Biological activity of Bordetella pertussis in lipopolysaccharide-resistant mice.

Effects of Bordetella pertussis organisms, such as adjuvanticity, induction of hypersplenia, and leukocytosis as well as modification of nonspecific resistance to infection and typical morphological response of lymphatic organs, were studied in the lipopolysaccharide-resistant C3H/HeJ mouse strain. It was shown that B. pertussis exerted all of these effects in C3H/HeJ mice, although the morphological response, hypersplenia, and modification of resistance to infection with Listeria monocytogenes in such animals were less pronounced than those in lipopolysaccharide-sensitive mouse strains. This

Molecular epidemiology of Bordetella pertussis in the Philippines in 2012-2014.

OBJECTIVES: The present study was designed to determine the genotypes of circulating Bordetella pertussis in the Philippines by direct molecular typing of clinical specimens. METHODS: Nasopharyngeal swabs (NPSs) were collected from 50 children hospitalized with pertussis in three hospitals during 2012-2014. Multilocus variable-number tandem repeat analysis (MLVA) was performed on the DNA extracts from NPSs. B. pertussis virulence-associated allelic genes (ptxA, prn, and fim3) and the pertussis toxin promoter, ptxP, were also investigated by DNA sequence-based typing. RESULTS: Twenty-six DNA

Evaluation of a tetraplex microsphere assay for Bordetella pertussis antibodies.

To increase testing efficiency, a microsphere-based multianalyte immune detection (MAID) system was developed to measure serum immunoglobulin G (IgG) and IgA recognizing two Bordetella pertussis antigens, pertussis toxin (PT) and filamentous hemagglutinin antigen (FHA). The assay was performed as two separate duplexes. One duplex measured IgG to PT and FHA, and the other measured IgA to PT and FHA. The two duplexes were then combined and analyzed as a tetraplex. The MAID system and an in-house enzyme-linked immunosorbent assay (ELISA) system were used to evaluate 100 sera from blood donors and

A new animal model of Bordetella pertussis infection and immunity.

Bordetella pertussis is a strictly human pathogen. Experimental infection of other animals can occur with large inoculating doses; rat, mice and primate models have been used to study pathogenesis and immunity. Recently, it was shown that newborn piglets are susceptible to B. pertussis. Lung pathophysiology of infected piglets was similar to that of human infants that develop bronchopneumonia. Piglets and infants are anatomically similar and maternal antibodies are transferred and secreted by a similar mechanism. This model could be valuable for studying the roles of passively and actively acq

Interaction of Bordetella pertussis with human respiratory mucosa in vitro.

The human respiratory tract pathogen Bordetella pertussis is the major cause of whooping cough in infants and young children, and also causes chronic cough in adults. B. pertussis infection damages ciliated epithelium in the respiratory tract. However, the interaction of the bacterium with the respiratory mucosa is poorly understood, and previous studies have either utilized animal tissue which may not be appropriate, or isolated cell systems which lack the complexity of the respiratory mucosa. We have studied the interaction of B. pertussis strain BP536 with human nasal turbinate tissue in an

Comparison of nasopharyngeal aspirates with swabs for culture of Bordetella pertussis.

Nasopharyngeal samples were collected from 117 children by aspiration from one nostril and by swab from the other one and cultured for Bordetella pertussis. Among 33 culture-positive specimens, there were 30 positive aspirates and 26 positive swab specimens. Aspirates are easily divided and saved for investigations with other assays which may further improve diagnostic sensitivity. As the aspiration technique was also preferred by nurses and parents, it was recommended and chosen for a planned pertussis vaccine efficacy trial to take place in Sweden from 1992 to 1995.

Simple and efficient method for clinical isolation of Bordetella pertussis.

Commercially available Amies transport medium with charcoal and three isolation media were tested to assess their efficiency in the clinical isolation of B. pertussis. First, the isolation rates of B. pertussis were compared between direct inoculation of nasopharyngeal specimens and inoculation after being stored in Amies transport medium for eight hours or less. The organism was detected in 43 specimens from 29 patients with at least one of the two inoculation methods. The comparative isolation rates were 81% (35 of 43) for both direct inoculation and transport medium. Second, nasopharyngeal

Naturally crystalline porin in the outer membrane of Bordetella pertussis.

The Gram-negative bacterium Bordetella pertussis is the agent responsible for whooping-cough, and much interest has focused on the functions, structures and immunological properties of the molecules exposed at its outer surface. We have found by electron microscopy that cells of two strains of B. pertussis are covered with a crystalline surface lattice. This lattice is not an extrinsic layer of high molecular weight glycoproteins, such as occur on many other bacteria, but is a natural crystal of an intrinsic membrane protein of 40,000 Mr. This molecule has been shown to be an anion-selective m

Genetic diversity of Bordetella bronchiseptica isolates obtained from primates.

Bordetella bronchiseptica is a highly contagious bacterial respiratory pathogen with a broad host range of wild and domesticated mammals that can cause a variety of clinical disease outcomes ranging from asymptomatic carriage to severe pneumonia. The goal of this study was to evaluate the genetic diversity of B. bronchiseptica isolates obtained from primates and evaluate the antimicrobial resistance harbored by these isolates. Two isolates were identified as belonging to B. bronchiseptica lineage II and 13 isolates represented new sequence types within B. bronchiseptica lineage I clonal comple

Proteomic comparison of epidemic Australian Bordetella pertussis biofilm cells.

Bordetella pertussis causes whooping cough, a severe respiratory infectious disease. Studies have compared the currently dominant single nucleotide polymorphism (SNP) cluster I (pertussis toxin promoter allele, ptxP3) and previously dominant SNP cluster II (ptxP1) strains as planktonic cells. Since biofilm formation is linked with B. pertussis pathogenesis in vivo, this study compared the biofilm formation capabilities of representative strains of cluster I and cluster II. Confocal laser scanning microscopy found that the cluster I strain had a denser biofilm structure compared to the cluster

Bordetella pertussis outer membrane vesicles impair neutrophil bactericidal activity.

Neutrophils constitute the primary defense against bacterial infections, yet certain pathogens express virulence factors that enable them to subvert neutrophils-mediated killing. Outer membrane vesicles (OMVs) have emerged as a secretory system through which bacteria deliver virulence factors to host cells. OMVs from Bordetella pertussis, the etiological agent of whooping cough, are loaded with most of bacterial virulence factors, including CyaA, which plays a key role in B. pertussis evasion of neutrophils bactericidal activity. In our study, we investigated the role of B. pertussis OMVs in b

Genomic epidemiology and multilevel genome typing of Bordetella pertussis.

Bordetella pertussis causes pertussis (or whooping cough), a severe respiratory infectious disease in infants, although it can be prevented by whole cell and acellular vaccines. The recent pertussis resurgence in industrialised countries is partly attributed to pathogen adaptation to vaccines, while emergence of antimicrobial resistance, specifically to macrolides in China, has become a concern. Surveillance of current circulating and emerging strains is therefore vital to understand the risks they pose to public health. Although the use of genomics-based typing is increasing a genomic nomencl

Genomic epidemiology of erythromycin-resistant Bordetella pertussis in China.

Macrolides such as erythromycin are the empirical treatment of Bordetella pertussis infections. China has experienced an increase in erythromycin-resistant B. pertussis isolates since they were first reported in 2013. Here, we undertook a genomic study on Chinese B. pertussis isolates from 2012 to 2015 to elucidate the origins and phylogenetic relationships of erythromycin-resistant B. pertussis isolates in China. A total of 167 Chinese B. pertussis isolates were used for antibiotic sensitivity testing and multiple locus variable-number tandem repeat (VNTR) analysis (MLVA). All except four iso

Macrolide-resistant Bordetella pertussis infection in newborn girl, France.

A macrolide antimicrobial drug was administered to a newborn with cough. On day 23 of hospitalization, macrolide-resistant Bordetella pertussis was isolated from nasopharyngeal aspirates. DNA sequencing and PCR-restriction fragment length polymorphism showed a 2047 A-to-G mutation in the 3 copies of the 23S rRNA gene. Monitoring for macrolide resistance is essential in infants <6 months of age.

Mechanisms of natural and vaccine-induced immunity to Bordetella pertussis.

Bordetella pertussis causes whooping cough (pertussis), a respiratory infectious disease that is resurgent despite high vaccine coverage. Research on the mechanisms of immunity to B. pertussis have demonstrated protective roles for innate immune cells, antibodies and T cells in immunity induced by natural infection. Studies in animal models have demonstrated that IL-17-secreting respiratory tissue-resident memory CD4+ T (TRM) cells and associated recruitment of neutrophils play a critical role in clearance of bacteria from nasal mucosa. However, current acellular pertussis (aP) vaccines, while

The complex role of nicotinic acid metabolism in the genus Bordetella.

Bacteria obtain nutrients and other essential factors from their immediate surrounding. For bacteria that reside in a specialized or limited niche range, their metabolism reflects adaptation to that environment(s). Bordetella pertussis and B. parapertussis have evolved from a B. bronchiseptica-like ancestor that survives within hosts and in the environment to become restricted to a single niche, the human respiratory tract. This restriction has shaped their metabolism. Here, the multifaceted role of nicotinic acid in Bordetella biology is reviewed. It serves as a precursor for the synthesis of

Host Specificity of Ovine Bordetella parapertussis and the Role of Complement.

The classical bordetellae are comprised of three subspecies that differ from broad to very limited host specificity. Although several lineages appear to have specialized to particular host species, most retain the ability to colonize and grow in mice, providing a powerful common experimental model to study their differences. One of the subspecies, Bordetella parapertussis, is composed of two distinct clades that have specialized to different hosts: one to humans (Bpphu), and the other to sheep (Bppov). While Bpphu and the other classical bordetellae can efficiently colonize mice, Bppov strains
