Dithiolethiones: a privileged pharmacophore for anticancer therapy and chemoprevention.

Dithiolethiones are five-membered sulfur-containing cyclic scaffolds that exhibit antioxidative, anti-inflammatory, antithrombic and chemotherapeutic activities. Dithiolethiones display the chemopreventive and cytoprotective effects by activating the antioxidant response element and mounting the transcription of cytoprotective phase II enzymatic machinery. In addition, several classes of dithiolethiones efficiently modulate the activities of proteins that play crucial roles in normal and cancer cells, including glutathione S-transferase, cyclooxygenases and master regulator NF-κB. The present

Root polysaccharides of Panax notoginseng alleviated melasma by regulating the Nrf2/ARE signaling pathway.

Panax notoginseng has gained attention for its potential efficacy and mechanisms in skin whitening and spot removal. In this study, we investigated the effects of pre-columnar saponins and polysaccharides from the roots, as well as pre-columnar saponins and flavonoids from the stems and leaves, in the prevention and treatment of melasma. We further explored their molecular mechanisms, focusing on the Nrf2/ARE signaling pathway. Inhibition of tyrosinase (TYR) activity and antioxidant activity assays showed that all four active compounds could effectively inhibit TYR activity and enhance antioxi

Ocotillol Derivatives Mitigate Retinal Ischemia-Reperfusion Injury by Regulating the Keap1/Nrf2/ARE Signaling Pathway.

Retinal ischemia-reperfusion (RIR) injury can lead to various retinal diseases. Oxidative stress is considered an important pathological event in RIR injury. Here, we designed and synthesized 34 ocotillol derivatives, then examined their antioxidant and anti-inflammatory capacities; we found that compounds 7 (C24-R) and 8 (C24-S) were most active. To enhance their water solubility, sustained release, and biocompatibility, compounds 7 and 8 were encapsulated into liposomes for in vivo activity and mechanistic studies. In vivo studies indicated that compounds 7 and 8 protected normal retinal str

The Contribution of the Nrf2/ARE System to Mechanotransduction in Musculoskeletal and Periodontal Tissues.

Mechanosensing plays an essential role in maintaining tissue functions. Across the human body, several tissues (i.e., striated muscles, bones, tendons, ligaments, as well as cartilage) require mechanical loading to exert their physiological functions. Contrary, mechanical unloading triggers pathological remodeling of these tissues and, consequently, human body dysfunctions. At the cellular level, both mechanical loading and unloading regulate a wide spectrum of cellular pathways. Among those, pathways regulated by oxidants such as reactive oxygen species (ROS) represent an essential node criti

Intralaboratory validation of four in vitro assays for the prediction of the skin sensitizing potential of chemicals.

Allergic contact dermatitis is induced by repeated skin contact with an allergen. Assessment of the skin sensitizing potential of chemicals, agrochemicals, and especially cosmetic ingredients is currently performed with the use of animals. Animal welfare and EU legislation demand animal-free alternatives reflected in a testing and marketing ban for cosmetic ingredients beginning in 2013. The underlying mechanisms of induction and elicitation of skin sensitization are complex and a chemical needs to comply several properties being skin sensitizing. To account for the multitude of events in the

Maternal Nutrition During Late Gestation and Lactation: Association With Immunity and the Inflammatory Response in the Offspring.

The immature immune system at birth and environmental stress increase the risk of infection in nursing pigs. Severe infection subsequently induces intestinal and respiratory diseases and even cause death of pigs. The nutritional and physiological conditions of sows directly affect the growth, development and disease resistance of the fetus and newborn. Many studies have shown that providing sows with nutrients such as functional oligosaccharides, oils, antioxidants, and trace elements could regulate immunity and the inflammatory response of piglets. Here, we reviewed the positive effects of ce

Possible Association of Nrf2/ARE and NFκB Response to Osteoblast Function in Spheroid Culture Induced by Low-Intensity Pulsed Ultrasound (LIPUS).

OBJECTIVE: NFκB negatively affects bone metabolism through inflammatory pathways, whereas Nrf2 benefits it by regulating antioxidant/detoxifying enzymes through antioxidant response element (ARE) activity. We investigated whether low-intensity pulsed ultrasound (LIPUS) enhances osteogenic differentiation in 3D culture and whether it affects osteoblastic differentiation-associated markers such as calcium and alkaline phosphatase (ALP), and the regulation of NFκB and/or ARE. METHODS: Murine MC3T3-E1 preosteoblasts were used to produce 3D-spheroids, which were treated with LIPUS. Time-dependent

Exposure to human immunodeficiency virus/hepatitis C virus in hepatic and stellate cell lines reveals cooperative profibrotic transcriptional activation between viruses and cell types.

UNLABELLED: Human immunodeficiency virus (HIV)/hepatitis C virus (HCV) coinfection accelerates progressive liver fibrosis; however, the mechanisms remain poorly understood. HCV and HIV independently induce profibrogenic markers transforming growth factor beta-1 (TGFβ1) (mediated by reactive oxygen species [ROS]) and nuclear factor kappa-light-chain-enhancer of activated B cells (NFκB) in hepatocytes and hepatic stellate cells in monoculture; however, they do not account for cellular crosstalk that naturally occurs. We created an in vitro coculture model and investigated the contributions of HI

Relationship between plasma uric acid levels, antioxidant capacity, and oxidative damage markers in overweight and obese adults: A cross-sectional study.

Obesity, a complex metabolic chronic illness, is commonly accompanied by an excessive production of free radicals, which influences the development of its comorbidities. Uric acid is commonly linked to pro-oxidant effects on human health. Though recent evidence suggests its potential antioxidant properties, it is possible that the increase in circulating uric acid levels is an adaptive protective response against the detrimental effects of excess free radicals and oxidative stress present in obese individuals. Hence, the purpose of this study was to evaluate the association between antioxidant

Protective effects of Nrf2-ARE activator on dopaminergic neuronal loss in Parkinson disease model mice: Possible involvement of heme oxygenase-1.

Parkinson disease (PD) is a neurodegenerative disorder characterized by a selective loss of dopaminergic neurons in the substantia nigra, and oxidative stress is thought to contribute to this pathogenesis. The nuclear factor erythroid 2-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway, which induces the production of antioxidant enzymes, is thereby a potential target for therapeutics to reduce neurodegeneration in PD. Previously, we identified TPNA10168 from a chemical library as an activator of the Nrf2-ARE pathway, and the present study examined the effects of TPNA10168 on

Lycopene protects against pressure overload-induced cardiac hypertrophy by attenuating oxidative stress.

Oxidative stress is considered an important pathogenic process of cardiac hypertrophy. Lycopene is a kind of carotenoid antioxidant that protects the cardiovascular system, so we hypothesized that lycopene might inhibit cardiac hypertrophy by attenuating oxidative stress. Phenylephrine and pressure overload were used to set up the hypertrophic models in vitro and in vivo respectively. Our data revealed that treatment with lycopene can significantly block pressure overload-induced cardiac hypertrophy in in vitro and in vivo studies. Further studies demonstrated that lycopene can reverse the inc

Unraveling the NRF2 confusion: Distinguishing nuclear respiratory factor 2 from nuclear erythroid factor 2.

In recent years, the acronym NRF2 has garnered significant attention in scientific discourse. However, this attention has occasionally led to confusion due to the existence of two distinct proteins sharing the same acronym: Nuclear Respiratory Factor 2 (NRF2), also known as GA-binding protein transcription factor subunit alpha (GABPA), and Nuclear Factor Erythroid 2-related Factor 2 (NFE2L2 or NRF2). This confusion has been highlighted in various scientific forums, including PubPeer and anonymous reader comments, where the confusion between the two proteins has been expressed. In this article,

Integrated Transcriptome and Metabolome Analysis of Mature Stage Sand Pear Fruit Response to High-Temperature Stress.

Sand pear is a fruit tree crop with high economic value, widely cultivated in East Asia. However, ripening fruits often suffer from high-temperature stress, which has adverse effects on the quality and yield of the fruit. In this study, we perform high-temperature treatment on mature stage 'Housui' pear fruits. The results showed that heat stress decreased fruit firmness and mineral elements, as well as lead to the flesh appearance of watercore. High temperature induces H2O2, MDA, and the antioxidant enzyme activity including SOD, APX, POD, and CAT were significantly increased. Transcriptome a
