Antifungal Activity of Methyl 2-Methylbutyrate Against Botrytis cinerea and the Induced Physiological, Transcriptional, and Metabolic Responses.

Gray mold, caused by the necrotrophic fungus Botrytis cinerea, is a devastating disease responsible for substantial global crop losses. The escalating threat of fungicide resistance and environmental concerns necessitates the development of eco-compatible control alternatives. In this study, we investigated the antifungal efficacy of methyl 2-methylbutyrate (M2M), a microbial volatile organic compound (mVOC), against B. cinerea, and elucidated the underlying physiological, transcriptomic, and metabolomic responses. Our results demonstrated that M2M significantly inhibited mycelial growth in a

β‑Cyclodextrin/Nerolidol Inclusion Complex as a Potential Ecofriendly Antifungal System against Botrytis cinerea in Postharvest Strawberries.

β-Cyclodextrin (β-CD) encapsulation may offer a strategic approach to overcome the high volatility of nerolidol (NE) for controlling Botrytis cinerea decay in postharvest strawberries. In this study, a β-cyclodextrin/nerolidol inclusion complex (β-CD/NE) was prepared by freeze-drying to improve the stability and performance of NE. The complex exhibited high complexation efficiency (96.71%) and nanoscale particle size (484.9 nm). FTIR, XRD, and thermal analyses evidenced strong molecular interactions and structural modifications, and an improvement in the thermal stability of the sesquiterpene,

Detection of SdhB mutations in Botrytis cinerea isolates from strawberry using a High-Resolution Melting (HRM) assay.

Botrytis cinerea, the causal agent of Botrytis fruit rot (BFR), is one of the most destructive pathogens of strawberry. In Florida, BFR management relies heavily on succinate dehydrogenase inhibitor (SDHI) fungicides; however, resistance associated with mutations in the SdhB subunit has increasingly been reported. This study developed and validated a high-resolution melting (HRM) assay for rapid detection of SdhB mutations directly from B. cinerea spores, and evaluated resistance frequencies to fluopyram, isofetamid, and pydiflumetofen across Florida strawberry fields. Two HRM primer sets succ

Evidence of copper (nano)formulation biotransformations triggered by Botrytis cinerea on grapevine leaves for targeted plant protection.

The present work aimed at unravelling if fungal inoculation on grapevine leaves could trigger the dissolution of foliarly deposited Cu (nano)formulations, and how this would impact Cu translocation in planta. Leaves of grapevine seedlings were exposed to 3.3 μg of Cu (5 μg cm-2). Formulations of contrasting solubilities (micro-sized conventional Bordeaux mixture, CuO-nanoparticles (CuO-NPs), and CuSO4) were applied to grapevine leaves, followed by the inoculation of Botrytis cinerea spores on top of the deposited Cu formulations. Nine days after Cu deposition, and six days post inoculation, Cu

ROS Stress and Cell Membrane Disruption are the Main Antifungal Mechanisms of 2-Phenylethanol against Botrytis cinerea.

2-Phenylethanol (2-PE), a common compound found in plants and microorganisms, exhibits broad-spectrum antifungal activity. Using Botrytis cinerea, we demonstrated that 2-PE suppressed mycelium growth in vitro and in strawberry fruit and reduced natural disease without adverse effects to fruit quality. 2-PE caused structural damage to mycelia, as shown by scanning and transmission electron microscopy. From RNA sequencing analysis we found significantly upregulated genes for enzymatic and nonenzymatic reactive oxygen species (ROS) scavenging systems including sulfur metabolism and glutathione me

CYP94-mediated jasmonoyl-isoleucine hormone oxidation shapes jasmonate profiles and attenuates defence responses to Botrytis cinerea infection.

Induced resistance to the necrotrophic pathogen Botrytis cinerea depends on jasmonate metabolism and signalling in Arabidopsis. We have presented here extensive jasmonate profiling in this pathosystem and investigated the impact of the recently reported jasmonoyl-isoleucine (JA-Ile) catabolic pathway mediated by cytochrome P450 (CYP94) enzymes. Using a series of mutant and overexpressing (OE) plant lines, we showed that CYP94B3 and CYP94C1 are integral components of the fungus-induced jasmonate metabolic pathway and control the abundance of oxidized conjugated but also some unconjugated deriva

Cloning and characterization of a glutathione S-transferase homologue from the plant pathogenic fungus Botrytis cinereadouble dagger.

Abstract A gene was cloned from Botrytis cinerea that encodes a protein homologous to glutathione S-transferase (GST). The gene, denominated Bcgst1, is present in a single copy and represents the first example of such a gene from a filamentous fungus. The biochemical function of GSTs is to conjugate toxic compounds to glutathione, thereby detoxifying the compound. In many other organisms, GST plays a role in chemical stress tolerance. We anticipated that GST functions for B. cinerea as a potential virulence factor, enabling the fungus to tolerate fungitoxic plant defence compounds. The express

Efficiency of nanoemulsion of essential oils to control Botrytis cinerea on strawberry surface and prolong fruit shelf life.

Strawberry fruit is highly susceptible to decay by fungi. The objective of this study was to determine if essential oils (EOs) or nano-emulsions (Nano-EM) of EOs from Thymus vulgaris (Th), Matricaria chamomilla (Mc), Pistacia atlantica (Pa), or Mentha longifolia (Me) could inhibit growth of strawberry spoilage fungi Botrytis cinerea and their effect, if any, on strawberry quality parameters. An In vitro study showed that Th and Me EOs had the same minimal inhibitory concentrations (MIC) of 0.021 % while the MICs for Mc and Pa EOs were 0.9 % and 1.5 %, respectively. Th and Me EOs were used for

Multi-method in vitro assessment of ultraviolet-C treatment against conidia and hyphal fragments of Botrytis cinerea.

UNLABELLED: Botrytis cinerea is a significant postharvest fungal pathogen responsible for substantial losses in a variety of fresh produce. Ultraviolet-C (UV-C) irradiation is a potential intervention that could reduce postharvest losses due to fungal disease. However, the diversity of fungal reproductive forms and lack of standardization in antifungal assessment methods complicate the evaluation of UV-C efficacy. This study evaluated the in vitro antifungal efficacy of UV-C against B. cinerea conidia and hyphal fragments (HF) treated in liquid suspension and on agar surfaces. B. cinerea viabi

Simultaneous Detection of Multiple Benzimidazole-Resistant β-Tubulin Variants of Botrytis cinerea using Loop-Mediated Isothermal Amplification.

Optimal disease management depends on the ability to monitor the development of fungicide resistance in plant pathogen populations. Benzimidazole resistance is caused by the point mutations of the β-tubulin gene in Botrytis cinerea, and three mutations (E198A, E198K, and E198V) at codon 198 account for more than 98% of all resistant strains. Although traditional methods remain a cornerstone in monitoring fungicide resistance, molecular methods that do not require the isolation of pathogens can detect resistance alleles present at low frequencies, and require less time and labor than traditiona

Antifungal Activity of Saponins from the Fruit Pericarp of Sapindus mukorossi against Venturia inaequalis and Botrytis cinerea.

The antifungal activity of an aqueous extract (AE) and the solid fraction of a chloroform-methanol fruit pericarp extract (CME) of Sapindus mukorossi resolved in water was tested for the first time against Venturia inaequalis and Botrytis cinerea-two important fungal pathogens worldwide. In the greenhouse, a CME (1% vol/vol) spray significantly reduced V. inaequalis symptoms and sporulation (99%) on apple seedling leaves (P ≤ 0.05). In field trials, applications of AE (1% vol/vol) reduced the disease severity of B. cinerea on grape, on average, by 63%. Extracts were fractionated by high-perfor

Overexpression of EXO70E2 in Arabidopsis thaliana Disrupts Normal Development and Enhances Susceptibility to the Necrotrophic Fungus Botrytis cinerea.

Background: The exocyst complex, a conserved hetero-octameric complex including the EXO70 subunit, is a pivotal regulator of various cellular and developmental processes in plants. Among the diverse EXO70 isoforms, the specific function of EXO70E2-a primary organizer of exocyst-positive organelles (EXPOs)-remains to be fully elucidated. Methods: Here, we investigated the functional impact of constitutive EXO70E2 overexpression in Arabidopsis thaliana. Results: We observe that EXO70E2 overexpression leads to severe etiolation and dwarfism, accompanied by dose-dependent inhibition of primary roo

Genomic and phenotypic characterization of Antarctic Pseudomonas/Aquipseudomonas sp. strains with cold-active antifungal activity against Botrytis cinerea.

Gray mold, caused by the psychrophilic necrotroph Botrytis cinerea, is a leading cause of postharvest losses, and its control is increasingly compromised by the spread of fungicide-resistant field populations and by the failure of conventional biocontrol agents to operate at cold-chain temperatures. Antarctic soils are a rational yet underexploited source of cold-active antagonists. Here we isolated cultivable bacteria from the rhizosphere of Deschampsia antarctica (King George Island) and screened them against B. cinerea at 22 °C and 0 °C. Of 57 bacterial isolates, 44 antagonized the pathogen

Physiological and image-based characterization of Botrytis cinerea infection progression in postharvest 'Haegeum' kiwifruit using interpretable deep learning.

Gray mold caused by Botrytis cinerea severely limits the postharvest storage and marketability of kiwifruit, creating a need for accurate and non-destructive infection assessment. In this study, a symptom based, ROI-aware, and interpretable deep-learning framework was developed to classify infection levels in golden-fleshed 'Haegeum' kiwifruit. Infection severity was categorized into four levels based on external visual symptoms: Level 1: no lesions; Level 2: slight discoloration; Level 3: localized mycelial growth; Level 4: extensive mycelial growth and tissue breakdown. The associated physio

A Trichoderma milRNA cross-species targeting chitin synthases genes is delivered by carbon nanomaterials to suppress Botrytis cinerea.

Botrytis cinerea is a globally destructive plant pathogen that threatens major agricultural crops. Although biocontrol agents are widely used against this fungus, their inconsistent efficacy reflects a limited mechanistic understanding, hindering the development of effective disease management strategies. In this study, we investigated the in-depth biocontrol mechanism of Trichoderma breve against B. cinerea on tomato plants and various fruits. We identified a new microRNA Tri-milR29 from T. breve. Tri-milR29 is delivered into B. cinerea via extracellular vesicles, enabling cross-species RNA i

A Rapid pH-Responsive Pyraclostrobin Delivery System with Enhanced Membrane Passing Property and Fungicidal Activity against Botrytis cinerea.

As the second most important fungal pathogen, Botrytis cinerea (B. cinerea) poses a serious threat to crop yields and agricultural safety. Pyraclostrobin (PYR), a broad-spectrum QoI fungicide, has been widely utilized since its launch in 2003. However, the inhibitory effects of both PYR technical and PYR formulations on B. cinerea are not outstanding. Even at a concentration of 50 μg/mL, the inhibition rates of B. cinerea by both PYR technical and PYR formulations remain below 85%. In this work, we prepared an acid-responsive Pickering emulsion encapsulating PYR (PYR@BTIB-PE), which completely
