The Pace of Biological Aging Predicts Nonspecific Chronic Low Back Pain Severity.

This study aimed to determine if and how the pace of biological aging was associated with nonspecific chronic low back pain (cLBP) and compare what measure of epigenetic age acceleration most strongly predicts cLBP outcomes. We used the Dunedin Pace of Aging from the Epigenome (DunedinPACE), Horvath's, Hannum's, and PhenoAge clocks to determine the pace of biological aging in 69 cLBP, and 49 pain-free controls (PFCs) adults, ages 18 to 85 years. On average, participants with cLBP had higher DunedinPACE (P < .001) but lower Horvath (P = .04) and Hannum (P = .02) accelerated epigenetic age than

Epigenetic Clocks and Their Prospective Application in the Complex Landscape of Aging and Alzheimer's Disease.

Nowadays, scientists are making efforts to elucidate the mechanisms involved in the phenotypic changes underlying the aging process in order to develop favorable therapeutical interventions. Epigenetic modifications, in particular DNA methylation, play a crucial role in the aging process, and this parameter has been used to set epigenetic clocks, algorithms that predict an individual's biological age based on a defined set of CpGs. In this review, we focus on the most recent literature to discuss the use of epigenetic clocks in the context of cognitive decline and dysregulation of Alzheimer's

Sociodemographic and Lifestyle Factors and Epigenetic Aging in US Young Adults: NIMHD Social Epigenomics Program.

IMPORTANCE: Epigenetic clocks represent molecular evidence of disease risk and aging processes and have been used to identify how social and lifestyle characteristics are associated with accelerated biological aging. However, most research is based on samples of older adults who already have measurable chronic disease. OBJECTIVE: To investigate whether and how sociodemographic and lifestyle characteristics are associated with biological aging in a younger adult sample across a wide array of epigenetic clock measures. DESIGN, SETTING, AND PARTICIPANTS: This cohort study was conducted using da

[Misaligned light entrainment causes metabolic disorders in Chrono knockout mice].

Most of the life forms on Earth have gradually evolved an endogenous biological clock under the long-term influence of periodic daily light-dark cycles. This biological clock system plays a crucial role in the orderly progression of life activities. In mammals, central circadian clock is located in the suprachiasmatic nucleus of the hypothalamus and the function of the biological clock relies on a transcription-translation negative feedback loop. As a negative regulator in this loop, the function of CHRONO is less known. To deeply explore the role of the Chrono gene in rhythm entrainment and p

Age-Related Changes in Circadian Rhythm and Association with Nutrition.

PURPOSE OF REVIEW: Considering the increase in life expectancy, there is a time-related decline in biological functions. Age-related changes are also observed in the circadian clock which directly leads to appropriate rhythms in the endocrine and metabolic pathways required for organism homeostasis. Circadian rhythms are affected by the sleep/wake cycle, environmental changes, and nutrition. The aim of this review is to show the relationship between age-related changes in circadian rhythms of physiological and molecular processes and nutritional differences in the elderly. RECENT FINDINGS: Nu

Biological aging across the metabolic dysfunction-associated steatotic liver disease spectrum: A systematic review.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a growing global health burden. Geroscience posits that accelerated biological aging is a key driver of chronic disease. We systematically reviewed the evidence to define the role of biological aging (BA) across the MASLD disease spectrum. METHODS: A systematic literature search of PubMed/MEDLINE and Embase was conducted from inception through August 2025 for observational studies in adults assessing validated BA measures (phenotypic age, Klemera-Doubal method, Homeostatic Dysregulation, epigenetic clocks

Chrononutrition in Critical Illness.

Circadian rhythms in humans are biological rhythms that regulate various physiological processes within a 24-hour time frame. Critical illness can disrupt the circadian rhythm, as can environmental and clinical factors, including altered light exposure, organ replacement therapies, disrupted sleep-wake cycles, noise, continuous enteral feeding, immobility, and therapeutic interventions. Nonpharmacological interventions, controlling the ICU environment, and pharmacological treatments are among the treatment strategies for circadian disruption. Nutrition establishes biological rhythms in metabol

Convergent latitudinal erosion of circadian systems in a rapidly diversifying order of fishes.

Biological clocks allow organisms to anticipate cyclical environmental changes, yet in high-latitude or deep-sea habitats, the diel cues that entrain these rhythms are often seasonally diminished or absent. Fishes of the order Perciformes have rapidly diversified across these arrhythmic ecosystems, raising the question of whether changes to circadian rhythms and biological clock genetic architecture are a component of their evolutionary success. Here, we used a comparative genomic approach to investigate patterns of core biological clock gene loss across 96 perciform and five outgroup species.

Rewinding the gut clock: from chronodisruption to targeted therapies.

Circadian rhythms orchestrate daily oscillations in physiology and behaviour, enabling optimal alignment between internal biological processes and environmental cycles. A hierarchical system centred in the suprachiasmatic nucleus and supported by peripheral clocks across tissues ensures temporal alignment of metabolic, immune and GI functions. Within the GI tract, local clocks drive rhythmic programmes that regulate digestion, absorption, motility, hormone secretion, immune responses and interactions with the gut microbiota. Feeding-fasting cycles and microbial metabolites act as powerful zeit

Biological aging and the hidden architecture of early-onset cancer risk.

In a Nature Medicine study, Tian et al. identify larger biological age gaps associated with early-onset lung, colorectal, and uterine cancers across birth cohorts. These findings position biological age as an integrative marker of physiological dysregulation. Future work combining aging clocks and mutational signatures could distinguish tumor-promoting states from tumor-initiating exposures.

Frailty in lung transplant recipients is associated with anemia and telomere dysfunction but independent of epigenetic age.

Frailty is a syndrome of vulnerability to stressors linked to worse outcomes before and after lung transplantation. However, the biological basis of this association is unknown. Biological correlates of aging include epigenetic reprograming, chronic inflammation, telomere dysfunction, and anemia. We hypothesized that these aging-associated biological processes would be associated with frailty in lung transplant recipients. In a nested case-control study, we compared 43 lung transplant recipients who were frail pretransplant and posttransplant with 43 nonfrail matched controls. We quantified pe

Biological aging of different blood cell types.

Biological age (BA) captures detrimental age-related changes. The best-known and most-used BA indicators include DNA methylation-based epigenetic clocks and telomere length (TL). The most common biological sample material for epidemiological aging studies, whole blood, is composed of different cell types. We aimed to compare differences in BAs between blood cell types and assessed the BA indicators' cell type-specific associations with chronological age (CA). An analysis of DNA methylation-based BA indicators, including TL, methylation level at cg16867657 in ELOVL2, as well as the Hannum, Horv

Regulation of circadian blood pressure: from mice to astronauts.

PURPOSE OF REVIEW: Circadian variation is commonly seen in healthy people; aberration in these biological rhythms is an early sign of disease. Impaired circadian variation of blood pressure (BP) has been shown to be associated with greater target organ damage and with an elevated risk of cardiovascular events independent of the BP load. The purpose of this review is to examine the physiology of circadian BP variation and propose a tripartite model that explains the regulation of circadian BP. RECENT FINDINGS: The time-keeper in mammals resides centrally in the suprachiasmatic nucleus. Apart f

The role of global DNA methylation and epigenetic aging in the suicidality of patients with schizophrenia.

Suicide is a leading cause of death among individuals with schizophrenia. Studies suggest that suicide attempters show increased DNA methylation and shorter telomere lengths, both linked to accelerated biological aging. Recently, DNA methylation has been used to estimate epigenetic aging and telomere length, the former of which serves as the most accurate measure of biological aging. In this study, we investigated whether patients with schizophrenia spectrum disorder (SSD) and a history of suicide attempts show greater biological aging than non-attempters. Epigenetic aging and telomere length

Trajectories of physical function and biological aging in generally healthy older adults with and without incident invasive cancer over a three-year follow-up: findings from the DO-HEALTH study.

Cancer is associated with biological aging and functional decline; however, few studies have simultaneously examined objective changes in physical function and biological aging in older adults who develop cancer. We therefore compared functional and accelerated aging in generally healthy adults with and without incident invasive cancer, using data from DO-HEALTH, a three-year, randomized controlled trial including 2152 participants (mean age: 74.9 years, 61.1% women), free of major health conditions. Functional aging was assessed by the Short Physical Performance Battery, handgrip strength, ga

Vascular Aging Across the Cardiovascular-Alzheimer's Continuum: Hemodynamics and Artificial Intelligence.

Predicting health trajectories and accurately measuring aging processes across the human lifespan remain profound scientific challenges. Assessing the effectiveness and impact of interventions targeting aging is even more elusive, largely due to the intricate, multidimensional nature of aging-a process that defies simple quantification. Traditional biomarkers offer only partial perspectives, capturing limited aspects of the aging landscape. Yet, over the past decade, groundbreaking advancements have emerged. Epigenetic clocks, derived from DNA methylation patterns, have established themselves

The role of Mediterranean diet adherence, smoking and their interactions in epigenetic age acceleration: A cross-sectional analysis of the Airwave cohort.

BACKGROUND: Epigenetic clocks are markers of biological ageing that may vary in their sensitivity to environmental and lifestyle exposures. To evaluate their utility as sensors of the human exposome, we investigated responses to two biologically relevant exposures with contrasting effects, the Mediterranean die and smoking. OBJECTIVES: We assessed the sensitivity of eleven epigenetic clocks to diet and smoking and evaluated whether Mediterranean diet adherence modifies associations between smoking and epigenetic ageing. METHODS: We analysed 928 participants (mean age 41 years, 59% male) from

Biological age associates with longitudinal frailty-related functional decline: the Health and Retirement Study.

Epigenetic clocks have emerged as markers of biological aging. Understanding their association with age-related functional decline may provide insights into DNA-mediated mechanisms underlying frailty-related functional decline and reveal which clocks best associate with accelerated functional decline. We therefore examined associations between established epigenetic clock measures and longitudinal trajectories of cognitive function, grip strength, and walking speed. We analyzed data from 4018 participants in the Health and Retirement Study with available DNA methylation data and up to 12 years

Epigenetic age predictors for non-invasive assessment of human skin.

Skin is both the most visible and most environmentally exposed organ, with apparent aging phenotypes. DNA methylation clocks faithfully capture the progression of aging, but so far have been limited to training on abundant in vitro material or invasively collected samples to generate narrow methylomes using microarray platforms. Here, we demonstrate that skin biological age can be measured directly from a person's face with superior accuracy, using non-invasive tape-stripping. We developed two clocks, MitraSolo, based on single CpGs, and MitraCluster, on regions, trained on the largest enzymat

Circadian rhythms in thrombosis and atherothrombotic events.

Biological circadian rhythms in living organisms are regulated by molecular clocks. Several of these clocks are present in blood vessels, peripheral tissues, and immune cells. There is strong evidence linking dysregulation of circadian rhythms to the development of cardiovascular disease. Dysregulation of circadian rhythms is believed to activate inflammatory processes at specific times of day, leading to an increased risk of thrombosis and atherosclerosis progression. Research into circadian clock genes and molecular networks has the potential to identify therapeutic targets to reduce cardiov
