhanced MRSA activity. The strategy was carried out through the formulation of liposomes by thin-layer film hydration methodology, containing phosphatidylcholine, cholesterol, oleic acid, the novel AMP, as well as vancomycin (VCM). Characterization of the AMPs and liposomes included HPLC and LCMS for pe

Development of Broad-Spectrum Antimicrobial Peptides through the Conjugation of FtsZ-Binding and Cell-Penetrating Peptides.

The rapid increase in bacterial resistance to conventional antibiotics has led to a great demand for novel antibacterial agents. Antimicrobial peptides (AMPs) are emerging as next-generation antimicrobial alternative drugs to conventional antibiotics because of their broad-spectrum antimicrobial activities and minimal potential for drug resistance induction. This work describes novel antimicrobial peptides (FtsZpcpp) synthesized through the conjugation of a cell penetration peptide ((RXR)4XB) to nonantimicrobial peptides (FtsZp). The FtsZp peptides were previously identified to bind FtsZ (flam

Imperfectly amphipathic design of α-helical antimicrobial peptides demonstrating potent antimicrobial activity with low toxicity.

Antimicrobial peptides (AMPs) are regarded as the most promising candidates for next-generation antimicrobial agents, with amphipathicity serving as a critical factor in de novo design or systematic optimization. Previous studies rarely investigated into imperfect amphipathicity scenarios. Therefore, we developed a series of peptides exhibiting diverse imperfect amphipathicity derived from the hybrid peptide P18 to explore the effect of different imperfect amphipathicity on peptides by evaluating their antimicrobial activity and toxicity. We found that imperfectly amphipathic peptides exhibite

Antimicrobial Peptide Signaling in Skin Diseases.

Antimicrobial peptides (AMPs) are important innate immune molecules at microbe-host interfaces. The biophysical properties of AMPs that facilitate direct killing of microbes have been extensively reviewed. In this article, we focus on how AMPs perform immunomodulatory functions through interaction with host receptors on epithelial, immune, and neuronal cell types. We summarize the current knowledge of known AMPs in the skin, the receptors that respond to AMPs, and the downstream intracellular signaling pathways. In the end, we discuss the roles of AMP signaling systems in skin diseases.

Strategies for improving antimicrobial peptide production.

Antimicrobial peptides (AMPs) found in a wide range of animal, insect, and plant species are host defense peptides forming an integral part of their innate immunity. Although the exact mode of action of some AMPs is yet to be deciphered, many exhibit membrane lytic activity or interact with intracellular targets. The ever-growing threat of antibiotic resistance has brought attention to research on AMPs to enhance their clinical use as a therapeutic alternative. AMPs have several advantages over antibiotics such as broad range of antimicrobial activities including anti-fungal, anti-viral and an

A novel chimeric peptide with antimicrobial activity.

Beta-lactamase-mediated bacterial drug resistance exacerbates the prognosis of infectious diseases, which are sometimes treated with co-administration of beta-lactam type antibiotics and beta-lactamase inhibitors. Antimicrobial peptides are promising broad-spectrum alternatives to conventional antibiotics in this era of evolving bacterial resistance. Peptides based on the Ala46-Tyr51 beta-hairpin loop of beta-lactamase inhibitory protein (BLIP) have been previously shown to inhibit beta-lactamase. Here, our goal was to modify this peptide for improved beta-lactamase inhibition and cellular upt

Fungicidal mechanisms of the antimicrobial peptide Bac8c.

Bac8c (RIWVIWRR-NH2) is an analogue peptide derived through complete substitution analysis of the linear bovine host defense peptide variant Bac2A. In the present study, the antifungal mechanism of Bac8c against pathogenic fungi was investigated, with a particular focus on the effects of Bac8c on the cytoplasmic membrane. We used bis-(1,3-dibutylbarbituric acid) trimethine oxonol [DiBAC4(3)] staining and 3,3'-dipropylthiacarbocyanine iodide [DiSC3(5)] assays to show that Bac8c induced disturbances in the membrane potential of Candida albicans. The antimicrobial peptide DEFB1 is associated with caries.

Genetics is an important component in the determination of individual susceptibility to caries and periodontal diseases. Since beta defensin 1 (DEFB1) localizes in the oral cavity, we tested if variation in DEFB1 is associated with caries and periodontitis. We analyzed 3 single-nucleotide polymorphisms in DEFB1 in DNA samples from unrelated individuals. Carrying a copy of the variant allele of the DEFB1 marker rs11362 (G-20A) increased the DMFT and DMFS scores more than five-fold. Also, carrying a copy of the variant allele of the DEFB1 marker rs179946 (G-52A) correlated with low DMFT scores.

Supramolecular Assembly of an Antimicrobial Peptide Into a Pore.

Antimicrobial peptides (AMPs) have long been ascribed the ability to form pores. These structures, some observed, others inferred, vary in their mechanisms of action, though most are associated with lysis. Here, we show that an AMP derived from a truncated staphylococcal δ-toxin can form functional pores that do not cause rapid lysis but, instead, we suggest that they act as self-transport systems. Specifically, a 17-amino acid AMP named STIP3-1 was observed to oligomerize and form a unique supramolecular helical assembly with an ∼14 Å pore. Substitutions to the STIP3-1 peptide abolished our a

Recent Advances in Antimicrobial Peptide Hydrogels.

Advances in the number and type of available biomaterials have improved medical devices such as catheters, stents, pacemakers, prosthetic joints, and orthopedic devices. The introduction of a foreign material into the body comes with a risk of microbial colonization and subsequent infection. Infections of surgically implanted devices often lead to device failure, which leads to increased patient morbidity and mortality. The overuse and improper use of antimicrobials has led to an alarming rise and spread of drug-resistant infections. To overcome the problem of drug-resistant infections, novel

Antimicrobial peptide resistance in Neisseria meningitidis.

Antimicrobial peptides (AMPs) play an important role as a host defense against microbial pathogens and are key components of the human innate immune response. Neisseria meningitidis frequently colonizes the human nasopharynx as a commensal but also is a worldwide cause of epidemic meningitis and rapidly fatal sepsis. In the human respiratory tract, the only known reservoir of N. meningitidis, meningococci are exposed to human endogenous AMPs. Thus, it is not surprising that meningococci have evolved effective mechanisms to confer intrinsic and high levels of resistance to the action of AMPs. T

Novel antimicrobial defensin peptides from different coleopteran insects (Coleoptera: Insecta): identification, characterisation and antimicrobial properties.

Antimicrobial peptides are crucial components of the immune systems of both vertebrates and invertebrates. Here, defensins, the most studied class of antimicrobial molecules in arthropods were investigated in four coleopteran insect species: Harpalus rufipes (DeGeer, 1774), Mylabris quadripunctata (Linnaeus, 1767), Sphaeridium marginatum (Linnaeus, 1758), and Ocypus mus (Brullé, 1832). The peptides synthesized with over 95% purity and their antimicrobial activities were evaluated by MIC test method. As a result, it was determined that Mylabris quadripunctata defensin (MqDef) showed high antimi

Multifaceted antimicrobial mechanisms of NCR147-derived peptides from Medicago truncatula.

INTRODUCTION: Antimicrobial peptides (AMPs), key components of innate immunity, offer broad-spectrum activity against diverse pathogens. In Medicago truncatula, over 700 nodule-specific cysteine-rich (NCR) peptides with highly diverse sequences and physicochemical properties are produced in the symbiotic cells of root nodules, where cationic members block bacterial cell division and display potent antimicrobial activity in vitro. In contrast, anionic NCRs typically lack antimicrobial effects, and NCR147-a neutral peptide-is the only known non-cationic NCR that shows weak bactericidal activity.

Deep Learning Enables Identification of Antimicrobial Peptides Through Mechanochromic Fingerprints.

Antimicrobial peptides (AMPs) are promising antibiotic alternatives, but their diverse modes of action make functional classification slow and labor-intensive. Here we introduce a rapid and scalable strategy for AMP identification that integrates low-cost, self-assembled polydiacetylene (PDA) sensors with hyperspectral imaging and deep learning. AMP-PDA interactions generate mechanochromic spectral fingerprints that capture subtle differences in affinity, conformation, and penetration depth in membranes. Convolutional neural networks (CNNs) trained on full spectral datasets accurately distingu

Discovery of potent low-toxicity antimicrobial peptides through diffusion modeling.

The rapid emergence of multidrug-resistant bacteria has created an urgent need for improved antimicrobial discovery and screening platforms. Here, we present ARCADIAMP, a generative and virtual screening platform that couples an iterative-learning discrete denoising diffusion probabilistic model with a two-stage Evolutionary Scale Modeling 2 (ESM2)-based antibacterial activity classifier to generate, classify, and prioritize potent AMPs with high activity, low toxicity, and favorable serum stability. Eight of the ten experimentally screened peptide candidates showed antimicrobial activity (MIC

Targeting Drug-Resistant Tuberculosis with Antimicrobial Peptides: Opportunities and Challenges.

INTRODUCTION: One of the main causes of infectious disease-related deaths globally is Mycobacterium tuberculosis (M.tb), the causative agent of tuberculosis (TB). Novel treatment strategies are now much more urgent due to the emergence of Extensively Drug-Resistant (XDR) and multidrug-resistant (MDR) strains. The purpose of this study is to investigate the potential of antimicrobial peptides (AMPs) as TB adjunctive therapeutic agents, with an emphasis on their immunomodulatory and direct antimycobacterial effects. METHODS: A summary of how antimicrobial peptides (AMPs) work was carried out, i

Performance optimization of ultrashort antimicrobial peptides through backbone amide cyclization.

Antimicrobial resistance demands therapeutics that combine potent activity with low resistance potential. Ultrashort antimicrobial peptides (AMPs) are attractive due to their manufacturability and tunability, but activity is often lost upon sequence minimization because of conformational instability. Here, we establish a conformational engineering strategy based on backbone amide cyclization to restore ultrashort AMP function. Using a snake venom-derived cathelicidin template ZY4, we identified a minimal Trp/Lys/Arg-rich motif and generated the ultrashort cyclic peptide WKR-cyl, which exhibits
