Do Astrocytes Play a Role in Intellectual Disabilities?

Neurodevelopmental disorders, including those involving intellectual disability, are characterized by abnormalities in formation and functions of synaptic circuits. Traditionally, research on synaptogenesis and synaptic transmission in health and disease focused on neurons, however, a growing number of studies have highlighted the role of astrocytes in this context. Tight structural and functional interactions of astrocytes and synapses indeed play important roles in brain functions, and the repertoire of astroglial regulations of synaptic circuits is large and complex. Recently, genetic studi

Morphometric analysis of astrocytes in brainstem respiratory regions.

Astrocytes, the most abundant and structurally complex glial cells of the central nervous system, are proposed to play an important role in modulating the activities of neuronal networks, including respiratory rhythm-generating circuits of the preBötzinger complex (preBötC) located in the ventrolateral medulla of the brainstem. However, structural properties of astrocytes residing within different brainstem regions are unknown. In this study astrocytes in the preBötC, an intermediate reticular formation (IRF) region with respiratory-related function, and a region of the nucleus tractus solitar

Astrocytes as new targets to improve cognitive functions.

Astrocytes are now viewed as key elements of brain wiring as well as neuronal communication. Indeed, they not only bridge the gap between metabolic supplies by blood vessels and neurons, but also allow fine control of neurotransmission by providing appropriate signaling molecules and insulation through a tight enwrapping of synapses. Recognition that astroglia is essential to neuronal communication is nevertheless fairly recent and the large body of evidence dissecting such role has focused on the synaptic level by identifying neuro- and gliotransmitters uptaken and released at synaptic or ext

Neuropharmacological effects of Phoneutria nigriventer venom on astrocytes.

Bites from genus Phoneutria (Ctenidae, Araneomorpha) are the second most frequent source of spider accidents in Southeast Brazil. Severe envenoming from Phoneutria nigriventer produces vision disturbance, tremor and convulsion, suggesting that the CNS is involved; however, the mechanisms by which P. nigriventer venom (PNV) affects the CNS remain poorly understood. The present study aimed to investigate whether PNV directly impairs astrocytes. Cultured astrocytes were exposed to PNV, and intracellular Ca(2+) release and signaling were measured (Fura-2/AM), Na(+)/K(+)-ATPase and Toll-like recept

Astrocytes influence the severity of spinal muscular atrophy.

Systemically low levels of survival motor neuron-1 (SMN1) protein cause spinal muscular atrophy (SMA). α-Motor neurons of the spinal cord are considered particularly vulnerable in this genetic disorder and their dysfunction and loss cause progressive muscle weakness, paralysis and eventually premature death of afflicted individuals. Historically, SMA was therefore considered a motor neuron-autonomous disease. However, depletion of SMN in motor neurons of normal mice elicited only a very mild phenotype. Conversely, restoration of SMN to motor neurons in an SMA mouse model had only modest effect

NDRG2 as a marker protein for brain astrocytes.

The protein NDRG2 (N-myc downregulated gene 2) is expressed in astrocytes. We show here that NDRG2 is located in the cytosol of protoplasmic and fibrous astrocytes throughout the mammalian brain, including Bergmann glia as observed in mouse, rat, tree shrew, marmoset and human. NDRG2 immunoreactivity is detectable in the astrocytic cell bodies and excrescencies including fine distal processes. Glutamatergic and GABAergic nerve terminals are associated with NDRG2 immunopositive astrocytic processes. Müller glia in the retina displays no NDRG2 immunoreactivity. NDRG2 positive astrocytes are more

Astrocytes: implications for neuroinflammatory pathogenesis of Alzheimer's disease.

Alzheimer's disease (AD) is a neurodegenerative disease with major clinical hallmarks of memory loss, dementia, and cognitive impairment. Neuroinflammation is involved in the onset of several neurodegenerative disorders. Astrocyte is the most abundant type of glial cells in the central nervous system (CNS) and appears to be involved in the induction of neuroinflammation. Under stress and injury, astrocytes become astrogliotic leading to an upregulation of the expression of proinflammatory cytokines and chemokines, which are associated with the pathogenesis of AD. Cytokines and related molecule

Structural remodeling of fibrous astrocytes after axonal injury.

Reactive astrocytes are a pathological hallmark of many CNS injuries and neurodegenerations. They are characterized by hypertrophy of the soma and processes and an increase in the expression of glial fibrillary acidic protein. Because the cells obscure each other in immunostaining, little is known about the behavior of a single reactive astrocyte, nor how single astrocytes combine to form the glial scar. We have investigated the reaction of fibrous astrocytes to axonal degeneration using a transgenic mouse strain expressing enhanced green fluorescent protein in small subsets of astrocytes. Fib

Endothelins Inhibit Junctional Permeability in Cultured Mouse Astrocytes.

Endothelins, a family of potent vasoconstrictor peptides initially characterized in peripheral tissues, have also been reported to be synthesized in the brain. In this structure several cell types, including astrocytes, endothelial cells and certain neurons, are potential targets for these peptides. In astrocytes, endothelins induce changes in the concentration of several second messengers (calcium, diacylglycerol, arachidonic acid, cAMP) known to be involved in the regulation of gap junction channels. Using the scrape loading/dye transfer technique we have observed that two isoforms of endoth

Occurrence of oligodendrocytes within astrocytes in demyelinating lesions.

We investigated the fine structural details of the presence of apparently newly formed oligodendrocytes within reactive astrocytes in white matter lesions obtained by biopsy from seven cases (3 multiple sclerosis (MS); 3 progressive multifocal leukoencephalopathy (PML); 1 with nonspecific reactive changes next to a sarcoid granuloma). Intact oligodendrocytes were found within astrocytic cytoplasm in two acute MS lesions and also in the reactive white matter lesion. The internalized cells appeared to lie within membrane-bound vacuoles. Formation of rudimentary junctions was observed between the

Astrocyte heterogeneity in brain metastases.

Brain metastasis is increasingly understood as a disease of reciprocal adaptation between metastatic cells and the brain microenvironment. Among host cells, astrocytes have emerged as central regulators of metastatic colonization, survival, immune remodeling, and treatment resistance. Recent studies are re-defining astrocytes to explore the functional implications of their cellular heterogeneity. Evidence in brain metastasis indicates that astrocyte responses are heterogeneous across anatomical space, stage of colonization, and tumor type. This framework helps explaining how distinct astrocyte

Cerebellar astrocytic alterations in depression.

Accumulating evidence suggests dysfunction of cerebellar-cerebral circuits in depression. However, the potential cellular and molecular alterations associated with depression in the cerebellum remain largely uncharacterized. While postmortem findings in the cerebral cortex indicate astrocyte dysregulation in depressed individuals who died by suicide (DS), the extent to which depression potentially alters cerebellar astrocytes is not well understood. In this study, two canonical astrocyte markers, glial fibrillary acidic protein (GFAP) and aldehyde Dehydrogenase-1 Family member L1 (ALDH1L1) wer

Astrocytic pathology in Alpers' syndrome.

Refractory epilepsy is the main neurological manifestation of Alpers' syndrome, a severe childhood-onset mitochondrial disease caused by bi-allelic pathogenic variants in the mitochondrial DNA (mtDNA) polymerase gamma gene (POLG). The pathophysiological mechanisms underpinning neuronal hyperexcitabilty leading to seizures in Alpers' syndrome remain unknown. However, pathological changes to reactive astrocytes are hypothesised to exacerbate neural dysfunction and seizure-associated cortical activity in POLG-related disease. Therefore, we sought to phenotypically characterise astrocytic patholog

New frontiers for astrocytic tumours.

Glioblastoma multiforme, the most common type of primary brain tumour, remains an unsolved clinical problem. A great deal of work has been done in an effort to understand the biology and genetics of glioblastoma multiforme, but clinically effective treatments remain elusive. It is well known that malignant gliomas develop resistance to chemo- and radiotherapy. In this review we evaluated the literature data regarding therapeutic progress for the treatment of astrocytic tumours, focusing our attention on new frontiers for glioblastoma. The research studies performed in in vitro and in vivo mode

Dual strategies for neural injury repair: Targeted modulation of reactive astrocytes and functional astrocyte transplantation.

Reactive astrogliosis-the adaptive structural and functional remodeling responses of astrocytes-is a biphasic response to neural injury. While initially protective, it often transitions into a chronic, detrimental state characterized by sustained neuroinflammation, blood-brain barrier disruption, and excitotoxicity, which collectively impede recovery. To address this, this review systematically compares the responses of reactive astrocytes across three major neural pathologies-traumatic brain injury, ischemic stroke, and spinal cord injury-identifying both conserved molecular targets that are

Factors determining the morphology and distribution of astrocytes in the cat retina: a 'contact-spacing' model of astrocyte interaction.

The retina provides a valuable opportunity to examine the interaction of astrocytes with neurones and vasculature, in adult tissue and in vivo. We have studied astrocytes in cat retina to delineate the interactions that determine their morphology and distribution. Their morphology varied with their interaction with surrounding cells, from a classic stellate shape to an elongated bipolar form associated with axon bundles. Evidence is presented that the distribution of astrocytes across the retina is determined by their morphology and by a previously unrecognised interaction between astrocytes,

Morphological Hallmarks of White Matter Astrocytes Across Development.

While the mechanisms underlying protoplasmic, or gray matter (GM), astrocyte morphogenesis have been extensively studied, far less is known about the development of fibrous, or white matter (WM), astrocytes. In particular, no study has comprehensively characterized the morphological development of WM astrocytes. To address this gap, we used an intracortical adeno-associated virus injection technique to sparsely label individual astrocytes in the mouse corpus callosum for morphological analysis across postnatal development. We found that astrocytes in the corpus callosum follow a distinct traje

Ethanol drinking involves astrocytes in male Wistar rats.

Astrocytes are the most abundant glial cells in the brain and an integrative component of the neural network. Studies have shown that ethanol altered expression of an astrocyte marker, i.e., glial fibrillary acidic protein (GFAP), in two key corticolimbic regions, the medial prefrontal cortex (mPFC) and nucleus accumbens (NAc). These regions comprise anatomically and functionally different subregions, i.e., the prelimbic (PL) and infralimbic (IL) cortex of the mPFC, the shell and core subregions of the NAc. However, ethanol effects on GFAP expression within these subregions remain largely unkn

Astrocytes in Parkinson's disease: Beyond support, toward therapy.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons in the substantia nigra. While neuronal dysfunction has traditionally been the focal point of PD research, growing evidence highlights the critical roles of astrocytes - the most abundant glial cells in the central nervous system - in PD pathogenesis and therapy. Targeting astrocytes offers a promising therapeutic avenue through astrocyte-to-neuron reprogramming, inducing A2 phenotypic polarization, suppressing oxidative stress, modulating metal ion deposition, enhancing neuro
