Betulinic acid restores imatinib sensitivity in BCR-ABL1 kinase-independent, imatinib-resistant chronic myeloid leukemia by increasing HDAC3 ubiquitination and degradation.

Although imatinib (IM) has been demonstrated to be an efficient treatment in chronic myeloid leukemia (CML), some patients still experience IM resistance and disease relapse. Through in vitro studies, we observed that HDAC3 levels were elevated in BCR-ABL1 kinase-independent, IM-resistant primary cells from CML patients and in IM-resistant K562 (K562R) cells and that downregulation of HDAC3 could enhance IM efficacy in K562R cells. Furthermore, betulinic acid (BA), a lupane-type pentacyclic triterpenoid saponin isolated from birch trees, restored IM sensitivity in the BCR-ABL1 kinase-independe

Betulinic Acid Protects From Bone Loss in Ovariectomized Mice and Suppresses RANKL-Associated Osteoclastogenesis by Inhibiting the MAPK and NFATc1 Pathways.

Osteoclasts with elevated bone resorption are commonly present in postmenopausal osteoporosis, and other osteolytic pathologies. Therefore, suppressing osteoclast generation and function has been the main focus of osteoporosis treatment. Betulinic acid (BA) represents a triterpenoid mainly purified from the bark of Betulaceae. BA shows multiple biological activities, including antitumor and anti-HIV properties, but its effect on osteolytic conditions is unknown. Here, BA suppressed receptor activator of nuclear factor-κB ligand (RANKL)-associated osteoclastogenesis and bone resorptive function

Betulinic acid is associated with miR-21 modulation, apoptosis and redox changes in breast cancer cells: an in vitro and in silico study.

Betulinic acid is defined as a hydrophobic pentacyclic triterpenoid primarily found in the bark of Betula alba, known for its anticancer activity through mechanisms such as upregulating proapoptotic proteins, modulating NF-kB, and inhibiting topoisomerase I. The current study aimed to explore the anticancer potential effect, molecular targets, and association with miR-21 modulation after BA treatment and its combination with doxorubicin against human triple-negative breast cancer (MDA-MB-231) cells. We determined drug cytotoxicity by MTT assay, and death mechanism by flow cytometry. Besides, t

Betulinic acid prevents high glucose‑induced expression of extracellular matrix protein in cardiac fibroblasts by inhibiting the TGF‑β1/Smad signaling pathway.

The proliferation and differentiation of cardiac fibroblasts (CFs) is central to cardiac fibrosis. Betulinic acid (BA) is an active compound isolated from the bark of the birch tree Betula spp. (Betulaceae) and has been shown to attenuate hepatic fibrosis. However, the effect of BA on the high glucose‑induced fibrosis response in CFs remains to be elucidated, therefore, the present study investigated the effect of BA on high glucose‑induced CFs and examined the possible mechanism underlying the effect of BA on CF transdifferentiation. CFs were pre‑incubated with various concentrations of BA fo

Targeted delivery to joints of the rheumatoid arthritis drug betulin using folic acid-coated nanoparticles based on human serum albumin.

Rheumatoid arthritis (RA) is a chronic inflammatory joint disease characterized by synovitis, cartilage and bone damage, and potential permanent disability. The conventional pharmacotherapies for rheumatoid arthritis, including non-steroidal anti-inflammatory drugs, glucocorticoids, and disease-modifying anti-rheumatic drugs, predominantly confer symptomatic palliation. Betulin, a pentacyclic triterpene derived from birch bark, has shown promise in treating RA. But it suffers from poor solubility, low oral bioavailability, short circulation time, and potential adverse effects. This study aimed

Design, synthesis and in vitro validation of bivalent binders of SARS-CoV-2 spike protein: Obeticholic, betulinic and glycyrrhetinic acids as building blocks.

SARS-CoV-2 is the virus responsible for the COVID-19 pandemic, which caused over 6.7 million deaths worldwide. The Spike protein plays a crucial role in the infection process, mediating the binding of the virus to its cellular receptor, angiotensin-converting enzyme 2 (ACE2), and its subsequent entry into target cells. Previous studies identified, through virtual screening, several natural products capable of binding to two distinct pockets of the Spike protein: triterpenoids binding to pocket 1 and bile acid derivatives binding to pocket 5. Building on these findings, our study advances the f

Optimization of betulinic and ursolic acids and phenolics extraction from endemic Rosa pisiformis using Box-Behnken design in relation to cytotoxic activities.

Despite its important pharmacological bioactivities, betulinic acid is still primarily obtained through extraction from heartwood and bark or synthesized synthetically, with less than 3% efficiency. Our endemic rose species, Rosa pisiformis (Christ.) D. Sosn., which is a new alternative source of betulinic acid and traditionally valued for its medicinal properties, was collected from its natural distribution in Gümüşhane province. The plant's organs, such as root, stem, leaf and fruit were air-dried and pulverized. The compounds were separately extracted using three different solvents (ethanol

Betulinic acid ameliorates lipopolysaccharide-induced intestinal barrier dysfunction in broilers by remodeling gut microbiota homeostasis and inhibiting the LYG2/NOD1/NF-κB signaling axis.

Betulinic acid (BA), a natural pentacyclic triterpenoid obtained from agroforestry by-products, possesses potent antioxidant, anti-inflammatory and intestinal-protective bioactivities. Our prior work verified that BA alleviated intestinal inflammatory damage in mouse and piglet models. Nevertheless, its protective effects and underlying molecular mechanisms against lipopolysaccharide (LPS)-evoked intestinal barrier dysfunction in broilers remain poorly understood. Under basal feeding conditions, dietary BA supplementation improved ileal mucosal integrity by elevating villus height and crypt de
