Cell line development and bioreactor process optimization for an atezolizumab biosimilar.

Checkpoint inhibitors are widely recognized immunotherapeutic drugs, known for their effectiveness in treating various cancers. Atezolizumab, targeting the immune checkpoint programmed death-ligand 1, is successfully used to treat several types of cancers. Atezolizumab is a potential biosimilar candidate due to its huge success in the clinic but there is no literature on its production process in mammalian cells. In this study, we generated a monoclonal cell line derived from recombinant Chinese hamster ovary DG44 cells to produce atezolizumab. The selected single clone was employed for media

Accelerated production of human epithelial organoids in a miniaturized spinning bioreactor.

Conventional static culture of organoids necessitates weekly manual passaging and results in nonhomogeneous exposure of organoids to nutrients, oxygen, and toxic metabolites. Here, we developed a miniaturized spinning bioreactor, RPMotion, specifically optimized for accelerated and cost-effective culture of epithelial organoids under homogeneous conditions. We established tissue-specific RPMotion settings and standard operating protocols for the expansion of human epithelial organoids derived from the liver, intestine, and pancreas. All organoid types proliferated faster in the bioreactor (5.2

The proliferation and differentiation of skeletal muscle stem cells are enhanced in a bioreactor.

Skeletal muscle (SKM) is the largest organ in mammalian body and it can repair damages by using the residential myogenic stem cells (MuSC), but this repairing capacity reduces with age and in some genetic muscular dystrophy. Under these circumstances, artificial amplification of autologous MuSC in vitro might be necessary to repair the damaged SKM. The amplification of MuSC is highly dependent on myogenic signals, such as sonic hedgehog (Shh), Wnt3a, and fibroblast growth factors, so formulating an optimum myogenic kit composed of specific myogenic signals might increase the proliferation and

EnduroBone: A 3D printed bioreactor for extended bone tissue culture.

Studies of the effects of external stimuli on bone tissue, disease transmission mechanisms, and potential medication discoveries benefit from long-term tissue viability ex vivo. By simulating the in-vivo environment, bioreactors are essential for studying bone cellular activity throughout biological processes. We present the development of an automated 3D-printed bioreactor EnduroBone designed to sustain the ex-vivo viability of 10 mm diameter cancellous bone cores for an extended period. The device is supplied with two critical parameters for maintaining bone tissue viability: closed-loop con

Large-scale cultivation of Leishmania infantum promastigotes in stirred bioreactor.

BACKGROUND & OBJECTIVES: Bioreactors are practical tools that are used for economical, time-conserving and large-scale production of biomass from cell cultivation. They provide optimal environmental conditions such as pH and temperature required for obtaining maximum amounts of biomass. However, there is no evidence in the literature on the large-scale cultivation of Leishmania infantum parasites in the bioreactor. Therefore, the present study was undertaken to develop a new approach for obtaining L. infantum biomass by using pH and temperature controllable stirred bioreactor and to compare pa

Bioreactor concepts for cell culture-based viral vaccine production.

Vaccine manufacturing processes are designed to meet present and upcoming challenges associated with a growing vaccine market and to include multi-use facilities offering a broad portfolio and faster reaction times in case of pandemics and emerging diseases. The final products, from whole viruses to recombinant viral proteins, are very diverse, making standard process strategies hardly universally applicable. Numerous factors such as cell substrate, virus strain or expression system, medium, cultivation system, cultivation method, and scale need consideration. Reviewing options for efficient a

Mechanisms of pathogenic virus removal in a full-scale membrane bioreactor.

Four pathogenic virus removal mechanisms were investigated in a full-scale membrane bioreactor (MBR; nominal pore size 0.04 μm): (i) attachment of virus to mixed liquor solids; (ii) virus retention by a just backwashed membrane; (iii) virus retention by the membrane cake layer; and (iv) inactivation. We quantified adenovirus, norovirus genogroup II (GII), and F+ coliphage in the influent wastewater, the solid and liquid fractions of the mixed liquor, return flow, and permeate using quantitative PCR (adenovirus and norovirus GII) and infectivity assays (F+ coliphage). Removal and fate of micropollutants in a sponge-based moving bed bioreactor.

This study investigated the removal of micropollutants using polyurethane sponge as attached-growth carrier. Batch experiments demonstrated that micropollutants could adsorb to non-acclimatized sponge cubes to varying extents. Acclimatized sponge showed significantly enhanced removal of some less hydrophobic compounds (log D<2.5), such as ibuprofen, acetaminophen, naproxen, and estriol, as compared with non-acclimatized sponge. The results for bench-scale sponge-based moving bed bioreactor (MBBR) system elucidated compound-specific variation in removal, ranging from 25.9% (carbamazepine) to 96

Anaerobic membrane bioreactor (AnMBR) for bamboo industry wastewater treatment.

Bamboo industry wastewater (BIWW) poses severe environmental problems because of its high organic matter content. In this study, anaerobic membrane bioreactor (AnMBR) was applied for BIWW treatment. During the start-up stage, the system presented an effective degradation with a final COD removal of 91%. Compared to the intermittent mode, a higher membrane rejection (45% COD, 60% NH3-N) was obtained when the system was operated continuously. N2 flushing was applied for membrane cleaning, and the cleaning efficiency was significantly influenced by the hydraulic retention time (HRT). While operat

Immobilization conditions of ketoreductase on enantioselective reduction in a gas-solid bioreactor.

The immobilization conditions of commercial ketoreductase for continuous enantioselective reduction in the gas-phase reaction were investigated with respect to the immobilization efficiency (residual activity and protein loading) and the gas-phase reaction efficiency (initial reaction rate, half-life, and enantioselectivity). For the analyses, ketoreductase was first immobilized by physical deposition on glass supports and the reduction of 2-butanone to (S)-2-butanol with the concomitant regeneration of NADH by 2-propanol was used as a model reaction. The optimal conditions of enzyme immobiliz

Performance of a submerged membrane bioreactor system for biological nutrient removal.

A pilot submerged membrane bioreactor coupled with biological nutrient removal was used to treat the primary effluent at a municipal wastewater treatment plant. Long-term experiments were conducted by varying hydraulic retention time from 6 to 8 hours and solids retention time from 20 to 50 days, respectively. The performance was assessed by monitoring key wastewater parameters, including chemical oxygen demand (COD), nitrogen, and phosphorus concentration in individual anoxic, anaerobic, aerobic, and membrane separation zones. Results showed that the tested system can consistently achieve COD

Demonstration of efficient trichloroethylene biodegradation in a hollow-fiber membrane bioreactor.

Rapid cometabolism of trichloroethylene (TCE) by pure cultures of Methylosinus trichosporium OB3b PP358 was demonstrated in a two-stage hollow-fiber membrane bioreactor over the course of 3 weeks. PP358 was grown in a continuous-flow chemostat and circulated through the shell of a hollow-fiber membrane module (HFMM), while TCE contaminated water (160 to 1450 micrograms/L) was pumped through the fiber lumen (fiber interior). In parallel-flow HFMM biological experiments, 82% to 89% of the influent TCE was removed from the lumen (5.1-min residence time) with 99% of the transferred TCE undergoing

A gas lift bioreactor for removal of contaminants from the vapor phase.

The cometabolic degradation of trichloroethylene (TCE) as a vapor by two aromatic-metabolizing pseudomonads was evaluated in an airlift reactor. These microorganisms were able to degrade 90 to 95% of TCE in air at concentrations at the reactor inlet of 300 to 4,000 mug/liter. Although exposure of the cells to high inlet concentrations of TCE (4 mg/liter) caused a decline in enzyme-specific activity and TCE removal efficiency, this loss in activity could be prevented or delayed by increasing the rate of cosubstrate addition. Under the appropriate operating conditions, the microorganisms were ab

Zero Nuisance Piggeries: long-term performance of MBR (membrane bioreactor) for dilute swine wastewater treatment using submerged membrane bioreactor in semi-industrial scale.

Effective aerobic/anoxic treatment of piggery manure wastewater was achieved in a real farm scale using a small piggery (72 pigs) with reuse of the treated water. The experimental procedure was followed for 9 months. Fresh manure (FM) is formed by daily flush on piggeries and biologically treated after centrifuge pre-treatment. For upgrade liquid/solid separation and pathogen retention in biological treatment, a membrane system was used with the aim of effluent reuse in flush. Despite an evolution of FM through time, centrifuge pre-treatment and bioreactor performances stayed at high level. An

Autotrophic and Mixotrophic Batch Processes with Clostridium autoethanogenum LAbrini in Stirred Tank Bioreactors with Continuous Gassing.

Simultaneous conversion of syngas and sugars is a promising approach to overcome limitations of syngas fermentation. Clostridium autoethanogenum LAbrini, obtained by adaptive laboratory evolution, is known to show improved autotrophic process performance. Under purely autotrophic conditions, C. autoethanogenum LAbrini exhibits substantially faster growth and biomass formation compared to the wild-type in fully controlled, stirred-tank bioreactors with a continuous gas supply. In mixotrophic processes, the pre-culture strategy has a significant impact on the growth and metabolic activity of C.

A new approach to off-gas analysis for shaken bioreactors showing high CTR and RQ accuracy.

BACKGROUND: Shake flasks are essential tools in biotechnological development due to their cost efficiency and ease of use. However, a significant challenge is the miniaturization of process analytical tools to maximize information output from each cultivation. This study aimed to develop a respiration activity online measurement system via off-gas analysis, named "Transfer rate Online Measurement" (TOM), for determining the oxygen transfer rate (OTR), carbon dioxide transfer rate (CTR), and the respiration quotient (RQ) in surface-aerated bioreactors, primarily targeting shake flasks.
