Dopamine-independent psychostimulant activity of a delta-agonist.

The effects of dopamine receptor agonists and antagonists on hyperlocomotion in mice induced by the nonpeptide delta-opioid receptor agonist (+)-4-[(aR)-a-((2S,5R)-4-allyl-2,5-dimethyl-1-piperazinyl)-3-methoxybenzyl]-N,N-diethylbenzamide) (SNC80) were investigated. SNC80 significantly increased locomotion (maximally at 2 mg/kg). In antagonism tests, naltrindole and naltriben completely attenuated this SNC80-induced hyperlocomotion, which suggests that SNC80-induced hyperlocomotion may be mainly mediated through delta-opioid receptors. Although haloperidol (dopamine D2-receptor antagonist) did

Does adenosine receptor blockade mediate caffeine-induced rotational behavior?

Caffeine, a competitive antagonist at adenosine receptors, produces contralateral turning in rats with unilateral 6-hydroxydopamine-induced lesions of the nigrostriatal pathway. Adenosine A2a and dopamine D2 receptors are negatively coupled in the striatum; blockade of these adenosine receptors is a possible mechanism of caffeine-induced rotational behavior. The present study had three objectives: 1) to evaluate the role of adenosine receptor blockade in the rotational behavior elicited by caffeine in rats. This was done by determining the dose-response functions for turning behavior induced b

Oligodeoxynucleotide antisense to the D1 dopamine receptor mRNA inhibits D1 dopamine receptor-mediated behaviors in normal mice and in mice lesioned with 6-hydroxydopamine.

The effects of intracerebroventricular (i.c.v.) administration of a 20-mer phosphorothioate oligodeoxynucleotide antisense to the D1 dopamine receptor mRNA (D1 antisense) on D1 dopamine receptor-mediated behaviors were studied in normal mice and in mice with unilateral 6-hydroxydopamine-induced lesions of corpus striatum. Treating mice with D1 antisense inhibited grooming behavior induced by the D1 dopamine receptor agonist SKF 38393 [1-phenyl-2,3,4,5-tetrahydro-1H-3 benzazepine-7,8-diol hydrochloride], the reduction in grooming being related to the amount and length of time D1 antisense was g

Determination of extracellular glutamate after local K+ stimulation in the striatum of non-anaesthetised rats after treatment with dopaminergic drugs--studies using microdialysis.

The present experiments were performed in order to investigate the effects of dopamine(DA)ergic drugs on the concentrations of extracellular glutamate (GLU) in the striatum of non-anaesthetised, freely moving rats by using microdialysis and to get further information about the interactions between glutamatergic and dopaminergic pathways. GLU was determined after pre-column derivatisation with o-phthaldialdehyde by HPLC and fluorescence detection. For increasing the fraction of extracellular GLU which is of neuronal origin, an enhanced release of this neurotransmitter was evoked by 100 mM K+ ad

Effects of the antiparkinson drug budipine on EEG activity in unrestrained rats.

Rats were stereotactically implanted with electrodes into four brain areas (frontal cortex, hippocampus, striatum, and reticular formation) to allow registration of intracerebral field potentials. Connection of the electrodes to a microplug fixed to the skull of the animals allowed wireless transmission of the signals using a four-channel telemetric device. The potentials were subjected to a frequency analysis in real time, the power spectra obtained were segmented according to previous experience. Even in the low-dose range, budipine affected the power within these frequency bands: at 2 mg/kg

Absence of D1 dopamine receptors that stimulate prolactin release in the rat pituitary.

It has been shown recently that SKF 38393-A (2,3,4,5-tetrahydro-7,8-dihydroxy-1-phenyl-1H-3-benzazepine-7-ol), a D-1 receptor agonist, possesses a prolactin-releasing effect in the rat, though the pituitary or central nervous system location of the receptors involved has not been clarified. The aim of our study was to elucidate this point. SKF 38393-A administered to freely moving adult female and male rats induced a striking, short-lived increase of basal prolactin levels. The prolactin stimulatory effect of SKF 38393-A was counteracted by pretreatment with SCH 23390, A D-1 receptor blocker.

Characterization of the antihypertensive activity of SK&F 86466, a selective alpha-2 antagonist, in the rat.

The effects of SK&F 86466 (6-chloro-N-methyl-2,3,4,5-tetrahydro-1-H-3-benzazepine), a potent and selective alpha-2 adrenoceptor antagonist, on blood pressure and heart rate were examined in normotensive and hypertensive rats. SK&F 86466 was approximately 10-fold more potent in lowering blood pressure in deoxycorticosterone acetate (DOCA)-salt hypertensive than in normotensive rats, when administered by i.v. infusion. A dose-related antihypertensive effect was observed in conscious DOCA-salt rats after p.o. doses of SK&F 86466 (2-15 mg/kg), with a duration of over 6 hr after the highest dose. O

Agonist-induced homologous and heterologous sensitization to D-1- and D-2-dependent contraversive turning.

A low dose of the D-1 receptor agonist SKF 38393 (2.0 mg/kg s.c.) 14 days post-lesion failed to induce contraversive turning in male Sprague-Dawley rats lesioned unilaterally in the medial forebrain bundle with 6-hydroxydopamine (6OHDA). Priming with a single administration of the specific D-2 agonist LY 171555 (0.2 mg/kg s.c. 3 days before), which itself elicited contraversive turning, made SKF 38393 (2.0 mg/kg s.c.) very active to produce contraversive turning. Priming with LY 171555 (0.2 mg/kg s.c. 3 days before) potentiated the contraversive turning in response to a low dose of LY 171555 (

The relaxing effect of SKF 38393 on the catch contraction of Mytilus smooth muscle.

The relaxing effect of SKF 38393 on the catch contraction in the anterior byssus retractor muscle (ABRM) of Mytilus and the effect of SKF 38393 on the cyclic AMP (cAMP) levels in the ABRM were investigated. The catch contraction was relaxed by SKF 38393 in a dose-dependent manner. The dose-response curves of SKF 38393 were shifted in parallel to the right by butaclamol (3 X 10(-6) and 6 X 10(-6) M) and haloperidol (3 X 10(-6) and 3 X 10(-5) M). Their pA2 values were 5.69 +/- 0.05 and 5.80 +/- 0.07. The cAMP levels in the ABRM were not altered by SKF 38393 at a concentration (10(-5) M) sufficie

Evidence for the nucleus accumbens as a neural substrate of heroin-induced immune alterations.

Administration of opioid drugs such as heroin produces several immunosuppressive effects, including decreases in natural killer (NK) cell activity, lymphocyte proliferative responses, and nitric oxide production. Interestingly, opioids have been shown to alter many immune parameters indirectly by modulating the immunoregulatory actions of the central nervous system. Recently, it has been demonstrated that morphine inhibits NK cell activity through a neural pathway that requires the activation of dopamine D(1) receptors in the nucleus accumbens shell. The present study examined whether the nucl

Prevention of hypoxic brain oedema by the administration of vasopressin receptor antagonist OPC-31260.

The numerous situations which can result in cerebral hypoxic damage occur in newborn infants and in the elderly. In research aimed at more effective therapeutic intervention in ischaemic disorders of the brain, the animal model used and the principles of the causal therapy should be better outlined. The effects of the non-peptide AVPR (V2) antagonist 5-dimethylamino-1-[4-(2-methylbenzoylamino) benzoyl]-2,3,4,5-tetrahydro-1H-benzazepine hydrochloride (OPC-31260) on the cerebral oedema induced by general cerebral hypoxia were studied in rats. Desensitization of the dopamine D1 and D2 receptor hetero-oligomer mediated calcium signal by agonist occupancy of either receptor.

When dopamine D1 and D2 receptors were coactivated in D1-D2 receptor hetero-oligomeric complexes, a novel phospholipase C-mediated calcium signal was generated. In this report, desensitization of this Gq/11-mediated calcium signal was demonstrated by pretreatment with dopamine or with the D1-selective agonist (+/-)-6-chloro-7,8-dihydroxy-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine hydrobromide (SKF-81297) or the D2-selective agonist quinpirole. Desensitization of the calcium signal mediated by D1-D2 receptor hetero-oligomers was initiated by agonist occupancy of either receptor subtype even t

cAMP-dependent reversal of opioid- and prostaglandin-mediated depression of the isolated respiratory network in newborn rats.

1. Membrane potential (Vm) and resistance (Rm) of ventral respiratory group (VRG) neurons were measured in the isolated brainstem-spinal cord from newborn rats during bath application of the opioid receptor agonists fentanyl or [D-Ala2, D-Leu5]-enkephalin (Ala-Leu-Enk) and of the prostaglandin E1 (PGE1). 2. PGE1 (0.1-3 microM) and fentanyl or Ala-Leu-Enk (1-50 microM) produced depression and, at higher doses, block of inspiratory nerve activity and respiration-related postsynaptic potentials. This apnoea was associated with hyperpolarization and Rm fall in 25% of thirty-two VRG neurons tested,

Effects of putative dopamine D3 receptor agonists, 7-OH-DPAT, and quinpirole, on yawning, stereotypy, and body temperature in rats.

7-OH-DPAT ((+/-)-2-(dipropylamino)-7-hydroxy-1,2,3,4-tetrahydronaphthalene) was recently identified as a dopamine receptor agonist having a > 100-, 1,000- and > 10,000-fold higher affinity for dopamine D3 than for D2, D4 and D1 receptors, respectively. Quinpirole (LY 171555) has also been reported to have a 113-fold greater affinity for dopamine D3 receptors than for D2 receptors. Therefore, we investigated the effects of these putative dopamine D3 receptor agonists on yawning, stereotypy and rectal temperature in rats (N = 424). 7-OH-DPAT and quinpirole administered subcutaneously (SC) at res

A Phase I Study of Pelabresib (CPI-0610), a Small-Molecule Inhibitor of BET Proteins, in Patients with Relapsed or Refractory Lymphoma.

PURPOSE: NF-κB, a transcription factor essential for inflammatory responses, is constitutively activated in many lymphomas. In preclinical studies, pelabresib (CPI-0610), an investigational (BET) bromodomain inhibitor, downregulated NF-κB signaling and demonstrated antitumor activity in vitro. Here we report the safety, pharmacokinetics, pharmacodynamics, and preliminary clinical activity from the first-in-human phase I study of pelabresib in patients with relapsed/refractory lymphomas (NCT01949883). Successful use of ivabradine in a 10-year-old patient with graft failure after heart transplantation.

We encountered a paediatric case of graft failure due to antibody-mediated rejection after heart transplantation in which ivabradine was effective. Inappropriate sinus tachycardia in denervated transplanted hearts is a good indication for ivabradine administration as beta-blockers have a limited efficacy. To our knowledge, this is the first report on the effectiveness of ivabradine in a paediatric heart transplant rejection case.
