Baicalin and berberine ultradeformable vesicles as potential adjuvant in vitiligo therapy.

0.5-1% of the world's population is affected by vitiligo, a disease characterized by a gradual depigmentation of the skin. Baicalin and berberine are natural compounds with beneficial activities, such as antioxidant, anti-inflammatory and proliferative effects. These polyphenols could be useful for the treatment of vitiligo symptoms, and their efficacy can be improved by loading in suitable carriers. The aim of this work was to formulate and characterize baicalin or berberine loaded ultradeformable vesicles, and demonstrate their potential as adjuvants in the treatment of vitiligo. The vesicle

Berberine as a Multitarget Therapeutic Agent: Insights from Molecular Docking Studies.

Natural compounds are crucial in pharmacological research for modulating numerous biological processes. This is particularly helpful in the case of complex diseases and in overcoming challenges (e.g., drug resistance and adverse effects). Berberine, an isoquinoline alkaloid extracted from plants such as Berberis, Coptis, and Hydrastis species, exhibits a wide range of activities, including antidiabetic, anticancer, anti-inflammatory, antimicrobial, and antiviral. However, due to its low oral bioavailability (poor absorption, high first-pass metabolism, along with rapid elimination), further me

Crystalline mate strategy for berberine hydrochloride: solvent-polarity-tuned co-crystallization.

Single-crystal X-ray diffraction is one of the most reliable techniques for elucidating the molecular structures, while it requires high-quality single crystals. The crystalline mate (CM) strategy has recently emerged as an effective method to induce rapid and selective co-crystallization. Here, we demonstrate that this strategy can be reliably extended to ionic systems using the neutral trinuclear silver pyrazolate complex Ag3Pz3 as a crystallization mate. By regulating solvent polarity, two co-crystals of silver pyrazolate berberine hydrochloride Ag3Pz3·BCl, SF (sandwich form) and NSF (non-s

Unlocking the antibacterial potential of berberine-thioether derivatives against plant canker.

BACKGROUND: Plant diseases have long posed a severe threat to global agricultural production. Among these, kiwifruit canker and citrus canker-caused by Pseudomonas syringae pv. actinidiae (Psa) and Xanthomonas axonopodis pv. citri (Xac), respectively-are quarantine diseases that lack effective green control agents. RESULTS: This work synthesized novel berberine-thioether derivatives, most of which showed potent antibacterial activity against Psa and Xac. The half-effective concentration values of compounds B-4 - B-6 against Xac (0.29, 0.37, and 0.39 μg mL-1, respectively) and of A-1 - A-6 aga

Berberine rescues PANoptosis-Driven hemophagocytic lymphohistiocytosis via cardiolipin-mediated mitochondrial stabilization.

Pyroptosis, apoptosis, and necroptosis can converge through a unified inflammatory cell death pathway termed PANoptosis, which plays a pivotal role in the pathogenesis of diverse inflammatory disorders. The isoquinoline alkaloid berberine (BBR) is known to suppress pyroptosis and NLRP3 inflammasome activation, yet its impact on the broader process of PANoptosis remains undefined. In this study, PANoptosis was induced in murine macrophages using poly(I:C) and LPS, and mechanistic insights were investigated by immunoblotting, immunofluorescence, and co-immunoprecipitation. The therapeutic potent

Overcoming Solubility Challenges: Micronization of Berberine Using the GAS Antisolvent Technique.

Berberine is a compound widely used in Chinese herbal medicine. It can potentially treat diabetes, cholesterol, and mental illnesses and has antimicrobial effects. However, its application is limited due to its low solubility in water, thus limiting its bioavailability. Micronization techniques are used to increase the surface area and improve the dissolution rate. The micronized particles were produced using the antisolvent GAS technique at a temperature of 35 °C and 80 bar. A solubility study with acetone, dichloromethane, ethanol, methanol, 1-butanol, and 1-propanol was carried out. Fourier

Purinergic P2X7 receptor involves in anti-retinal photodamage effects of berberine.

Berberine (BBR) is a Chinese herb with antioxidant and anti-inflammatory properties. In a previous study, we found that BBR had a protective effect against light-induced retinal degeneration in BALB/c mice. The purinergic P2X7 receptor (P2X7R) plays a key role in retinal degeneration via inducing oxidative stress, inflammatory changes, and cell death. The aim of this study was to investigate whether BBR can induce protective effects in light damage experiments and whether P2X7R can get involved in these effects. C57BL/6 J mice and P2X7 knockout (KO) mice on the C57BL/6 J background were used.

Green-synthetized selenium nanoparticles using berberine as a promising anticancer agent.

OBJECTIVE: The chemo-preventative and therapeutic properties of selenium nanoparticles (SeNPs) have been documented over recent decades and suggest the potential uses of SeNPs in medicine. Biogenic SeNPs have higher biocompatibility and stability than chemically synthesized nanoparticles, which enhances their medical applications, especially in the field of cancer therapy. This study evaluated the potential of green-synthetized SeNPs by using berberine (Ber) as an antitumor agent and elucidated the mechanism by which these molecules combat Ehrlich solid tumors (ESTs). METHODS: SeNPs containin

Berberine Prevents Disease Progression of Nonalcoholic Steatohepatitis through Modulating Multiple Pathways.

The disease progression of nonalcoholic fatty liver disease (NAFLD) from simple steatosis (NAFL) to nonalcoholic steatohepatitis (NASH) is driven by multiple factors. Berberine (BBR) is an ancient Chinese medicine and has various beneficial effects on metabolic diseases, including NAFLD/NASH. However, the underlying mechanisms remain incompletely understood due to the limitation of the NASH animal models used. Methods: A high-fat and high-fructose diet-induced mouse model of NAFLD, the best available preclinical NASH mouse model, was used. RNAseq, histological, and metabolic pathway analyses w

Berberine Delays Onset of Collagen-Induced Arthritis through T Cell Suppression.

There is evidence that berberine (BBR), a clinically relevant plant compound, ameliorates clinically apparent collagen-induced arthritis (CIA) in vivo. However, to date, there are no studies involving the use of BBR which explore its prophylactic potential in this model of rheumatoid arthritis (RA). The aim of this study was to determine if prophylactic BBR use during the preclinical phase of collagen-induced arthritis would delay arthritic symptom onset, and to characterize the cellular mechanism underlying such an effect. DBA/1J mice were injected with an emulsion of bovine type II collagen

Berberine reduces endothelial injury and arterial stiffness in spontaneously hypertensive rats.

Background: Changes in circulating endothelial microparticles (EMPs) and endothelial progenitor cells (EPCs) are considered as a new perspective reflection of the endothelial injury and repair status. Our previous studies have demonstrated that berberine improved endothelial function and arterial stiffness in healthy subjects. In this study, we further investigated the effects of berberine on regulating the circulating EMPs and EPCs, and preventing endothelial dysfunction and arterial stiffness in spontaneously hypertensive rats (SHRs). Methods: Twenty male SHRs were randomly divided into two

Characterization, pharmacokinetics, and hypoglycemic effect of berberine loaded solid lipid nanoparticles.

The high aqueous solubility, poor permeability, and absorption of berberine (BBR) result in its low plasma level after oral administration, which greatly limits its clinical application. BBR solid lipid nanoparticles (SLNs) were prepared to achieve improved bioavailability and prolonged effect. Developed SLNs showed homogeneous spherical shapes, small size (76.8 nm), zeta potential (7.87 mV), encapsulation efficiency (58%), and drug loading (4.2%). The power of X-ray diffraction combined with (1)H nuclear magnetic resonance spectroscopy was employed to analyze chemical functional groups and th
