Monocarboxylate transporters: past, present, and future.

We review here the 14 members of the Monocarboxylate transporter family (MCTs), their relationship based on sequence homology. The range of substrates transported by different members of this family extends from the standard monocarboxylate metabolites, lactic and pyruvic acids, to aromatic amino acids and thyroid hormones. The family is denoted Solute Carrier Family 16, or SLC16, among 43 SLC families constituting more than 300 members, which are annotated regularly at the website http://www.bioparadigms.org/slc/intro.htm. MCTs classically transport metabolites across plasma membranes with di

Biosynthesis and Export of Bacterial Glycolipids.

Complex carbohydrates are essential for many biological processes, from protein quality control to cell recognition, energy storage, and cell wall formation. Many of these processes are performed in topologically extracellular compartments or on the cell surface; hence, diverse secretion systems evolved to transport the hydrophilic molecules to their sites of action. Polyprenyl lipids serve as ubiquitous anchors and facilitators of these transport processes. Here, we summarize and compare bacterial biosynthesis pathways relying on the recognition and transport of lipid-linked complex carbohydr

Effects of amide creatine derivatives in brain hippocampal slices, and their possible usefulness for curing creatine transporter deficiency.

The creatine/phosphocreatine system carries ATP from production to consumption sites and buffers the intracellular content of ATP at times of energy deprivation. The creatine transporter deficiency syndrome is an X-linked disease caused by a defective creatine transporter into the central nervous system. This disease is presently untreatable because creatine lacking its carrier cannot cross neither the blood-brain barrier nor the cell plasma membranes. Possible strategies to cure this condition are to couple creatine to molecules which have their own carrier, to exploit the latter to cross bio

The SLC31 (Ctr) copper transporter family.

Copper is essential for many copper-dependent processes, including mitochondrial oxidative phosphorylation, free-radical detoxification, pigmentation, neurotransmitter synthesis, and iron metabolism. The identification of proteins for high affinity copper uptake and export has greatly expanded our understanding of cellular copper homeostasis. Copper export in human cells is mediated by the ATP7A and ATP7B P-type ATPases, which are, respectively, affected in the genetic disorders of copper metabolism, Menkes disease and Wilson disease. A different class of transporter known as the SLC31 or Ctr

Semi-metric optimal transport enables robust spatial multi-omics integration.

Integrating heterogeneous spatial multi-omics data to uncover underlying biological and pathological mechanisms remains a major challenge, while existing approaches generally lack mathematically justified distances for comparing complex spatial tissues. We introduce Spatial Optimal Transport (SpaOT), a spatial multi-omics integration framework based on a theoretically grounded semi-metric formulation of unbalanced optimal transport. Utilizing a total-variation-relaxed Fused Partial Gromov-Wasserstein strategy, SpaOT establishes a mathematically consistent distance geometry that enables stable

The fall and rise of active chloride transport: implications for regulation of intraocular pressure.

Early study of transepithelial salt transfer focused on Cl(-) and not Na(+), partly because Cl(-) was readily measureable. The advent of flame photometry and tracer techniques brought Na(+) to the fore, especially since short-circuited frog skin (Rana temporaria) produces baseline net movement of Na(+) and not of Cl(-). Zadunaisky was among the first to describe what is currently termed secondary active Cl(-) transport, helping stimulate interest in Cl(-) handling by other tissues, notably the thick ascending limb of the loop of Henle important in renal counter-current multiplication. Transport of organic anions across the basolateral membrane of proximal tubule cells.

Renal proximal tubules secrete diverse organic anions (OA) including widely prescribed anionic drugs. Here, we review the molecular properties of cloned transporters involved in uptake of OA from blood into proximal tubule cells and provide extensive lists of substrates handled by these transport systems. Where tested, transporters have been immunolocalized to the basolateral cell membrane. The sulfate anion transporter 1 (sat-1) cloned from human, rat and mouse, transported oxalate and sulfate. Drugs found earlier to interact with sulfate transport in vivo have not yet been tested with sat-1.

Dynamic coordination engineering of gallium biomaterials: A biological accessibility framework for therapeutic biofunction.

Gallium (Ga)-based biomaterials have emerged as versatile therapeutic platforms owing to the iron-mimicking properties and broad biological activities of gallium. Coordination engineering regulates Ga speciation, stability, transport, and therapeutic activation; however, optimizing individual physicochemical properties does not necessarily improve biological performance. Excessive coordination stability may restrict ligand exchange and therapeutic activation, whereas insufficient stability may cause premature dissociation, nonspecific exposure, and reduced target delivery. In this Review, we p

In Vitro and Ex Vivo Studies on the Absorption and Distribution of β-Cyclodextrin Polymer.

Background: Cyclodextrin (CD) polymers have attracted increasing attention due to their favourable drug delivery properties and broad pharmaceutical applicability. While the bioavailability and biological behaviour of native cyclodextrins have been extensively investigated, considerably less information is available regarding modified cyclodextrin polymers. Therefore, the present study aimed to investigate the permeation and cellular uptake of an epichlorohydrin-crosslinked β-cyclodextrin polymer using multiple in vitro and ex vivo models. Methods: Fluorescently labelled β-cyclodextrin polymer

Gut-eye axis and transporter-mediated strategies: enhancing retinal uptake of dietary nutrients.

The blood-retinal barrier (BRB) stringently restricts the delivery of dietary nutrients to retinal tissues, presenting a major bottleneck for targeted nutritional intervention. This review systematically delineates three core pathways through which food-derived bioactive compounds overcome the limited delivery efficiency of the BRB: transporter-mediated active transport across the barrier, biotransformation by gut microbiota and the liver into bioactive metabolites with superior bioavailability, and gut-retina axis-mediated remote regulation of retinal homeostasis. An innovative focus is place

An artificial model of ATP-binding cassette mediated transport.

Active drug efflux is a critical bottleneck in multidrug-resistance (MDR) in cancers, bacterial infections and in neurological disease. Preclinical acellular permeation assays do not predict efflux (active) transport, and cell-based assays may be confounded by the complexity and heterogeneity of cells which can make efflux data analysis challenging. We report here a cell-free platform as a model for functional multidrug resistance (MDR) efflux pump that mimics the inner membrane of Escherichia coli (E. coli). The system comprises a PE:PG:CL (67:23.2:9.8 mol%) lipid bilayer reconstituted with A

The Systems Biology of Drug Metabolizing Enzymes and Transporters: Relevance to Quantitative Systems Pharmacology.

Quantitative systems pharmacology (QSP) has emerged as a transformative science in drug discovery and development. It is now time to fully rethink the biological functions of drug metabolizing enzymes (DMEs) and transporters within the framework of QSP models. The large set of DME and transporter genes are generally considered from the perspective of the absorption, distribution, metabolism, and excretion (ADME) of drugs. However, there is a growing amount of data on the endogenous physiology of DMEs and transporters. Recent studies-including systems biology analyses of "omics" data as well as

Salt and water transport across alveolar and distal airway epithelia in the adult lung.

Substantial progress has been made in understanding the role of the distal airway and alveolar epithelial barriers in regulating lung fluid balance. Molecular, cellular, and whole animal studies have demonstrated that reabsorption of fluid from the distal air spaces of the lung is driven by active sodium transport. Several different in vivo, in situ, and isolated lung preparations have been used to study the mechanisms that regulate fluid transport in the normal and injured lung. Catecholamine-dependent and -independent regulatory mechanisms have been identified that modulate fluid transport,

Effects of divalent cations on diffusion dynamics of biological water confined between lipid membranes.

Biological water is an ionic solution containing both monovalent and divalent ions. However, the effects of divalent ions on the dynamics of biological water remain largely unknown. Here, we investigate how the transport dynamics of water molecules nanoconfined between lipid membranes depends on the concentration of calcium (Ca2+) and magnesium (Mg2+) ions by using molecular dynamics simulations and the generalized transport equation for biological water. We find that the diffusion coefficient of biological water monotonically increases with Ca2+ ion concentration but exhibits a largely opposi
