[Somite patterning and segregation of different somite lineages].

The somite is a transient embryonic mesodermal structure, found only in vertebrates. In amniotes, somites give rise to the dermis of the back, to the striated skeletal muscles of the trunk and limbs and to the vertebral column and ribs. Segregation of these different lineages is linked to the establishment of two somitic polarity axes, a dorso-ventral one and a medio-lateral one. While the establishment of the former relies essentially on extrinsic cues, that of the latter obeys to both intrinsic and extrinsic mechanisms. Concerning the environmental cues regulating the establishment of both a

BMPRIA is a promising marker for evaluating ganglion cells in the enteric nervous system--a pilot study.

Congenital disorders of the enteric nervous system (ENS) comprise a large group of conditions characterized by abnormalities in the number, size, or location of enteric ganglia. Their diagnosis requires careful histological evaluation of intestinal biopsies to determine the presence and morphology of these cells. Based on the recently discovered role of bone morphogenetic proteins (BMPs) in ENS development, we examined the expression of the ligands, BMP2 and BMP4, and their receptors, BMPRIA, BMPRIB, and BMPRII, during formation of the human ENS. The spatiotemporal expression pattern of these

Ridge bone maintenance in human after extraction.

PURPOSE: The aim of this study was to evaluate, clinically and histologically, the tissues formed in human alveolar sockets filled with bovine morphogenetic protein/bovine organic matrix (BOM) and absorbable membrane (AM) immediately after extraction. MATERIALS: Forty-six human alveolar sockets, exhibiting buccal bone defects were selected for this study. Group 1 received no biomaterial to serve as control. Sockets from group 2 were filled with bovine bone morphogenetic protein (bBMP) associated with bOM. The association of bBMP/bOM/AM filled the alveolar defects from group 3. AM was placed o

Structure and expression of Xenopus tropicalis BMP-2 and BMP-4 genes.

The Xenopus tropicalis BMP-2 and BMP-4 genes have been cloned and sequenced. A comparison with the corresponding genes from X. laevis reveals that the BMP-4 genes are conserved at a higher extent than the BMP-2 genes. This is especially evident for the intron sequences, but is also reflected by the exon sequences. While the amino acids of X. tropicalis and X. laevis BMP-4 proteins diverge at about 4%, those of BMP-2 proteins diverge at about 7%. By reverse transcriptase polymerase chain reaction analyses and whole mount in situ hybridizations, we demonstrate the temporal and spatial expression

Misexpression of noggin leads to septal defects in the outflow tract of the chick heart.

BMP-2 and BMP-4 are known to be involved in the early events which specify the cardiac lineage. Their later patterns of expression in the developing mouse and chick heart, in the myocardium overlying the atrioventricular canal (AV) and outflow tract (OFT) cushions, also suggest that they may play a role in valvoseptal development. In this study, we have used a recombinant retrovirus expressing noggin to inhibit the function of BMP-2/4 in the developing chick heart. This procedure resulted in abnormal development of the OFT and the ventricular septum. A spectrum of abnormalities was seen rangin

Bone morphogenic protein-4 expression in vascular lesions of calciphylaxis.

Calciphylaxis is characterized by an extensive media-calcification of cutaneous and subcutaneous arterioles and capillaries. Recent studies have provided evidence that vascular calcification is a process with similarities to bone metabolism. Bone morphogenic protein-4 (BMP-4) is physiologically involved in bone development and repair. The presence of BMP-4 in atherosclerosis and in sclerotic heart valves led us to suggest that BMP-4 is also involved in calciphylaxis. A 47-year-old male patient developed end-stage renal failure due to chronic glomerulonephritis. He has had two kidney transplant

BMP pathways are involved in otic capsule formation and epithelial-mesenchymal signaling in the developing chicken inner ear.

The vertebrate inner ear consists of a complex labyrinth of epithelial cells that is surrounded by a bony capsule. The molecular mechanisms coordinating the development of the membranous and bony labyrinths are largely unknown. Previously, using avian retrovirus encoding Noggin (RCAS-Noggin) or beads soaked with Noggin protein, we have shown that bone morphogenetic proteins (BMPs) are important for the development of the otic epithelium in the chicken inner ear. Here, using two additional recombinant avian retroviruses, dominant negative and constitutively active forms of BMP receptors IB (BMP

Polyphenols as Modulators of the BMP Signaling Pathway to Counter Phenotypic Transformation of Colonocytes.

The intestinal bone morphogenetic protein (BMP) pathway counteracts the Wnt pathway, which is overactivated in 90% of colorectal cancers (CRC) due to mutations in the adenomatous polyposis coli (APC) gene. Our study aimed to identify a polyphenol that acts on the colonic BMP pathway and to evaluate its ability to mitigate the harmful effects of 4-hydroxynonenal (HNE), a dietary genotoxic molecule, on normal colonocytes. We first evaluated the effect of 10 polyphenols on the expression of BMP pathway actors using a normal mouse colonocyte and fibroblast culture model. Apigenin (Apig) and resver

Investigation of the Role of BMP2 and -4 in ASD, VSD and Complex Congenital Heart Disease.

Congenital heart malformations (CHMs) make up between 2 and 3% of annual human births. Bone morphogenetic proteins (BMPs) signalling is required for chamber myocardium development. We examined for possible molecular defects in the bone morphogenetic protein 2 and 4 (BMP2, -4) genes by sequencing analysis of all coding exons, as well as possible transcription or protein expression deregulation by real-time PCR and ELISA, respectively, in 52 heart biopsies with congenital malformations (atrial septal defect (ASD), ventricular septal defect (VSD), tetralogy ofFallot (ToF) and complex cases) compa

Nonbiased Molecular Screening Identifies Novel Molecular Regulators of Fibrogenic and Proliferative Signaling in Myxomatous Mitral Valve Disease.

BACKGROUND: Pathological processes underlying myxomatous mitral valve degeneration (MMVD) remain poorly understood. We sought to identify novel mechanisms contributing to the development of this condition. METHODS AND RESULTS: Microarrays were used to measure gene expression in 11 myxomatous and 11 nonmyxomatous human mitral valves. Differential gene expression (thresholds P<0.05; fold-change >1.5) and pathway activation (Ingenuity) were confirmed using quantitative reverse transcriptase polymerase chain reaction and immunohistochemistry. lmo4a Contributes to Zebrafish Inner Ear and Vestibular Development via Regulation of the Bmp Pathway.

BACKGROUND: In vertebrates, the development of the inner ear is a delicate process, whereas its relating molecular pathways are still poorly understood. LMO4, an LIM domain-only transcriptional regulator, is drawing an increasing amount of interest for its multiple roles regarding human embryonic development and the modulation of ototoxic side effects of cisplatin including cochlear apoptosis and hearing loss. The aim of the present study is to further explore the role of lmo4a in zebrafish inner ear development and thus explore its functional role. METHODS: The Spatial Transcript Omics DataB

Phenotype-specific differences in bone morphogenetic protein ligand expression and intracellular signaling pathway activation in chronic rhinosinusitis.

PURPOSE: Chronic rhinosinusitis (CRS) comprises distinct phenotypes with divergent inflammatory and tissue remodeling patterns. Bone morphogenetic proteins (BMPs) are key regulators of airway repair; however, it remains unclear whether phenotypic differences between CRS with nasal polyps (CRSwNP) and without nasal polyps (CRSsNP) are associated with BMP ligand abundance or downstream pathway activity. We investigated phenotype-specific BMP/Smad pathway patterns and their potential relevance to airway remodeling. METHODS: Sinonasal tissue samples were obtained from patients with CRSwNP (n = 20

Reduced BMP signaling results in hindlimb fusion with lethal pelvic/urogenital organ aplasia: a new mouse model of sirenomelia.

Sirenomelia, also known as mermaid syndrome, is a developmental malformation of the caudal body characterized by leg fusion and associated anomalies of pelvic/urogenital organs including bladder, kidney, rectum and external genitalia. Most affected infants are stillborn, and the few born alive rarely survive beyond the neonatal period. Despite the many clinical studies of sirenomelia in humans, little is known about the pathogenic developmental mechanisms that cause the complex array of phenotypes observed. Here, we provide new evidences that reduced BMP (Bone Morphogenetic Protein) signaling

Genes affecting tooth morphogenesis.

The development of dentition is a fascinating process that encompasses a complex series of epithelial-mesenchymal interactions involving growth factors, transcription factors, signal receptors and other soluble morphogens. It is not surprising that such a complex process is prone to disturbances and may result in tooth agenesis. Initial discoveries indicating that the homeo-domain protein MSX1 and the paired-domain transcription factor PAX9 are causative genes in tooth morphogenesis were made in mice. Both genes are co-expressed in dental mesenchyme and either one, when homozygously deleted, r

[Ex-vivo gene therapy with BMP-4 for critically sized defects and enhancement of fracture healing in an osteoporotic animal model].

Fractures in osteoporotic bones or segment defects are problematic bone lesions with a reduced biological capability of regeneration. We tested the hypothesis that cell-mediated ex vivo gene therapy to deliver BMP4 can heal critically sized defects and improve bone healing in osteoporotic rats. Primary muscle-derived cells were isolated from the hindlimb muscle of rats and retrovirally transduced to express bone morphogenic protein 4 (BMP4). The bone formation was evaluated following local application of these cells in critically sized defects and in fractures of osteoporotic bones. Radiograph

BMP-4 response in wild-type and craniosynostotic rabbit bone cells.

BACKGROUND: Craniosynostosis results from improper regulation of bone formation. Investigations of cells derived from patients with craniosynostosis suggest that craniosynostotic bone-derived cells have increased osteogenic or proliferative capacities compared with other cells. Research into the pathogenesis of craniosynostosis using cells derived from children has been hindered by small sample sizes and inappropriate control cell populations. The authors hypothesized that cells derived from suture-associated regions of bone from craniosynostotic rabbits were more osteogenic and proliferative

Inappropriate cadherin switching in the mouse epiblast compromises proper signaling between the epiblast and the extraembryonic ectoderm during gastrulation.

Cadherin switching from E-cadherin (E-cad) to N-cadherin (N-cad) is a key step of the epithelial-mesenchymal transition (EMT) processes that occurs during gastrulation and cancer progression. We investigate whether cadherins actively participate in progression of EMT by crosstalk to signaling pathways. We apply ectopic cadherin switching before the onset of mouse gastrulation. Mutants with an induced E-cad to N-cad switch (Ncadki) die around E8.5. Severe morphological changes including a small epiblast, a rounded shape, an enlarged extra-embryonic compartment and lack of the amnion, combined w

RNA interference of Bmp-4 and midface development in postimplantation mouse embryos.

INTRODUCTION: Clefts of the lip and palate result when the midfacial processes fail to fuse during embryonic development. Many of the cellular and molecular events underlying the fusion of these midfacial processes are unknown. There is evidence from our laboratory and others that Bmp-4 is a signal that regulates the process of fusion in the midface. RNA interference (RNAi) is a relatively new technique used to knock down select RNA levels--and ultimately protein levels--by using short RNA strands complementary to target mRNA in the cytoplasm. This was the first investigation to use RNAi to st

Making up the numbers: The molecular control of mammalian dental formula.

Teeth develop in the mammalian embryo via a series of interactions between odontogenic epithelium and neural crest-derived ectomesenchyme of the early jaw primordia. The molecular interactions required to generate a tooth are mediated by families of signalling molecules, which often act reiteratively in both a temporal and spatial manner. Whilst considerable information is now available on how these molecules interact to produce an individual tooth, much less is known about the processes that control overall tooth number within the dentition. However, a number of mouse models are now starting
