A deep-sea hydrothermal vent worm detoxifies arsenic and sulfur by intracellular biomineralization of orpiment (As2S3).

The alvinellid worm Paralvinella hessleri is the only animal that colonizes the hottest part of deep-sea hydrothermal vents in the west pacific. We found P. hessleri accumulates exceptionally high level of toxic element arsenic (>1% of wet weight) and tolerated elevated concentrations hydrogen sulphide. Using advanced microscopy, elementary analysis, and genomics and proteomics approaches, we identified a previously unrecognized arsenic-sulfide biomineralization process in P. hessleri. Our data suggest that arsenic accumulates within epithelial cell granules, where it likely reacts with sulphi

Increased neurotoxicity of arsenic in methylenetetrahydrofolate reductase deficiency.

A 16-year-old girl from Surinam presented with mental deterioration and severe paraparesis with areflexia and bilateral Babinski signs. Laboratory examination showed a hyperhomocysteinemia that was caused by 5,10-methylene-tetrahydrofolate reductase (MTHFR) deficiency. In addition, urine samples contained large amounts of arsenic. An open bag with the pesticide copper acetate arsenite was found to be the source of exposure. In remethylation defects such as MTHFR deficiency, the concentration of methyldonors is severely reduced. As arsenic is detoxified by methylation, we suggest that the MTHFR

Two cases of deliberate elemental arsenic ingestion with no apparent toxicologic consequences.

INTRODUCTION: Arsenic is a metalloid that is highly toxic in its inorganic and organic forms. Ingestion of elemental arsenic is rare. CASE REPORTS: A 33-year-old woman ingested elemental arsenic 10 g purchased online. She experienced brief gastrointestinal upset but was otherwise asymptomatic. She underwent chelation with succimer until it became apparent that she had no toxicologic consequences. She remained asymptomatic at 19-day follow-up. Her spot urine inorganic arsenic concentrations were >5,000 µg/L, 109 µg/L, and 87 µg/L at 12 days, 36 days, and 19 days post-ingestion, respectively. A

Role of H3K18ac-regulated nucleotide excision repair-related genes in arsenic-induced DNA damage and repair of HaCaT cells.

Arsenic is an environmental poison and is a grade I human carcinogen that can cause many types of damage to the body. The skin is one of the main target organs of arsenic damage, but the molecular mechanisms underlying arsenic poisoning are not clear. Arsenic is an epigenetic agent. Histone acetylation is one of the earliest covalent modifications to be discovered and is closely related to the occurrence and development of tumors. To investigate the role of acetylated histone H3K18 (H3K18 ac) in arsenic-induced DNA damage, HaCaT cells were exposed to sodium arsenite (NaAsO2) for 24 h. It was f

Phytoremedial effect of Tinospora cordifolia against arsenic induced toxicity in Charles Foster rats.

Arsenic poisoning is one of the most serious health hazards of recent times. It has been estimated that more than 200 million people of about 105 countries in the world are affected due to arsenic poisoning. Except mitigation, there is no such mode by which the population can be prevented from being exposed to arsenic. Tinospora cordifolia (T. cordifolia) is widely used in the folk medicine system for the treatment of various diseases. Hence, the aim of the present study was to investigate the antidote effects of ethanolic extract of T. cordifolia stem against arsenic induced hepato-renal toxi

Chinese Medicine Might Be A Promising Way for A Solution to Arsenic Nephrotoxicity.

Arsenic from environmental contamination is a risk factor for kidney disease, and the clinical use of arsenic also triggers a new concern that the potential kidney damage caused by exposure to clinical doses of arsenic trivalent. So far, the undergoing mechanisms contributing to arsenic nephrotoxicity mostly remain unclear, and universally accepted methods for preventing this complication are limited too. Ancient Chinese medical scientists recognized the toxicity of arsenic long ago, and there were some records of Chinese herbs against arsenic poisoning in ancient books of Chinese medicine. In

Computational Analysis of Differentially Expressed Genes in Arsenic-Induced Carcinogenesis and Their Effect on Human Repair Mechanisms.

Arsenic poisoning significantly elevates the risk of cancer and other chronic illnesses. The goal of this research is to identify important genes whose expression changes in response to arsenic toxicity, and the molecular pathways affected by arsenic, using computational analysis of arsenic toxicity profiles. This approach will computationally identify and analyze genes whose expression changes in response to arsenic, thereby elucidating the heightened risk of carcinogenesis in arsenic-exposed individuals. This work employed high-throughput arsenic toxicity profiles to computationally identify

Poisoning of animals in the Los Angeles area with pesticides during 1977.

During 1977, there were 289 pesticide exposure incidents involving animals that were handled by the Thomas J. Fleming Memorial Poison Information Center in Los Angeles. Almost all of this service was provided to veterinarians and most of the incidents involved dogs. Cases handled by this center are considered typical for small animal practices in urban areas of California. Ingestion was the major route of exposure. As recently as 10 years ago, arsenic, strychnine, and phosphorus were major causes of such poisonings. During 1977, the n-methylcarbamates (27%), anticoagulants (19%), and organo-ph

Health importance of arsenic in drinking water and food.

Arsenic is a toxic metalloid of global concern. It usually originates geogenically but can be intensified by human activities such as applications of pesticides and wood preservatives, mining and smelting operations, and coal combustion. Arsenic-contaminated food is a widespread problem worldwide. Data derived from population-based studies, clinical case series, and case reports relating to ingestion of inorganic arsenic in drinking water, medications, or contaminated food or beverages show the capacity of arsenate and arsenite to adversely affect multiple organ systems. Chronic arsenic poison

Arsenic Analysis in the Petroleum Industry: A Review.

The presence of arsenic in natural gas and liquid hydrocarbons is of great concern for oil companies. In addition to health risks due to its toxicity as well as environmental issues, arsenic is responsible for irreversible poisoning of catalysts and clogging of pipes via the accumulation of As-containing precipitates. To address these problems and to better design treatment units, robust methods for the analysis of arsenic and its compounds in oil streams are required. In addition, the use of feedstocks as a novel source of energy is becoming increasingly important. Most biomasses used as feed

Study on the Mechanism of Arsenic-Induced Lung Injury Based on SWATH Proteomics Technology.

Chronic arsenic poisoning is a global health problem that affects millions of people, and studies have found that long-term ingestion of arsenic-containing compounds can lead to lung damage, but the exact mechanism is unknown. In this study, Sprague-Dawley (SD) rats were used as the research object, and the proteomic analysis method based on sequential window acquisition of all theoretical fragment ions (SWATH) was used to detect the changes in the expression levels of related proteins in the lung tissue of arsenic-exposed rats, and to explore the mechanism of arsenic compound-induced lung inj

Clinical and environmental aspects of arsenic toxicity.

Arsenic is widely distributed throughout the animal and plant kingdoms and our environment where sources can be natural or anthropogenic. Agricultural uses of arsenic have declined recently, but it still has well-defined roles in industry. Small amounts of arsenic are metabolized in a variety of ways and are largely rapidly methylated and excreted by man and animals. Poisoning can occur and may follow an acute or chronic course. Toxic manifestations in man occur at the cellular level and may appear in many organ systems. Specific effects can often be demonstrated in the skin and in the vascula
