Genetic variation between Biomphalaria alexandrina snails susceptible and resistant to Schistosoma mansoni infection.

Much effort has been made to control schistosomiasis infection in Egypt. However, enduring effects from such strategies have not yet been achieved. In this study, we sought to determine the genetic variability related to the interaction between Biomphalaria alexandrina snails and Schistosoma mansoni. Using RAPD-PCR with eight (10 mers) random primers, we were able to determine the polymorphic markers that differed between snails susceptible and resistant to Schistosoma mansoni infection using five primers out of the eight. Our results suggest that the RAPD-PCR technique is an efficient means b

Genetic markers between Biomphalaria glabrata snails susceptible and resistant to Schistosoma mansoni infection.

The analysis of the genetic variability related to susceptibility to Schistosoma mansoni infection in the vector of the genus Biomphalaria is important in terms of a better understanding of the epidemiology of schistosomiasis itself, the possible pathological implications of this interaction in vertebrate hosts, and the formulation of new strategies and approaches for disease control. In the present study, the genetic variability of B. glabrata strains found to be resistant or susceptible to S. mansoni infection was investigated using DNA amplification by random amplified polymorphic DNA-polym

Sequential histological changes in Biomphalaria glabrata during the course of Schistosoma mansoni infection.

Biomphalaria glabrata, highly susceptible to Schistosoma mansoni, were seen to shed less and less cercariae along the time of infection. Histological examination kept a close correlation with this changing pattern of cercarial shedding, turning an initial picture of no-reaction (tolerance) gradually into one of hemocyte proliferation with formation of focal encapsulating lesions around disintegrating sporocysts and cercariae, a change that became disseminated toward the 142nd day post miracidial exposure. Findings were suggestive of a gradual installation of acquired immunity in snails infecte

Lectin binding to cells of Schistosoma mansoni sporocysts and surrounding Biomphalaria glabrata tissue.

The binding of different lectins to the surface of mother and daughter sporocysts of Schistosoma mansoni (Trematoda) and to cells of its intermediate host Biomphalaria glabrata (Gastropoda) was investigated. The test system consisted of several biotin-labeled lectins, an avidin-biotin-peroxidase complex and 3,3'-diaminobenzidine. The fixatives used were Formalin, Bouin's and Zenker's solutions; unfixed material was also studied. Most lectins reacted equally with host tissue and parasite tissue. However, receptors for Ulex europaeus I (most probably fucose) were only demonstrated on daughter sp

Schistosoma mansoni (Gezira) infection in Biomphalaria pfeifferi snails from Gezira and Jebel Marra.

The comparative susceptibility of Biomphalaria snails from Gezira and Jebel Marra to S. mansoni (Gezira) was investigated. Groups of laboratory-bred snails from the two localities were exposed to 0, 1, 2, 3 and 4 miracidia. The growth rate of the snails was followed over the experimental period by measuring the snails' shell size. Sub-groups of infected snails were examined for daughter sporocyst counts by crushing of the snails. Other groups of exposed snails were maintained until cercariae were shed. The exposed snails grew faster than the controls and growth was not dependent on the number

Behavioral response of Biomphalaria glabrataexposed to a sublethal concentration of Euphorbia miliivar. hislopii latex.

The Euphobia milii var. hislopii latex has been tested in the control of schistosomiasis but its action in the locomotor activity of Biomphalaria glabrata is unknown. The objective of this work was to study the locomotor and reproductive behaviors of B. glabrata exposed to E. milii var. hislopii latex. For this, 96 snails were individually exposed to the latex (LC50 - 0.53 mg / L) for 24 hours. The specimens were submitted to biomonitoring for image analysis to record the locomotor parameters at different times: before exposure (control), one day post exposure (group 1 d-p-e) and 30 days post

An immunohistochemical analysis of peptidergic neurons apparently associated with reproduction and growth in Biomphalaria alexandrina.

Peptide hormones and neurotransmitters involved in reproduction and growth have been studied extensively in certain gastropod molluscs, such as Lymnaea stagnalis and Aplysia californica. The present study employs antisera that have been used to study peptidergic neurons in those species to probe the central nervous system of another gastropod, Biomphalaria alexandrina, an intermediate host of the parasitic trematode that causes schistosomiasis in humans. Whole mount preparations of central ganglia were stained immunohistochemically, and several populations of neurons appeared to be homologous

Biomphalaria camerunensis as a viable alternative intermediate host for Schistosoma mansoni in southern Cameroon.

BACKGROUND: Intestinal schistosomiasis due to Schistosoma mansoni was mapped in Cameroon in the 1990s and preventive chemotherapy launched since 2005. A situation analysis conducted in 2011 revealed an increase in schistosomiasis transmission, especially in the equatorial part of the country, despite the fact that Biomphalaria pfeifferi, the main intermediate host of this parasite, is now scarce in many foci. Biomphalaria camerunensis, restricted to the equatorial part of the country, is considered as a less suitable host for S. mansoni due to it resistance to the parasite, although exhibiting

Sequence and structural variation in the genome of the Biomphalaria glabrata embryonic (Bge) cell line.

BACKGROUND: The aquatic pulmonate snail Biomphalaria glabrata is a significant vector and laboratory host for the parasitic flatworm Schistosoma mansoni, an etiological agent for the neglected tropical disease schistosomiasis. Much is known regarding the host-parasite interactions of these two organisms, and the B. glabrata embryonic (Bge) cell line has been an invaluable resource in these studies. The B. glabrata BB02 genome sequence was recently released, but nothing is known of the sequence variation between this reference and the Bge cell genome, which has likely accumulated substantial ge

Molecular and morphological identification of Biomphalaria species from the state of São Paulo, Brazil.

DNA barcoding and morphological characters were used to identify adult snails belonging to the genus Biomphalaria from 17 municipalities in the state of São Paulo, Brazil. The DNA barcode analysis also included twenty-nine sequences retrieved from GenBank. The final data set of 104 sequences of the mitochondrial cytochrome oxidase I (COI) gene was analyzed for K2P intraspecific and interspecific divergences, through tree-reconstruction methods (Neighbor-Joining, Maximum Likelihood and Bayesian inference), and by applying different models (ABGD, bPTP, GMYC) to partition the sequences according

Usnic acid potassium salt: an alternative for the control of Biomphalaria glabrata (Say, 1818).

In Brazil, the snail Biomphalaria glabrata is the most important vector of schistosomiasis due to its wide geographical distribution, high infection rate and efficient disease transmission. Among the methods of schistosomiasis control, the World Health Organization recommends the use of synthetic molluscicides, such as niclosamide. However, different substances of natural origin have been tested as alternatives for the control or eradication of mollusks. The literature describes the antitumor, antimicrobial and antiviral properties of usnic acid as well as other important activities of common

Spatial distribution of Biomphalaria spp., the intermediate host snails of Schistosoma mansoni, in Brazil.

Schistosomiasis mansoni remains an important parasitic disease of man, endemic in large parts of sub-Saharan Africa, the Middle East, South America and the Caribbean. The aetiological agent is the trematode Schistosoma mansoni, whereas aquatic snails of the genus Biomphalaria act as intermediate hosts in the parasite life cycle. In Brazil, the distribution of Biomphalaria spp. is closely associated with the occurrence of schistosomiasis. The purpose of this study was to map and predict the spatial distribution of the intermediate host snails of S. mansoni across Brazil. We assembled snail "pre

Molecular epidemiology of Brazilian Biomphalaria: a review of the identification of species and the detection of infected snails.

The three vector species of Schistosoma mansoni in Brazil, Biomphalaria glabrata, B. tenagophila and B. straminea show different susceptibility levels to the trematode besides a wide geographical distribution. The identification of such molluscs is important to further understand the disease epidemiology. Considering the fact that morphological identification may become difficult or even impossible under particular circumstances, the use of molecular-based methods have permitted the generation of more consistent information concerning the population structure of Biomphalaria furthering knowled

[Susceptibility of Biomphalaria tenagophila from the Paraná and Uruguay River basins to Schistosoma mansoni].

To study the possibility that schistosomiasis might be able to spread into the basins of the Paraná and Uruguay rivers, 1,711 snails were experimentally exposed to infection with Schistosoma mansoni. These snails were laboratory-bred progeny of Biomphalaria tenagophila collected from 15 populations in the geographic area under consideration. Three strains of S. mansoni were used: BH2, adapted to B. glabrata, and SJ and SJ2 adapted to B. tenagophila. None of the 543 snails exposed to the BH2 strain became infected, nor did any of the 668 exposed to SJ. However, of the 500 exposed the SJ2, 4 of

[Immunologic and parasitologic aspects of Biomphalaria tenagophila infected by Schistosoma mansoni and other Digenea].

The behavior of Biomphalaria tenagophila amoebocytes was studied in infections produced by Schistosoma mansoni and other Digenea. The resistance to superinfection was also verified in mixed infections. Data on amoebocyte phagocytic activity, on the number of amoebocytes in hemolymph, and on amoebocyte tissue reactions were obtained and electrophoretic and immunodiffusion examinations of the hemolymph were carried out. It was concluded that the snails infected with Digenea show resistance to superinfection with S. mansoni. Apparently sporocysts are not destroyed by the action of amoebocytes. An

380 B. glabrata infected with 1 miracidium of S. mansoni produced 87 positive snails (22,9%), 38 of which sending out male cercariae and 49 female cercariae. The snails belonged to 3 experimental series kept at 27 degrees and 24 degrees C for 15 weeks under continuous lighting. There was no difference between the proportions of both sexes. The survival of the positive snails was favoured by the male sex. The shedding of female cercariae was greater than shedding of male. During those 15 weeks, the cercarial shedding of both sexes showed large and recurring changes. A certain correlation seems
