The sex-biased chromatin modifier SMC1A promotes autoimmunity by shaping inflammatory pathways in patients with SLE.

A strong female bias is characteristic of systemic lupus erythematosus (SLE), the prototypic systemic autoimmune disease. Here, through an unbiased transcriptome analysis, we report a pronounced female-biased expression of the cohesin complex subunit SMC1A, a genome architectural factor, in monocytes from SLE patients compared to those from healthy individuals or patients with ankylosing spondylitis, a non-sex-biased autoimmune disorder. Integration of SMC1A binding, chromatin activity, and accessibility in lupus-like monocytes reveals extensive SMC1A redistribution to active enhancers of immu

User experiences, unmet needs, and access barriers to lower limb orthoses in India.

PURPOSE: Assistive products can enhance the social functioning of an individual with disability and could offer support for meaningful engagement in society. The research paper focuses on users' perspective to identify ease and comfortability of use of assistive products which enhances mobility. Improved mobility is an important tool to enable people with disabilities' integration into society and in the working environment. The study focused particularly on use of lower limb orthoses (commonly referred to locally as walking callipers) by people with locomotor disabilities having functional li

A gap in perceived accessibility to play spaces for physical activity in Arizona elementary schools.

BACKGROUND: The Physical Activity Guidelines for Americans states all children should be provided with various physical activity opportunities; however, school play spaces are often unaccommodating to students with disabilities and this limitation may be exacerbated in lower-income schools. OBJECTIVE: The purpose of the study was to compare elementary school play space accessibility among children with and without disabilities at lower- and higher-income schools. METHODS: This cross-sectional study utilized an online survey assessing the perceived accessibility of play spaces for students wi

Concurrent validity of the Swedish version of the life-space assessment questionnaire.

BACKGROUND: The Life-Space Assessment (LSA), developed in the USA, is an instrument focusing on mobility with respect to reaching different areas defined as life-spaces, extending from the room where the person sleeps to mobility outside one's hometown. A newly translated Swedish version of the LSA (LSA-S) has been tested for test-retest reliability, but the validity remains to be tested. The purpose of the present study was to examine the concurrent validity of the LSA-S, by comparing and correlating the LSA scores to other measures of mobility. METHOD: The LSA was included in a population-b

The role of chloride ions in serotonin transport.

The human serotonin (5-HT+) transporter SERT facilitates 5-HT+ transport into cells by coupling it to Na+ symport and K+ antiport. Although extracellular Cl- is also essential for transport, Cl- cotransport has been disputed, raising questions about the role of Cl- ions and why they are required. We show that Cl- gradients do not impact 5-HT+ accumulation, indicating that Cl- does not provide a driving force for uptake and arguing against stoichiometric Cl- symport. The presence of Cl- had only a small effect on Na+-mediated cytoplasmic pathway closure but markedly reduced the accessibility of

Effectiveness of the International Symbol of Access and inclusivity of other disability groups.

BACKGROUND: The International Symbol of Access (ISA) is recognized world-wide for designating and identifying areas which are wheelchair accessible, however its meaning has evolved to include both restricted use and universal accessibility. OBJECTIVE: This study seeks to investigate the effectiveness of the ISA in representing individuals of all impairment types. METHODS: A mixed-method survey was disseminated in the U.S. and internationally to persons without self-identified impairment and individuals of various impairment group types, including mobility, vision, hearing, and cognitive impa

Work problems and accommodations reported by persons who are postpolio or have a spinal cord injury.

A number of studies have documented early functional declines in persons with a disability. The purpose of this study was to document (1) whether employees who are aging with their disability have experienced new work problems as a consequence of functional declines and (2) whether their work problems are being accommodated adequately. Ninety-six individuals with a disability (50 who are postpolio and 46 who had a spinal cord injury) were interviewed by phone. Each had worked at least 5 years postonset and was either currently working or unemployed for less than 5 years at the time of the inte

Implications of the Americans with Disabilities Act of 1990 for elderly persons.

Because the prevalence of disability increases proportionately with age, the expanding population of older adults is potentially the largest single group to benefit from the Americans With Disabilities Act of 1990 (ADA) (Public Law 101-336). One fourth of elderly persons have functional disabilities acquired through age-related chronic and acute conditions. The ADA guarantees older adults with disabilities equality in the workplace and the community. It also enhances their opportunities to continue living independently as long as possible. Applications of the ADA for elderly persons with funct

Structural principles underlying the evolution of SWI/SNF chromatin remodelers.

Adenosine 5'-triphosphate-dependent switch/sucrose nonfermentable (SWI/SNF)-family chromatin remodelers regulate genome accessibility by repositioning nucleosomes, yet how these complexes diversified across evolution while preserving a common remodeling mechanism has remained unresolved. Here, we determine cryo-electron microscopy structures of the SWI/SNF and RSC (remodels the structure of chromatin) complexes from the thermophilic fungus Chaetomium thermophilum in apo and nucleosome-bound states, providing an evolutionarily informative structural bridge between budding yeast and metazoan rem

Societal context-dependent multi-modal transportation network augmentation in Johannesburg, South Africa.

In most developing countries, formal and informal transportation schemes coexist without effective and smart integration. In this paper, the authors show how to leverage opportunities offered by formal and informal transportation schemes to build an integrated multi-modal network. Precisely, the authors consider integration of rickshaws to a bus-train network, by taking into account accessibility and societal constraints. By modelling the respective networks with weighted graphs, a graph augmentation problem is solved with respect to a composite cost taking into account constraints on the use
