Exploring insulin aggregation at neutral pH and its inhibition via caffeic acid: a biophysical and bioinformatics analysis.

This study aimed to induce fibril formation in human insulin under physiological conditions and to investigate the inhibitory potential of caffeic acid (CA) on these fibrils in-vitro. Various techniques including circular dichroism (CD) spectroscopy, Thioflavin T, ANS fluorescence, Rayleigh light scattering (RLS), and turbidity analysis were conducted to elucidate fibril formation and CA inhibition potential. Fibril formation in insulin was induced by heating (37 °C) and agitation (600 rpm) significantly increased (27.01-fold) the ThT binding after 120 h of incubation. Aggregation also increas

High-performance caffeic acid-based superhydrophilic membranes for efficient removal of microplastics-oil co-contaminants.

Microplastic-oil co-contaminants have raised high environmental concerns due to their persistence and synergistic toxicity. However, conventional separation membranes, typically designed for single-pollutant removal, are ineffective against such complex systems. Moreover, the strong interfacial interactions between microplastics and oil promote aggregate formation, leading to severe membrane fouling and compromised separation performance. Herein, we present a robust, antifouling, and superhydrophilic membrane fabricated via sodium periodate-assisted co-deposition of caffeic acid and ε-polylysi

Caffeic Acid Mitigates Behavioral and Biochemical Alterations in Lead-induced Neurotoxicity in Rats With Possible Involvement of TFEB.

Lead (Pb) neurotoxicity is characterized by persistent cognitive and motor impairments that arise from converging disturbances in mitochondrial function, oxidative balance, and neuroinflammatory signaling. Increasing evidence suggests that these pathological outcomes are closely linked to disruption of Transcription Factor EB (TFEB), a key regulator of autophagy-lysosomal pathways and mitochondrial quality control. Impaired TFEB function can promote the accumulation of dysfunctional mitochondria and perpetuate oxidative and inflammatory cascades, thereby exacerbating Pb-induced neurodegenerati

Experimental Evidence of Caffeic Acid's Neuroprotective Activity in Alzheimer's Disease: In Vitro, In Vivo, and Delivery-Based Insights.

Background and Objectives: Alzheimer's disease (AD) is a complex neurodegenerative disorder marked by cholinergic deficits, oxidative stress, amyloid-β (Aβ) aggregation, and tau hyperphosphorylation. Caffeic acid (CA), a naturally occurring hydroxycinnamic acid, has emerged as a promising neuroprotective candidate due to its antioxidant, anti-inflammatory, and enzyme-inhibitory properties. This review systematically evaluates recent in vitro and in vivo evidence regarding the therapeutic potential of CA in AD models and examines the impact of delivery systems and derivatives on its efficacy an

Caffeic acid borate functionalized linear polyglycidol for targeted siRNA delivery in the treatment of primary biliary cholangitis.

Interfering with protein arginine methyltransferase 5 (PRMT5) gene expression with siRNA targeting is a promising strategy for precision primary biliary cholangitis (PBC) therapy, yet achieving effective and specific delivery of therapeutic agents to the liver remains challenging. In this study, we develop a caffeic acid borate derived linear polyglycidol polymer micelle CAPG, which can efficiently deliver siRNA to liver. The degradation of borate and hydrazone bonds in the high-ROS and acidic environment of inflammatory cells triggers micelle disintegration, facilitating siRNA release. Compre

Caffeic acid alleviates cerebral ischemic injury in rats by resisting ferroptosis via Nrf2 signaling pathway.

There are few effective and safe neuroprotective agents for the treatment of ischemic stroke currently. Caffeic acid is a phenolic acid that widely exists in a number of plant species. Previous studies show that caffeic acid ameliorates brain injury in rats after cerebral ischemia/reperfusion. In this study we explored the protective mechanisms of caffeic acid against oxidative stress and ferroptosis in permanent cerebral ischemia. Ischemia stroke was induced on rats by permanent middle cerebral artery occlusion (pMCAO). Caffeic acid (0.4, 2, 10 mg·kg-1·d-1, i.g.) was administered to the rats

A facile synthesis of CuBi2O4 hierarchical dumbbell-shaped nanorod cluster: a promising photocatalyst for the degradation of caffeic acid.

The present study reports on the synthesis of Cu-bismuth oxide (CuBi2O4)-based nanorods by using a simple co-precipitation method for the photocatalytic degradation of caffeic acid (CA). The incorporation of Cu metal ions during the synthesis of CuBi2O4 nanorods might be advantageous to avoid the aggregation and control the leach out of metal ions. The calculated bandgap values of ~ 1.04, 1.02, and 0.94 eV were observed for CuBi2O4 with different amounts of Cu 1.0, 0.50, and 0.25 g, respectively. Varying the quantity of Cu metal ions easily tuned the bandgap value within the CuBi2O4-based nano

The Anti-Biofilm Efficacy of Caffeic Acid Phenethyl Ester (CAPE) In Vitro and a Murine Model of Oral Candidiasis.

Candida albicans is the main fungal species associated with the development of oral candidiasis. Currently, therapeutic options for these infections are limited by the adverse effects of antifungal drugs and by the emergence of drug resistant strains. Thus, the development of new antifungal agents is needed for the prevention and treatment of oral Candida infections. Caffeic acid phenethyl ester (CAPE) is a natural compound from propolis polyphenolic groups that exhibits many pharmacological properties. In this study, we investigated whether CAPE can have antifungal and immunomodulatory effect

Neuroprotection of retinal cells by Caffeic Acid Phenylethyl Ester(CAPE) is mediated by mitochondrial uncoupling protein UCP2.

Oxidative stress due to mitochondrial produced reactive oxygen species is a major cause of damage seen in many retinal degenerative diseases. Caffeic acid phenylethyl ester (CAPE) is protective agent in multiple tissues and is reported to have anti-oxidant properties. Systemically applied CAPE protected retinal ganglion cells from ischemic injury induced by increased intraocular pressure. CAPE provided complete protection for ARPE19 retinal pigment epithelial cells against tert-butyl hydrogen peroxide and reduced both basal and LPS-stimulated ROS production. The major effect of CAPE was mediat

A Novel Butylated Caffeic Acid Derivative Protects HaCaT Keratinocytes from Squalene Peroxidation-Induced Stress.

Squalene is a major sebum lipid which is easily peroxidized by ultraviolet A (UVA) irradiation, generating products that can stress keratinocytes. Prevention of squalene photooxidation with antioxidants could potentially help defend skin from environmental stress. Previously, we have systematically characterized butylated caffeic acid (BCA) as a more lipophilic alternative of caffeic acid (CA). The current study aimed to test the hypothesis that the lipophilic property of BCA makes it a good candidate as a skin antioxidant. The UVA-induced peroxidation of squalene and the antioxidative activit

Synergistic Antibacterial Effects of Chitosan-Caffeic Acid Conjugate against Antibiotic-Resistant Acne-Related Bacteria.

The object of this study was to discover an alternative therapeutic agent with fewer side effects against acne vulgaris, one of the most common skin diseases. Acne vulgaris is often associated with acne-related bacteria such as Propionibacteriumacnes, Staphylococcusepidermidis, Staphylococcusaureus, and Pseudomonasaeruginosa. Some of these bacteria exhibit a resistance against commercial antibiotics that have been used in the treatment of acne vulgaris (tetracycline, erythromycin, and lincomycin). In the current study, we tested in vitro antibacterial effect of chitosan-phytochemical conjugate

Targeting ASC in NLRP3 inflammasome by caffeic acid phenethyl ester: a novel strategy to treat acute gout.

Gouty arthritis is caused by the deposition of uric acid crystals, which induce the activation of NOD-like receptor family, pyrin domain containing 3(NLRP3) inflammasome. The NLRP3 inflammasome, composed of NLRP3, the adaptor protein ASC, and caspase-1, is closely linked to the pathogenesis of various metabolic diseases including gouty arthritis. We investigated whether an orally administrable inhibitor of NLRP3 inflammasome was effective for alleviating the pathological symptoms of gouty arthritis and what was the underlying mechanism. In primary mouse macrophages, caffeic acid phenethyl este

Caffeic acid phenethyl ester prevents 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced neurodegeneration.

Parkinson's disease is associated with the loss of dopaminergic neurons in the substantia nigra and decreased striatal dopamine levels. We now report that caffeic acid phenethyl ester (CAPE), an active component of propolis, attenuated dopaminergic neurodegeneration and dopamine loss in the MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) mouse model of Parkinson's disease. The neuroprotective effect of CAPE was associated with marked reductions in inducible nitric oxide synthase (iNOS) and caspase 1 expression. Additionally, CAPE inhibited MPP+-induced neurotoxicity in vitro and directly i

Inhibition of store-operated Ca2+ entry channels and K+ channels by caffeic acid phenethylester in T lymphocytes.

The increase of cytoplasmic Ca2+ concentration (Delta[Ca2+]c) in response to antigenic stimulation is a critical step of signals activating immune responses. In addition, the voltage-gated K+ channels (Kv) in T lymphocytes draw attention as an effective target of immune-modulation. Caffeic acid phenethyl ester (CAPE), an active component of propolis, shows strong anti-inflammatory effects and T cell suppression. Although various mechanisms have been suggested for the action of CAPE, the effects of CAPE on intracellular Ca2+ signaling and ion channels are unknown. Here we investigated the effec

Caffeic acid protects human peripheral blood lymphocytes against gamma radiation-induced cellular damage.

In the present study, we investigated in vitro radioprotective potential of caffeic acid (CA), a naturally occurring catecholic acid against gamma radiation-induced cellular changes. Different concentrations of CA (5.5, 11, 22, 44, 66, and 88 microM) were incubated with lymphocytes for 30 min prior to gamma-irradiation, and micronuclei (MN) scoring and comet assay were performed to fix the effective concentration of CA against gamma-irradiation. Among all concentrations, 66 microM of CA showed the optimum protection by effectively decreasing the MN frequencies and comet attributes. From the ab

Design, synthesis, and biological evaluation of novel multifunctional caffeic acid-piperazine derivatives for the management of Alzheimer's disease.

Alzheimer's disease (AD), the most prevalent form of dementia, is marked by progressive memory impairment, cognitive decline, reduced acetylcholine level, oxidative stress, amyloid-β (Aβ) aggregation, and disturbances in metal homeostasis. Our earlier work on caffeic acid-based multifunctional inhibitors prompted us to explore structure-activity relationships (SARs) to improve BChE cholinesterase inhibition while maintaining or enhancing the multifunctional activity of the previously identified lead molecule 12d. For this purpose, the glycinamide linker was rationally replaced with benzylpiper
