Pityriasis rubra pilaris: a clinico-pathological and therapeutic study with special reference to histochemistry, autoradiography, and electron microscopy.

Five patients with pityriasis rubra pilaris (PRP) were analyzed by means of light and electron microscopy as well as by histochemistry and autoradiography. The results were compared with findings in psoriasis vulgaris. In PRP we found a moderate increase of the labeling index of epidermal cells, a highly increased labeling index of dermal infiltrating cells, and a mild spongiosis, and in the stratum granulosum, a decreased number of tonofilaments and an increased number of keratinosomes. The horny layer in PRP showed a pronounced histochemical and electron microscopical parakeratosis, even whe

Osteometry and autoradiography of the long bones of skeletons of women exposed to internal radiation at 13--19 years of age.

Lengths of long bones of skeletons were examined in 25 women first exposed to large skeletal doses of radiation (alpha particles from radium) at the age of 13--19 years. Mean lengths did not differ significantly between two subgroups based on age at first exposure to radiation (i.e., 13--16 vs. 17--19 years). Autoradiographs of femora of some women who ingested radium at 13-15 years of age showed evidence for bone growth when blood levels of radium were low (i.e., after ingestion of radium). These findings indicate no detectable effect of large skeletal doses of radiation on growth in adolesce

Ex vivo delta opioid receptor autoradiography: CNS receptor occupancy of two novel compounds over their antihyperalgesic dose range.

Discovered as part of an effort to identify delta opioid (DOPr or DOR) agonist analgesics, JNJ-20788560 and JNJ-39204880 exhibited high DOR affinity, with K(i) values of 1.7 and 2.0nM, respectively, and were selective for DOR over the mu opioid receptor (MOPr or MOR), with 596- and 122-fold selectivity, respectively. Both compounds stimulated DOR but not MOR induced GTPgammaS binding and were effective antihyperalgesic agents in the complete Freund's adjuvant model of thermal hyperalgesia in the rat, with oral ED(50) values of 13.5 and 35mg/kg, corresponding to plasma levels of 1 and 9microM,

Strongyloides ratti: migration study of third-stage larvae in rats by whole-body autoradiography after 35S-methionine labeling.

In order to clarify the migration pathway of Strongyloides ratti, Wistar rats were given 5,000 35S-labeled infective larvae subcutaneously and killed at 10, 15, 20, 25, 30, 40, and 50 hr postinfection. Prior to inoculation, the specific radioactivity level was assessed in the labeled larvae using a scintillation counter. The frozen rat specimens were sectioned at 50 microm, and the sections were freeze-dried and mounted on X-ray film in darkness. The labeled larvae appeared as dark spots on the film after 14 days of exposure. The infected larvae remained at the inoculated site (lower abdomen)

Quantitative autoradiography reveals regionally selective changes in dopamine D1 and D2 receptor binding in the genetically dystonic hamster.

Dystonia has been proposed to be caused by abnormal input from thalamus to premotor cortex due to altered activity of the striatum projecting by way of the globus pallidus and substantia nigra pars reticulata to the thalamus. However, in the case of idiopathic dystonia, i.e. the most common form of dystonia in humans, there is only limited evidence to support such a neuroanatomic concept. In view of the problems of studying the pathophysiology of idiopathic dystonia in patients, genetically determined animal models of idiopathic dystonia may be used as a practical means of studying brain dysfu

Pharmacological analysis of 125I-Bolton and Hunter labelled eledoisin binding sites in rat spinal cord by quantitative autoradiography.

Using the technique of quantitative autoradiography it has been possible to investigate and compare the pharmacological characteristics of 125I-Bolton-Hunter conjugated eledoisin (125I-BHE) binding sites in rat spinal cord with those in the rat cortex. 125I-BHE specific binding sites were discretely localised in the outer layers of the rat spinal cord. The rank order of affinity of unlabelled tachykinins in competing for 125I-BHE specific binding sites in rat spinal cord (NKB greater than ELE greater than NKA greater than SP) was identical to that found in the rat cortex suggesting the presenc

Effects of chlorpromazine on plasma membrane permeability and fluidity in the rat brain: a dynamic positron autoradiography and fluorescence polarization study.

Antipsychotic drugs have been widely used in psychiatry for the treatment of various mental disorders, but the underlying biochemical mechanisms of their actions still remain unclear. Although phenothiazine antipsychotic drugs have been reported to directly interact with the peripheral plasma membrane, it is not known whether these drugs actually affect plasma membrane integrity in the central nervous system. To clarify these issues, we investigated the effect of chlorpromazine (CPZ), a typical phenothiazine antipsychotic drug, on plasma membrane permeability in fresh rat brain slices using a

A detailed mapping of histamine H1-receptors in guinea-pig central nervous system established by autoradiography with [125I]iodobolpyramine.

[125I]Iodobolpyramine, a potent and selective histamine H1-receptor antagonist derived from mepyramine, was used to generate light microscopic autoradiograms on sections of guinea-pig brain and spinal cord. Histamine H1-receptors were labelled with high sensitivity over a low background as determined using mianserin or other H1-receptor antagonists as competing agents. An atlas of H1-receptors was established using five sagittal sections and 39 frontal sections, the latter serially prepared at 50 micron intervals. Labelled areas were identified by comparison with corresponding, classically sta

Localization by autoradiography of viral proteins in the parenchymal cells of the liver during frog virus 3 induced hepatitis of mice.

Frog virus 3 inoculated into mice induces an acute degenerative hepatitis. This hepatitis is of toxic origin since the virus is unable to multiply at 37 degrees C. The Kupffer cells, which are the target cells for FV3, reveal the presence of viral particles, viral DNA and proteins. Although the hepatocytes present early and drastic nuclear lesions, viral particles were never observed in these cells. Viral proteins however but not DNA, could be found inside parenchymal cells.

[Changes in the liver and brain after portacaval and modified portacaval end-to-side anastomosis: histology, autoradiography and clinical studies].

Histological studies were done in order to investigate the early and late alterations of the liver and brain in rats after establishment of portacaval anastomosis and modified portacaval anastomosis. The latter procedure allows pancreatic blood to flow into the liver beside portacaval anastomosis. Shunting of portal blood into the systemic circulation through a portacaval anastomosis resulted in hemo-dynamically induced central lobular necrosis of the liver. Necroses and atrophy were less pronounced after modified portacaval anastomosis. The results indicate that optimal perfusion and direct a

Parathyroid hormone biosynthesis. Correlation of conversion of biosynthetic precursors with intracellular protein migration as determined by electron microscope autoradiography.

The formation of parathyroid hormone (PTH) in the parathyroid gland occurs via two successive proteolytic cleavages from larger biosynthetic precursors. The initial product coded for by PTH mRNA is pre-proparathyroid hormone (PreProPTH), a polypeptide of 115 amino acids. Within 1 min of synthesis, the polypeptide, proparathyroid hormone (ProPTH), is formed as a result of the proteolytic removal of the NH2-terminal 25 amino acids from Pre-ProPTH. After a delay of 15-20 min, the NH2-terminal six-amino acid sequence of ProPTH is removed to give PTH of 84 amino acids. To investigate the subcellula

Imaging Synaptic Vesicle Protein SV2C with 18F-UCB-F: An In Vitro Autoradiography and In Vivo NHP PET Study.

The synaptic vesicle protein SV2C, predominantly found in the basal ganglia, has been associated with Parkinson's disease through genetic studies. It plays a crucial role in regulating dopamine release and has been shown to be disrupted in PD animal models and brain tissues from PD patients. In the context of PD-related synaptopathy, SV2C may serve as a potential imaging target for monitoring disease progression and response to treatment. [18F]UCB-F is a radioligand binding to SV2C developed by UCB. Preliminary autoradiography and PET studies in rats showed that [18F]UCB-F displays a brain dis

Exploring the GLP-1-GLP-1R axis in porcine pancreas and gastrointestinal tract in vivo by ex vivo autoradiography.

INTRODUCTION: Glucagon-like peptide-1 (GLP-1) increases insulin secretion from pancreatic beta-cells and GLP-1 receptor (GLP-1R) agonists are widely used as treatment for type 2 diabetes mellitus. Studying occupancy of the GLP-1R in various tissues is challenging due to lack of quantitative, repeatable assessments of GLP-1R density. The present study aimed to describe the quantitative distribution of GLP-1Rs and occupancy by endogenous GLP-1 during oral glucose tolerance test (OGTT) in pigs, a species that is used in biomedical research to model humans. RESEARCH DESIGN AND METHODS: GLP-1R dis

[3H]-aldosterone binding sites (type I receptors) in the lateral wall of the cochlea: distribution assessment by quantitative autoradiography.

Mineralocorticoids, such as aldosterone, are steroids that enhance Na+ retention and K+ excretion in ion-transporting epithelial tissues through regulation of Na, K-ATPase. Previous studies suggest that aldosterone may regulate labyrinthine ion transport through up-regulation of Na, K-ATPase sites, a process mediated by high-affinity aldosterone (type I) receptors. In the present study, information concerning density and distribution of aldosterone binding sites in cochlear lateral wall tissues was determined by quantitative autoradiography using [3H]-aldosterone and RU-28362, a glucocorticoid

Muscarinic receptor M1 and M2 subtypes in the human eye: QNB, pirenzipine, oxotremorine, and AFDX-116 in vitro autoradiography.

Muscarinic cholinergic agents are used to lower intraocular pressure in the medical management of glaucoma and subtypes of muscarinic receptors have now been recognised in many tissues including the eye. To localise muscarinic receptors and their M1 and M2 subtypes in the human eye, in vitro ligand binding and autoradiographic techniques with densitometric quantitation on postmortem eye sections were used. Direct demonstration of dopamine D1-like receptor sites in the ciliary body of the rabbit eye by light microscope autoradiography.

The pharmacological characteristics and the anatomical localization of [3H]-SCH 23390 in sections of the ciliary body of the rabbit eye were analyzed using a radioreceptor assay and autoradiographic techniques. [3H]-SCH 23390 was bound to sections of rabbit ciliary body in a manner consistent with the labelling of D1-like receptor sites. The dissociation constant (Kd) was 0.62 nmol/l, while the maximum binding capacity (Bmax) was 117 +/- 9 fmol/mg tissue. Light microscope autoradiography revealed [3H]-SCH 23390 binding sites within the epithelium of the ciliary processes, which is the ocular s

Distribution of alpha latrotoxin receptor in the rat brain by quantitative autoradiography: comparison with the nerve terminal protein, synapsin I.

Tissue slice autoradiography was employed to reveal the brain distribution of the receptor for alpha latrotoxin, the presynaptic neurotoxin of the black widow spider venom. The receptor distribution pattern was compared with that of a marker protein for nerve endings, synapsin I, a phosphoprotein known to be present within nerve terminals. The alpha latrotoxin receptor and synapsin I were detected in gray matter-containing regions but their relative amounts were not constant. In the cerebral cortex and in the caudatum their distribution was similar, while in the hippocampus they were both abun

Whole-body autoradiography of [3H]dihydrostreptomycin in guinea pigs and rats: the labelling of the inner ear in relation to other tissues.

[3H]Dihydrostreptomycin was given intramuscularly to young pigmented guinea pigs and rats. Whole-body autoradiography, combined with densitometric measurement of the blackening of the autoradiograms, and liquid scintillation counting were used to determine the levels of radioactivity in the inner ear in relation to blood and other tissues. It was found that there was an accumulation of radioactivity in the perilymph of both the cochlear and vestibular parts of the labyrinth. The labelling of the endolymph was weak.
